        버찌 분말을 첨가하여 제조한 국수의 품질특성

        This study was carried out to investigate the quality characteristics and storage of noodle with added flowering cherry fruit powder. Wet noodles were prepared at concentration of 1, 2, and 3% (w/w) of the fruit based on flour weight. Moisture content of the noodles with or without flowering cherry fruit powder was 33.09~33.50%. Ca, K and Mg contents of flowering cherry fruit noodle were increasing at increasing concentrations of flowering cherry fruit powder. Cooked weight, volume, and water absorption decreased with increased fruit powder, whereas turbidity increased. For Hunter's color values of noodles, L (lightness) and b (yellowness) values decreased with increasing concentration of flowering cherry fruit powder, whereas a (redness) value increased. For mechanical characteristics of the noodles, adhesiveness of cooked noodle with flowering cherry fruit powder were higher than those of control. The pH of flowering cherry fruit noodle was lower than that of control during storage. Bacterial counts of wet noodle with flowering cherry fruit powders were lower than those of the control on the 10th day of storage at $5^{\circ}C$. From the sensory evaluation of 20's, texture, taste, and overall preference were not significantly different between the control and the fruit groups, but the color scores of 2% and 3% flowering cherry fruit noodle were significantly higher than that of 1% fruit noodle (p<0.05). In 40's sensory evaluation, color and overall preference scores of flowering cherry fruit noodle were significantly higher than those of control. In conclusion, the results of this study were suggested that the addition of flowering cherry fruit powder in combination with flour was tended to improve antimicrobial effects during storage when compared to control.

        Objectives: To investigate the effect of LH receptor in folliculogenesis, we confirm the expression level of LH receptor (LH-R) mRNA in human granulosa cells (GCs) and its expression levels were analyzed by comparison to embryo developmental rate and pregnancy rate. Materials and Methods: GCs were obtained at the time of oocyte retrieval from the patients undergoing IVF-ET program. The patients were divided into two groups: Group I (n=20) is poor responder (retrieved oocyte(s)$\leq$3ea), Group II (n=80) is normal responder (retrieved oocytes>3ea). After the extraction of total RNA, semiquantitative RT-PCR was performed and the expression level of LH-R mRNA was normalized by $\beta$-actin. Statistical analysis was performed by using $X^2$ test, Student's t-test and Pearson correlation. Results: In Group II, the relative values of LH-R mRNA (0.680 vs. 0.463, p<0.005) and pregnancy rate (54.7% vs. 23.1%, p<0.05) were significantly higher than in Group I. Number of retrieved oocyte(s) was gradually increased when the expression of LH-R mRNA was increased (p<0.05). But the quality of retrieved oocyte and transferred embryo were not related with the expression of LH-R mRNA. When the pregnancy rate was compared with FSH only group and FSH combined with hMG group in the ovarian stimulation protocol, FSH combined with hMG group was significantly higher than FSH only group in Group I (37.5% vs. 0%), and the expression of LH-R mRNA was significantly higher in hMG combined group than FSH only group (p<0.05). Conclusion: Expression level of LH-R mRNA has important role in ovarian function related with the response to gonadotrophin in human folliculogenesis. Furthermore these data might provide the evidence that additional use of hMG is helpful to poor responders.

        최근 일반 건강인에 대한 매독 혈청 검사 양성율 - 1995년

        Background: The VDRL is used as a screening test for syphilis worldwide. In addition, reactive rates of the VDRL test are used to estimate the incidence of syphilis. The prevalence of syphilis reported in Korea varies according to the study population, district and time. Reports on prevalence of syphilis indicate that reactive rates of the serologic test for syphilis showed an increasing tendency in the 1960s and has steadily decreased in the 1990s. We have already reported VDRL positive rates periodically since the late 1970s in similar areas with similar population groups and methods in order to standardize the results for accurate analysis of the prevalence of syphilis in Korea. Objective : This study was performed utilizing the VDRL test in order to find out if the decreasing trend of syphilis prevalence is still continuing. Methods : The ser ologic tests for syphilis were carried out on 13,426 apparently normal adults aged twenty-years-old or over(8,250 blood donors in the Seoul area, 2,461 physical examinees examined at Severance Hospital, Yonsei University and 2,715 pregnant women at delivery at Severance Hospital, Yonsei University ) from January to December 1995. We hve performed the VDRL quantitative test in VDRL-reactive persons among blood donors and pregnant women. Results : The reactive rate of the serum VDRL test was 0.2% in 13,426 healthy adults(blood donors.0.2%, physical examinees:0,2%, pregnant women:0.3%). There was a statistically decreasing tendency compared to 0.4% in 1990(p<0.001). The VDRL quantitative test resulted in levels below the 1:2 titer in 92% of the VDRL-reactive physical exarninees and pregnant women. Conclusion . From the comparison of VDRL reactive rates for syphilis in an apparently normal Korean population obtained by the present author group since 1977, i.e., 2.5% in 1977, 1.1% in 1981, 0.6% in 1986, 0.4% in 1990 and 0.2% in 1995, it is clear that the prevalence of syphilis has been rapidly decreasing since the mid 1970s in Korea.         The possibility of rapid quantitative analysis of AV(Acid Value), Ⅳ(Iodine Value) and fatty acids in vegetable oils with NIRS(Near-infrared spectroscopy) was evaluated. A calibration equation calculated by MPLS regression technique was developed and correlation coefficient of determination for AV, Ⅳ, C16:0, C16:0, C16:1, C16:2, C16:6, and C20:0 content were 0.9727, 0.997, 0.9805, 0.942, 0.9987, 0.9994, 0.9966, and 0.975 respectively. According to the data obtained from validation study, R² of contents of perilla, corn, soybean, rapaseed oils were 0.897, 0.993, 0.935, 0.707, 0.994, 0.996, 0.984, 0.798, SEP of contents of 0.185, 1.367, 0.899, 0.640, 1.498, 1.360, 0.476, 0.076 by MPLS. The results indicate that the NIRS procedure can potentially be used as a non-destructive analysis method for the rapid and simple measurement of AV, Ⅳ and fatty acids in vegetable oils.

        경기도내에서 분리한 캠필로박터 제주니균의 유전적특성 및 항생제내성 연구

        Campylobacter jejuni는 사람에서 매우 중요한 식중독 원인균의 하나이며, 2010년 경기도내 병원을 내원한 설사 환자와 4번의 식중독 outbreak에서 42균주를 분리하였다. 본 연구에서는 분리된 C. jejuni 42균주의 유전적 특성과 혈청형, 항균제 내성율을 분석하였다. hipO 종특이 유전자(100%), cdtB 독소 유전자(100%)가 검출되었고, gyrA 돌연변이 유전자(95.2%)가 PCR 실험결과 검출되었다. gyrA 돌연변이 유전자는 ciprofloxacin 내성과 연관이 깊으며, 디스크 확산법에 의해 실험한 결과 gyrA 돌연변이 유전자가 검출된 40균주(95.2%)에서 실제 ciprofloxacin에 대해 내성을 보였다. 분리된 42균주 C. jejuni에 대한 gyrA 유전자의 염기서열을 분석한 결과 ciprofloxacin에 내성을 가진 40균주에서 아미노산 서열이 ACA (트레오닌)에서 ATA (이소루신)으로 돌연변이 되어있음을 확인하였다. 하지만 대체 치료제인 erythromycin과 azithromycin에 대해서는 97.6% (41균주) 감수성을 보였다. 또한 C. jejuni의 혈청형을 분석한 결과 4가지 타입으로 분류되었는데, HS2(B), HS3(C), HS4(D), HS19(O)로 확인되었다. 도내에서 분리한 C. jejuni 42균주의 유전형을 rep-PCR과 PFGE로 확인한 결과 PFGE에 의해서는 12 cluster, rep-PCR에 의해서는 11 cluster의 유전형으로 구분되었다. Campylobacter jejuni is one of important food-borne pathogens causing human gastroenteritis. We isolated 42 strains of C. jejuni from diarrhea patients and 4 food-poisoning outbreaks in 2010, Gyeonggi-do. In this study, 42 strains were tested for genetic characteristics, the serotype distribution and antimicrobial resistant rate. The presence of hipO (100%), cdtB (100%), and mutated gyrA (95.2%) genes was detected in C. jejuni by polymerase chain reaction (PCR). Detection of mutated gyrA gene correlated with ciprofloxacin resistance. Forty isolates had mutated gyrA gene and were actually resistant to ciprofloxacin. Furthermore, comparing the gyrA DNA sequence data, ciprofloxacin-resistant isolates had a mutation of the DNA sequence from ACA (threonine) to ATA (isoleucine). But 41 strains (97.6%) of patient isolates were susceptible to erythromycin and azithromycin. A total of 35.7% among 42 C. jejuni isolates were identified into 4 different serotypes. The serotype distribution of C. jejuni strains were shown to be HS2(B), HS3(C), HS4(D), HS19(O). To investigate the genotypes of C. jejuni isolated in Gyeonggi province, repetitive sequence polymerase chain reaction (rep-PCR) analysis and SmaI-digested pulsed-filed gel electrophoresis (PFGE) profile analysis were performed. From the PFGE analysis of 42 C. jejuni strains, 12 clusters of PFGE profile were obtained. On the other hand, 11 clusters of rep-PCR profile were obtained from 42 strains of C. jejuni.

        경기도에서 분리한 병원성대장균의 역학적 특성 및 PFGE, 항생제 내성 연구

        병원성 대장균은 최근 3년간 경기도에서 발생한 학교급식식 중독의 주된 원인균으로서 그 역학적 특성을 정리하고 PFGE typing과 항생제 감수성 시험으로 분리균주의 특성을 알아보고자 하였다. 2010년부터 2012년까지 19건의 학교급식 식중독에 대한 역학조사보고서를 분석한 결과, 급식의 주된 운영방식은 직영(18건, 95%)이며, 고등학교에서 주로 발생하였다. 계절적 요인으로는 13건(65%)이 6월부터 9월에, 특히 여름방학 후 학기 초인 8월말부터 9월에 더 많이 발생하였다. 수요일(7건, 37%)과 목요일(7건, 37%)에 최초환자가 발생되어, 목요일 7건(37%) 금요일 5건(26%)에 주로 신고가 이루어 졌다. 추정위험에는 월요일(4건, 21%), 화요일(7건, 37%), 수요일(4건, 21%)에 노출되었고, 추정원인체로서 가능성이 높은 식품은 김치류가 50%(5건)를 차지하였다. 분리된 98균주는 EPEC, ETEC, EAEC, EHEC가 각각 50%, 34%, 15%, 1%이며, 항생제 감수성 시험 결과 ampicillin, nalidixic acid, sulfamethoxazole/trimethoprim, tetracycline에 대하여 각각 40%, 37%, 24%, 19%의 내성을 나타내었고, 2, 3세대 cephalosporins계, cabarpenem계, aminoglycosides계, 2세대 quinolones계 항생제에는 감수성이 있었으며, cefalotin, ampicillin/sulbactam, chloramphenicol에는 각각 29%, 25%, 6%가 중간내성을, 그리고 70%에서 한 가지 이상의 항생제에 내성이 관찰되었다. PFGE 분석결과, 57.6%의 similarity로 8개의 group과 32개의 profiles로 분류되었다. 개별적인 소규모 식중독에서 항생제 감수성과 유전자형의 연관성을 찾기가 어려웠으며, 동시에 발생한 대규모 식중독의 경우, 항생제 내성과 PFGE 유형이 매우 유사하여 동일 기원으로부터 유래한 병원성대장균에 의해 식중독 사고가 발생한 것으로 추정이 가능하였다. This study was conducted to survey the epidemiological characteristics and the isolated strains for pathogenic E. coli which was the major causative organisms for food poisoning occurred at school food services in the Gyeonggi-do area during the past three years. We investigated 19 accidents of food-borne disease outbreaks by pathogenic E. coli at school food services from 2010 to 2012. Food-borne disease outbreaks by pathogenic E. coli were usually occurred at direct management type (18 accidents, 95%) and high schools. For the seasonal factors, 13 accidents (65%) were occurred in June to September, especially the end of August and September after the summer holidays. The first patients were occurred on Wednesday (7 accidents, 37%) and Thursday (7 accidents, 37%), and they were mainly reported on Thursday (7 accidents, 37%) and Friday (5 accidents, 26%). The exposure of risk was estimated in Monday (4 accidents, 21%), Tuesday (7 accidents, 37%) and Wednesday (4 accidents, 21%), and kimchi (5 accidents, 50%) was estimated as the food of the high risk responsible for the outbreaks. 98 isolates of pathogenic E. coli consisted of PEC (50%), ETEC (34%), EAEC (15%), and EHEC (1%). The antibiotic resistance of pathogenic E. coli showed in the descending order of ampicilline (40%), nalidixic acid (37%), trimethoprim/sulfamethoxazole (24%), and tetracycline (19%). The antibiotics of second and third generation cephalosporins, cabarpenem, aminoglycosides, and second generation quinolones had antimicrobial susceptibilities and cefalotin, ampicillin/sulbactam and chloramphenicol showed medium resistance at 29%, 25%, and 6% respectively, and 70% of isolates were resistant to more than one antibiotic. By the PFGE analysis, they were classified into nine major groups and 31 profiles with 57% pattern similarity. It was very difficult to find the correlation of antimicrobial susceptibilities and genotype in the small scale-food poisoning, but the similarity of antimicrobial resistance and PFGE patterns in the large scale-food poisoning enabled the outbreaks to estimate the same pathotype of E. coli derived from identical origins.
