        작약(Paeonia lactiflora Pall.) 동아의 경정배양을 통한 기내증식

        작약동아의 정아 및 액아의 기내배양에 의한 유묘의 증식에 필요한 배양조건을 구명코자 실험한 결과는 다음과 같다. 정아배양의 경우, 생장조절재의 조성에 무관하게 두지방종에서 100% 신초가 신장하였으나 생장은 의성지방종 은 NAA 0.1 mg/L 단용배지, 영천지방종은 NAA 0.01 mg/L 단용배지에서 가장 양호하였다. 액아배양의 경우, NAA 0.01 mg/L와 zeatin 5.0 mg/L 혼용배지에서 의성지방종은 100%, 영천지방종은 50%의 가장 높은 신초신장율을 보였고 신초의 생장도 양호한 경향이었다. 정아 및 액아배양으로부터 유도된 신초는 vermiculite를 지지물로 한 NAA 0.1 mg/L 첨가배지에서 30.0%의 발근율을 보였으나 뿌리의 생장은 양호하였다. The experiment was conducted to identify the optimal in vitro propagation condition for P. lactiflora Pall. Through apical shoot tip and axillary shoot tip culture of winter bud. When apical shoot tip and axillary shoot tips excised from winter bud were cultured on MS medium supplemented with various concentrations of plant growth regulators, all the apical shoot tips elongated regardless of the composition of the medium but axillary shoot tips responded differently. Shoot of 'Uisong' local cultivate was well elongated in the medium containing 0.01mg/L NAA. Frequency of shoot formation and subsequent shoot growth in axillary shoot tip culture were promoted in the medium containing 0.01 mg/L NAA and 5.0mg/L zeatin. 30% of the elongated shoots were vigorously rooted on the medium containing 0.1mg/L NAA with vermiculite as a support medium.

      • Petunia Hybrida 의 엽내조직으로부터 원형질체 유리 및 배양후 Callus 형성에 미치는 요인

        The study was conducted to improve protoplast isolation and fusion efficiency of mesophyll of Petunia hybrids $quot;Cascade red.$quot; The protoplast culture was also attempted to determine 1-naphtalene acetic acid concentration added to basal medium, 8p-Kao and Michayluk(8p-KM), for colony and callus formation. Preplasmolysed mesophyll of Petunia hybrids was well digested in enzyme solution containing 0.5% macerozyme R-10, 2.0% cellulase Onozuka R-10, 0.01M MES buffer, and 0.7M mannitol adjusted pH to 5.8, and protoplasts with debris were washed with 0.6M mannitol solution once and purified with 0.7M sucrose solution twice. When PEG solution had been eluted with eluting solution which was adjusted pH to 9.0-10.5 after 30% PEG solution treatment, fusion frequency was highly developed. Callus was formed in 8p-KM medium containing 1.0ppm 2,4-D, 0.5ppm BA and 1.0ppm NAA, and 0.1M mannitol which was added as osmoticum.

      • 일선관료제에 대한 비교연구 : 파출소와 동사무소를 중심으로

        본 논문은 일선관료제의 한국적 의미를 파악하고 이러한 일선관료제가 조직내부 또는 조직외부와의 관계 속에서 어떠한 제약조건을 가지고 있는지를 연구하는데 초점을 둔다. 특히 개념적으로는 동일한 일선관료제라 할지라도 그것이 어떤 조직맥락과 환경맥락에 직면해 있느냐에 따라 각 기관은 각기 다른 제약조건에 직면에 있으며 이로 인해 각기 다른 차별적인 조직형태와 관료행태를 보일 수 있다는 문제의식을 갖고 접근한다. 본 논문에서는 이러한 문제의식에 기반하여 일선관료조직 중 파출소와 동사무소를 선정하고 두 조직이 지니고 있는 제약조건을 공통점과 차이점이라는 시각에서 파악하고 그러한 제약조건하에서 어떠한 조직형태와 관료행태가 차별적으로 나타나는지 논의한다. 이상의 논의를 통해 일선관료가 본원적인 기능 즉, 대민지향적 업무를 수행할 수 없는 구조적인 제약점들을 파악할 수 있을 것이다. 본 논문이 가질 수 있는 함의는 우선 한국적 맥락에서의 일선관료제를 분석해 봄으로써 서구에서 논의되는 일선관료제와 우리나라 일선관료제가 가지는 차변성을 파악할 수 있다는 점이며, 다음으로 이러한 기본시각에 근거하여 우리나라 일선관료가 가지는 제약요인을 나름대로 파악할 수 있다는데 있다. 이상의 논의를 통해서 단순히 기계적으로만 일선관료제를 파악하여 효과성의 측면의 개혁을 강조하는 근자의 경향에 논박을 가하고자 한다.

        EFFECT OF PARAMETERS RELATED TO CHANNEL AND DESICCANT ON THE SIZE OF DESICCANT ROTOR

        The desiccant rotor is the most essential component of desiccant cooling system, but one of its drawbacks to spread out is its rotor size. To reduce the size of rotor the analysis of rotor performance is crucial. Systematic examination on the effect of desiccant and channel geometrics has been conducted based on the numerical program previously developed. Considered parameters related to channel geometrics are channel shape and cross-section area of channel, and parameters related to desiccant are mass fraction, heat capacity, density, maximum water uptake and separation factor of isotherm. Considerable reduction of rotor size is expected by adjusting the parameters.

        순환골재의 품질평가를 위한 시험방법 개선에 관한 실험적 연구

        This study investigates the saturation level of surface dryness, quantity of adhesive mortar, and the alien substance content of recycled aggregates for concrete to develop an adequate quality testing method for understanding the properties of recycled aggregates, which differ greatly from preexisting aggregates. For tests that measure the saturation level of surface dryness, where detail methods are applied differently according to the tester, various testing methods from across world were compared and analyzed. This study revealed that when measuring the saturation level of surface dryness of a certain sample, aggregates must be supplemented immediately whenever the height of the sample becomes lower than the measuring mold, and allowing the tamper to free fall on the sample will provide the most accurate results. When measuring the quantity of adhesive mortar of recycled aggregates for concrete, an acid solution was used, and since the quantity of adhesive mortar increases as the particle sizes gets smaller, the sample for testing should represent the entire granularity. Sulfuric acid solution is adequate for immersion, and the concentration should be 20% for best results. According to the alien substance content measurement, which was examined by the naked eye, the error range caused by the difference in particle size was neglectable, and therefore the sample should be 2.5~5.0mm in size concerning the accuracy and measuring time. Also, for coarse recycled aggregates, the sample should amount to 1kg for measuring alien substance content by the naked eye, which proves that assortment by the naked eye is the most adequate method for measuring the alien substance content of a recycled aggregate.         재생온도가 저온인 경우 제습로터의 재생부/제습부 면적비율과 회전속도 최적화

        The desiccant rotor is the most essential component of desiccant cooling system, but its design relies on manufacturer’s experience and principles are not yet clear in spite of a lot of theoretical/experimental work published. The mathematical modeling of desiccant rotor needs solution of coupled partial differential equations of heat and mass transfer. In this study, numerical program is developed and validated using a real desiccant rotor. The calculation results are in reasonable agreement with the experimental data and other available numerical results. Optimization of desiccant rotor on the condition of low regeneration temperature are investigated. The optimal rotor speed at which the process outlet humidity becomes minimized, shows same as that of the system optimization. Compared to high regeneration temperature, broad is the range of optimal speed of low regeneration temperature. Systematic analysis on the optimal area ratio of regeneration to dehumidification section has also been conducted.
