rnpB 유전자 발현에 RNA 이차구조가 미치는 영향

E. coli rnpB gene의 발현에 영향을 미치는 M1 RNA structural gene의 부분을 조사하였다. P-1 promoter 에서 합성되는 RNA의 5' 부분이 이루는 이차구조를 분석한 결과 +1 에서 부터 +72 까지에 존재하는 hairpin loop 구조가 rnpB gene의 발현에 중요한 역할을 하는 것으로 밝혀졌다.

성장속도에 따른 대장균 rnpB 유전자 발현의 조절작용

Growth rate-dependent expression of the rnpB gene encoding M1 RNA, the RNA component of Escherichia coli RNase P, was examined using a hybrid transdcuing λ phage. The hybrid transducing λ phage was constructed carrying the lacZ gene fused to the promoter for the rnpB gene. The amounts of β-galactosidase were analyzed in the E. coli lysogen carrying this transducing phage grown in different growth media to see dependence of the rnpB transcription on growth rate. The synthesis of β-galactosidase from the rnpB-lacZ fusion increased with growth rate. This result suggests that the rnpB promoter activity is dependent on the growth rate.

釜山市民의 經濟意識에 관한 調査硏究 : 釜山地域 대학생의 經濟倫理를 중심으로

본 연구는 부산시민 특히 부산지역 대학생들의 경제의식 및 경제윤리수준에 관한 조사 분석연구로서 그 중요한 결과를 요약하면 다음과 같다. 첫째, 근면에 관한 의식은 근면의 가치를 높이 평가하는 기존의 연구와 유사한 결과이며, 세대간 지역간 차이도 미미하였으나 직업에 관한 의식을 사회적 가치보다는 보수를 중시하는 것으로 나타났다. 둘째, 소비의식은 도시적 특성을 반영하여 유행과 브랜드에 따른 구매경향이 강했으며 절약의식의 경우 구세대가 자신들보다 절약의식이 강하다고 평가하였다. 가설검증의 결과를 보면 대학생의 윤리의식을 결정하는 주요통제변수로서 교육적인 요인 즉 종교적 배경, 보호자의 직업 ·생활수준 등이 성별이나 성장배경보다 더 중요한 것으로 나타났으며, 부산지역 대학생의 경우 상대적으로 직업의식과 소비의식이 근면 ·절약 ·협동의식에 비해 낮은 것으로 나타났다.

REGULATION OF M1 RNA GENE EXPRESSION IN E. COLI

M1 RNA의 상위 염기배열이 M1 RNA 촉진제로 부터의 갈락토키나제 합성에 미치는 영향을 조사하기 위하여 GalK 발현 벡타에 클론된 E. coli M1 RNA 유전자의 촉진제 영역을 조사하였다. M1 RNA 유전자가 효율적으로 발현되기 위해서는 -40 에서 -60 까지의 염기배열이 필요하지만 -60 위의 염기배열음 중요한 역할을 하지 않았다. 또한 C5 단백질이 클론된 M1 RNA 유전자에서 부터의 M1 RNA 합성을 감소시킨다는 결과를 얻었다. 이는 M1 RNA 합성에 억제시스템이 존재한다는 것을 시사한다. The promoter region of the E. coli M1 RNA gene cloned into the GalK expresstion vector was analyzed to examine whether upstream sequences affected the level of galactokinase from M1 RNA promoter. Although the sequences between -40 and -60 were required for the efficient expression of the M1 RNA gene, the upstream sequences of -60 showed no significant effects on the expression. We also show that functional C5 protein decreased M1 RNA synthesis from the cloned M1 RNA gene, which implies the presence of repression system in M1 RNA synthesis.

성인학습자의 평생학습 참여경험 및 자기주도 학습이 자기효능감에 미치는 영향

The purpose of this study was to investigate the influence of adult learner's lifelong learning participation experience and self-directed learning on self-efficacy. For achieving the purpose, lifelong learning participation questionnaire, self-directed learning test, and self-efficacy test were administrated using 376 adult learners. The collected data was statistically processed using descriptive statistics analysis, correlation analysis, and multiple regression analysis. The main results were as the follows. First, adult learners who had lifelong learning participation experience were less than those who had no it. Second, there were statistically significant correlations between the total of self-directed learning and the total of self-efficacy and between subscale of self-directed learning and subscale of self-efficacy mostly. Finally, after controlling adult learner's gender, age, and educational level, previous lifelong learning participation experience statistically significantly influenced only self-regulated efficacy, subscale of self-efficacy. And self-directed learning generally influenced self-efficacy statistically and significantly. In particular, among subscales of self-directed learning, creative approach, responsibility acceptance of learning, attachment to learning, and future-oriented influenced the total of self-efficacy, academic efficacy, and self-regulated efficacy statistically and significantly. The discussions on the main results and suggestions for further research were provided. 본 연구는 성인학습자들을 대상으로 평생학습 참여경험 유무와 자기주도 학습이 자기효능감에 미치는 영향을 검증하는데 그 목적이 있다. 이러한 연구목적을 달성하기 위하여 성인학습자 376명을 대상으로 평생학습 참여 질문지, 자기주도 학습 검사, 자기효능감 검사를 실시하였다. 수집된 자료는 기술통계분석, 상관분석, 중다회귀분석을 사용하여 통계 처리하였다. 주요 결과를 살펴보면 첫째, 평생학습 참여경험이 있는 성인학습자들이 없는 성인학습자들보다 적었다. 둘째, 성인학습자의 자기주도 학습과 자기효능감 간의 관계를 알아본 결과, 자기주도 학습 전체와 자기효능감 전체 간, 자기주도 학습 하위요인과 자기효능감 하위요인 간 대부분에서 통계적으로 유의한 상관이 있는 것으로 나타났다. 셋째, 과거 평생학습 참여경험 유무와 자기주도 학습이 자기효능감에 미치는 영향을 알아보기 위해 중다회귀분석을 실시한 결과, 성인학습자의 성별, 연령, 학력을 통제한 후에 과거 평생학습 참여경험 유무는 자기효능감 하위요인 중 자기조절 효능감에만 통계적으로 유의한 영향을 미쳤다. 그리고 대체로 자기주도 학습은 자기효능감에 통계적으로 유의한 영향을 미치는 것으로 나타났다. 특히, 자기주도 학습 하위요인 중 독창적 접근, 학습의 책임성 수용, 학습에 대한 애착, 미래지향성이 자기효능감 전체, 학업적 효능감, 자기조절 효능감에 통계적으로 유의한 영향을 미치는 것으로 나타났다. 주요 연구결과에 대한 논의와 향후 연구를 위한 제언을 제시하였다.

The DNA sequences required for heat shock regulated expression were examined in Drosophila melanogaster. Various hsp70-YP1 fusion constructs where D. melanogaster hsp70 gene and YP1 gene were used as a heat shock promoter and as a target gene, respectively, were transfected into D. melanogaster Schneider line 2 cells and their heat inducibility was determined by Northern analysis. The sequences containing two HSEs between -89 and -38 of the hsp70 were not sufficient to induce YP1 RNA upon heat shock whereas the sequences between -89 and +34 including the TATA box and the transcription start region of the hsp70 besides the HSEs were able to drive YP1 transcription during heat shock. The small deletion of the sequences near the transcription start region of the YP1 gene was also observed to repress the heat induced transcription of YPl RNA by the sequences between -89 and +34 of the hsp70.

Biosynthesis of Plasmid-borne M1 RNA Transcripts and Their Effect on the Escherichia coli rnpB Gene Expression

M1 RNA transcripts synthesized from plasmids were analyzed and their effect on the Escherichia coli rnpB gene expression was examined to see whether the overexpressed M1 RNA transcripts affect rnpB pormoter activity. Whereas abnormal M1 RNA transcripts were metabolically unstable, normal M1 RNA transcripts were accumulated. The accumulated M1 RNA transcripts showed no signigicant effect on the galactokinase synthesis directed by the rnpB promoter. This result suggests that M1 RNA itself might not be involved in the regulation of the rnpB gene expression for RNase P biosynthesis.

The small deletion of the sequences near the transcription start region of the YP1 gene was also observed to repress the heat induced transcription of YP1 RNA by the sequences between -89 and +34 of the hsp70. 초파리 Drosophila melalnogaster에서 고열충격 유발을 조절하는데 필요한 DNA 염기배열을 조사하였다. hsp 70 유전자를 고열충격 촉진자로, YP1 유전자를 표적유전자로 하여 hsp70-YP1 융합유전자를 제조하고 이들 융합유전자를 이용하여 초파리 Schneider line 2 세포를 형질변환시키고 Northern 분석을 함으로써 hsp70-YP1 융합유전자의 고열충격 유발성을 결정하였다. 두개의 HSE (heat shock element) 가 존재하는 hsp70 유전자의 -89에서 -38까지 염기배열만으로는 고열충격에 의해 YP1 RNA를 유발시킬 수 없었지만 두 개의 HSE 이외에 hsp70 유전자의 TATA 박스와 전사시작점을 포항하고 있는 -89에서 +34까지 염기배열은 고열충격에 의해 YP1 RNA를 유발시킬 수 있었다. 또한 YP1 유전자의 전사시 작정 근처의 염기배열이 조금 결실되었을 때 hsp70 유전자의 -89에서 +34까지 염기배열에 의한 YP1 RNA의 고열충격 유발이 억제되는 것이 관찰되었다.

대장균에서 많은 수의 플라스미드에 존재하는 rnpB 유전자 전사의 성장기의 따른 조절 작용

The Escherichia coli rnpB gene encodes M1 RNA, the catalytic component of RNase P which is a processing enzyme for tRNA maturation. Biosynthesis of M1 RNA is known to be stringently regulated. The rnpB transcription under different growth phases of E. coli was examined using the truncated rnpB in the multicopy plasmids which could generate the metabolically unstable M1 RNA transcript. Biosynthesis of this RNA was directly analyzed during the transition from the exponential phase of growth to the stationary phase. During the transition the rnpB transcription became repressed. The discriminator motif CGCC adjacent to the transcription start-point of the rnpB gene was altered to the TATT using site-directed mutagenesis. The modified promter containing the mutated discriminator was at least partially rescued from this growth phase-dependent repression of the rnpB transcription. The results clearly show that the discriminator motif play a key role in control of the rnpB transcription under different growth phases.
