Ethanolic Extract from Rubus coreanus Enhanced Swimming Capacity in Mice

Swimming enhancement effects of ethanol extract from ripe Rubus coreanus (RCE) fruits were investigated in mice. An Ex-RCE group was orally administered 1 g/kg body weight/day of RCE. An Ex-Control group received distilled water for 28 days. Exhaustive swimming times were significantly (p<0.05) prolonged in Ex-RCE mice, compared with Ex-Control mice. Ex-RCE mice had lower lactate and higher glucose levels after exhaustive swimming. Plasma non-esterified fatty acid levels in Ex-RCE mice were significantly (p<0.05) increased during swimming, compared with Ex-Control. Lower plasma triglyceride levels were shown in Ex-RCE mice after swimming. Ex-RCE mice exhibited higher muscle glycogen and lower malondialdehyde levels, compared with Ex-Control mice. RCE is a useful agent for enhancement of the swimming capacity and amelioration of physical fatigue via a decrease in lactate accumulation, and supply of energy sources via glycogen sparing and facilitation of fat use, and reduction of lipid peroxidation levels through swimming.
Isolation of Antioxidant with Exercise Enhancing Potential from Pseudosasa japonica Leaves

An antioxidative compound with effective exercise-enhancing potential was isolated from the ethanolic extract of Pseudosasa japonica leaves. The ethanolic extract was partitioned in the order of hexane,chloroform, ethyl acetate, and water. Of the 4 fractions, the ethyl acetate-fraction (PJE-E) showed profound scavenging activity on superoxide anion- and ABTs cation radicals. Thin layer chromatography (TLC) and high performance liquid chromatography (HPLC) were conducted to isolate a compound with antioxidative activity on exercise endurance capacity from the PJE-E. The structure of the resulting compound was identified as ferulic acid by 1H/13C nuclear magnetic resonance (NMR). This ferulic acid was orally administered to mice for 18 days and its effect on exercise endurance capacity was investigated using an adjustablecurrent water pool. Compared to the control group, a 1.8fold increase in swimming time was observed in the ferulic acid-administered mice. Results indicate that ferulic acid from P. japonica leaves might contribute to the potent exercise-enhancing effect.

The protective activity of a mixture of aqueous and ethanolic extracts from Houttuynia cordata Thunb, Nelumbo nucifera G. leaves, and Camellia sinensis seed (HNC) was evaluated in C57BL/6 mice. Pretreatment with HNC prevented the elevation of serum aspartate aminotransferase and alanine aminotransferase caused by ethanol-induced hepatic damage. The HNC-treated mice showed significantly lower triglyceride levels, reduced CYP2E1 activity, and increased antioxidant enzyme activities and lipogenic mRNA levels. These results suggest that HNC might be a candidate agent for liver protection against ethanol-induced oxidative damage, through enhancement of antioxidant and antilipogenic activity.
동과 분획물이 3T3-L1 지방세포 분화 억제에 미치는 영향

본 연구에서 동과 물추출물의 계통분획을 통해 획득된 세가지 분획물인 핵산 분획(BHHH), 클로로포름 분획(BHHC), 에틸아세테이트 분획(BHHE)들을 3T3-L1 분화과정 중에 처리한 후, Oil Red O 염색법에 의한 lipid accumulation, 지방구내 triglyceride 함량을 평가하고, free glycerol release 함량과 adipogenesis와 관련된 transcription factor들의 발현 함량을 비교하여 동과 물추출물 중 anti-adipogenesis 활성 분획물을 밝히고, 이 분획물의 작용 메커니즘을 규명하고자 하였다. 50 ㎍/㎖ 농도의 BHHC와 BHHE의 처리는 분화된 지방세포 내 지질 축척을 11%와 13%로 낮추었다. 지방세포 내 중성지방(TG)의 함량은 동일 농도의 각 분획물에서 21%와 16%로 낮게 나타났다. TG 함량의 감소와 지방구내 지질 축적의 감소, 즉 anti-adipogenesis 메커니즘을 밝히기 위해 free glycerol 분비량을 평가하였다. 동일 농도의 BHHC와 BHHE에서 각각 13%와 17% 감소되어 나타났다. BHHC와 BHHE는 세포가 분화하는 동안 PPARγ, C/EBPα, leptin의 mRNA 발현을 억제하는 것으로 나타났다. 특히 BHHE의 경우 각 transcriptional factor들의 발현을 45%, 67%, 35%로 현저히 억제시키는 우수한 anti-adipogenetic 소재로 나타났다. 이에 BHHE는 항비만 기능성 소재로 활용될 수 있을 것으로 판단된다. The effects of three fractions, hexane (BHHH), chloroform (BHHC), and ethyl acetate (BHHE), from water extract of Benincasa hispidaon the underlying mechanisms of adipogenesis were investigated in 3T3-L1 cells. Intracellular lipid droplets were stained with Oil Red O dye and quantified. Compared to control, lipid accumulation significantly decreased by 11% and 13% upon treatment with BHHC and BHHE, respectively at a concentration of 50 ㎍/㎖. Intracellular triglyceride (TG) levels were also reduced by 21% and 16%, respectively, at the same concentration. To determine the mechanism behind the reductions in TG content and lipid accumulation, glycerol release and expression levels of adipogenic marker genes were measured. The levels of free glycerol released into culture medium increased by 13% and 17% upon treatment with BHHC and BHHE, respectively. In subsequent measurements using real-time polymerization chain reaction, the mRNA levels of PPARγ, C/EBPα, and leptin significantly decreased upon treatment with BHHE (45%, 67%, and 35%) in comparison with non-treated control. These results suggest that BHHE inhibits adipocyte differentiation by blocking PPARγ, C/EBPα, and leptin gene expression in 3T3-L1 cells, resulting in reduced lipid accumulation, increased glycerol release, and intracellular triglycerides.
