        Multivariate Characterization of Common and Durum Wheat Collections Grown in Korea using Agro-Morphological Traits

        Developing improved wheat varieties is vital for global food security to meet the rising demand for food. Therefore, assessing the genetic diversity across wheat genotypes is crucial. This study examined the diversity of 168 durum wheat and 47 common wheat collections from 54 different countries using twelve agro-morphological parameters. Geumgang, a prominent Korean common wheat variety, was used as a control. Both qualitative and quantitative agronomical characteristics showed wide variations. Most durum wheats were shown to possess dense spikes (90%), while common wheats showed dense (40%) or loose (38%) spikes, with yellowish-white being the dominant spike color. The majority of the accessions were awned regardless of wheat type, yellowish-white being the main awn color. White or red kernels were produced, with white kernels dominating in both common (74%) and durum (79%) wheats. Days to heading (DH) and days to maturity (DM) were in the ranges of 166–215 and 208–250 days, respectively, while the culm length (CL), spike length (SL), and awn length (AL) were in the ranges of 53.67–163, 5.33–18.67, and 0.50–19.00 cm, respectively. Durum wheats possessed the shortest average DH, DM, and SL, while common wheat had the longest CL and AL (p < 0.05). Common wheats also exhibited the highest average one-thousand-kernel weight. Hierarchical cluster analysis, aided by principal component analysis, grouped the population into seven clusters with significant differences in their quantitative variables (p < 0.05). In conclusion, this research revealed diversity among common and durum wheat genotypes. Notably, 26 durum wheat and 17 common wheat accessions outperformed the control, offering the potential for developing early-maturing, high-yielding, and lodging-resistant wheat varieties.

        국제미작연구소(International Rice Research Institute)의 종자 건전성 관리체계와 우리나라 적용 가능성

        Seedborne pathogens can reduce the seed quality and have the potential to function as inoculum in the field and to cause severe damage to crop production and seed for international trade. The International Rice Research Institute (IRRI) which develops new rice varieties and rice crop management techniques operates Seed Health Unit (SHU) in cooperation with the Plant Quarantine Service of the Philippines. SHU ensures that seed lots meet the quarantine requirements of the Philippines and the recipient countries. It conducts seed health testing by dry seed inspection, washing test, blotter test, or agar plate test. Seed health testing has been done on more than 500,000 rice seed lots, and a total of more than 80 species of fungi were detected. Seeds are treated by physical, chemical, or both methods to avoid introduction and spread of seedborne pathogens. The seed health management system of IRRI can be introduced to South Korea in appropriate way for circumstances. It will improve the quality of the plant germplasms and promote the aquisition of the plant genetic resource by exchange of the high-quality germplasms. 종자 건전성(seed health)은 종자 또는 식물체에 유익하지 못한 영향을 미칠 수 있는 생물체의 감염상태를 의미한다. 이들 생물체는 종자의 품질에 직접적인 피해를 줄 수 있으며, 감염 종자는 식물체에 병을 유발하고 포장에서의 발병을 확산시킴으로써 대규모의 경제적 피해를 유발할 수 있다. 상업용 종자의 해외증식 및 국제교역, 종자 유전자원의 해외분양 및 상호교환이 활발해짐에 따라 이러한 위험성이 더욱 커지고 있으며 더불어 종자 건전성 관리의 중요성 또한 증가하고 있다. 다양한 벼 종자 유전자원을 보존하고 연구용으로 분양하는 국제미작연구소(International Rice Research Institute, IRRI)는 유전자원의 건전성 확보를 위하여 종자건전성연구실(Seed health unit)을 두어 체계적인 관리시스템을 운영하고 있다. 필리핀 식물산업국 산하 식물검역소로부터 권한을 부여받아 필리핀 국내로 도입되거나 해외로 분양되는 유전자원에 대하여 종자처리 및 건전성 모니터링, 종자 건전성 검사 및 데이터베이스 관리 업무를 수행하고 있다. 대상 병원균에 따라 육안검사, 수세현탁액 검사, 습지법 검사, 한천배지 검사 등을 적용하고 있으며, 감염종자는 온탕처리와 같은 물리적 처리와 살균제 또는 훈증제와 같은 화학적 처리를 수행한다. 국제미작연구소의 건전성 관리시스템을 우리나라 벼 종자 유전자원의보존·관리에 선별 적용하고, 더불어 종자소독방법과 저장 전 관리체계를 확립함으로써 벼 유전자원의 품질을 제고할 수 있을 것이다. 또한 국제협력 및 공동연구 등 신뢰구축을 통해 유용한 해외 유전자원을 확보하는 데 기여할 것으로 기대된다.

        성인 여성의 외상 후 중도장애 경험에 대한 자전적 내러티브 탐구

        Objectives The purpose of this study is to understand meaning of my own experience by exploring the experiences of post-traumatic disability experienced in adulthood with autobiographical narrative inquiry. Methods Research puzzle was built what life story do I form when it comes to experiencing post-traumatic disability. Based on data such as my own memories, diaries and memos, I reconstructed the narrative in consideration of the three-dimensional narrative exploration space, “Temporality, Sociality, and Place”. The meaning of the experience was derived according to the narrative exploration procedure of Clandinin and Connelly(2000). Results As a result of the study, First, the story of living as a relational being was reconstructed into six narratives: “A body trapped in a body, a voice that came in an urgent moment, choosing to be a mother rather than a daughter, numerous Samaritans who constantly became my neighbor, reaching out with new invitations, and a family who was with me through a long tunnel.” Second, the story of living in relation to the body was reconstructed into four narratives: “A broken jar, I am not disabled, a flawed pottery is beautiful, and I wake up to the fountain of life, not a jar.” The meaning of the experience is revealed through the exploration of the story lived after the disability in adulthood. First, “the scales are peeled off in my eyes and I see them right away,” second, “invitation to connect to my neighbors and the world beyond me,” and third, “a journey to accept self-acceptance and life as a whole.” Conclusions Through this autobiographical narrative inquairy, I hope that the understanding of the experience of living with disability in adulthood and damaged bodies will extend beyond the understanding of individual humans to the understanding of the lives of the disability in adulthood. And I made a suggestion based on the research results.

        상추 국외 도입 유전자원의 시들음병 저항성 검정

        국내 시설재배 상추의 시들음병에 대한 안정적 재배를 위해 농가에 보급 가능한 저항성 자원을 탐색하기 위하여 12개 국가에서 육성한 111개 도입자원의 시들음병 저항성 정도를 조사하였다. 1. 병원균 접종 후 14일째 공시한 저항성 표준품종인 ‘뚝섬적축면’과 감수성 ‘미풍포찹적축면’을 포함한 공시 자원들의 병징이 뚜렷하게 나타남. 2. 시들음병 저항성 정도 범위는 전혀 고사되지 않은 자원( 발병정도 0)부터 전체 고사하는 자원(발병정도 4) 까지 넓게 분포하여 전체 111개 자원에 대한 평균 발병정도는 2.6±0.19 이었음. 3. 전체 자원 중 IT 294125 등 7개 자원이 병징이 전혀 나타나지 않았고, 저항성 기준으로 정한 발병정도 0인 개체비율이 80%이상이고 발병정도가 3이상인 개체비율이 20% 이하인 자원은 IT 289569 등 16개 자원이었음. 4. 저항성을 보인 16개 자원 중 IT 289569 등 미국 원산이 11개, 터키 원산이 IT296680 등 4개, 중국 원산이 IT 301287로 1개이었음. 1) 상추 유형으로는, 결구상추가 IT289569 등 8개, 잎상추는 IT 296679 등 5개, 로메인 상추가 IT296680 등 2개, 줄기상추 1개(IT 301287) 5. 본 연구결과 시들음병 저항성을 보인 16개 자원은 시들음병 발생이 높은 농가에서 이용 가능한 품종으로 생각되나, 병 저항성 검정이 유묘 단계에서 이루어졌기 때문에 생육 전반의 종합적인 특성을 고려하여 우수자원을 선발하는 후속연구 필요 This study was conducted to get information of Fusarium wilt resistance for introduced lettuce cultivars from 12 countries. 111 developed cultivars introduced from 12 countries showed intense and broad resistance symptom. The withered rates of the 111 accessions widely ranged from 0(no symptom) to 4(completely died caused by Fusarium wilt) index. The average of pathogenicity of all evaluated accessions was 2.6±0.19. Among them, 7 accessions including IT 294125 showed strong resistance(100 percent survival from Fusarium wilt) and 16 accessions including IT 289569, showed high level of resistance that is higher than 80% of 0 level and lower than 20 % of 3 or 4 level to young plants evaluated. Among the 16 accessions, 11 accessions were developed from U.S.A, 4 accessions from Turkey, and 1 from China. The 16 accessions consisted of 8 crisphead, 5 leaf lettuce, 2 romain type, and 1 stem type. As a result, 16 accessions which showed high level of resistance to Fusarium wilt can be possible to be used by commercial farmers and breeders. However, research related to the exploration of appropriate materials( accessions) for breeding cultivars with variable characters, good quality, high functional components, high consumer acceptability, etc. should be continued, considering pathogenicity test was conducted in young stage.

      • 들깨 국내 재래종 및 육성종 유전자원의 균핵병 저항성 검정

        Background : Sclerotinia rot, caused by a fungus Sclerotinia sclerotiorum, is one of the serious and unpredictable yield losses in perilla (Perilla frutescens) leaf production in Korea. Screening disease resistant genetic resources is necessary to develop disease-resistant cultivars and conduct related research. Methods and Results : A Total of 150 perilla accessions, including 123 Korean landraces and 27 cultivars developed in Korea, were evaluated for resistance to Sclerotinia rot (Sclerotinia sclerotiorum) using detached leaf inoculation technique. Sclerotinia sclerotiorum isolate KACC40457 was inoculated at the seedling stage (five to six leaves). For detached leaf method, a mycelial plug was placed fungus-side down on the main leaf vain and incubated at 22 ± 1℃ on moistened paper towel in a plastic box. Three Korean landraces, including IT117036, IT117106, and IT117110, and cultivar IT229431 showed 100% of resistance ratio (no. of plants showed below 1 ㎝ of lesion size/total evaluated plants × 100). Seven accessions including five landraces, IT117080, IT117107, IT117048, 117042, 117029, and two cultivaers, IT276225 and IT213781, showed high level of resistance that is higher than 80% of resistance ratio Conclusion : 11 accessions which showed strong and moderate level of resistance to Sclerotinia rot could be possibly used by breeders, farmers, and researchers to produce new disease resistant cultivars and use them commercially. However, research related to the exploration of appropriate materials (accessions) for breeding cultivars with good quality, high functional components, high consumer acceptability, etc. should be continued, considering pathogenicity test was conducted in young stage.

        파절된 도재면에 대한 수종의 도재 수리 시스템의 인장결합강도

        Statement of problem. Dental ceramics exhibit excellent esthetic property, compressive strength, chemical durability biocompatibility and translucency. However, it suffers from inherent brittle fractures. Various techniques and materials for intraoral porcelain repair has been suggested. Purpose. This study is to compare the tensile bond strength of four commonly used porcelain repair systems (Vivadent, Bisco, Ulttadent, Voco) and to insure the best system for the clinical application to the fractured porcelain. Materials and methods. A total of fifty specimens were fabricated. Specimens were stored in $37^{\circ}C$ distilled water for 7 days and thermocycling was performed(1000 cycles), and subjected to a tensile force parallel to the repair resin and porcelain interface by use of an Universal Testing Machine. Result. 1. Voco showed the highest tensile bond strength. In decreasing order, the tensile bond strength of the other materials was as follows : Ultradent, Bisco, Vivadent. 2. There was a statistically significant difference between the porcelain repair systems(Voco, Ultradent > Bisco, Yivadent) (p<0.05). 3. SEM examination of prepared porcelain surfaces revealed that the surface treated with Voco showed brittle fracture. However, Ultradent, Bisco and Vivadent showed ductile fracture. 4. All specimens treated with four porcelain repair systems showed adhesive failure between porcelain and composite resin.

        배추 아시아 도입 유전자원의 국내 재배포장에서 수집한 뿌리혹병 균주에 대한 저항성 반응

        Clubroot, caused by Plasmodiophora brassicae, is one of the most crucial disease in Kimchi cabbage. Screening disease resistant genetic resources is necessary to develop disease resistant cultivars and conduct related research. We have evaluated the development of clubroot to the 120 genetic resources of Kimchi cabbage introduced from World Vegetable Center and five Asian countries using isolate of Plasmodiophora brassicae collected in Haenam fields in Jeollanam-do Province, Rep. of Korea. This isolate was determined race 2 using differential varieties reported by Kim et al., 2016. IT100384 and IT305623 showed strong resistance, lower than disease index (DI) 1.0. IT100385, 100439, and 135407 showed moderate resistance (1.0<DI<2.0). The rest except above 5 accessions showed susceptible. As a result, 5 accessions which showed strong and moderate level of resistance to the isolate of Plasmodiophora brassicae could be possibly used by breeders and researchers to produce new disease resistant cultivars of Kimchi cabbage. However, research related to the exploration of appropriate materials (accessions) for breeding cultivars that are resistant to variable pathogens need to be conducted. In addition, several characters such as consumer acceptability, need to be taken into consideration. 배추의 뿌리혹병 저항성 품종 개발 및 관련 연구에 활용할 수 있는 자원 탐색을 목적으로 세계채소센터(World Vegetable Center, 대만 소재)와 아시아 지역 5개국에서 도입한 120 유전 자원을 대상으로 전라남도 해남군 배추 재배포장에서 발병한 뿌리혹병균(레이스 2)을 접종하여 저항성 반응을 조사함 1. 발병정도가 1 이하로 저항성 반응을 보인 자원은 IT100384와 IT305623로 2개이고, 발병정도가 1 초과 2 미만 으로 중도 저항성 반응을 보인 자원은 IT100385, 100439, 135407로 3개임. 그 외 자원은 발병정도가 2 이상으로 감수성 반응을 보임 2. 저항성 반응을 보인 IT100384는 발병정도는 0.3으로 거 의 증상을 보이지 않았으며, IT305623은 발병한 식물체가 발 견되지 않아 가장 저항성이 높은 자원으로 나왔음. 3. 뿌리혹병균에 대해 감수성 반응을 보인 자원들 중 발병 정도가 4.0으로 병 증상이 아주 심한 자원이 IT100349 등 23개이고, 발병정도가 3.0에서 4.0 미만인 자원이 84개로 가장 많았고, 2.0에서 3.0 미만인 자원은 8개임 4. 본 연구결과 배추 뿌리혹병 발병정도가 낮아 저항성 및 중도 저항성 자원으로 예측되는 5개 자원에 대해서, 병 검정 이 유묘 단계에서 이루어졌고 한 개의 균으로 검정하였기 때 문에 향후 다양한 균주를 이용하여 재평가함으로 활용가치가 높은 유전자원을 제공할 수 있도록 계속 연구 필요.

        Taxonomic Position of Korean Isolates of Rhizoctonia solani Based on RAPD and ITS Sequencing of Ribosomal DNA

        Taxonomic position of 46 Korean isolates of Rhizoctonia solani which were classified into nine intraspecific groups by anastomosis and cultural characteristics was analyzed by randomly amplified polymorphic DNA (RAPD)and sequence analyses of the internal transcribed spacer (ITS) regions of ribosomal DNA. All the isolates within each group showed highly similar band patterns in RAPD. The ITS regions of the isolates within the same groups showed a high level of sequence similarity above 96.0% whereas similarities among different groups were below 94.4%. When compared with several reference strains of R. solani from foreign countries, all the Korean isolates were clustered with the foreign isolates belonging to the same groups in the phylogenetic tree. All six Korean strains of AG-4 were identified as HG-1out of 3 subgroup of AG-4. We discussed taxonomic position of Korean isolates of R. solani and showed that sequence analysis with ITS regions could be a rapid and useful method for identification of intraspecific group of R. solani.
