N/A Labeling Index on Mouse Epidermal Cells and the Effect of Ultraviolet Light Rediation Jai II Youn, M.D., Bang Soon Kim, M.D., Jin Ho Chung, M.D. Department of Dermatiology, College of Medicine, Seoul National University, Seoul, Korea We investigated the labeling index(LI) on mouse epidermal cells and the effect of ultraviolet light radiation(UVR). At one hour after injection of tritiated thymidine, UVB 100mJ/㎠ using a high pressure mercury arc was irradiated. Skin biopsy was done at 24 hours and 48 hours after UVR> Microautoradiography was done to check the labeling index. The results are summarized as follows, 1. LI on mouse epidermal cells was 5.4±1.7% after 24 hours, 2.6±1.5% after 48 hours. LI after 24 hours and 48 hours had decreased slightly but there was no difference between before and after UVR This result suggest that the changes in DNA synthesis had recovered by 24 hours after UVR. This recovery persisted at 48 hours after UVR radiation.

Background: We predicted that t.he self reporting questionnaire proposed by Fitzpatrick might not be appropriate for classifyin~v; the skin phoiotype in Korean people. Objective : We classified the skin phototypes in Korean people by measuring the minimal erythema dose (MED) of UVB on t.he basis of MEDs of skin type proposed by Pathak and Fitzpatrick. Methods : Two hundred and eighty-four male Korean rnedical students participated in this study. We irradiated UVB with Waldmann UV 800 and measured MEDs after 24 hours. Results : The mean MED was 70.1+21.3 mJ/cm2. Most of the MEDs were 70, 60, 50 mJ/cm in the order of frequency. On the basis of the skin type proposed by Pathak and Fitzpatrick, the most frequent phototype was the skin type V(56.3%) by MED. 20.8% and 13.8% of the subjects eorresponded to the skin types UV and III, respectively. 9.1% of the subjects fell into the UV sensitive group, skin type l and II. Conclusion : Using these results as groundwork, simple and precisely applicable classification of skin phototype in Korean people needs to be established. (Kor J Dermatol 1996;34(6): 893-897)

This study was undertaken to investigate the effect of superoxide dismutase(SOD) to sunburn cell production and development of UV-induced ear swelling reaction in mouse skin after ultraviolet irradiation. In this study, a total of 60 ICR female albino haired mice were used and divided into two groups, A(UVB: 150mJ/cm) and B(UVB:300mJ/cm). Groups of mice were injected intravenouly with SQD(300mJ,/1000cm) just befare UVR and after completion of UVR. The results were as follows . 1. The number of sunburn cells was significantly decreased by injection of SOD (300ug.1000ug)(p<0.05). 2. The number of sunburn cells in a group of mice which was given SOD 1000ug was significantly decreased more than in SOD 300pg(p<0.05). 3. Ear swelling reaction was not significantly suppressed by injection of SOD(300 Pa. 100SC)(p<0 05)

N/A We have investigated the effect of PUVA(8-methoxypsoralen and ultraviolet A) on the viability and DNA synthesis in human lymphocyte cultures after stimulation with phytohemagglutinin(PHA) using various concentrations of 8-MOF(0㎍/㎖, 0.01㎍/㎖, 0.1㎍/㎖, 1㎍/㎖) and doses of UVA radiations(0J/㎠, 0.1J/㎠, 0.5J/㎠, 1J/㎠). The results were as follows ; 1. There was a UVA dose-dependent decrease in the viability of lymphocytes at every 8-MOP concentrantion. In the 0.01㎍/㎖ 8-MOP treated group, there was a statistically significant decrease in viability from 0.5J/㎠ UVA in the 3 and 5 day cultures. In 0.1㎍/㎖ 8-MOP group, there was a significant decrease in culture viability from 2J/㎠ UVA at 1 day. 0.5J/㎠ UVA at 3 days, 0.1J/㎠ UVA at 5 days and in the 1㎍/㎖ 8-MOP group, from 1J/㎠ UAV at 1 day, 0.1J/㎠ UVA at 3, and 5 days. 2. There was a 8-MOP dose-dependent decrease in viability of lymphocytes. In the 0.1J/㎠ UVA exposed group, there was a statistically significant decrease in viability from 1㎍/㎖ 8-MOP in the 3 day old culture, and 0.1㎍/㎖ 8-MOP at 5 days. In the 0.5J/㎠, 1J/㎠ and 2J/㎠ UVA exposed groups, there was a significant decrease in viability from 0.1㎍/㎖ 8-MOP in the 3 and 5 day old cultures. 3. There was a UVA dose-dependent decrease in DNA synthesis after stimulation with PHA in lymphocytes. In the 0.01㎍/㎖ 8-MOP treated group, there was a significant decrease in DNA synthesis from 0.5J/㎠ UVA, and in 0.1㎍/㎖ 8-MOP, and 1㎍/㎖ 8-MOP groups, there was a decrease from 0.1J/㎠ UVA. 4. There was a 8-MOP dose-dependent decrease in DNA synthesis. In the 0.1J/㎠ UVA exposed group, there was a significant decrease in DNA synthesis from 0.1㎍/㎖ 8-MOP and in 0.5J/㎠, 1J/㎠, 2J/㎠ UVA groups, from 0.01㎍/㎖ 8-MOP.

N/A We have observed the dark effect of 8-methoxypsoralen(8-M(P) on the viability and DNA synthesis in human lymphocyte cultures after stimulaticn with phytohemagglutinin (PHA) in the absence of ultraviolet A radiation. The concentratioiis of 8-MOP was 0.5-32 ug/ml. We have also measured the LDH activity in supernatant. of lymphocyte cultures treated with 8-MOP. The results were as follows: 1. There was no 8-MOP dose-dependent decrease in the viability of lymphocytes up to 8- MOP 32pg/ml. 2. There was a 8-MOP dose-dependent decrease in PHA-induced DNA synthesis of lymphocytes from the concentration of 8-MOP 2 ug/ml. 3. There was a time-dependent decrease in PHA-induced DNA synthesis of lymphocytes at the concentration of 8-MOP 32 ug/ml. 4. There was no LDH release in supernatant of lymphocyte ciltu es after incubation with 8-MOP up to 8-MOP 32ug/ml. (Kor J Dermatol 992;30(5):601-607)

The allopurinol hypersensicivity syndrome is a rare, but life thereaning immunologic reaction of allopurinol therapy, characterized by multiple abnormalities such as fever, rash, decreased renal function, hepatocellular injury, leukocytosis, and eosinophila. It may require prolonged hospitalization and occasionally involves residual morbidity. Three patients developed erythematous skin eruption three to five weeks after beginning therapy with allopurinol. The clinical, laboratory, and histologic findings of these patients were compatible with a allopurinol hyperensitivity syndrome. (Kor J Dermatol 1995; 33(1): 130-134)

Background : The erythemal response and delayed tanning of the skin to UV radiations which are used as diagnostic phototest and guideline of phototherapy are different according to the races and light sources. Objective : The MED and MMD induced by UVB radiation were measured in 130 normal young adult Koreans. Methods : In this study, a high pressure mercury are lamp(Burdick UV-800) and a sunlight fluorescent lamp(Waldmann UV 800) were used as UVB light sources. Multiple sites of the lower baek or buttock skin were irradiated with an increasing dose by a constant, anount. The minimal doses of erythema response and delayed tanning of the skin were assessed visaually at 24 hours and 7 days after irradiation, respectively. Results : MED and MMD of Burdick UV-800 are 18.0+9.8mJ/cm(mean+S.D.), 29.7+12.5mJ/cm, respectively. MED and MMD of Waldmann UV 800 are 70.8+28.5mJ/cm and 91.2+33.1mJ/cm, respecively. The most frequent MED by Burdick UV-800 is 10mJ/cm and those by Waldmann UV 800 are 50m J/cm and 60m J/cm. Conclusion : In this study MED and MMD of young adult Koreans by two kinds of UVB light sources were assessed. The results : how that MED is less than MMD and the levels of MED under which the photosensitivity is suggested are 5.6mJ/cm by Burdick UV-800 anr 32.4mJ/cm by Waldmann UV 800. Background: There are few epidemiological studies on patch test with additional batteries in Korea. Objective : We report the results of patch tests with additional batteries which are relatively uncommonly used in contact dermatitis patients. Methods : Standard patch testing was performed with Finn chamber on Scanpor tape. We used additional batteries including cosmetic and fragrance series which are commonly used, and the others including 14 batteries(so-called other batteries). Results : Over the last 8 years, 719 patients(male 184, female 535) were tested in our contact, dermatitis clinic. 178 patients(24.8%) were tested with both Standard and other batteries, In other batteries, rubber additive series were most frequently used(9.2%), and textile colours finish(4.3%) and hairdressing series(3.6%) were used in order of frequency. Rarely performed batteries were(meth) acrylate ser ies, photographic chemicals series, wood set, oil & cooling fluid series, and various allergens. Patients tested with steroid batteries did not show positive reactions. Conclusion : We suggest that 6 batteries((meth)acrylate series, photographic chernicals series, wood set, oil & cooling fluid series, various allergens, steroid battery) have law testing frequencies and positive rates. A more organized multicenter study is required. (Kor J Dermatol 1997;35(2): 279-283)
