W. Somerset Maugham said in The Summing Up, "The ordinary is the writer's richer field. Its unexpectedness, its singularity, its infinite variety afford unending material. The great man is too often all of a piece; it is the little man that is a bundle of contradictory elements." It dawned on me that he might have applied the 'contradictory elements' to his stories. And I began to think it interesting and useful to find out the 'contradictory elements' in his stories. As I read one story after another, I found them in many of his stories. Such elements as morality and immorality, modesty and immodesty, love and hate, conversion, treachery and disguise are clearly seen in his stories. For example, a missionary of morality committed an immoral act in Rain. A spinster who denied immorality and was reputed as a virtuous woman eloped with a man in The Round Dozen. An educated woman who always wore an identification disc in memory of her dead husband stole into a man's bed in The Razor's Edge. Contradictory elements of morality and immorality are clearly seen. A woman committed adultery with a friend of her husband's and had her husband killed by her lover in Footprints in the Jungle. A woman had illicit intercourse with a footman, pretending to lead a solitary life in. The Human Element. A newly married wife had secret intercourse with her father-in-law in Woman of Fifty. Contradictory elements of modesty and immodesty are obviously seen. A woman loved a man and killed him when she found that he had another woman in The Letter. A chief sailor who had hated the captain's girl fell in love with her in The Four Dutchmen. A drunkard and rascal who had hated a missionary girl decided to marry her in The Vessel of Wrath. A wife of an artist who cursed her husband when he left her showed herself off as a wife of a genius in The Moon and Sixpence. An agent killed a girl whose love he had dearly won in The Hairless Mexican. A woman of an artist who had hated her husband's fellow artist finally fell in love with him in The Moon and Sixpence. Elements of love and hate are apparently seen. A girl who had killed a man to save her lover's life eloped with another lover in Honolulu. A man who loved his wife and adored her literary activities eloped with the cook in The Creative Impulse. A woman who had been so devoted to her husband as to require a double bed when invited to a friend's home for a weekend eloped with a young lover in Virtue. A white man who had lived with a native woman for ten years married a white woman in The Force of Circumstance. Conversion to new lovers are manifestly seen. A youth betrayed his bosom friend and married his friend's girl in A Man with a Conscience. A boy broke the promises he had made with his father in The Facts of Life. A postman paid court to a college girl, but when she had prepared herself to marry him, he refused to marry her in Episode. A woman who had promised to help her friend in marriage broke her promise in The Razor's Edge. We can see obvious elements of treachery. A man who had worked as a carwasher hid his past and pretended to have come from a gentle family in The Lion's Skin. The couple who had once been criminals disguised themselves as gentle people in The Happy Couple We can see apparent elements of disguise. In brief, elements of morality and immorality, modesty and immodesty, love and hate, conversion, treachery and disguise are obviously shown in many of W. Somerset Maugham's stories. Conclusively we can say many of his stories have 'contradictory elements' in them.

들뢰즈의 영원회귀와 예수의 영원회귀성

The intellectual tradition of the western philosophy have varied the concept of the transcendental three times in its course of history: in ancient era, the transcendental denoted ‘above’ as Plato’ s form, in modern era, ‘within the faculty of human’ as in Kant’s transcendental pure form of time and space, and 12 categories, and in late modern era, under the subject or object as in Deleuzian transcendental empiricism. Friedrich Nietzsche was the predecessor of all thoughts of “the depth.” The eternal return enabled Nietzsche to demonstrate repetition of difference, and such thought led him to expel any thoughts under the supervision of identity. In Deleuze’s philosopy, the thoughts of the depth works as a metaphysical principle of the eternal repetition of difference. Unlike Nietzsche, whose philosophical system is largely indebted to the concept of will to power, Deleuze substituted its place with Freud’s concept of death instinct in Difference and Repetition. The death is the ground principle of difference and repetition in his philosophy. The history of the theology has developed in the similar manner: from logos theology as the thought of above to the biblical studies as the thought of the depth. The depth in biblical study denotes the oral tradition of Jesus. K.L Schmidt located this depth by spotting the verses where the chronological and biographical inconsistencies among the Synoptic Gospels were found. He found out that the oral tradition preceded the complete genere of Gospel transcendentally. Moreover, W.G. Kümmel contended that the arrangement and memories of Jesus in the oral tradition have transposed and changed through the reproduction of Gospel. The author of Matthew and Luke have changed the order originally arranged by Mark. Gospel changed its arrangement of the oral tradition to develop each different narrative of Jesus. Therefore, the repetition of life of Jesus presupposes the repetition of genre of Gospel, and the repetition of genre of Gospel presupposes the repetition oral tradition. Through the repetitions, the death and resurrection of Jesus is repeated differently. The revelation of Jesus’ identity proceeds with this dice throw. This paper deals with the tradition of German biblical studies in Deleuze’s perspective, mainly through his early thoughts developed until 1968 서양 철학에서 초월적인 것(the transcental)은 크게 3번 변주한다. 지상 ‘위’(above)를 가리키는 플라톤의 이데아, 인간 ‘안’(within)을 가리키는 대상 인식을 위한 칸트의 12가지 범주라는 논리학적 형식과 시공간의 순수 형식 그리고 들뢰즈의 ‘아래’(under)를 상징하는 초월론적 경험론(Transcendental Empiricism). 서양 철학은 초월을 천상에서 깊이로 인지하기 시작했다. 서양의 철학에서 이 깊이를 가장 먼저 인식한 사람은 프리드리히 니체이다. 니체는 영원회귀 개념을 통해서 오로지 차이나는 반복만이 존재함을 증명하고, 영원한 동일성을 자신의 철학체계에서 추방했다. 들뢰즈 철학에서 이 깊이는 영원한 “차이”를 반복가능하게 해주는 형이상학적 근거로서 작동한다. 니체에게 영원회귀의 작동원리가 힘들간의 강자와 약자를 구별하는 힘에의 의지들간의 영원한 싸움의 반복으로서 표상되었다면, 󰡔차이와 반복󰡕의 들뢰즈는 힘에의 의지의 자리에 프로이트의 죽음 개념을 놓았다. 들뢰즈 철학에서 죽음은 반복의 원리이자, 삶의 근거이다. 같은 방식으로 서양의 신학은 예수를 천상적 이데아의 현신으로 이해하는 로고스 신학에서, 성서 아래에 있는 구전들을 인식하기 시작했다. 구전은 성서학의 ‘깊이’이다. 칼 루드비그 슈미트는 이 깊이를 공관복음서간의 시간적-공간적 차이를 통해서 발견했다. 하나의 복음서라는 작품 이전에 작품은 ‘초월론적’ 근거가 되는 구전들의 존재를 발견했다. 그리고 이 구전들에 담긴 예수에 관한 기억들이 그대로 보존된 것이 아니라 차이를 만들어가면서 보존된다는 것을 게오르그 빌헬름 큄멜은 밝혔다. 구전들이 일정한 서사적 순서로 배치되면 복음서라는 장르가 탄생한다. 최초로 형성된 마가복음서는 구전이 일정한 형태로 배치된 첫 번째 복음서라는 장르다. 그에 뒤따라 나타난 마태복음서와 누가복음서는 마가복음서의 “차이와 반복”이다. 장르의 반복은 구전의 재배치를 전제하고, 장르의 해체는 일정한 서사 안에서 이해된 예수의 ‘죽음’을 의미한다. 예수는 마가의 구전 배치, 마태의 구전 배치, 누가의 구전배치로 다른 죽음과 다른 부활을 반복한다. 예수의 정체는 복음서라는 장르의 반복을 전제하고, 복음서라는 장르의 반복은 구전의 반복을 전제한다,이것이 예수의 주사위 놀이다. 이 논문은 위와 같은 방식으로 들뢰즈의 1968년도까지의 사유를 토대로 20세기 독일 성서학의 존재론적 근거를 마련한다.

실시간 항공 인터컴을 위한 향상된 항공전자 인터페이스 전처리 기법

항공기 인터컴 장비는 조종사, 부조종사, 동승자뿐만 아니라 여러 라디오 채널을 통해 음성을 송수신하는 기능을 담당하고 있다. 또한, 특정 경고, 주의 사항 조건에서 음성 경고음을 재생함으로써 탑승자들에게 즉각적인 상황인지 기능을 제공한다. 이러한 기능들을 성공적으로 수행하기 위해서 인터컴 장비는 임베디드 시스템 기반의 실시간성 보장이 필수이고 실시간성 보장을 위해 대표적으로 인터럽트 방식과 서점형 스케줄링 방식을 사용한다. 하지만 이러한 기법에 대한 단점으로 실시간성이 저하되며 이를 극복하기 위해 사용자의 최적화가 요구되나 하드웨어 및 운영체제에 의존성으로 인해 제한적이다. 이를 위해 본 학위 논문에서는 인터컴 장비의 실시간성 제약사항을 분석하고 연동 데이터 처리 관점과 이벤트 처리관점이라는 두 가지 관점으로 실시간성 향상기법을 제안하였다. 연동데이터 처리관점의 제안은 무의미하며 과도한 Transient 발생으로 인한 실시간성을 저하는 요인을 제거하는 것이고, 이벤트 처리관점의 제안은 실질적으로 데이터 처리에서 유효한 데이터만 취득하고 나머지는 정제하는 것이다. 제안한 두 가지 기법에 대해서 설계 반영 전과 반영 후를 정량적으로 비교함으로써 실시간성에 대한 향상 결과를 정량적으로 입증하였다. The Intercom in aircraft takes the role to provide audio communication among relative personnel such as pilots, copilots, crew and the radio communicators. In addition the Intercom provides the aural warnings and cautions so that crews intuitively identify the situation awareness under particular warning and caution conditions. In order to successfully perform these functions, the Intercom should guarantee real time capabilities based on embedded system but the capabilities are degraded under several constraints. In this paper, after analyzing factors of realtime constraints enhancement designs are suggested with two viewpoints which are the interface data handling and the event handling respectively. And then the two suggestions are verified quantitatively comparing between before design and after design.

북한 집단주의의 형성 및 변천에 관한 연구

This thesis on the formation and changes of North Korean Collectivism of which general idea is the way of communists' life helps to learn how the collective feelings among people and the proletarian political power have been made and changed. This study is done by a kind of historical research which has been introduced by researchers on North Korean politics and sociology. First of all, the historical research is interested in the universal and unique stream of the history in such socialist nation as North Korea. For example, the universal history of North Korean socialism has been unfolded by showing that the Soviet socialist systems of which ideology was called Marxism-Leninism have been transferred to North Korea since it established its own government in the north of the peninsula. On the other hand, the unique history of North Korean political power has been revealed by such totalitarian ideologies as 'Juche ideology', 'Sun-goon political ideology', etc. The unique history has been spotted by the adoration of the highest leader who has monopolized all of the power. According to this, North Korean Collectivism has been formed and changed over several decades especially from the collectivism sprout age in Japanese colony. Japanese colonial socialists, laborers, and farmers somewhat experienced the proletarian collective union. After that early North Korean Collectivism was inclined to the Soviet collectivism which came true for the first time in the world. It was characterized by Lenin's collectivism including his 'party-nation theory' that the communist party and nation represented the proletarian interests as well as by the original proletarian collectivism. Since that, North Korean Collectivism, however, has been changed into totalitarianism in which the adoration of the leader , the secret police organization and its terror system, the monopoly of the political party and the mass media, and so on are found. The changes of North Korean Collectivism are characterized by the change of the main body in the proletarian revolution. In other words, North Korea has changed the labor class which is called proletariate and which is originally the main body in the proletarian revolution into the subordinate body. The downfall of the proletariate in North Korea is specified by the domination of the communist party and its prime leader. In addition to that, 'Sun-goon politics' which made the People's Liberation Army appear on the stage of the political power openly changed the army into the political body. This means that the army is no longer a device of the proletarian revolution but the main body of the revolution just like the communist party and the people. North Korean Collectivism has had some noticeable characteristics through these changes of its own. First, like the above written passage, North Korean Collectivism turned into totalitarianism in which every power is in the hands of the highest leader. Secondly, North Korean Collectivism is found to have its operation principles such as suffering the national difficulty, learning the national ideologies, and magnifying and reproducing of collective thinking. These principles make the people strained by the pressure from the foreign hostile countries, every day collective activities, customary inspections, etc. Thirdly, there are two functions of North Korean Collectivism. One is unifying the society and the other is controlling the society. In the last place, two weaknesses of North Korean Collectivism such as potential individualism and ruling stratum's irrationality are found in interviews with people to escape from North Korea. In conclusion North Korean Collectivism is in danger of resigning its true character that is the proletarian cultural identity or exclusion. Global socialist sphere's downfall, economic difficulties, the question of the leadership or of the governor, the potential individualism among people, and so on threaten even the traditional collective system by Marx and Lenin. Collective farms, for example, and food distribution are not preferred to individual farms and food trade in market. Consequently, the government is going to force people to expose themselves to more propaganda about collectivism than before.

2015 개정 수학 교과서의 '도전 수학'에 대한 초등 교사의 인식 및 실태 분석

본 연구의 목적은 2015 개정 교육과정에서 강조하고 있는 교과 역량 신장 및 수학과 교육과정의 목표를 달성하기 위해서 도입된 2015 개정 수학과 교육과정에 따른 초등 5~6학년군 수학 교과서의 ‘도전 수학’은 어떠한 역할을 하고 있는지, 어떻게 운영되고 있는지, 과정 중심 평가에 어떻게 활용되고 있는지, ‘도전 수학’을 지도하기 위해 필요한 교사 지원은 어떠한 것들이 있는지에 대한 교사의 인식을 분석하여 ‘도전 수학’ 차시의 교수·학습 활동에 도움을 줄 수 있는 기초 자료를 제공하고 교육과정 개정 방향 모색에 도움을 주는 데 있다. 본 연구를 위해 5, 6학년 교사들을 대상으로 하여 설문지를 통한 조사 연구 방법을 적용하였고, ‘도전 수학’에 대한 교사의 인식, 지도 실태, 지도를 위해 필요한 지원 방안을 분석한 결과는 다음과 같다. ‘도전 수학’에 대한 교사의 인식에서 ‘도전 수학’에 대한 교사의 관심은 높지 않지만, ‘도전 수학’이 필요하다고 생각하는 교사의 비율이 높았다. 교사들은 2009 개정 수학과 교육과정의 ‘문제 해결’ 차시와 ‘도전 수학’의 차이를 인식하고 있는 편이고, 2015 개정 수학과 교육과정에서 강조하는 부분을 인식하고 있었다. 교사들은 ‘도전 수학’ 차시는 수와 연산, 도형, 규칙성, 자료와 가능성, 측정 영역의 순으로 지도에 효과적이라고 생각하였고, 그에 대한 이유로 다양한 풀이 방법을 활용하여 수학적 사고력을 높일 수 있고, 실생활 관련 주제로 학생들이 수학의 유용성을 인식할 수 있기 때문이라고 생각했다. ‘도전 수학’ 차시는 문제 해결, 추론, 창의·융합, 정보처리, 의사소통, 태도 및 실천 역량의 순으로 그것을 기르는 데 효과적이라고 생각했다. ‘도전 수학’ 차시 지도의 실제 부분에서 교사들이 ‘도전 수학’을 지도할 때 선호하는 지도 방법은 ‘해결할 시간을 주고 다 같이 확인하기’, ‘각자 해결할 시간을 주고 부진아를 집중 지도하기’, ‘학생들 스스로 해결하고 서로 가르치게 하기’의 순으로 나타났다. ‘도전 수학’ 차시가 한 차시 분량으로 적당하다고 생각하는 교사는 절반 이상이었고, 학생들에게 도전의 수준이 대체로 높거나 적절하다고 생각하는 비율이 각각 절반 정도로 나타났다. ‘도전 수학’ 학습이 학생들에게 주는 긍정적인 효과로 수학적 사고력 증진을 가장 많이 선택하였고, 수학적 기능 숙달을 다음으로 많이 선택하였다. ‘도전 수학’을 지도하면서 어려운 점은 학생들의 학습 능력 차이와 학습 내용과 학생 수준의 차이, 학생 흥미 부족, 수업 자료의 부족의 순으로 나타났는데, 이러한 이유로 수업을 재구성하여 진행하기도 하고 희망 연수에 의견을 제시하기도 했다. 교사용 지도서에 안내된 평가 안내사항에 대해 인식하지 못하고 있는 교사가 절반 가까이 있었고, ‘도전 수학’ 차시에서 과정 중심 평가를 실시해보지 않은 교사가 많았다. 다른 차시에서의 평가로 성취기준에 도달할 수 있고 평가를 실시하기에 난이도가 높아 피드백에 대한 부담이 있는 것으로 나타났다. ‘도전 수학’ 차시 지도를 위해 필요한 지원 부분의 결과로 2015 개정 교육과정에 대한 연수에 참여한 경험이 있는 교사는 절반 미만이었고, 이 중 40%의 교사는 연수가 도움이 된다고 하였다. ‘도전 수학’에 관련된 연수 경험은 대부분 없었다. ‘도전 수학’ 차시와 관련하여 희망하는 연수 내용은 학생들의 개인차 극복 방안, 수업 지도 사례, 교과 역량의 이해 및 지도 방안, 과정 중심 평가 사례, 차시에 관련된 수학적 지식, 수학 교구 활용 방법, 수업 재구성 방법의 순으로 나타났고, 실제로 적용할 수 있는 방안들에 대한 희망이 많았다. 절반 이상의 교사들은 교사용 지도서가 ‘도전 수학’ 차시의 지도에 도움이 된다고 생각하고 있고, 수업의 흐름과 핵심 발문, 차시에서 강조하는 교과 역량을 주로 참고하는 것으로 나타났다. 이와 같은 연구 결과를 통해 다음과 같은 결론을 얻을 수 있다. ‘도전 수학’이 학생들의 수학적 사고력과 수학 교과 역량을 기르는 데 효과적이라고 인식하고 ‘도전 수학’의 필요성을 제기하는 교사가 많지만, ‘도전 수학’ 차시를 지도하는 데 여러 가지 어려움이 따르고 있으므로 ‘도전 수학’ 차시의 도입 목표를 달성하기 위해서는 다음과 같은 노력이 필요하다. 다양한 학습자의 수준과 요구를 반영할 수 있도록 다양하고 흥미로운 주제의 수준별 학습 자료와 지도 방법의 개발 및 보급에 힘써야 할 것이다. 교과서를 개발하는 단계와 시범 적용하는 단계에서 다양한 상황이 모두 반영될 수 있도록 연구·시범학교의 운영을 늘려서 다양한 의견을 수렴해야 한다. 교육과정에 대한 전달 연수 체계를 확립하고 선진적인 강사 시스템을 도입하며 교사가 부담 없이 연수에 참여할 수 있도록 방문 연수 등의 형태로 연수 참여율을 높이는 방안을 연구해야 한다. 그리고 교사들 간에 수업 지도 사례, 수업 방법 등의 공유가 활발히 이루어질 수 있는 소통의 장을 마련해주어야 한다. 담임교사의 업무 경감을 위해 노력해야 하고 교사가 전문성을 키울 수 있도록 교사 공동체 중심의 연수를 적극 지원하며 교사에게 수업의 자율성을 보장해 주어야 한다. 수업에 필요한 수학 교구를 구비하여 수업에 활용할 수 있도록 해 주어야 한다. 교사는 ‘도전 수학’에 관심을 가지고 ‘도전 수학’에 대한 전문성을 키우기 위해 교사용 지도서를 적극적으로 활용하거나 교육과정 연수에 적극적으로 참여해야 하고, 자신의 지도 사례나 과정 중심 평가 사례 등을 적극적으로 공유하여 교육과정의 목표가 실현될 수 있도록 힘써야 할 것이다.

HHT를 이용한 간극이 있는 회전체의 고장진단

Partial rub and looseness are common faults in rotating machinery due to the clearance between the rotor and the stator. These problems are the cause of malfunctions in rotating machinery since those create strange vibrations coming from impact and friction. However, non-linear and non-stationary signals due to the impact and friction are difficult to identify. Therefore, exact time and frequency information are necessary. For those purpose, newly developed time-frequency analysis method, HHT(Hilbert-Huang Transform) is applied to the signals of partial rub and looseness. Comparison of conventional signal processing methods such as FFT, STFT and CWT has been made to verify the effectiveness of fault diagnosis using HHT. The results show that the impact signals are generated regularly when the partial rub is occurred, and then intermittent impact and friction are occurred as the increase of the looseness. Time and frequency information is represented exactly by using HHT.Consequently the features of rotor system with clearances are identified clearly.

한국인 불어학습자들을 대상으로 한 설문조사에서 명확하게 드러났듯이, 이들은 불어의 세 가지 핵심 과거시제인 복합과거/반과거/단순과거를 명확히 구분하여 올바로 사용하는데 상당히 큰 어려움을 나타내고 있다. 이러한 문제점에 입각하여 본 논문은 특히 이 세 종류의 과거시제와 연관된 불어교육 측면을 다루고 있다. 이 논문에서는 이론적 방법론의 확립을 위하여 우선 동사와 밀접한 관계를 맺고 있는 시상의 일반적인 문제에 접근하였으며, 아울러 하나의 과거시제밖에 가지고 있지 않는 한국어특성이 불어학습자들을 혼돈시키므로 현대 한국어의 시제와 시상에 관한 분석을 발화이론에 입각하여 행하였다. 왜냐하면 이러한 분석을 통하여 한국인 불어학습자들이 행하는 오류의 일반적인 특성을 파악할 수 있기 때문이다. 다음으로 불어의 시제체계에 관한 언어학자들의 다양한 견해를 살펴보았는데, 이들 중 특히 E.BENVENISTE의 발화이론과 H.WEINRICH의 텍스트언어학이론이 언어학이론의 언어교육에의 활용측면상 가장 큰 장점을 나타냄을 관찰할 수 있었다. 왜냐하면 상호 관련되어 기능하는 불어의 시제체계는 단순한 하나의 문장수준으로는 설명되어질 수 없기 때문이다. 이와 같은 특성상 본 논문은 언어학적 바탕으로서 발화이론과 텍스트이론을 동시에 차용하였다. 상기와 같은 이론적 고찰단계를 마친 후, 본 논문은 한국인 학습자들을 위한 이러한 이론의 불어교육에의 활용을 시도하였다. 불어의 복합과거/반과거 및 단순과거/반과거 쌍의 효율적이고 기능적인 교수 및 학습활동을 위하여 설명도식과 함께 "Premier plan (1차측면)"과 "Second plan (2차측면)"의 개념을 도입하였다. 이와 같이 이론의 효율적인 적용을 위한 설명단계를 거친 후, 이의 활용을 위해 단계별로 구성된 연습문형을 제시하였다. 연습문형은 학습자의 이해도 및 학습난이도에 따라 점진적으로 발전하도록 총 여섯 단계로 구분되었으며, 이 문형들은 이해하기 쉬운 불어로 쓰여진 '광고문안, 인터뷰기사, 서간문, 샹송' 등의 다양한 유형의 텍스트로 구성되었다. 이 연구는 어떤 특정 언어학 주제에 관해 이론적인 고찰과 분석을 하는 연구가 아니라 불어의 과거시제 체계를 학습하는 한국인 학습자들을 위해 그들이 부딪치는 한 가지 어려운 문제에 공헌을 해 보고자 하는 외국어 교육학적 관점의 연구였으며, 이러한 종류의 연구를 통하여 외국어 교수자들에게는 그들의 어려운 임무를 경감시키고, 학습자들에게는 그들의 의사소통 능력의 향상을 위해 도움을 줄 수 있을 것이다.

Evaluation of real-time PCR assay for diagnosis of Tuberculosis

결핵 (TB)은 결핵균 (Mycobacterium tuberculosis, MTB)에 의해 주로 발생하고, 결핵균은 4개의 다른 마이코박테리아 종과 함께 결핵균 복합체 (Mycobacterium tuberculosis complex, MTBC)에 속한 병원성 박테리아 종입니다. MTBC에 포함되지 않은 마이코박테리아는 비결핵항산균 (nontuberculous mycobacteria, NTM)으로 알려져 있으며 폐렴을 비롯한 여러 폐 질환을 유발한다. 최근 결핵으로 의심되는 호흡기 환자의 검체에서 NTM의 발생이 증가하여 결핵의 진단 및 예방 접종에 있어 MTBC와 NTM의 정확한 구분과 개별 탐지가 중요하다. 결핵의 진단에 사용되고 있는 여러 가지 방법 중에서 실시간 PCR(Real-time PCR)은 신속성, 정확성 및 취급 용이성 때문에 MTBC 및 NTM 검출에 자주 이용된다. 최근 실시간 PCR을 이용하여 결핵균 및 비결핵항산균을 진단할 수 있는 새로운 키트가 출시되었고, 이 연구에서 우리는 객담, 기관지 세척 및 배양 검체를 대상으로 하여 새로운 키트에 대한 분석적 성능 및 임상적 성능을 평가하였다. 분석 성능을 평가하기 위해 표준 DNA 샘플을 사용하여 검출 한계 (LOD), 반응성 및 반복성 테스트를 수행하였고, 임상적 성능시험을 평가하기 위해 612 개의 샘플을 수집하여 국내 대형병원에서 임상 시험을 실시했다. LOD는 probit analysis (95 % positive)에 의해 MTBC는 0.584 copies / μL 와 NTM은 47.836 copies / μL로 확인되었다. 반응성 시험은 설정한 모든 박테리아 DNA를 검출하였고, 재현성 시험에서 0.36에서 1.59까지의 변동 계수(Coefficient Variation, CV)를 확인함으로써, 반복된 시험에서 일정하고 안정된 결과가 확인되었다. 임상 시험에서 민감도와 특이도는 MTBC와 NTM 각각 98.6 % -100 %와 98.8 % -100 %였다. 이 결과로 인해 결핵의 진단에 있어 평가 대상 키트의 유용함을 입증하였다. Tuberculosis (TB) is mainly caused by Mycobacterium tuberculosis (MTB), which is a pathogenic mycobacterial species grouped under Mycobacterium tuberculosis complex (MTBC) with four other pathogenic mycobacterial species. The mycobacteria not included in MTBC are known as nontuberculous mycobacteria (NTM), and cause several pulmonary diseases including pneumonia. Currently, NTM occurrences in TB-suspected respiratory specimens have increased, due to which, precise detection of MTBC and NTM is considered critical for the diagnosis and vaccination of TB. Among the various methods available, real-time PCR is frequently adopted for MTBC/NTM detection due to its rapidness, accuracy, and ease of handling. In this study, we evaluated a new real-time PCR kit for analytical and clinical performance on sputum, bronchial washing, and culture specimens. For assessing its analytical performance, limit of detection (LOD), reactivity, and repeatability test were performed using DNA samples. To evaluate clinical performance, 612 samples were collected and clinically tested at a tertiary hospital. LOD was confirmed as 0.584 copies/μL for MTBC and 47.836 copies/μL for NTM by probit analysis (95% positive). For the reactivity test, all intended strains were detected and, in the repeatability test, stable and steady results were confirmed with coefficient of variation ranging from 0.36 to 1.59. For the clinical test, sensitivity and specificity were 98.6%-100% and 98.8%-100% for MTBC and NTM, respectively. The results proved the usefulness of the kit in TB diagnosis. Furthermore, it could be adopted for the assessment of vaccine efficacy.
