      • 급격한 pH 변화에 대한 꺽지(Coreoperca herzi)조직내 젖산탈수소효소의 순응상

        The metabolism of lactate dehydrogenase(EC 1.1.1.27, LDH) in tissues of Coreoperca herzi acclimated to acute change of pH were studied by measurement of enzyme activity and native polyacrylamide gel electrophoresis. LDH A₄, B₄, A₂B₂, C₄ and isozyme containing subunit C were expressed but A₃B and AB₃ isozyme were absent. The LDH activity was decreased largely in heart tissue and skeletal muscle and was increased in the kidney and liver.. When the pH was increased, the LDH activity was changed to maximum state within 2-6 hours and was restored to the base level within 8-12 hours. The activity of LDH[ isozymes showed large variation in the kidney and eye tissues, but was changed a little in the heart tissue. The activity of subunit that has higher activity among subunit A and B was increased and that of the subunit having lower activity was decreased. The metabolism of eye tissue was regulated by the decrease - of C4 isozyme activity and the increase of B₄ isozyme. Therefore, LDH was played role as pyruvate reductase in skeletal muscle, and as lactate oxidase in other tissues. The change of LDH to the increase of pH might be the acclimation process to control the metabolism.

      • 고등학교 정치교과서의 정치교육내용 비교 분석 : 『정치·경제』·『정치』교과서의 비교를 중심으로 『Politics·Economics』and 『Politics』

        The purpose of this study is to compare and analyze the political education contents in a viewpoint of conservative and innovative function between 『Politics』and 『Politics·Economics』of the high school textbooks, those have been used in 5th and the 6th curriculum. First of all, this study made an analytic model about the political education context of the high school textbooks on the basis of many theories about the political education for students. Second, it made criteria about the usefulness and the harmony between conservative and innovative function for political education in a socio-functional point of view, and also, briefly reviewed the process of undergoing changes in the social studies curriculum until now. Third, according to the criteria, it examined the contents of the politics textbooks of the high school and analysed the frequencies of contents in each political education areas. The contents of political education in 『Politics·Economics』 showed more tendencies of a rightist direction than those in 『Politics』. In conclusion, the study is suggesting it is critical for students to teach organization culture, discipline and to maintain the society, but the recent political education needs to teach them both conservative and innovative area to a adjust themselves to a changing society.

      • Mark Twain의 비관주의와 도덕적 이상주의 (1)

        The main purpose of this writing is to consider Mark Twain as a serious writer. All too often he has been misunderstood as one merely giving witty comments on life to entertain his readers. Another line of thinking doing scant justice to Mark Twain is examplified in Maxwell Geismer's Mark Twain: An American Prophet, in which the author concludes that Twain's thoughts reveal him to be a “natural and unconscious Marxist.”In this article I take issue with such positions. Mark Twain's ideals and thoughts in his social criticism are characterized by his firm belief in American ideals, not by his “natural and unconscious Marxism.”Like Walt Whitman, Mark Twain was convinced that American democracy offered the best climate, not only for material progress, but also for spiritual and moral development. This is divided into three chapters. The First Chapter, entitled “The Humanist Quarrelling with God,”deals with Mark Twain's religious background, his criticism of the Christian conception of God and of “Christian Science”in particular, and his deterministic view of the universe. The Second Chapter, entitled “Mark Twain's Negative Views of Man”deals with the view of man as presented in his writings. The chapterends with the discussion of the limitations of deterministic views of man, also entertained by Twain. The Third Chapter, entitled “Mark Twain's Moral Idealism,”discusses Mark Twain's conscience and ideals, his denunciation of all forms of prejudice, and his pursuit of truth and honesty. Though much of Mark Twain's writings are overshadowed by his mechanistic determinism, Mark Twain is at heart a romantic idealist. This dichotomy is a unique feature of Mark Twain's thoughts. In spite of his religious skepticism, he clung firmly to his belief in a Supreme Being, who had created the universe and whose majesty and wisdom are revealed in physical nature. Seeing nobility and goodness in individuals, he regarded the human race collectively as stupid, mean, and despicable. And yet, his heart was filled with pity and sympathy for doomed mankind, the victim of forces beyond his control, thus admitting a glimmer of hope for human betterment. Hence, the ambivalence of Twain's views of man.

      • 생쥐(Mus musculus) 젖산탈수소효소 동위효소들의 정제

        The lactate dehydrogenase(EC 1.1.1.27, LDH) isozymes of muscle and mixture of liver. kidney and brain tissue in mouse(Mus rnusculus) were purified by affinity chromatograpy. LDH A₄, A₃B. A₂B₂ and B₄ isozymes were purified using Prep Cell system separately. The purified LDH isozymes were identified with homogeneity. The inhibition pattern of LDH A₄, A₃B and A₂B₂ isozyme by pyruvate was detected in the same rate. Therefore, we suggested that the structure of LDH A₄, A₃B and A₂B₂ isozyme seems to be similar.

      • Philip Roth의 『偉大한 美國小說』(1973)

        Although hard to pinpoint the exact date, there was a strange obsession among American novelists toward the end of the 19th century: that they would write a Great American Novel. In 1903, Frank Norris commented on this strange, yet understandable energy: "The Great American Novel is not extinct like the Dodo but mythical like the Hippogriff...." As late as in 1973, Philip Roth quotes this dedication in his novel, The Great American Novel. There have been many American novels since the fin'desie`cle, but such an obssessive dream can hardly be realized, because such a novel should include all the aspects of American life, including geography, sociology, economics, politics, religion, psychology, etc. No one can possibly write the total spirit of American life. John Dos Passos and Thomas Wolfe, in their fanatic desire to express arid capture such totality, failed topresent the "assimilated experience," a sine qua non of literature. Based upon the historical phenomenan, the purpose of this article is to investigate how Philip Roth has treated the Idea of the Great American Novel and how his version of The Great American Novel can be related to the presentation of the zeitgeist of the 60's, the absurdist novel, and the black humor that has been prevalent in American literature, as in European literature, since the end of the World War II. For this purpose, in Section II, some attempt has been made to clarify how the novelist reduces the recent social confusion and restlessness in the form of novels. In Section III, the decade of "demythologizing of American myths," in particular of the baseball myth" has been discussed. The conditions of modern America are so grotesque that the only way to interpret and tolerate is through fun dominated by farce and fantastic illusions. The Great American Novel is an excellent exemplar of this. Several book reviewers were angry at the time of its publications As shown in Roth's My Life as a Man, his moral concerns are apparently lying under the surface of comicality in the Great American Novel. Smitty is a strangely unsatisfying character; however, when we come to see that he serves as "a kind of passageway from the imagenary that seems real to the real that seems imaginary, a continuum between the credible incredible and incredible credible," we come to see the American intellectual climate and the plight of the novelist in it. The reader should, in this respect, respond with deeper understanding and sympathy toward Roth's sixth novel. The Great American Novel.

        Mark Twain의 兩面性

        The purpose of this article is to eliminate the prevalent notion that Mark Twain became suddenly pessimistic after experiencing personal misfortunes toward the end of his life. Many critics, it is true, have been concerned with the duality of Mark Twain in terms of biographical or structural considerations. However, we encounter, with the subject of duality, one of Mark Twain's most persistent insights into man's nature. Therefore, the subject of Mark Twain's duality has been traced from The Prince and the Pauper (1882), in which he began developing the concept of his duality as a narrative device with thematic implications. The dark side of Man's nature is concretely shown in The Mysterious Stranger, Letters from the Earth, “The Damned Human Race,” and “What is Man?” This growing pessimism led him to speculate about the dichotomy of good-and-evil in relation to man's nature, Basically he found that the duality of good-and-veil is caused by the disparity between man's “low” impulses, drives, and instincts and “high” social and moral code. This disparity was obvious to some extent even in Huck Finn, Pudd'nhead Wilson, and other works. Therefore, we can conclude that the was interested in this duality even when his humor seemed light-hearted in the minds of many readers. His preoccupation with the dark side of man's nature, with him from the very beginning, was not merely a new development toward the end of his life.

      • Ceruloplasmin의 정제 및 안정성에 관한 연구

        The activity of Ceruloplasmin(EC 1.16.3.1, CP) in human plasma was measured in several conditions in order to study the stability of CP. The CP was partially purified by polyethyleneglycol 4000 and Sephadex G-200 chromatography. And the Cp was determined using a polyacrylamide gel electrophoresis with o-dianisidine stain. The activity of CP in human plasma was stabilized for 10days at 4 ℃ and frozen plasma was stabilized for 15days at -35℃ . The activity of CP was more stabilized in 0.05M sodium acetate buffer(pH 5.3) than 0.075M potassium phosphate buffer(pH 6.5). In addition, the activity of CP was stabilized in 0.05M sodium acetate buffer(pH 5.3) containing 0.225M NaCl.

      • Mark Twain의 社會批評

        It is my thesis that Mark Twain's social criticism was firmly based on his Americanism, not on such ideas as are proposed by Maxwell Geismar, who seems to suggest that Mark Twain declare the fall of his country. I think this is too much. Twain's social criticism was influnced, first of all, by his intense Americanism, the love of his country. In fact, Twain's strong Americanism showed itself in many ways; at times, he became quite jingoistic. His bitter and serious criticism of the American civilization is nothing but his explosion of the conflict between appearance and reality of democracy. Although Twain could find nobility and goodness in individuals, he regarded the human race collectively as stupid, mean, and despicable. He hated its sham and pretense, its hypocrisy, its vain pretensions, its cruelty and injustice, its lust for killing, its moral cowardice, its peversity. Twain's "damned human race" attitude was most pronounced in his later years when he was beset by a number of personal tradegies, traces of it can be found in his early sketches. These traces have been collected in this paper. Unedrneath the sparkling humor lay important moral and social criticism. It was unfortunate that he was regarded as a professional "funny man" without no particular literary ability except his unique sense of humor. His masterful use of language, the sharpness of descriptive detail, his vivid portrayal of character in his writings are to be emphasized; yet, his role as moralist and social critic should be pushed to the front, since his writings present much incisive comment on human society. Twain's acceptance of the philosophy of mechanistic determinism was never a complete one, and as a result there always remained a strange dichotomy in his view of man. On the other hand, his heart was filled with pity and sympathy for doomed mankind, the victim of forces beyond his control. He also raged at the human race, thus tacitly admitting a glimmer of hope for human betterment. The ambivalence of Mark Twain's view of man is reflected in his writings. But at heart Twain was also a romantic idealist and he believed that man's ideal could be trained upwards within the limits prescribed by his make. It was this glimpse of hope for mankind which prodded him into his role as moralist and social satirist. In this paper, Twain's Americanism, his ideal of democracy, his ideas for individual and society and his strong hatred for monarchy hove been investigated. He is a sharp and critical examiner of life, and his writings abound in incisive comment; however, he seldom has the patience to get to the core of the matter. This is his weakness as a social critic. As a result he frequently jumps to conclusions before he has thoroughly explored all aspects of the problem. His hasty judgements frequently lead him to hold contradictory views simultaniously. Thus, for instance, he repeatedly states his determinstic concept of man as a machine, but at the same time he reiterates his belief in the possibility of fostering higher ideas through training. In conclusion, I would like to add that Mark Twain's moral and social criticism suffers from his hit-and-run technique. This defect in him gives the impression of superficial thinking, even though he is a more profound thinker than most people realize.
