Alpha2-macroglobulin (alpha2-M) has been shown to have a variety of activities. One of those activities is the suppression of immune response. Characterization of the immunosuppressive factor secreted by a T cell hybridoma showed that alpha2-M was produced and secreted from the T cell hybridoma. alpha2-M was produced abundantly from the T cell hybridoma when cultured as ascites. The isolation and identification of the alpha2-M were studied using affinity chromatography and N-terminal amino acid sequencing. The extended observations were that the alpha2-M produced by the T cell hybridoma suppresses mixed lymphocyte reaction.

수종 Flavonoid의 항허피스바이러스효과

To search for less toxic antiherpetic agents, the inhibitory effects of twelve kinds of flavonoids including chrysin, quercetin, quercitrin, rutin, fisetin, gossypin, kaempferol, morin, naringenin, naringin, hesperetin and hesperidin on the plaque formation of herpes simplex virus type 1 (HSV-1) and type 2 (HSV-2) in Vero cells were examined by plaque reduction assay in vitro. Some flavonoids tested in this study showed potent antiherpetic activity, reducing intracellular replication of herpes simplex viruses when Vero cell monolayers were infected and subsequently cultured in medium containing flavonoids. Naringenin showed the most potent antiviral activity against HSV-1 with selectivity index (SI) of 19.1 and hesperetin showed the most potent antiviral activity against HSV-2 with SI of 9.8. These results suggest that some flavonoids may be a potential therapeutic agent for the treatment and prevention of herpes simplex virus infections.

한국인 시각 장애우 환자의 복약지도 증진을 위한 점자용 보조라벨 개발의 필요성과 개발방법 제시

All pharmacists must provide the drug consultation whenever dispense drugs to patients by the Korean Pharmacy Law. Drug consultation is very important procedure for increasing pharmacotherapy. Because it maximizes the therapeutic effects or/and minimizes adverse drug reaction during the drug therapy. However, it is not easy to do because of the dynamic and hectic pharmacy environment. Especially, if someone has a disabling body function, they required more time and efforts to perform consultation by pharmacist. Currently several auxiliary labels for helping drug consultation are using in pharmacy practice but not for disabling patients. Therefore we developed the total 53 auxiliary labels with size of 0.7 cm (width) and 1 cm (length) by Braillewriter letters for blind patients. This research has been performed for total 12 months (Mar. 15ts, 2007$\sim$Feb. 25th, 2008) and the developing methods are consisted of 4 steps: 1) selection of essential informations, 2) simplification of information, 3) changing for Braillewriter letters, 4) application and revising by blindness patients. Also the labels are consisted of 12 for adverse reactions and precautions, 8 for directions, 2 for storages, 9 for duration, 9 for dosage forms, and 12 for common names. After developed those labels, we revised those labels by discussion with 2 blind people. In conclusion, the new auxiliary labels for blind patients can increase therapeutic effects and decrease risks from pharmacotherapy besides decreasing of pharmacist's work load in the future.

천연 퀘루세틴이 수종 항생물질의 항균력에 미치는 병용효과

As part of our search for less toxic antimicrobial agents from natural resources. rutin was isolated from Sophora japonica and then hydrolyzed to quercetin. Antimicrobial activity of quercetin was tested in vitro against five kinds of gram positive and ten kinds of gram negative bacteria by serial broth dilution method. Among fifteen kinds of bacteria tested, the antimicrobial activity of quercetin was the most potent against Proteus vulgaris showing minimal inhibitory concentration(MIC) of 125 ${\mu}$g/ml. To investigate the effect of antimicrobial combinations of quercetin with four kinds of antibiotics (ampicillin, cefazolin, oxytetracycline and chloramphenicol). the fractional inhibitory concentration index (FICI) was determined by checkerboard assay for each strain. The antimicrobial combinations of quercetin with four kinds of antibiotics resulted in synergism in one instance, additive effect in four instances, but no antagonism was observed.

다중 중합효소 연쇄반응을 이용한 DNA 바이러스의 동시검출

Epstein-Barr virus (EBV), cytomegalovirus (CMV), hepatitis B virus (HBV), parvovirus B19 (B19)등 4종의 바이러스는 인체에 감염을 일으키는 병원체로서 DNA를 유전물질로 함유한다. 각 바이러스 유전자의 염기서열을 분석하여 EBV CMV, HBV의 pol 유전자와 B19의 ns 유전자에 특이적으로 결합할 수 있는 primer를 설계 제작하고 단일 시험으로 4종의 바이러스를 동시에 검출할 수 있는 다중 중합효소 연쇄반응(Multiplex PCR)법을 확립하였다. Primer 염기서열, PCR 반응조성물의 농도, PCR 반응시간 및 온도조건을 최적화하여 민감도를 증대시킴으로써, 단일 시험으로 5-10 분자수의 유전물질까지 검출이 가능하였다. 또한 4종의 바이러스 사이에 교차반응이 일어나지 않았으며 생체시료를 이용한 시험에서도 특이성과 민감도가 유지됨을 확인하였다. 그러므로 본 연구에서 확립한 다중 중합효소 연쇄반응은 세포배양액 또는 생체 시료에 감염된 4종 DNA 바이러스진단에 효율적으로 이용할 수 있을 것이라고 판단된다. We describe a multiplex PCR method that can detect and differentiate simultaneously four different kinds of DNA viruses, Epstein-Barr virus (EBV), cytomegalovirus (CMV), hepatitis B virus (HBV) and parvovirus B19 (B19). Primers for the multiplex PCR reaction were designed to amplify specific regions of the EBV (pol), CMV (pol), HBV (pol) and B19 (ns) viral genomes and used to simultaneously detect individual viruses. In order to achieve optimal sensitivity and specificity for multiplex PCR, the thermo-cycling parameters, primer sequences, and concentration of each reaction components were optimized systematically. The sensitivity of the detection method ranged between 5 and 10 copies of viral genome with a mixture of multiple primer pairs. Furthermore, this highly sensitive test showed no cross-reactivity among the four viruses. Thus, the results obtained in this study provide evidence that the assay system is a good tool for supporting the diagnosis of viral infection and contamination.

가정에서 미사용된 약물처리에 대한 평가

Most of families in Korea keep some medicines that has been expired or not been used for a long time, and these medicines could be causing environmental or social problems. Currently many people concerned about their health and therefore show a tendency to visit many health facilities. It eventually brings to remain more medication in home The aim of this study is to determine the drug waste minimization and drug disposal in Korean home and provide the proper guideline for them. This study were performed by visiting total randomly selected 140 homes at the Seoul area from July 23rd, 2008 to July 31st, 2008 by questionnaire. This survey comprised total 6 parts and total 22 questions. In results, 77 (55.0%) among 140 families answered to this survey. 89.6% of families keep unused medicine in the home. Most were tablet (66.3%) and syrup (19.2%) dosage type. They usually kept the unused medicine in the room (41.6%) or no certain place. 76.6% families reused the medicine and used for themselves. The reason for reuse is similar symptoms by their decision. The reason for medicine was left over is not following the prescription's direction (31.3%). They mostly dispose the unused medications by throwing into a trash can (71.4%). In conclusion, many families keeps unused medication and it could cause health and environmental problems.

초유에 함유되어 있는 면역조절물질인 MIEF가 B 세포의 분화에 미치는 영향

The levels of maternal immunity enhancing factor(MIEF), which is an immunomodulatory protein identified from bovine colostrum, were determined by indirect competitive enzyme-linked immunosorbent assay(ELISA) for the colostrum and normal milk collected during the first two weeks of lactation. The mean concentration of MIEF in the colostrum of the first day of lactation was 109mcg/ml, and fell from the third day of lactation to 3-4mcg/ml. The molecular weight of the purified MIEF determined by reducing SDS-PAGE and TSK G2000SW column chromatography was 22,000 and 24,000 daltons, respectively, showing that MIEF is a monomeric peptide in its native form. To examine the capacity of MIEF to induce differentiation of B lymphocytes, human tonsillar lymphocytes were cultured in the presence of different concentrations of MIEF, and then antibody secreting cells were enumerated by enzyme-linked immunospot(ELISPOT) assay. When added to cultures of human tonsillar lymphocytes, MIEF induced differentiation of resting B lymphocyte to antibody secreting plasma cells as efficiently as LPS.

잔나비걸상버섯 수용성물질의 항바이러스효과

In order to find less toxic antiviral agents from basidiomycetes, EA, the water soluble substance, was prepared from the carpophores of Elfvingia applanata (Pers.) Karst. EA was examined for antiviral activity against five strains of pathogenic viruses such as encephalomyocarditis virus (EMCV), vesicular stomatitis virus (VSV) Indiana and New Jersey strains, influenza A virus (Flu A), and varicella zoster virus (VZV) in vitro. Antiviral activity was evaluated by plaque reduction assay. Among five strains of viruses tested, EA exhibited the most potent antiviral activity against VSV Indiana strain with 50% effective concentration $(EC_{50})$ of 0.104 mg/ml in Vero cells, and its selectivity index (SI) was 36.5. EA was also examined for the virucidal activity, antiviral activity in preincubation on VSV Indiana strain in order to examine possible mode of antiviral activity. Preincubation of Vero cells with EA did not confer protection against VSV, however, prolonged exposure of cells to EA inhibited the replication of virus dose-dependently. In virucidal activity, the titer of infectious virus did not decrease significantly.

나린게닌의 항균력에 미치는 관련 후라보노이드의 병용효과

As part of our search for less toxic antimicrobial substances from natural resources, naringin was isolated from feels of Citri fructus and then hydrolyzed to naringenin. The antimicrobial activity of naringenin was examined by measuring the minimal inhibitory concentration(MIC) against fourteen species of bacteria. The antimicrobial activity of naringenin in combination with rutin or hesperetin was evaluated by checkerboard method. Among fourteen species tested, the antimicrobial activity of naringenin was the most prominant against Staphylococcus aureus and Shigella boydii showing MIC of 100mcg/ml for both species. Combinations of naringenin with rutin or hesperetin showed synergism against several species of bacteria, but no antagonism was observed.
