        The ultrastructural study of the adrenal medulla has been investigated in normal Sprague-Dawley rats from neonates to adults. Adrenal glands were fixed in ice-cold, 4% paraformaldehyde and 0.5% glutaraldehyde and processed for routine electron microscopic procedures. In the adrenal medulla of the neonate, chromaffin cells contained a mixture of epinephrine and norepinephrine cell granules. Between second and eighth weeks distinct epinephrine and norepinephrine chromaffin cells were evident in the adrenal medulla and myelinated nerve fibers appeared and synaptic nerve endings were matured.

        This study was performed in male Sprague-Dawley rats to investigate effects of ceftezole sodium(CTZ) on Brunner's gland. Rats were treated intraperitoneally with CTZ at 1000mg/kg/day for 7 consecutive days. 1)Alcian blue(pH 2.5)-P.A.S stain: In Brunner's gland of the treated group, there was a decrease in red and blue color reactivity. This means that weakly sulfated acid mucin and neutral mucin especially decreased. 2)EM study: In acinus of Brunner's gland of the treated group, wide and irregular intercellular spaces and small sized secretory vesicles were observed. But any other distinct changes in the cytoplasmic organelles were not detectable. Consequently, it was suggested that CTZ influences the mucopolysaccharides synthesis in Brunner's gland.

      • 흰쥐 Chromaffin 세포의 분화에 대한 형태학적 연구

        The development of rat adrenal medulla was studied at the morpholgical and immunocytochemical level with particular emphasis placed on pathways of the movement and activities of the tyrosine hydroxylase. In the chromaffin cells, tyrosine hydroxylase, dopamine-beta-hydroxylase and PNMT are necessary to catecholamines metabolism. Of them tyrosine hydroxylase is rate-limiting enzyme of the catecholamines synthesis. The author studies the morphological changes of the adrenal medulla and immunocytochemical distinctions for tyrosine hydroxylase during the differentiation of the chromaffin cells. The adrenal glands were obtained from fetal rats at the 14th, 16th, 18th and 20th days of gestation and 1st, 3rd postnatal days and these paraffin sections were stained by hematoxylin-eosin for general morphological changes. And immunocytochemical study for the expression of tyrosine hydroxylase of the chromaffin cells was done by ABC methods. The results obtained were as follows. 1. Maturation of the adrenal medulla occurred by the pheochromoblasts migrating to a central position of the cortex anlagen. At 3th postnatal day, most chromaffin cells were situated in central location and the formation of capsular structure was finished. 2. Expressions of the tyrosine hydroxylase showed high activities at the time just before birth.

        모바일 앱 기반 자동굴절력계의 임상성능 분석

        Purpose : The mobile app-based auto refractometerb (NETRA refraction mobilized, NR), developed by EYENETRA in the United States, is a device that measures refractive errors by operating the device directly by the test subject, and in order to confirm the clinical performance of NR, the measured values of subjective refraction (SR) and HRK-8000 were compared and analyzed. Methods : This study recruited 124 patients (248 eyes) with myopia or myopic astigmatism, no systemic disease or eye disease under treatment, and no history of ophthalmic surgery from the customers of M optical store in Gyeonggi-do. The tests were conducted in the order of NR, HRK-8000, and SR to obtain a refractive error measurement result. The SPSS program (version 26) was used for statistical analysis, and a case of p<0.050 was judged statistically significant. Results : Spherical, cylindrical, and spherical equivalent refractive powers were measured to have the highest myopia in NR measurements (p<0.01), and the average value compared to SR showed a lager difference in NR than HRK-8000, out of the standard deviation range. In the ±5° range, the astigmatic axis of NR was 41.4% concordant with the SR astigmatic axis, whereas HRK-8000 was 90.8% concordant. The 95% limits of agreement range of spherical, cylindrical, and spherical equivalent of refractive powers for SR were +2.01∼-2.72 D, +1.69∼-2.20 D, and +1.89∼-2.81 D in NR, +0.84∼-0.96 D, +0.42∼-0.73 D, and +0.81∼ -1.09 D in HRK-8000. Therefore, the range of agreement of NR was wider than that of HRK in all variables, resulting in low accuracy. Conclusion : In light of the findings mentioned above, it is judged that NR will be difficult to replace the existing automatic refractometer, and it is believed that accuracy needs to be improved in order to be used as a preliminary test in clinical practice. 목적 : 미국의 EYENETRA 회사가 개발한 모바일 앱 기반 이동형 자동굴절력계(NR)는 피검사자가 직접 기기를 작동하여 굴절이상도를 측정하는 기기로, 그 임상성능을 확인하기 위해, 고정형 자동굴절력계(HRK-8000)와자각적 굴절검사(subjective refraction, SR) 측정값을 비교 분석하고자 하였다. 방법 : 경기도 소재 M 안경원의 고객 중, 치료 중인 전신질환 및 안질환이 없고, 안과 수술을 받은 적이 없는근시 또는 근시성 난시 124명(248안)을 대상으로 하였다. 굴절이상도 측정값을 얻기 위하여 NR, HRK-8000과SR 순서로 검사를 진행하였다. 통계처리는 SPSS program(version 26)을 사용하였고, p<0.050인 경우를 통계적으로 유의한 것으로 판단하였다. 결과 : 구면, 원주, 등가구면 굴절력은 NR 측정값의 근시도가 가장 높게 측정되었고(p<0.010), SR에 대한 NR 의 평균값은 HRK-8000보다 차이가 크게 나타나 표준편차 범위를 벗어났다. SR에 대한 난시축 일치도는 ±5° 범위에서 NR이 41.4% 일치하였고, HRK-8000은 90.8% 일치하여 NR의 난치축 일치도가 낮게 나타났다. SR에대한 NR과 HRK-8000의 정확도를 확인하기 위해 95% 일치 한계(LOA)를 비교한 결과 구면, 원주굴절력과 SE 성분의 NR 범위는 각각 +2.01∼-2.72, +1.69∼-2.20와 +1.89∼-2.81 D로 나타났고, HRK-8000 범위는 각각 +0.84∼-0.96, +0.42∼-0.73와 +0.81∼-1.09 D로 나타났으며, 모든 변수에서 NR의 범위가 넓어 정확도가 낮은 것으로 확인되었다. 결론 : 이상의 결과를 종합하면, 기존의 자동굴절력계를 대체하기는 어려울 것으로 판단되었고, 임상에서 예비검사로 활용되기 위해서는 정확도가 개선될 필요가 있다고 사료된다.

      • 흰쥐 정소 간질세포의 발생에 관한 형태학적 연구

        It is well known that interstitial cell of Leydig is developed from the mesenchymal cell in the intestitial tissue of the testis during fetal period, but it gradually regressed until puberty when mature Leydig cell is derived from the mesenchyme of interstitial tissue of fetal Leydig cell. The function of the Leydig cell is synthesis and secretion of testosterone which influences on differentiation of male geritalia at fetal period, and revealation of male secondary sexual characteristies and differentiation of sperm at puberty. To investigate the differentiation of interstitial cell in interstitium and ultrastructural changes of interstitial cell in rat testis and ultrastructural changes of interstitial cell in rat testis and relation to testosterone secretion, the author has studied this experiment. Fetal rats on the 16th, 18th and 20th gestational days and rats on the 1st, 3rd, 7th day, 2nd, 3rd and 8th week after birth were used as experimental animals. Some of obtained testis were stained with hematoxylin and eosin for light microscopic study, and other specimens were double stained with uranyl acetate and lead citrate for electron microscopic study. The results were as follows: 1. On the 16th day of pregnancy, a few of round eosinophilic Leydig cells are firstly observed in the interstitium of testis in fetal rat. On the 18th and 20th day of pregnance, 1st and 3rd day of neonatal life, a number of Leydig cells forming cellular cord were observed. On the 1st and 2nd week after birth, number and size of Leydig cell are markedly reduced but on teh 3rd week after birth Leydig cells are reappeared. On the 8th week after birth distribution of Leydig cells is similar to that in adult rat testis. 2. On the 18th and 20th day pregnancy, and 1st and 3rd day of neonatal life, circular or oval shaped mitochondria matrix were observed in the Leydig cell. On the 1st and 2nd week after birth, giant mitochondria and mitochondria with disorganized mitochondrial crista were revealed. On the 3rd and 8th week after birth, appearances of mitochondria were similar to that in Leydig cell of the rat testis on the 20th day of pregnancy. 3. Numerous and round cisternae of smooth endoplasmic reticulum were observed in Leydig cell of rat testis on the 18th and 20th day of pregnancy. 1st and 3rd day of neonatal life, and 1st week after birth . Horseshoe-shaped deformity and reduction in number of cisternae of smooth endopalmic reticulum were revealed in Leydig cells on the 2nd week after birth. Numerous and round cisternae of smooth endoplasmic reticulum were seen on the 3rd and 8th week after birth. 4. A few lipid droplets were revealed in Leydig cell of rat testis on the 18th day of pregnancy, but many lipid droplets were observed in Leydig cells of rat testis on the 20th day of gestation, 1st and 3rd day of neonatal life, and 1st week after birth. Numbers of lipid droplets were gradually decreased after 2nd week of postnatal life. 5. Many autophagic vacuoles were observed in Leydig cells would be firstly differentiated from the mesenchymeal cells in interstitium of fetal rat testis on the 16th day of pregnancy, and proliferated from the 18th day of pregnancy to the 1st day of neonatal life, and then gradually regressed before the puberty, but at the begining of puberty adult type of Leydig cell be proliferated.
