Thiazolidinediones (TZD), which are widely used as insulin sensitizers, and fibrates, which are lipid-lowering drugs, are used in the treatment of dyslipidemia that commonly accompanies diabetes. Several reports suggest elevated levels of high-density lipoprotein (HDL) cholesterol, but the paradoxical reduction of HDL cholesterol level during single or combined TZD and fibrate therapies has been occasionally reported. Herein, we report a case of paradoxical decrease in HDL cholesterol and apolipoprotein A-1 levels during rosiglitazone and fenofibrate treatment for the first time in Korea. The patient was a 56-yr-old man presenting with type 2 diabetes mellitus and dyslipidemia. His HDL cholesterol and apolipoprotein A-1 levels returned to normal after the cessation of fenofibrate therapy. Since diabetes is an established risk factor of cardiovascular diseases, low HDL cholesterol can be a key cause of concern for patients with diabetes. Therefore, HDL cholesterol level should be determined before and after starting TZD and/or fibrate therapy in diabetic patients. (Korean J Lab Med 2010;30:17-9)
t(8;21) 잠재 염색체 변화로 인해 RUNX1-RUNX1T1 재배열을 보인 근네배수체증 급성골수백혈병 1예

Tetraploidy or near-tetraploidy is a rare cytogenetic abnormality found in AML, and is divided into primary and secondary forms. The secondary tetraploidy or near-tetraploidy found in AML is known to be specifically associated with t(8;21). In this case report, FISH analysis detected RUNX1-RUNX1T1 gene rearrangement in the absence of cytogenetic abnormality of t(8;21), which suggests the presence of unvailed t(8;21). This is the first case report of tetraploidy or near-tetraploidy AML with cryptic RUNX1/RUNX1T1 in Korea. Although the prognosis of tetraploidy or near- tetraploidy with t(8;21) is known to be poor, this patient shows a relatively good clinical course compared to other reported cases. 네배수체증 및 근네배수체증은 AML에서 드물게 나타나는 염색체 이상으로 일차성과 이차성으로 구별되며 이차성 네배수 체증 또는 근네배수체증 AML은 특히 t(8;21)과 연관이 있는 것 으로 알려져 있다. 본 증례에서는 FISH 검사결과에서 RUNX1- RUNX1T1 유전자 재배열이 보여 염색체 검사에서 발견되지 않 은 t(8;21)의 존재를 시사하였다. 본 증례는 네배수체증 또는 근네배수체증에 동반된 잠재(cryptic) RUNX1-RUNX1T1이 발견된 국내 첫 보고이며, t(8;21) 을 동반한 네배수체증 또는 근네배수체증 AML의 예후는 불량 한 것으로 알려져 있으나 이 환자는 다른 예들에 비해서 상대적 으로 좋은 예후를 보이고 있다.
유아의 연령별로 요구되는 교사의 특성에 대한 종사자와 어머니의 기대 연구

본 연구는 유아의 연령에 따라 요구하는 교사의 인구통계학적 배경변인의 특성 및 교사의 자질 특성에 대한 기대가 다른지를 원장ㆍ교사 등 종사자와 어머니를 대상으로 조사ㆍ분석하는 것을 목적으로 하였다. 설문조사로 수집한 자료를 분석한 결과 원장ㆍ교사 등 종사자들은 유아의 연령에 따라 요구되는 교사의 특성을 다르게 인식하고 있다는 사실이 밝혀졌다. 어머니의 경우 기대하는 교사의 특성이 유아의 연령에 따라 달라야 한다고 생각하는 점은 동일하였으나 구체적인 경향은 종사자의 것과 다소 다르게 나타났다. 본 연구는 유아교육 기관의 원장, 교사, 어머니들이 유아의 연령에 따라 요구하는 교사의 특성에 대한 기대가 다르다는 사실을 입증하였다는 의의가 있다. 본 연구 결과를 바탕으로 학년초에 담임을 배정하거나 교육과정을 진행할 때 유아의 연령에 따라 요구되는 교사의 특성을 적절하게 반영할 수 있어야 하겠다.

Bone turnover normally occurs in a highly regulated manner throughout life. It involves two different kinds of cells, osteoclasts and osteoblasts. Osteoclasts, the monocyte-macrophage lineage cells, resorb bone and osteoblasts deposit bone. Perturbations to these processes underlie skeletal disorders, such as osteoporosis. To understand the cellular and molecular mechanisms of bone biology and for the identification and characterization of new drug targets and therapies, a mouse co-culture system of osteoblasts and hematopoietic cells is established, in which osteoclasts are formed in response to 1 a ,25-dihydroxyvitamin D3 (1 a ,25(OH)₂D₃) and prostaglandin E₂ (PGE₂). Using this co-culture system, the bacterial metabolite n-butyrate, which has well-known anti-inflammatory effects, is shown to stimulate or inhibit osteoclastogenesis depending on the concentration. These results demonstrate a novel property of w-butyrate that may modulate osteoclastogenesis via osteoblasts.

Background: Since the recent introduction of radioimmunotherapy (RIT) using antibodies against cluster of differentiation (CD) 45 for the treatment of lymphoma, the clinical significance of the CD45 antigen has been increasing steadily. Here, we analyzed CD45 expression on lymphocyte subsets using flow cytometry in order to predict the susceptibility of normal lymphocytes to RIT. Methods: Peripheral blood specimens were collected from 14 healthy individuals aged 25-54 yr. The mean fluorescence intensity (MFI) of the cell surface antigens was measured using a FACSCanto II system (Becton Dickinson Bioscience, USA). MFI values were converted into antibody binding capacity values using a Quantum Simply Cellular microbead kit (Bangs Laboratories, Inc., USA). Results: Among the lymphocyte subsets, the expression of CD45 was the highest (725,368±42,763) on natural killer T (NKT) cells, 674,030±48,187 on cytotoxic/suppressor T cells, 588,750±48,090 on natural killer (NK) cells, 580,211±29,168 on helper T (Th) cells, and 499,436±21,737 on B cells. The Th cells and NK cells expressed a similar level of CD45 (P=0.502). Forward scatter was the highest in NKT cells (P<0.05), whereas side scatter differed significantly between each of the lymphocyte subsets (P<0.05). CD3 expression was highest in the Th and NKT cells. Conclusions: NKT cells express the highest levels of CD45 antigen. Therefore, this lymphocyte subset would be most profoundly affected by RIT or pretargeted RIT. The monitoring of this lymphocyte subset during and after RIT should prove helpful. IL-1 의 신호전달에 관여하는 TRAF6 결합 motif의 기능적 중요성

Crystal structure of TRAF6 in complex with TRAF6-binding sites from CD40 was previously determined. The structure revealed a distinct TRAF6-binding groove of CD40, the key structural determinant of interaction. The structural information leads to a proposed TRAF6-binding motif. This allows the identification of TRAF6-binding sequences in the hIRAK protein, whose functional requirement in IL-1 mediated signal transduction is further demonstrated using site-directed mutagenesis. The mutational effects of hIRAK on the down-stream NF-kB signaling shows the importance of the TRAF6 interface for signaling by IL-1.
