      • 五倍子가 Alloxan에 의한 實驗的 糖尿病 마우스의 膵臟 glucokinase 및 hexokinase活性에 미치는 影響에 關한 硏究

        Glucokinase와 hexokinase는 간과 췌장에서 인슐린에 의해 혈당이용을 증대시키며 혈당농도를 감지하여 인슐린 분비를 조절하는 효소로서 인슐린비의존성당뇨병(NIDDM)의 병인과 밀접한 관계가 있는 것으로 알려져 있다. 또한, alloxan은 glucokinase의 활성부위에 존재하는 -SH기와 경쟁적으로 결합하여 효소활성을 저해함으로서 당뇨병을 유도한다. 본 연구에서는 당뇨병치료의 전통한방약재로서 사용되고 있는 五倍子추출물의 효과를 해명하기 위하여 alloxan유발 실험적 당뇨마우스의 glucokinase와 hexokinase의 활성을 검토한 겨과, alloxan주사에 의해 체중과 공복시의 glucose 및 insulin 분비가 증가되었으나, 五倍子투여로 공복시의 insulin분비가 감소하였으며 체중과 공복시의 혈당은 유의성은 없었으나 대조군에 비하여 감소하였다. 그러나, 혈당수치가 정상군에 비하여 현저하게 증가되었으나 五倍子투여로 현저히 감소하였으며 Insulin치의 상승과 분비지연도 五倍子투여군에서는 정상군과 유사한 경향을 보였다. 한편, 혈중glucokinase와 hexokinase의 활성은 alloxan주사에 의해 현저히 감소되었으나, 五倍子 투여로 유의성 있게 상승되었다. 이상의 결과로, 五倍子는 alloxan으로 유발된 실험적 당뇨병에서 포도당인 산화 효소인 glucokinase와 hexokinase의 활성을 증가시키는 것으로 나타나 당뇨병의 치료에 임상적인 효과가 있음을 시사하였다. The in vivo effect of an aquous extract from Rhois Galla (R-G) on glucokinase and hexokinase activities has been examined in mice having diabetes mellitus induced by alloxan, which specifically bind to -SH group of the enzymes and inhibits the enzyme activities. After 1 week of alloxan injection, the levels of serum glucose and insulin secretion were dramatically increase, however, the insulin secretion was decreased with administration of R-G in alloxan-treated mice. Alloxan injection allowed the serum glucose level increased and the level was decreased by R-G administration. Furthermore, it has been observed that R-G was effective in recovering the levels of insulin secretion in the alloxan-treated diabetic mice. Enzyme activities of the glucokinase and hexokinase were decreased by alloxan treatment. In contrast, R-G administration to the mice allowed proportional increasing. These results strongly suggested that R-G is highly effective in treatment of diabetes mellitus induced by alloxan.

        허혈(虛血)/재관류(再貫流) 뇌(腦) 조직(組織)에서 녹용(鹿茸) 약침액(藥鍼液)의 항산화작용(抗酸化作用)에 대한 연구(硏究)

        Cervus elaphus, being known to reinforce Kidney, have tested to study the effects concerning damages of brain tissue induced by lipid peroxidation. In vitro, the level of lipid peroxide in brain tissue was decreased proportinally according to dose by Cervus elaphus extract solution for aqua-acupuncture (CESAA). It was much more decreased, when lipid peroxidation was induced with Fe(II). And, it was seen proportinally decrease according to the dose of CESAA on xanthine oxidase activities and type conversion ratio. However, I can not find special changes about aldehyde oxidase activities. And, I had observed the effects of CESAA on damages of rat's brain following ischemia and reflow. Before ischemia was caused, CESAA was applied 0.2 ml per 250 g through femoral vein in ischemia and reflow group and normal sailine was applied in normal group. Ischemia was caused by cervical artery's clamp for 30 min and reflowed by clamp remove after 15 min. It was increased on the content of lipid peroxidation, activies and type conversion ratio of xanthine oxidase following ischemia and reflow. However, they were decreased when CESAA was pre-appllied. However, it could not seen special changes on aldehyde oxidase activities, either. In conclusion. CESAA recovers the damage of brain due to ischemia and reflow by decreasing the lipid peroxidation through decreasing of xanthine oxidase activies and type conversion ratio.

        청아환(靑娥丸)에 의한 활성(活性) 산소류(酸素類)의 소거(消去) 작용(作用)과 항산화(抗酸化) 효소계(酵素系)의 활성(活性) 증가(增加) 효과(效果)에 대(對)한 연구(硏究)

        This study was undertaken to examine the effect of Cheongahwan(CAH), being known to reinforce Kidney-yang, on the activities of endogenous antioxidant enzymes and the production of oxygen free radicals in the kidney tissues. Alterations in enzyme activities were observed after in vivo treatment in rats. CAH caused a significant increase in the activities of superoxide dismutase (SOD), glutathione peroxidase and glutathione S-transferase. But catalase activity was not significantly altered by CAH. Treatment in vitro of CAH decreased the production of oxygen free radicals in a dose-dependent fashion. These results suggest that CAH stimulate the activities of antioxidant enzymes and inhibit directly the production of oxygen free radicals. These effects of CAH may contribute to prevent the oxygen free radical-induced impairment of cell function.

      • Effect of Rhois Galla Extract on Glucokinase and Hexokinase Activities of Alloxan-Induced Diabetes Mellitus Mice

        The in vivo effect of an aquous extract from Rhois Galla (R-G) on glucokinase and hexokinase activities has been examined in mice having diabetes mellitus induced by alloxan, which specifically bind to -SH group of the enzymes and inhibits the enzyme activities.         신장(腎臟) 조직(組織)에서 제조의 항산화(抗酸化) 효과(效果)의 기전(機轉) 연구(硏究)

        This study was carried out to examine mechanisms by which Holotrichia (HTC) produces protective effect against renal cell injury. HTC extraction (5%) prevented $H_2O_2(50mM)$-induced LDH release and lipid peroxidation in renal cortical slices. When slices were treated with 5% HTC extraction, cellular glutathione content and the superoxide dismutase activity were not altered in control and $H_2O_2$-treated tissues. When slices were treated with 50 mM $H_2O_2$, the catalase activity was significantly inhibited, which was completely restored by addition of 5% HTC. Treatment of slices with 5% HTC extraction increased the glutation peroxidase activity in $H_2O_2$-treated tissues. These results suggest that HTC prevents oxidant-induced cell injury and lipid peroxidation by increasing the activities of catalase and glutathione peroxidase in renal cortical slices.

        이정환(二精丸)이 노화과정에 미치는 영향

        Objectives: It is well known that aging and aging-related diseases are linked to the increased level of oxidative stress caused by reactive oxygen species(ROS) and reactive nitrogen species(RNS). Nonprotein-SH decreases during aging, while substances such as ROS, nitric oxide(NO), peroxynitrite($ONOO^-$), myeloperoxidase(MPO), and dityrosine show a significant increase. This study investigated the effect of Ichungwhan on the aging process by examining its effect on the generation of the above-mentioned substances. Methods: Four comparison groups of SD rats were used. They were 6 month-old rats, 24 month-old rats, and 24 month-old rats fed with food containing 0.1% and 0.3% of Ichungwhan extract. The amount of NO, $ONOO^-$, and ROS in the rats' kidneys were examined using a fluorescence microplate reader. The reagents used for this purpose include: dihydrorhodamine 123 (DHR 123), 2',7' -dichlorodihydrofluorescein, diacetate(DCFDA), and 4,5-diaminofluorescein(DAF-2). A spectrophotometer was used to investigate the reactivity of nonprotein-SH and myeioperoxidase(MPO), using reagents such as trichloroacetic acid(TCA) and tetramethylbenzidine(TMB). The amounts of MPO protein and dityrosine were measued by western blot. Results: The observed effects of Ichungwhan on rats were as follows: increase of nonprotein-SH; effective decrease of RNS level by suppression of the generation system of $ONOO^-$ and NO; decrease of ROS level; decrease of the MPO reactivity and the subsequent reduction of amount of MPO protein; retardation of dityrosine formation. It can be hypothesized, therefore, that Ichungwhan affects both the earlier and later phases of the molecular inflammatory process, and retards the aging process. Conclusions: Empirical evidence in this study supports a role for Ichungwhan in generation mechanisms of aging process-linked substances ROS, NO, $ONOO^-$, nonprotein-SH, MPO and dityrosine. Affects contrary to the aging process observed in rats beg further empiricism to investigate potential application of Ichungwhan as a medication for age-related diseases in humans.
