Human adenovirus type 5 (hAd5) vectors have been demonstrated to be useful vehicles for gene expressions in animals. However, it has not been reported whether hAd5 transduction might be hampered in the sera of livestock animals inRepublic of Korea. We collected 205 samples of livestock animals, such as pig (n=84), cattle (n=84), and goat (n=37)in Korea. The neutralizing antibody (NAb) titers to hAd5 virus were less than 15 in most of samples. Only 8% of goatsamples had a NAb titer of 15 or 30. Thus, we showed that hAd5 virus was not neutralized in sera from cattle, pig, andgoat, and suggest that the hAd5 vector could be used for the effective delivery of vaccines or proteins in livestockanimals in the field.

Foot-and-mouth disease (FMD) is the most contagious disease of mammals. The use of inactivated vaccine can be chosen to prevent or control FMD. However, vaccination against one serotype of FMDV doses not cross-protect against other serotypes and may not protect fully against some strains of the same serotype. Appropriate selection of vaccine strain is an important element in the control of FMD. The immunity of vaccine antigens should be matched against newly circulating viruses. The phylogenetic analysis of serotype Asia1 reported from China, Mongolia, North Korea and Russia since 2005 shows that they are all classified into genetic group V, but the strain, Asia1/Shamir (ISR/89) which have been used as a vaccine strain in Korea, is clustered into different genetic group. So, in this study the serological relationship between the isolate (Asia1/MOG/05; MOG) and the Shamir strain was determined by ELISA and virus neutralization test. Even though the matching value of the virus (MOG) against the vaccinated sera in target animals was not so high, the vaccinated animals elicited antibodies enough for protection after vaccinated once or twice. Conclusively, we suggest that the vaccine containing Asia1/Shamir antigen could protect the genetic group V strains circulating in East Asia currently if vaccinated twice or the more.

Foot-and-mouth disease (FMD) is a severe vesicular disease of cloven-hoofed animals including domesticated ruminants and pigs. Acute clinical signs may be mild in sheep and goats but are associated with lameness in pigs and mouth lesions with vesicles in cattle. The required condition for a successful pathogen appears to be the ability to counteract both the host innate and adaptive immune response. FMD virus (FMDV) inhibits the induction of antiviral molecules and interferes with the secretory pathway in the infected cell. The surface expression of Major Histocompatibility Complex (MHC) class I molecules is reduced in infected cells. Thus, the ability of the host to recognize and eliminate virus infected cells is decreased. Furthermore, FMDV infection results in a rapid, but transient lymphopenia,reducing the number of T and B cells, and affecting T cell function. The virus appears to premature apoptosis-mediated cell death because it has a very short replication cycle and is able to rapidly produce large amounts of virus. FMDV engages the host protective response at multiple steps to ensure its effective replication and pathogenesis. This review describes the recent pathological and immunological studies to overcome the powerful abilities of FMDV to counteract defense mechanism of host.
Infection of Foot and Mouth Disease of Serotype A in Farm-raised Deer in Korea

This report describes a case of foot and mouth disease (FMD) in farm-raised Sika deer. FMD serotype A occurred in six cattle farms in January 2010 since the outbreak of pig cases 2002 in South Korea. In addition to the six cattle farms where the disease occurred during January, positive reactions to FMD antibodies were found in two heads of deer on March at a deer farm in serological tests intended to lift the movement restriction of the susceptible animals imposed within risk zone. In the specimens collected from 12 heads for confirmatory tests in the same farm, no virus was detected in antigen tests and neutralising antibody titers in all raising deer in the farm were relatively high. So it seems likely that the animals had been infected with FMD at least one month earlier when they were found.

Bovine enteroviruses (BEVs) were separated into two groups, BEV-1 and BEV-2. BEVs, found in cattle worldwide, usually cause asymptomatic infections and are excreted in the feces of infected animals. Antibodies against BEV have been found in different species including human, cattle, sheep, goats, dogs, horses and monkeys in the world. This study aimed to investigate prevalence of the neutralizing antibodies for BEVs in human and animals in Korea. Antibodies against BEV-1 in humans, cattle, pigs, goats, horses and dogs were shown to be 46.8%, 48.3%, 70.6%, 11.5%, 11.5% and 6.3% respectively. Also, antibodies against BEV-2 were shown to be 98.7%, 68.1%, 89.2%, 59.4%, 9.4% and 96.9% respectively. We found that the neutralizing antibodies against these viruses are common in Korea. The prevalences of antibodies against BEV-1 were lower than those against BEV-2 in humans and in all animals except horses. These results showed that the BEV is considered endemic in cattle in many regions in Korea.

Foot-and-mouth disease (FMD) is a highly contagious disease affecting cloven-hoofed animals and causes severe economic loss and devastating effect on international trade of animal or animal products. Since FMD outbreaks have recently occurred in some Asian countries, it is important to understand the relationship between diverse immunogenomic structures of host animals and the immunity to foot-and-mouth disease virus (FMDV). We performed genome wide association study based on high-density bovine single nucleotide polymorphism (SNP) chip for identifying FMD resistant loci in Holstein cattle. Among 624532 SNP after quality control, we found that 11 SNPs on 3 chromosomes (chr17, 22, and 15) were significantly associated with the trait at the p.adjust <0.05 after PERMORY test. Most significantly associated SNPs were located on chromosome 17, around the genes Myosin XVIIIB and Seizure related 6 homolog (mouse)-like, which were associated with lung cancer. Based on the known function of the genes nearby the significant SNPs, the FMD resistant animals might have ability to improve their innate immune response to FMDV infection.

Despite nation-wide immunization with O, A, and Asia 1 type vaccines in Republic of Korea, foot-and-mouth disease type O occurred again in July 2014 after three years and three months. This virus was a Mya-98 strain of the Southeast Asian topotype and was most similar to the identified type that circulated in East Asia in 2014. This was new virus with the deletion of 23 amino acids in 3A/3B1 region and low pathogenic property.

Foot-and-mouth disease (FMD) virus exists in seven serotypes and is known to be a highly contagious disease that is hard to eradicate from the world. The O, A, Asia1 and SAT2 serotypes commonly infected cattle, sheep and goats during 2007~2009 throughout the world. In particular, the outbreak of the Asia1 serotype in China appeared in all areas from 2005 and is still present. Surprisingly, in 2009, Taiwan reported the first outbreak of the type O serotype since 2001. Then type A appeared in China for the first time since the early 1960s. The virus shows a close relationship to the viruses from Southeast Asia suggesting one or more recent introductions into China in the OIE reports. Recently the subtype of A/Iran05 spread to nearby countries exhibiting genomic evolution. The use of molecular epidemiology is an important tool in understanding and consequently controlling the FMD virus. The phylogenetic analysis with VP1 gene was especially useful for molecular epidemiological studies and showed the same pattern which matches with serotype classification. This paper describes basic information about the disease, and the serotype-specific characteristics and evolution to perform molecular epidemiological analysis. Furthermore, we show the importance of the genetic evolution on the FMD serotypes in global surveillance and molecular epidemiology of FMD for outbreak investigation.
