Surface plasmon resonance imaging microarray for fusion protein expression analysis

Surface plasmon resonance (SPR) is an optical technique used to detect the specific binding of unlabeled bio-molecules onto molecules attached to gold thin films by measuring changes in the index of refraction upon adsorption. The SPR technique can be expanded to SPR imaging and used for the high-throughput analysis of bioaffinity interactions by fabricating protein arrays on gold surfaces. A SPR imaging system was constructed and used to detect the affinity-tagged recombinant proteins expressed in Escherichia coli. In this study, we introduce a robust technique to rapidly detect the hexahistidine (His6)-, glutathione-S-transferase (GST)-, and maltose-binding protein (MBP)- fused proteins expressed in E. coli using SPR imaging measurement. The cell lysates were spotted on gold thin films coated with 11-mercaptoundecanol (MUOH)/dextran derivatized with Ni(II)- iminodiacetic acid (IDA-Ni(II)), glutathione, or cyclodextrin. After briefly washing the gold chip, SPR imaging measurements were carried out in order to detect the bound affinity-tagged fusion proteins. Using this new approach, rapid high-throughput expression analyses of the affinity-tagged proteins were obtained.
성인애착유형과 공감수준에 따른 부부 갈등해결전략의 차이

This study investigates how adult attachment styles and empathy levels influence on couples in choosing their conflict resolution strategies. The sample for this research consists of 135 couples (270 persons) who reside in Seoul and Kyung-gi areas. The data was analyzed using cluster analysis and two-way ANOVA to compare the differences between couples’ conflict resolution strategies, depending on their attachment styles and empathy levels. The results suggest four adult attachment types: secure type(39.3%), dismissing type(23.0%), preoccupied type(20.7%), and fearful type(17.0%). The interaction effects between adult attachment styles and empathy levels were found in compromising and integrating strategies. Specifically, secured, dismissing, and preoccupied types used more compromising and integrating strategies when their empathy levels were high, whereas fearful type showed no significant differences in using compromising and integrating strategies regardless of empathy level. Implications of the study results on couple counseling were discussed and further study topics were suggested. 본 연구는 부부가 갈등을 해결하기 위한 전략을 선택할 때 성인애착유형과 공감수준에 따라 어떠한 차이를 나타내는지 알아보기 위해 실시되었다. 이를 위해 서울 및 경기지역의 부부를 대상으로 총 135쌍(270명)의 자료를 분석하였다. 성인애착유형을 알아보기 위해 불안과 회피의 두 차원으로 K-평균 군집분석을 실시하였고, 성인애착유형과 공감수준에 따른 갈등해결전략의 차이를 알아보기 위해 이원변량분석을 실시하였다. 연구결과 성인애착유형은 불안과 회피차원의 높고 낮음에 따라 안정형(39.3%), 거부형(23.0%), 몰입형(20.7%), 두려움형(17.0%)인 네 가지 유형의 집단으로 분류되었다. 성인애착유형과 공감수준에 따른 갈등해결전략은 절충전략과 협력전략에서 유의미한 상호작용 효과가 나타났다. 즉 안정형, 거부형, 몰입형은 공감수준이 높을 때 절충전략과 협력전략의 사용이 높게 나타난 반면 두려움형은 공감수준에 따라 절충전략과 협력전략의 사용에 큰 차이를 보이지 않았다. 이러한 결과를 토대로 본 연구의 시사점과 후속연구를 위한 제언을 논의하였다.

Boron is a weakly-ionized species, which is hard to be removed completely by conventional water filtration processes for ultrapure water, such as ion exchange, reverse osmosis technologies. Alternatively, polymer based chelating agents are of great interest for boron removal due to their high boron adsorption capacity, provided by multiple hydroxyl groups. For multiple functional groups, glycidyl methacrylate(GMA) was introduced onto the surface of porous polymer beads via ATRP in the presence of divinylbenzene (DVB) as a crosslinking agent. Effect of crosslinking agent on the grafting yield and the morphological changes was investigated. Boron adsorption capacity of the fuctionalized beads was studied in therms of their grafting yield and porosity.

Alternatively, polymer based chelating agents are of great interest for boron removal due to their high boron adsorption capacity, provided by multiple hydroxyl groups. Here, core-shell type polymer particles were prepared via surface initiated polymerization to accommodate more N-methyl-D-glucamine (NMDG) groups. The core polymer beads were prepared though suspension polymerization of vinylbenzyl chloride (VBC) and divinylbenzene (DVB), and poly(glycidyl methacrylate) (PGMA) chains were grown from the core particles by ATRP. The grafted chain length was controlled by polymerization time, and their boron adsorption capacity was investigated after the functionalization with (NMDG).

URC는 기존 로봇에 네트워크 및 정보 기술을 접목한 지능형 서비스 로봇의 새로운 개념으로서, 언제, 어디서나 나와 함께 하며, 나에게 필요한 서비스를 제공하는 네트워크기반 로봇이다. URC 개념이 구현되기 위해서는 유비쿼터스 네트워크 또는 센서 네트워크, 고성능 로봇용 서버 등과 같은 하드웨어 인프라가 구축되어 있어야 하며, 이러한 인프라 상에서 구동되는 소프트웨어 인프라가 필요하다. 본 고에서는 URC의 소프트웨어 인프라의 구현에 요구되는 많은 기술 중 특히 상황인식 컴퓨팅 기술에 대해 논의하고 ETRI에서 개발중인 상황인식 미들웨어인 CAMUS에 대해 소개하고자 한다.
