        Immunolocalization of Runx2 and Osterix in the Developing Periodontal Tissues of the Mouse

        Runx2 and Osterix, the transcription factors for osteoblast differentiation, are known as fundamental factors to regulate the development of calcified tissues. However, the biological functions of these factors in the development of the periodontal tissues remain unclear. In this study, we investigated the distribution of Runx2 and Osterix during periodontal tissue development of the mice. Mandibles from 14-day-old mice were prepared for paraffin section. Serial sections of the mandible containing 1 st molar tooth germs were obtained as a thickness of 7 μm. Some sections were stained with hematoxylin and eosin. Others were used for immunohistochemistry for PCNA, Runx2, and Osterix. Epithelial cells in growing end of Hertwig’s epithelial root sheath (HERS) and mesenchymal cells adjacent to the growing end of HERS expressed PCNA. Undifferentiated mesenchymal cells and hard tissue forming cells like cementoblasts and osteoblasts in early stage of differentiation expressed Runx2. Fully differentiated cementoblasts and osteoblasts secreting matrix proteins expressed Osterix. However, the cells terminated the matrix formation did not express Osterix. Periodontal ligament cells expressed Runx2 and Osterix. Pulp cells expressed Runx2 only.These results suggest that Runx2 and Osterix might regulate the differentiation of cementoblasts in the same manner as osteoblasts. Runx2 might participate in the process of cementoblast differentiation in early stage, whether Osterix might regulate the maturation and matrix synthesis of the cells.

      • 청소년시설 대표의 진정성 리더십과 활동 참여 청소년에 대한 관계 분석

        본 연구는 청소년시설 대표의 진정성 리더십이 활동 참여 청소년의 만족도에 미치는 영향을 검증하고, 청소년지도사의 인식 만족도를 매개로 하여 그 효과를 알아보고자 하였다. 이를 위하여 청소년들의 접근성이 용이하여 이용하기 편리한 청소년수련관, 청소년문화의집, 청소년활동진흥센터 등을 선정, 충북, 세종, 충남의 세 지역의 청소년시설 중 30개 시설을 선정하였다. 현재 그곳에서 근무하고 있는 청소년지도사 279명과 선정된 기관에서 최근 6개월 이내에 활동에 참여한 경험이 있거나 참여 중인 청소년 308명을 대상으로 실시하였으며 SPSS 22.0과 빈도분석과 신뢰도와 타당도에 대한 Cronbach’s α 기준값 0.6 이상으로 설정하여 신뢰성 판단을 하였으며, AMOS 22.0 프로그램을 이용하여 구조 방정식모형을 통한 연구문제 검증과 청소년지도사의 인식 만족도는 청소년시설 대표의 진정성 리더십과 활동 참여 청소년만족도 관계에서 매개 효과에 관한 정확한 검증을 위해 Sobel-test 분석을 실시하였다. 결과, 청소년지도사의 인식 만족도와 청소년시설 대표의 진정성 리더십은 활동 참여 청소년의 만족도에 영향을 주는 주요 예측요인임을 확인하였다. 또한, 구조적 관계모형을 바탕으로 청소년시설 대표의 진정성 리더십의 영향력보다는 청소년들과 직접 상호작용하는 청소년지도사의 인식 만족도가 훨씬 더 큰 영향력을 가지고 있음을 알 수 있었다. 특히 청소년시설 대표의 진정성 리더십이 활동에 참여하는 청소년들과 많은 상호작용을 하며 청소년지도사의 인식 만족도를 매개 변인으로 작용하여 효과를 밝혔다는데 그 의의가 있다. The purpose of this study was to examine the effects of the leadership of youth facilities representatives on the satisfaction of the participating youth and to investigate the effects of the satisfaction of youth. For this purpose, the researcher selected youth training center, youth culture center, and youth activity promotion center that are easy to access by young people. the researcher selected 30 youth facilities in three areas: Chungbuk, Sejong, and Chungnam. The survey was conducted on 279 youth leaders currently working there and 308 youth who have participated in activities for the past six months at selected institutions. A total of 587 people were surveyed by online method and mail delivery using measurement tools for the influence of the leader of the youth facilities on their authenticity leadership and satisfaction with the youth leader's activities, and the satisfaction perceived by the youth leader in guiding them. The results of this study are as follows. Firstly, the results of the research hypothesis were adopted as a result of analysis of the research hypothesis through the structural equation model in the effect of the authenticity leadership of the youth facility representatives on the perception satisfaction of the youth instructor and the satisfaction of the participating youth. Secondly, it was analyzed by using Pearson correlation coefficient to verify the correlation between the lesions in the relationship between the leader of the youth facility, the awareness satisfaction of the youth leader, and the youth satisfaction of participating in the activity, and it was found that there was a good correlation between the two. In this study, it was found that the self-awareness of the authenticity leadership of the representative of the youth facility was the highest correlation between the job self and colleagues of the youth leader's satisfaction with the perception. The relationship between the leader of youth facilities' authenticity leadership and participation in activities and the satisfaction of youth's satisfaction with the program, the facility's satisfaction, and the leader's satisfaction with the transparent relationship, and the job and the remuneration's satisfaction with the leader's satisfaction in relation to the awareness satisfaction of youth leaders' participation in activities. Thirdly, the results of the sobel-test analysis were conducted for accurate verification of how the perceived satisfaction of the youth leader in the relationship between the leader of the youth facility and the activity participation youth satisfaction level, and it was found that the perceived satisfaction of the youth leader was significant and played a partial role in the relationship between the leader's authenticity leadership and the activity participation youth satisfaction level. As the result of this study, it was confirmed that the awareness satisfaction level of the youth leader and the authenticity leadership of the representatives of youth facilities were the main predictors affecting the satisfaction level of the youth who participated in the activity. Also, based on the structural relationship model, it was also shown that the awareness satisfaction level of the youth leader interacting directly with the youth rather than the authenticity of the youth facility's representative. Especially, it is meaningful that the authenticity leadership of the youth facilities representatives interacted with the youth participating in the activities. Through this research, the influence of the representative's authenticity leadership in youth facilities on the satisfaction of youth participation and awareness among youth leaders was known. However, it was difficult to find a prior study that was handled directly, so, various youth facilities presented authenticity leadership considering representatives and middle managers.

        청소년시설 대표의 진정성 리더십이 활동 참여 청소년의 만족도에 미치는 영향 - 청소년지도사의 인식 만족도를 매개로 -

        목적: 청소년시설 대표의 진정성 리더십이 활동 참여 청소년의 만족도에 미치는 영향을 검증하고, 청소년지도사의 인식 만족도를 매개로 한 그 효과를 알아보고자 하였다. 방법: 청소년들의 접근성이 용이하여 이용하기 편리한 청소년수련관, 청소년문화의집, 청소년활동진흥센터 등 청소년시설 중 30개 시설을 선정하여 청소년지도사 279명과 선정된 기관에서 최근 6개월 내에 활동에 참여한 경험이 있거나 참여 중인 청소년 308명을 대상으로 실시하였다. 결과: 청소년시설 대표의 진정성 리더십과 활동에 참여한 청소년의 만족도에 미치는 영향, 청소년지도사가 인식하는 만족도에 대하여 측정도구를 사용하여 총587명의 자료를 최종 분석에 사용하여 다음과 같은 결과를 얻었다. 첫째, 청소년시설 대표의 진정성 리더십이 청소년지도사의 인식 만족도와 활동 참여 청소년 만족도에 미치는 영향에서 연구가설에 대하여 구조방정식모형으로 분석한 결과 정(+)에 영향을 미치는 것으로 모두 채택 되었다. 둘째, 청소년시설 대표의 진정성 리더십과 청소년지도사의 인식 만족도, 활동 참여 청소년 만족도 관계에서 변인간 모두 양호한 상관관계가 있으며, 청소년시설 대표의 진정성 리더십의 자아인식은 청소년지도사의 인식 만족도와의 상관관계에서 직무자체와 동료가 가장 높았다. 청소년시설 대표의 진정성 리더십과 활동 참여 청소년 만족도 상관관계에서는 투명한 관계성이 프로그램 만족 ,시설 만족, 지도자 만족 모두에서 높았다. 또한 청소년지도사의 인식 만족도와 활동 참여 청소년 만족도의 상관관계에서는 직무자체와 보수, 시설 만족과 지도자 만족이 높은 것으로 나타났다. 셋째, 청소년시설 대표의 진정성 리더십과 활동 참여 청소년 만족도 간의 관계에서 청소년지도사의 인식 만족도는 유의하였으며 부분매개역할을 하는 것으로 나타났다. 결론: 이에 청소년지도사의 인식 만족도와 청소년시설 대표의 진정성 리더십은 활동 참여 청소년의 만족도에 영향을 주는 주요 예측요인임을 확인하였다. 또한 구조적 관계모형을 바탕으로 청소년시설 대표의 진정성 리더십의 영향력 보다는 청소년들과 직접적으로 상호작용하는 청소년지도사의 인식 만족도가 훨씬 더 큰 영향력을 가지고 있음을 알 수 있었다. Purpose: The purpose of this study was to verify the effect of the youth facility representative's authentic leadership on the satisfaction of the youth participating in the activity, and to investigate the effect of the youth instructor's perception satisfaction as a medium. Methodology: Youth centers, youth culture centers, youth activity promotion centers, and other youth facilities that are easily accessible are selected, and 279 youth instructors and selected institutions have participated in or participated in activities within the last 6 months. It was conducted with 308 youths who are in the process. Findings: A total of 587 data were used for the final analysis using a measuring tool for the authentic leadership of youth facility representatives, the impact on the satisfaction of the youth who participated in the activities, and the satisfaction perceived by youth instructors, and the following results were obtained. Firstly, the results of the research hypothesis were adopted as a result of analysis of the research hypothesis through the structural equation model in the effect of the authenticity leadership of the youth facility representatives on the perception satisfaction of the youth instructor and the satisfaction of the participating youth. Secondly, it was found that the self-awareness of the authenticity leadership of the representative of the youth facility was the highest correlation between the job self and colleagues of the youth leader's satisfaction with the perception.         고농도 포도당이 뼈모세포와 치주인대세포의 세포자멸사에 미치는 영향에 관한 연구

        This experiment was designed to clarify the effect of extracellular glucose on the osteoblasts and periodontal ligament cells. The cells were incubated for 24 and 48 hours with ${\alpha}$-MEM including 1,000 mg/L (control group) and 4,500 mg/L (experimental group) of glucose. Then, the expressions of caspase-3, p38 MAPK, JNK-1, and ERK-1 were examined using Elisa assay and Western blot. The results were as follows: 1. The expression of caspase-3 and p38 MAPK was increased by the high extracellular glucose in both cells. 2. The expression of caspase-3 and p38 MAPK was increased greatly in the periodontal ligament cells than the E1 cells by the high extracellular glucose. 3. The expression of JNK-1 was increased by the high extracellular glucose in both cells. 4. The expression of ERK-1 was not changed by the high extracellular glucose in both cells. These results suggest that extracellular glucose at high concentrations may inhibit the periodontal regeneration process increasing cellular apoptosis. And p38 MAPK and JNK-1 pathway may be the most responsible intracellular pathway rather than ERK-1.

        배양된 사람 치주인대세포와 골수유래간엽줄기세포의 분화에 미치는 법랑기질유도체 (Enamel Matrix Derivative, EMD)의 영향

        Introduction: Enamel matrix derivative (EMD) is a protein which is secreted by Hertwig root sheath and plays a major role in the formation of cementum and attachment of peridontium. Several studies have shown that EMD promoted the proliferation and differentiation of preosteoblasts, osteoblasts and periodontal ligament cells in vitro: however, reports showing the inhibition of osteogenic differentiation by EMD also existed. This study was designed to simultaneously evaluate the effect of EMD on the two cell lines (human mesenchymal stem cells: hMSC, human periodontal ligament derived fibroblasts: hPDLCs) by means of quantitative analysis of some bone related matrices (Alkaline phosphatase : ALP, osteopontin ; OPN, osteocalcin ; OC). Materials and Methods: hMSCs and hPDLCs were expanded and cells in the 4${\sim}$6 passages were adopted to use. hMSc and hPDLCs were cultured during 1,2,7, and 14 days with 0, 50 and 100 ${\mu}g/ml$ of EMD, respectively. ALP activity was assessed by SensoLyte ALP kit and expressed as values of the relative optical density. Among the matrix proteins of the bony tissue, OC and OPN were assessed and quantification of these proteins was evaluated by means of human OC immunoassay kit and human OPN assay kit, respectively. Results: ALP activity maintained without EMD at $1,2^{nd}$ day. The activity increased at $7^{th}$ day but decreased at $14^{th}$ day. EMD increased the activity at $14^{th}$ day in the hPDLCs culture. In the hMSCs, rapid decrease was noted in $7^{th}$ and $14^{th}$ days without regard to EMD concentrations. Regarding the OPN synthesis in hPDLCs, marked decrease of OPN was noted after EMD application. Gradual decrease tendency of OPN was shown over time. In hMSCs, marked decrease of OPN was also noted after EMD application. Overall concentration of OPN was relatively consistent over time than that in hPDLCs. Regarding the OC synthesis, in both of hPDLCs and hMSCs, inhibition of OC formation was noted after EMD application in the early stages but EMD exerted minimal effect at the later stages. Conclusion: In this experimental condition, EMD seemed to play an inhibitory role during the differentiation of hMSCs and hPDLCs in the context of OC and OPN formation. In the periodontium, there are many kinds of cells contributing to the regeneration of oral tissue. EMD enhanced ALP activity in hPDLCs rather than in hMSCs and this may imply that EMD has a positive effect on the differentiation of cementoblasts compared with the effect on hMSCs. The result of our research was consistent with recent studies in which the authors showed the inhibitory effect of EMD in terms of the differentiation of mineral colony forming cells in vitro. This in vitro study may not stand for all the charateristics of EMD; thus, further studies involving many other bone matrices and cellular attachment will be necessary.
