지렁이 (Eisenia andrei)의 중장에서 내생의 endoglucanase 유전자의 전체 염기 서열을 동정하였다. ORF의 길이는 1,371 bp이며, 456개의 아미노산으로 번역된다. NCBI에 등록된 가재와 흰개미의 cellulase 및 endo-$\beta$-1, 4-glucanase와 50-51%의 유사성을 보이며, 활성 부위가 잘 보존되어 있었다. 계통수 분석에서는 다른 동물 분류군에서 밝혀진 GHF9 그룹의 cellulase와 근연관계가 없음이 확인되었다. Endogeneous endoglucanase (EC 3.2.1.4) cDNA was cloned from a representative species (Eisenia anderi) of the earthworm family Lumbricidae. Endoglucanase from the midgut of the earthworm is composed of 456 amino acids and belongs to glycosyl hydrolase family 9 (GHF9), sharing high homologies (50-51 %) with those of selected termite and crayfish. This endoglucanase consists of three consensus catalytic domains found in most microbial cellulases. A phylogenetic tree was constructed using the amino acid squence data matched through the BLASTX program and showed that GHF9 families could be divided into four groups of arthropoda, bacteria, plant and annelida.

It might be at the age of Queen Chinseong that the present 5eongju District began to be freed from the rule of the Central Government. And then, it might be inferred, when Later Packjae attacked and occupied 10 castles or so south of II-sung Commencery at the 11th year of King Hyogong, that Seongju began to be influenced by Later Packjae. At that time, however, Later Packjae was unable to keep on holding the territory which it took from Sillla, so the present Seongju District was ruled by the lord who extended his power and called himself a General. After that, it is obvious that Seongju District was influenced againby Later Packjae from about 4th year of King Kyoungmyoung(920 A.D.), equal to the period when Koryeo and Later Packjae was in the rivals each other. But Seongju became to be under the rule of Korea, aftert Ryangmun, General of Pyuckjin Comrnendery, yielded to Koryung in the ?th year of King Kyoungmyoung(924). In the first year of King, Kyoungae(924), Koryeo and Later Packjae committed a battle for a long period at Jomoo Castle, and General Lee in Later Packjae, local gentry in Seongju District, yielded to Koryeo, which turned the battle favorable to Koryeo. After the Battle of Gongsan, offering a route of retreat to King, Wang Keon who lost in the battle, he contributed much to King, Wang Keon, On one hand, when Lee Chong-eon, General of Pyuckjin Commendery, fought against Koryeo at the Battle of Chomul Castle, his son, Lee Young joined in the battle and served greatly to gain victory for Koryeo. King, Wang Keon rewarded him far his distinguished services at the battle, and appointed him as General Boneup and as Samjungdaekwang. What Lee Neung-il had achieved, can be revealed from a lot off history books written at the middle period of Lee Dynasty, but his name, life and achievements are not written in the Samgaoksaki and Koryeosa which are the basic historical materials. Therefore, the writer concluded that Lee Neung-il might be considered as Lee Neung-mun who can be traced and identified from the basic historical documents, who yielded to Koryeo in the second year of King Kyoungae(925) and was appointed as Seangsangun and ruled Kyaungsan Commendery.
Linkage Analysis를 이용한 다발성 내분비 선종 Type 2A의 유전자 진단

연구 배경 : 다발성 내분비선종(MEN)2A는 상염색체성 우성 유전의 형태를 취하면서 여러 내분비 기관에서 다발성으로 종양이 발생하는 질환으로서 조기 진단이 중요하다. 이와 관련하여 전통적으로 사용되는 pentagas-trin 자극 시험등의 생화학적 검사에는 여러 문제가 있으므로, 최근에는 MEN 유전자와 close linkage 상태에 있는 marker gene을 이용하여 진단을 하려는 연구가 시도되고 있다. 이 연구가 성공적으로 수행되먼 MEN 2A의 분자 생물학적 이해에도 도움을 줄 수 있으며, 임상적으로는 매년 생화학적 검사를 받지 않고도 유아기 또는 prenatal period에 진단이 가능해진다. 연자들은 한 MEN type 2A 가족에서 MEN 2A 유전자와 매우 가깝다고 알려진 RBP3와 D10S15 유전자좌의 poly-morphism 즉 RFLP와 이의 유전양식을 조사함으로써 MEM 2A의 유전자 진단을 시도하였다. 방법 : MRN 2A 가족의 DNA를 phenol-chlorform 법으로 추출하고 Bg1Ⅱ, MSPⅠ 제한효소로 절단한 후 agarose gel로 전기영동를 시행한 후 Southern trans-fer를 실시한 다음 각각 IRHP. H4, MCK2 탐식자와 함께 hybridization 시켰다. 결과 : 1) proband는 Bg1Ⅱ 제한효소 절단시 6.3 kb band를 갖는 대립 유전자의 homozygote로 판명되었다. 2) 표현형 상 MEN 2A가 있었던 proband의 어머니는 6.3kb, 4.3kb band를 갖는 heterogygote였으며 따라서 6.3kb band를 갖는 대립 유전자가 MEN 2A 유전자와 linkage 상태에 있고 그 haplotype이 어머니로부터 proband에게 넘어 왔으리라 추정되었다. 3) MEN 2A 표현형을 가지고 있지 않던 여동생은 6.3kb band의 homozygote로서 역시 어머니의 MEN 2A 유전자를 물려받았을 것으로 생각되었다. 4) D10S15 유전자좌의 경우에서는 MspⅠ 절단 후 RFLP 분석 결과 전원이 2.4kb band의 homozygote로서 유전자 진단에 도움이 되지 못하였다. 결론: MEN 2A 유전자와 매우 근접해 있는 유전자좌에서의 RFLP의 분석은 early 또는 prenatal period에서의 진단을 가능케 하리라고 사료되었다. Background Multiple endocrine neoplasia type 2A (MEN 2A) gene has been shown to be in linkage with genetic loci near the centromere of chromosome 10. Polymorphic DNA probes for the region are now available permitting the use of restriction fragment length polymorphism (RFLP) to identify carriers of the gene for this disease. We attemped genetic diagnosis of MEN 2A by RFLP analysis of RBP3 and D10S15 loci located near the centromere of chromosome 10. Methods: DNA was extracted from the leukocytes of family members with MEN 2A using phenol-chlorform method, digested with BgⅢ and MspI, subjected to electrophoresis and Southern blotting, and hybridized with radiolabelled IRBP3. H4 and MCK2 probes. Results: 1) A IRBP3 allele yielding 6.3 kb band when digested with BgⅢ was supposed to be in linkage with MEN 2A in this family. 2) Sister of the proband was a homozygote of 6.3 kb band and thought to have MEN 2A gene. 3) RFLP analysis of D10S15 locus after MspI digestion was not contributory to the diagnosis in this family because all family members were homozygous for a D10S15 allele yielding 2 . 4 kb when digested with MspI. Conclusion: RFLP analysis of the loci located near the centromere of chromosome 10 would make presymptomatic diagnosis of MEN 2A possible.
