        Molecular Characterization and Infectious cDNA Clone of a Korean Isolate of Pepper mild mottle virus from Pepper

        A Korean isolate of Pepper mild mottle virus (PMMoVKr) was isolated from a diseased hot pepper plant and its biological and molecular properties were compared to that of PMMoV-J and PMMoV–S. The genomic RNA of PMMoV-Kr consists of 6,356 nucleotides. The nucleotide and amino acid sequences identities of four viral proteins and two noncoding regions among PMMoV-Kr, PMMoV-S and PMMoV-J were 96.9% to 100.0% and 97.5% to 98.6%, respectively. Full-length cDNA amplicon of PMMoV-Kr was directly amplified by RT-PCR with a set of 5'-end primer anchoring T7 RNA promoter sequence and 3'-end virus-specific primer. Capped transcript RNAs from the full-length cDNA clone were highly infectious and caused characteristic symptoms of wild type PMMoV when mechanically inoculated to systemic host plants such as Nicotiana benthamiana and pepper plants.

        High-performance ZnS@graphite composites prepared through scalable high-energy ball milling as novel anodes in lithium-ion batteries

        A ZnS@graphite composite was developed using a high-energy mechanical milling (HEBM) for use as ananode in lithium-ion batteries. Compared to previous studies on ZnS-based electrodes prepared usingdifferent processes, the HEBM is advantageous because of its simplicity, scalability, and greaterperformance. In this work, we mainly focused on thefinding of optimal carbon-based matrix that cansignificantly enhance the performance of ZnS-based composite electrode. In comparison with other typesof ZnS-based composites, ZnS@graphite exhibited superior performances with a reversible capacity of444 mA h g 1 after 300 cycles for the half cell (capacity retention of 71% compared to that of the 2ndcycle), 117 mA h g 1 after 50 cycles for the full cell (energy density of 363 W h kg 1), and excellent ratecapability (80% capacity retention at 3000 mA g 1 compared with that at 100 mA g 1). This is attributedto higher surface area (associated with solid-lubrication of graphite), homogeneous mixing, and highelectrical conductivity as confirmed by various characterizations including BET, HRTEM, SEM, and sheetresistance measurements. The origin of the improved cyclic stability and rate capability of ZnS@graphiteover other ZnS-based electrodes was further investigated by CV, EIS, and ex situ SEM.

        Construction of Infectious cDNA Clone of a Chrysanthemum stunt viroid Korean Isolate

        Chrysanthemum stunt viroid (CSVd), a noncoding infectiousRNA molecule, causes seriously economic losses ofchrysanthemum for 3 or 4 years after its first infection. Monomeric cDNA clones of CSVd isolate SK1 (CSVd-SK1) were constructed in the plasmids pGEM-T easyvector and pUC19 vector. Linear positive-sense transcriptssynthesized in vitro from the full-length monomericcDNA clones of CSVd-SK1 could infect systemicallytomato seedlings and chrysanthemum plants, suggestingthat the linear CSVd RNA transcribed from the cDNAclones could be replicated as efficiently as circularCSVd in host species. However, direct inoculation ofplasmid cDNA clones containing full-length monomericcDNA of CSVd-SK1 failed to infect tomato and chrysanthemumand linear negative-sense transcripts fromthe plasmid DNAs were not infectious in the two plantspecies. The cDNA sequences of progeny viroid in systemicallyinfected tomato and chrysanthemum showed afew substitutions at a specific nucleotide position, butthere were no deletions and insertions in the sequencesof the CSVd progeny from tomato and chrysanthemumplants.

        Genetic Diversity of a Natural Population of Apple stem pitting virus Isolated from Apple in Korea

        Apple stem pitting virus (ASPV), of the Foveavirus genusin the family Betaflexiviridae, is one of the most common viruses of apple and pear trees. To examine variability of the coat protein (CP) gene from ASPV, eight isolates originating from 251 apple trees, which were collected from 22 apple orchards located in intensiveapple growing areas of the North Gyeongsang and North Jeolla Provinces in Korea, were sequenced and compared. The nucleotide sequence identity of the CP gene of eight ASPV isolates ranged from 77.0 to 97.0%, while the amino acid sequence identity ranged from 87.7 to 98.5%. The N-terminal region of the viral CP gene was highly variable, whereas the C-terminal region was conserved. Genetic algorithm recombinationdetection (GARD) and single breakpoint recombination(SBP) analyses identified base substitutions between eight ASPV isolates at positions 54 and 57 and position 771, respectively. GABranch analysis was used to determine whether the eight isolates evolved due to positive selection. All values in the GABranch analysis showed a ratio of substitution rates at non-synonymous and synonymous sites (dNS/dS) below 1, suggestive of strong negative selection forces during ASPV CP history. Although negative selection dominated CP evolutionin the eight ASPV isolates, SLAC and FEL tests identified four possible positive selection sites at codons 10, 22, 102, and 158. This is the first study of the ASPV genome in Korea.

        Morphology and molecular phylogeny of harmful dinoflagellate Pseudocochlodinium profundisulcus from Korean coastal area

        The morphological features of germling cells were examined to identify an unspecified resting cyst (described as Cochlodinium cf. polykrikoides-like resting cyst) in the Korean coastal area. LSU rRNA gene sequences were also obtained from a strain established from the germling cells. The resting cysts isolated from Korean coastal sediment were characterized as being brown in color, having a large dark-red body, and fibrous lobed ornaments. The germling cells were ellipsoidal with an irregular outline and had an open comma-shaped ASC (apical structure complex), a wide and deep cingulum, and a deep sulcus. These morphological features were consistent with those of previously described harmful dinoflagellate Pseudocochlodinium profundisulcus. The molecular phylogeny revealed that the germling cells and P. profundisulcus were conspecific. Based on these morphological and phylogenetic data, this study documents the occurrence of P. profundisulcus in a Korean coastal area for the first time.

        고압산소와 물리치료의 복합 적용이 노인의 만성 요통 완화에 미치는 영향

        Purpose: To investigate the effects of hyperbaric oxygen therapy (HBOT) on chronic low back pain in elderly patients with impaired functioning in daily life. Methods: 35 subjects with chronic low back pain were randomly allocated into two groups, HBOT group and Placebo group. We applied TENS and mechanical massage to the lower back in both groups. Then 35% oxygen at 1.4 ATA was given to the HBOT group and atmospheric oxygen at 1 ATA to the placebo group. Results: Comparing the HBOT group with the placebo group after performing interventions six times in 2 weeks, a statistically significant difference in the Visual Analogue Scale (VAS) and Oswestry Disability Index (ODI) scores was observed between the two groups. There was a statistically significant difference in VAS and ODI scores (56% and 34.97% respectively) between the HBOT group and the placebo group. A statistically significant difference was ascertained in the degree of variables VAS and ODI in the HBOT group. Conclusion: Application of HBOT in chronic low back pain could be beneficial for reducing pain and could help in functional recovery.

      • 국내에서 1989년부터 2001년 사이에 세 번 돌발 유행한 홍역의 역학 및 임상적 비교

        목 적 : 1989~1990, 1993~94, 2000~01년 3차례 홍역 유행 시기의 홍역 환아들에서 역학 및 임상적 차이가 있는가를 알아보고자 하였다. 방 법 : 3차례의 홍역 유행시 입원한 환아 총 502명의 의무기록지를 후향적으로 분석하였다. 환아군을 유행 시기에 따라 I군(1989~90년, 116명), II군(1993~94년, 127명) 및 III군(2000~01년, 227명)으로 나눈 후 각 군간의 역학적, 임상적 특징에 대해 비교 분석하였다. 결 과 : 연령 분포에서 2세 미만의 비율은 각 군에서 각각 61%, 58% 및 57%로 통계학적으로 유의한 차이를 보이지 않았다. 그러나 2세 이상의 연령분포에서 I군의 2~5세 비율이 21%로, II군의 6~9세 비율이 28% 및 II군의 10세 이상 비율이 21%로 각 군의 같은 연령군에서 유의한 차이를 보였다. 남녀비, MMR 예방 접종률, 총 발열일 및 간염의 빈도에서는 3군간에서 차이를 보이지 않았다. 총 입원일(P=0.019)과 합병증 발생률(P=0.012)에서 III군에 비해 I군에서 유의한 차이를 보였다. 결 론 : 3차례의 연속적으로 유행한 홍역에서, MMR 접종을 받았으나 홍역에 이환된 경우도 있었고, 또한 발생 연령이 세월이 지나감에 따라 유의하게 증가하는 것이 관찰되었다. 이는 홍역 예방접종의 이차적 실패의 가능성도 있음을 시사하는 것으로 향후 지속적인 관심과 연구가 필요할 것으로 사료된다. Purpose : We evaluated and compared clinical and laboratory characteristics of patients with measles in three epidemics(1989~90, 1993~94, and 2000~01) in Daejeon, Korea. Methods : Retrospective analyses were performed using medical records of 520 patients with measles at the Catholic University of Korea, Daejeon St. Mary's Hospital during three epidemics. We divided the subjects into three groups, i.e., those who admitted during 1989~1990(group I, 116 patients), those during 1993~1994(group II, 127 patients), and those during 2000~2001(group III, 277 patients). We compared clinical, demographic and laboratory characteristics among these 3 groups. Results : In age distribution, ratios of under 2 years of age in three groups were 61%, 58% and 57%, respectively with no statistical differences. However there are differences in age distribution above 2 years of age, as 21% in 2~5 years of group I, 28% in 6~9 years of group II, and 21% above 10 years of group III. No statistical differences were present between 3 groups in the male to female ratio, MMR vaccination rate in above 2 years of age, duration of fever, incidence of hepatitis. Hospitalization days(P=0.019) and rate of complications(P=0.012) were longer and higher in group I than in group III. Conclusion : In three epidemics, the second peak age group(except 0~1 year) of children above 2 years of age who had mostly received MMR vaccination showed a trend for increased age with increasing time(statistical difference). This result suggest that secondary vaccine failure may have a role in each epidemics. So, if measles outbreaks is happened in the future, we will have consideration in this aspect.

        오이모자이크바이러스에 대한 무 육종 계통 저항성 평가

        In order to evaluate CMV resistance of the radish inbred lines, leaves of twenties radish (Raphanus sativus L.) inbred lines were mechanically inoculated with a strain of Cucumber mosaic virus (CMV) systematically infecting radish plants, respectively. The inoculated radish plants were incubated at 22°C±3°C and resistance assessment was examined using symptom development for 4 weeks. Based on the reactions of differential radish inbred lines, 11 radish lines were produced mosaic, mottling, yellowing and severe mosaic symptoms by CMV infection. These results were confirmed by RT-PCR analysis of CMV coat protein gene, suggesting that CMV is responsible for the disease symptoms. In contrast, 9 resistant radish lines did not induce systemic mosaic symptoms on upper leaves and chlorosis in stem tissues for 4 weeks, showing that the radish plants did not express any symptoms by 8 weeks. Further examination of CMV infection in the 9 resistance-candidate radish lines showed no CMV infection in systemic leaves of the 9 radish lines. Taken together, these results suggest that the 9 radish lines are highly resistant to CMV.

        건강장애 학생의 교육 참여와 부모의 자녀 교육 경험

        연구목적: 본 연구에서는 건강장애 학생과 부모의 교육 참여 경험을 바탕으로 이들의 어려움과 요구에 대해 살펴보고자 하였다. 연구방법: 연구 목적 달성을 위해 건강장애 학생 6명과 부모 6명에 대해 개별 심층면담의 방법으로 연구를 진행하였으며 이를 통해 수집된 자료를 바탕으로 귀납적으로 주제를 도출하는 전통적 내용분석방법을 적용하여 자료를 분석하였다. 연구결과: 건강장애 학생의 교육 참여 경험은 정서적 영역, 또래 관계 영역, 학습 및 학교생활 영역의 3가지 주제에서 7개의 하위 내용이 도출되었고, 건강장애 학생 부모의 교육 참여경험은 역할 수행 영역, 대인 관계 영역, 학습 및 학교생활 영역에서 총 7개의 하위 내용이 도출되었다. 건강장애 학생과 부모는 개인적 경험들을 바탕으로 교육 참여에 대한 다양한 어려움과 요구를 가지고 있었으며 학생과 부모의 경험에서 공통적인 경험들이 있기도 했고 강조되는 부분들에서 차이를 보이기도 했다. 또한 이러한 결과들은 매우 광범위하면서도 전체적으로는 유기적이며 상호적인 관계로 연결되어 있음을 알 수 있었다. 결론: 본 연구는 건강장애 학생과 부모의 직접적인 목소리를 통해 그들의 경험을 전달하고 이를 통해 교육 참여 과정에서 이들이 겪는 어려움과 요구를 탐색하고자 했다는 것에 의의가 있으며 추후 건강장애 학생과 관련된 질적 연구의 기초적인 역할을 할 것으로 기대한다. Purpose: The purpose of this study was to investigate the difficulties and needs of students with health impairment based on their experience of participating in education. Method: 6 students with health impairment and each of their mother was studied by means of individual in-depth interviews. The collected data were analyzed by applying traditional content analysis method. Results: First, the sub-contents of the students with health impairment were derived from the three subjects of emotional domain, peer relationship domain, learning and school life domain. From the three domains, a total of seven sub-contents were derived. Students with health impairment and their parents had a variety of difficulties and demands for participation in their education based on their personal experience. There were some common parts in the experiences of the students and the parents, and there were also different parts that were emphasized. Also, these results were very broad and overall, they are connected with organic and mutual relations. Conclusion: The results of this study were intended to convey their experiences through the direct voice of students with health impairment and their parents and to seek out the difficulties and needs that they experience during their participation in education.
