      • 천식 마우스모델에서 Human Cytotoxic T Lymphocyte-associated Antigen 4-immunoglobulin (Hu.CTLA4-Ig) 융합단백의 기도 염증과 기도과민성 억제

        Background: CD4+ T helper cells which differentiate into the Th2 cells are crucial for the initiation and progression of allergic asthma. Human Cytotoxic T Lymphocyte-associated Antigen 4-immunoglobulin (CTLA4-Ig) has been widely used in order to block T-cell costimulation in an array of experimental animal models of T-cell-mediated diseases. Objective: To evaluate whether human CTLA4-Ig fusion protein inhibits airway inflammation in a murine model of OVA-induced asthma. Method: Six-weeks-old BALB/c mice were used and airway inflammation was induced by OVA. Human CTLA4-Ig fusion protein was administered intravenously before and after sensitization with OVA. Airway responsiveness to methacholine was measured after antigen challenge. Total IgE, OVA-specific IgE, IgG1 and IgG2a in serum samples were assessed by ELISA. Bronchoalveolar lavage (BAL) and lung section were also performed. Result: In mice which were treated with the CTLA4-Ig fusion protein before and after sensitization OVA, airway hyperresponsiveness and eosinophil BAL were markedly decreased. In mice which were with human CTLA4-Ig before sensitization with serum total IgE level were more significantly reduced that of positive control mice. Peribronchial inflammatory infiltration was desreased by the treatment of CTLA4-Ig before and after sensitization with OVA. Conclusion: These results suggest that human CTLA4-Ig fusion protein may inhibit eosinophilic airway inflammation and airway hyperresponsivness in OVA-sensitized airway challenged mice.         고소득층 여성의 의복구매행동특성에 관한 연구

        This study gives information for the enterprise to map out a positive marketing strategy and facilitate a new product development by researching and analyzing the specific character of the high income woman-brackets. In order to do that , based upon the materials of the National Statistical office, I made a selection of the high income brackets, subdividng their scholastic ability into four sets. The subject of the study was the women from twenties to sixties among customers of a high class dress shop in Taejeon. I made up 600 questionnaires for them, using 441 of these for the statistical analysis and making available the correlation analysis, the primary factor analysis, and χ^2-test. This study is classified into the type of women customers` purchasing clothes who are the high-income brackets, intended to study a special quality of purchasing according to income. 1. The purchasing manner of the high-income brackets customers is exposed the luxurious vain purchasing manner, the rational reasonable manner, and idiosyncrasy individual manner in order. 1) The consumer of the high-income brackets in general prefer to a high class clothes. In the contrast, low-income brackets prefer to rational reasonable consumption in spending money on clothes. 2) The favorite consumption manner of the high income brackets is the luxurious vain purchasing manner. This consumption values the more symbolic means the things show than owing the goods. 2. The higher standard of education and younger tends to be high in the luxurious, vain purchasing. 3. The unreasonable purchasing show an high irrelation at the impulse buying and self-satisfaction. 4. Self-satisfaction shows the highest interrelation than any other variations in the concern of clothes. 5. The symbolic images which exerts an influence upon purchasing manner are the notable elements of constituting as growing older. 6. The high standard of education prefers to the rational resonable purchasing, but on the other hand shows a high rate at the unreasonable individual purchasing manner.

        마우스 EAE, GVHD 질환에서 CTLA4Ig 융합단백의 면역치료 효과

        Background: CTLA4 (CD152), which is expressed on the surface of T cells following activation, has a much higher affinity for B7 molecules comparing to CD28, and is a negative regulator of T cell activation. In contrast to stimulating and agonistic capabilities of monoclonal antibodies specific to CTLA-4, CTLA4Ig fusion protein appears to act as CD28 antagonist and inhibits in vitro and in vivo T cell priming in variety of immunological conditions. We've set out to confirm whether inhibition of the CD28-B7 costimulatory response using a soluble form of human CTLA4Ig fusion protein would lead to persistent inhibition of alloreactive T cell activation. Methods: We have used CHO-$dhfr^-$ cell-line to produce CTLA4Ig fusion protein. After serum free culture of transfected cell line we purified this recombinant molecule by using protein A column. To confirm characterization of fusion protein, we carried out a series of Western blot, SDS-PAGE and silver staining analyses. We have also investigated the efficacy of CTLA4Ig in vitro such as mixed lymphocyte reaction (MLR) & cytotoxic T lymphocyte (CTL) response and in vivo such as experimental autoimmune encephalomyelitis (EAE), graft versus host disease (GVHD) and skin-graft whether this fusion protein could inhibit alloreactive T cell activation and lead to immunosuppression of activated T cell. Results: In vitro assay, CTLA4Ig fusion protein inhibited immune response in T cell-specific manner: 1) Human CTLA4Ig inhibited allogeneic stimulation in murine MLR; 2) CTLA4Ig prevented the specific killing activity of CTL. In vivo assay, human CTLA4Ig revealed the capacities to induce alloantigen-specific hyporesponsiveness in mouse model: 1) GVHD was efficiently blocked by dose-dependent manner; 2) Clinical score of EAE was significantly decreased compared to nomal control; 3) The time of skin-graft rejection was not different between CTLA4Ig treated and control group. Conclusion: Human CTLA4Ig suppress the T cell-mediated immune response and efficiently inhibit the EAE, GVHD in mouse model. The mechanism of T cell suppression by human CTLA4Ig fusion protein may be originated from the suppression of activity of cytotoxic T cell. Human CTLA4Ig could not suppress the rejection in mouse skin-graft, this finding suggests that other mechanism except the suppression of cytotoxic T cell may exist on the suppression of graft rejection.

        Background: Atopic dermatitis (AD) is the initial step of allergic march that progresses to airway allergic disease. There has been much interest in the interruption of the allergic march, but the results are not promising. CpG oligodeoxynucleotide (ODN) has been reported to have both preventive and therapeutic effects on the development of asthma in mouse models. Objective: To identify the development of airway inflammation after having atopic dermatitis-like skin lesions induced by topical application of Dermatophagoides farinae (Df), and to evaluate the preventive effect of CpG ODN on the development of airway inflammation. Method: We injected 5 μg of Df intradermally 8 times for 2 weeks into 8-week-old NC/Nga mice and then challenged them with 3 times of Df intranasally. CpG ODN with or without Df was injected subcutaneously before airway challenge. Airway hyperreactivity was determined by the change inenhanced pause (Penh) during methacholine provocation. Lung tissue and bronchoalveolar lavage specimens were evaluated. Result: Topical Df application induced AD-like skin lesion microscopically, but not macroscopically. NC/Nga mice with topical Df application developed more severe airway inflammation upon intranasal Df challenge than those with topical PBS application. However, the mice with CpG ODN injection before airway challenge showed reduced airway inflammation. The levels of Df-specific IgG1 were increased in the mice with both topical Df application and intranasal Df challenge but were decreased in the mice with CpG ODN injection. Conclusion: These results suggest that CpG may inhibit the development of airway inflammation from those with previous AD-like skin lesions induced by Df.         최근 노후화와 기능적 요건을 만족하지 못하여 불용되는 원통형 사일로의 해체 수요가 증가되면서 원통형 사일로의 해체에 대한 관심이 부각되고 있다. 본 연구에서는 전도공법에 의한 원통형 사일로의 발파해체를 수행하였다. 발파해체 결과, A 원통형 사일로는 예측된 방향으로 정확히 전도되었으며, B 원통형 사일로의 실제 전도방향은 예측방향과 약간의 차이를 보였다. 이것은 하단부 기둥과 링거더 지지부가 힌지 라인으로 설계되었으나, 실제 거동에서는 링거더 지지부만이 힌지 라인의 역할을 하였기 때문이다. 충격진동의 경우에는 예측범위 이하로 측정되었다. Recently the demand of demolition for the unnecessary cylindrical silo structure is increasing due to deterioration and unsatisfactory functional conditions and the issue of demolition is becoming a major highlight. This case study introduced the explosive demolition of the cylindrical silo structure by felling method. The results of explosive demolition conducted on cylindrical silo structure using the felling method show, A silo had collapsed precisely according to estimated direction but in case of B silo there was a minor difference. The lower columns and ring girder support was designed to the hinge line but in reality the lower columns of the silo did not do its structural support role and only the ring girder support did its role successfully. As for the impact vibration, most of the measurements were within the estimated range.

        철골구조물 발파해체 시공사례

        최근 노후화와 기능적 요건을 만족하지 못하여 불용하는 철골구조물의 해체 수요가 증가하면서 철골구조물 해체에 대한 관심이 부각되고 있다. 본 시공사례는 구조적 노후화와 기능적 요건을 만족하지 못하는 철골구조물인 ore bin과 coke bin 해체에 발파해체공법 중 전도공법을 적용하였으며, 철골구조물을 발파 절단하기 위해 장약용기를 사용하였다. 발파해체 결과, ore bin과 coke bin은 예측된 방향으로 정확히 전도되었으며, 주변 시설물에 피해 없이 발파해체를 완료하였다. Recently, the demand for demolition for the unnecessary steel frame structure is increasing due to deterioration and unsatisfactory functional conditions and the major highlights of demolition issues. This execution case was intended to describe an application of the felling method as one about the suggested method for explosive demolition method of ore bin and coke bin facilities, which were steel frame structures. And we used the charging container for blast cutting of the steel frame structure. As a result of the explosive demolition, the ore bin and coke bin had collapsed precisely according to the estimated direction. And the explosive demolition was completed without causing any damage to surrounding facilities.

      • 성인 아토피피부염 환자에서 Lactobacillus rhamnosus (Lcr35)의 면역학적 변화

        Background: Probiotics have recently been reported as a means of the prevention and treatment of atopic dermatitis (AD). However, detailed immune mechanisms involved in this effect remain speculative. The aim of this study was to investigate the effects of Lactobacillus rhamnosus (Lcr35) on symptoms of atopic dermatitis and cytokines production in peripheral blood monocytes (PBMCs). Method: Lcr35 was given to 5 adult patients with mild AD (2 males and 3 females; median SCORAD score, 15.2) for 4 weeks. We examined the SCORAD score and blood samples obtained in the initial, second, fourth and eighth weeks. We observed changes in production of IL-4, IFN-γ and IL-10 as well as responses to cell proliferation after stimulating PBMCs with Con-A. Result: There was no relevant significance in the SCORAD score before and after the treatment. PBMC proliferation was inhibited in the second and fourth weeks and was restored 1 month after the cessation of medication. IL-4 was decreased after the medication and persistently decreased after the cessation. IFN-γ was increased in the second week but decreased in the fourth week. IL-10 was increased in the second week (P=0.008) and showed variant patterns in the fourth week. Conclusion: The results of this study suggest that Lcr35 supplementation appears to modify cytokine production in PBMCs of AD patients. Further studies are needed to confirm this conclusion.         산업용 플랜트 기초의 발파해체 시공사례

        산업용 플랜트의 기능적·노후화에 따른 해체 수요가 증가하고 있으며, 시간적·공간적 환경위해요소를 최소화하기 위해 발파해체공법 또는 기계식 해체공법 및 발파해체공법을 혼용한 해체공법의 적용이 증가하고 있다. 본 시공사례에서는 철근콘크리트 구조방식으로 건설된 플랜트 기초를 해체하기 위해 발파해체공법을 적용하였다. 그 결과로서 플랜트 기초의 적정한 목표 파쇄가 이루어졌으며, 주변 시설물에 대한 피해가 없음을 확인하였다. The number of a industrial plant that must be demolished due to functional and structural deterioration has been increased. There is an increasing application of explosive demolition or explosive demolition combined with mechanical demolition to minimize temporal and spatial environmental hazardous factors created during the process of demolition. In this case study, to demolish the industrial plant foundation, which is a reinforced concrete structure, the explosive demolition technique was conducted. As a result of the explosive demolition, the overall crushing of plant foundation structure was satisfactory, and the explosive demolition was completed without causing any damage to surrounding facilities.
