        유기폐자원을 이용한 고도하수처리 대체탄소원 개발 및 실플랜트 적용

        The purpose of this research was evaluated economical effect to apply alternative external carbon source. Conventional activated sludge process in municipal wastewater treatment plant was adapted and introduced to Biological nutrient removal processes to meet the newly enforced effluent quality standard for nutrient removal in Korea. Low COD/NH₄^(+)-N ratio and higher nutrient concentration of influent characteristics force to inject external carbon source for denitrifying recycled nitrate. In the most case, methanol was used as external carbon source. But Methanol is expensive and very dangerous in handling. So we could find cheaper and safer external carbon source substituted methanol in last study. This alternative external carbon source is named RCS(recoverd carbon source) and a by-product of fine chemical product at chemical plant. When RCS was applied real municipal wastewater treatment plant, average 55~65% of T-N removal efficiency, 8.8mg/l of effluent T-N concentration, 11.3mg/l of effleunt COD concentration were obtained without effluent COD increase as against used methanol. To apply RCS in municipal wastewater treatment plant obtain approximately 74.5% expenditure cost reduction in comparison with methanol dosage cost.

        애드거 앨런 포의 소설 「아서 고든 핌 이야기」의 공간묘사에 대한 지리학적 분석

        이 연구의 목적은 애드거 앨런 포의 소설 「아서 고든 핌 이야기」의 공간묘사에 대해 지리학적 관점에서 고찰하는 것이다. 이를 위해 먼저 소설에 언급된 지리적 내용을 항해가 진행되는 시간 순으로 살펴보았다. 둘째, 동시대에 간행된 지도를 통해남극 탐사 자료를 인용하는 부분의 공간적 맥락을 살펴보았다. 셋째, 남극권을 넘어 항해하는 경로를 지도 상에 표시하고, 항해경로의 공간적 맥락을 살펴보았다. 넷째, 포가 활용한 지리적 자료의 출처와 인용 의도를 분석했다. 마지막으로 소설의 말미에 언급된남극의 하얀 장막과 지구공동설과의 관련성을 살펴보았다. 연구결과 심스의 지구공동설과 모렐의 항해기를 모티브로 공간묘사가이루어졌음을 확인했다 The purpose of this study is to consider the spatial description of Poe's The Narrative of Arthur Gordon Pym, from a geographic point of view. To do this, we first looked at the geographic content mentioned in the novel in chronological order. Secondly, the spatial context of the citation of Antarctic exploration data was examined through contemporary published maps. Third, the route of sailing over the Antarctic Circle is displayed on the map published in the same period, and the spatial context of the route is examined. Fourth, the source and citation intentions of the geographic data Poe used were analyzed. Finally we considered the relationship between the white curtain of the South mentioned at the end of the novel and Hollow earth theory. The results confirm that the spatial description of the novel is based on Symmes’ hollow earth theory and Morrell's journal.

      • Oligonucleotide microarray를 이용하여 10-Hydroxycamptothecin이 처리된 사람 백혈병 HL-60 세포에서의 유전자 발현양상의 분석

        Background Camptothecin is an antitumor alkaloid that has been isolated from the Chinese tree, Camptotheca acuminata.. DNA topoisomerase I has been firmly established as the molecular target of the camptothecin group of anticancer drugs. These drugs include 10-hydroxycamptothecin(10-CPT), topotecan, 9-aminocamptothecin, and irinotecan. Many of them are now in clinical trials and are showing activity against a wide variety of solid human malignancies. DNA chip microarray technology can be used to establish associations between characteristic gene expression patterns and molecular responses to drug therapy. This study was conducted to determine whether pretreatment of HL-60 human leukemia cells with the topoisomerase I - directed drug 10-CPT regulates gene expression and to investigate the activity of topoisomerase mediated by 10-CPT in nuclear extract from HL-60 human leukemia cells. Methods We have conducted experiments on cell cytotoxicity in drug-treated cellls, topoisomerase purification, enzyme assay using agarose gel electrophoresis and oligo chip microarray analysis, respectively. Results Treatment of HL-60 cells with 10-CPT resulted in marked inhibition of type I topoisomerase activity. 10-CPT IC(50) values was 8 nM for HL-60 cells. Thirty-one genes that were up-regulated(18 genes) and down-regulated(13 genes) in 10-CPT treated HL-60 cells were found. Conclusion Our results suggest that the various expression profiles of newly identified genes in 10-CPT treated human leukemia cells by oligonucleotide microarray can be potentially used as novel therapeutic markers.

        미나르 지도의 지도학적 특성과 주제도 발달에 미친 영향

        미나르는 19세기의 가장 창의적인 지도제작자로 알려져 있다. 그러나 그의 지도에 대한 연구는 지도학분야에서는 거의 이루어지지 않고 있다. 본 연구에서는 미나르의 지도의 지도학적 특성과 그의 지도가 주제도 발달에 미친 영향을 고찰하는 것을 목적으로 한다. 이를 위해 먼저 미나르의 지도가 19세기 주제도 발달사에서 가지는 의미를 고찰하였다. 그의 형상적이고 개락적인 지도는 프랑스의 국토정책 의사결정을 위한 것으로 지도학적 정확성보다는 신속하게 인지가 가능하도록 일반 메시지 전달에 중점을 두고 제작되었다. 둘째, 미나르 지도 중 유선도와 단계구분도와 도형표현도, 그리고 나폴레옹의 1812년 러시아 원정 지도의 지도학적 특성을 살펴보았다. 셋째, 미나르의 지도가 이후의 주제도 발달과 지리적 시각화에 미친 영향을 살펴보았다. 미나르는 유선도와 파이 원 지도를 발전시켰는데, 그의 지도 제작 방식은 이후의 프랑스 통계 지도첩 제작에 직접적인 영향을 미쳤다. Charles Joseph Minard is known as the most creative mapmaker of the nineteenth century. But research on his maps are scarcely done in cartographic domain. The objective of this paper is to investigate the cartographic characteristics of maps of Minard and their influence on the development of thematic mapping. For this, this paper begins by considering the development background of the first half of the nineteenth century thematic cartography. Minard’s figurative and approximative maps were intended for French national land policy decision-making, so they were produced to convey general message rather than cartographic accuracy. Secondly, cartographic representation techniques of flow map, choropleth map, proportional symbol map, and Napoleon’s 1812 Russian campaign were examined. Thirdly, The influence of his mapping style on thematic maps and geovisualization were considered. He is considered as the developer of flow map and pie-chart map. His mapping style was adopted by the statistical atlases of France.

      • Effect of Mistletoe on Activity of NK cell in Endometriosis

        The purpose of this study is to investigate the effect of mistletoe on NK cell cytotoxicity and its mechanism of action on NK cells in the endometriotic peritoneal fluid. NK cell line was incubated in peritoneal fluids from women between 20 and 40 years of age (divided into two groups according to the disease stage: stage 1 & 2 - group A, stage 3 & 4 - group B) who underwent laparoscopic surgery from January 2011 to December 2011, and the cytotoxicity of NK cells against target cells (K562 cells) was evaluated. The cytotoxicity of NK cells before mistletoe treatment was compared with the cytotoxicity of NK cells after mistletoe treatment, and the effect of mistletoe on NK cell apoptosis was evaluated. In addition, the expression of CD107a, which directly participates in exocytosis of cytotoxic granules, was compared. The cytotoxicity of NK cells showed a significant difference between the control peritoneal group, the endometriosis group A and group B (75.55%, 69.59% and 63.8%, respectively). The cytotoxicity of NK cells was diminished according to the stage of endometriosis (p=0.032). The cytotoxicity of NK cells before and after treatment with 200 ng/ml mistletoe was 81.64% vs 87.75% (p=0.012) in the control peritoneal fluid group, 78.30% vs 86.40% (p=0.003) in the endometriosis group A, and 73.67% vs 84.54% (p=0.024) in group B, respectively. No significant difference was observed between NK cell apoptosis before treatment with mistletoe and NK cell apoptosis after treatment with mistletoe in all groups; 1.94% vs 1.93% in the control peritoneal fluid group, 1.74% vs 1.77% in group A, and 1.49% vs 1.27% in group B (p=0.373). The expression of CD107a was significantly increased in each group after mistletoe treatment. The expression of CD107a before and after mistletoe treatment was 0.70% vs 1.67% (p=0.012) in the control peritoneal fluid group, 0.59% vs 1.10% (p=0.002) in group A, and 0.79% vs 1.40% (p=0.014) in group B, respectively. Mistletoe directly influenced exocytosis of cytotoxic granules by increasing the expression of CD107a in NK cells, and consequently there was a recovery of cytotoxicity of NK cells. Therefore, it can be expected that mistletoe will be significantly effective in the treatment of endometriosis

      • Human c-myc 원암유전자에서 Benzo(a)pyrene 이 DNA Topoisomerase Ⅱ에의한 DNA 절단에 미치는 영향

        In a previous study, we have found that benzo(a)pyrene(BP) and it's metabolites specifically bind to the DNA topoisomerase Ⅱ fraction of mouse fibroblast C3H/l0T1/2 cell cultures. The enzyme was predicted to be a primary target of BP causing malignancy. We therefore investigated, in the present study, changes in DNA cleavage by various concentrations of BP bound to DNA topoisomerase Ⅱ in the human c-myc protooncogene. DNA topoisomerase Ⅱ was purified by glycerol gradient centrifugation from murine L1210 leukemia cells. Supercoiled pBR322 DNA was completely reversed to a relaxed form in the presence of 160ng of purified DNA topoisomerase Ⅱ. In BP-treated Hi-ndⅢ/XbaI cutting c-myc DNA, DNA cleavage was most apparently increased in the upstream of exon 1, the effect being particularly pronounced with 0.1㎛ BP. It is indicated that stimulation of the DNA topoisomerase Ⅱ induced DNA cleavage sites in the c-myc by BP act on selective region, which increased cleavage at specific sites may induce gene expression of abnormal pattern.
