        재생불량성 빈혈의 병태생리에서 Fas 항원과 Apoptosis의 역할

        Background: Clinical observations and laboratory studies have supported an immune basis for most acquired aplastic anemias, with the majority of patients responding to immunosuppressive therapy. Fas, a member of the tumor necrosis factor (TNF) receptor superfamily is a critical downregulator of cellular immune responses. Proinflammatory cytokines like interferon gamma (IFN-${\gamma}$) and TNF-${\alpha}$ can induce Fas expression and render hematopoietic progenitor cells susceptible to Fas-induced growth suppression and apoptosis. Methods: In order to investigate the involvement of apoptosis in the pathogenesis of aplastic anemia (AA), we measured the expression of Fas antigen and caspase-3 on bone marrow (BM) mononuclear cells (MNCs) of AA in the presence or absence of IFN-${\gamma}$, TNF-${\alpha}$, or macrophage inflammatory protein 1-${\alpha}$ (MIP-$1{\alpha}$). Results: We confirmed that AA BM MNCs were more apoptotic and highly expressed Fas antigen than normal donors. Stimulation by IFN-${\gamma}$, TNF-${\alpha}$, or MIP-$1{\alpha}$ increased Fas antigen and caspase-3 expression in AA BM MNCs than BM MNCs of normal donors. Anti-Fas monoclonal antibody enhanced IFN-${\gamma}$, TNF-${\alpha}$, or MIP$1{\alpha}$ mediated caspase-3 expression in BM MNCs of normal donors. Among these three cytokines, IFN-${\gamma}$ enhanced apoptosis most strongly via Fas-caspase-3 pathway. Conclusion: These results suggest that Fas signal pathway may play a role in the pathophysiology of aplastic anemia and negative hematopoietic regulators like IFN-${\gamma}$ can induce apoptosis of bone marrow progenitors in part by Fas induction.

        초정밀가공기를 이용한 SIL 렌즈의 절삭특성

        The aspherical lenses are used as objective lens of optical pickup. To examine the design factor the sample product is made before manufacturing of injection mould of lens. The optimum cutting condition of PMMA lens sample with ultra precision SPDT, the roam spindle speed, the depth of cut, the feedrate are found. The demanded surface roughness 100m Ra, aspherical form error $0.5{\mu}m$ P-V for aspherical lens of optical data storage device are satisfied.

        동해남부연안 해양환경특성 시공간적 변화

        동해남부연안에서 해수 중 해양환경특성의 시공간적 분포양상을 살펴보기 위해서 2012년 5월부터 2013년 2월까지 계절별로 현장조사를 실시하였다. 표층수 중 영양염류(용존무기질소, 용존무기인, 용존무기규소)의 농도는 봄과 여름에 비해 수직혼합이 활발하게 일어나는 가을과 겨울에 높게 나타났다. 여름에 chlorophyll a 농도가 높게 나타나 이 시기에 낮은 영양염 농도는 밀도약층의 강화로 인한 아표층으로부터 영양염류의 공급 감소와 식물플랑크톤의 광합성에 의한 소비로 판단된다. 반면 봄에는 chlorophyll a 농도가 낮게 나타나 봄에 표층수 중 낮은 영양염 농도는 육상 및 아표층으로부터 영양염류의 공급 감소에 의한 것으로 판단된다. 조사기간 동안 표층수와 저층수 중 용존무기질소와 용존무기인의 비는 각각 연평균 15.6, 14.8로 유사하였으나, 표준편차는 각각 13.6, 4.2로 표층수가 저층수에 비해 상대적으로 크게 나타났다. 특히, 봄에 용존무기질소에 대한 용존무기인의 비가 상대적으로 낮게 나타나(평균 $8.35{\pm}4.67$) 이 시기에 용존무기질소가 식물플랑크톤의 성장에 제한인자로써 작용했을 것으로 판단된다. In order to elucidate the spatiotemporal variations of marine environmental parameters, we collected seawater samples in the south-western region of the East Sea in May, August, and November 2012 and February 2013. The concentrations of dissolved inorganic nutrients (dissolved inorganic nitrogen, phosphorus, and silicate) in surface seawater during the summer season were lower than those during autumn and winter seasons, which the mixed layer is deeper. The low nutrient concentration in spring and summer seasons seems by consumption of dissolved inorganic nutrients by phytoplankon photosynthesis (high chlorophyll a concentration) and the limited supply of dissolved inorganic nutrients from subsurface layer having high nutrients. The low nutrient concentration during spring season seems to be related to the limited supply of dissolved inorganic nutrients from land and subsurface layer because the concentration of chlorophyll a was low. The DIN:DIP ratio was a wide range of average $15.6{\pm}13.6$ in the surface seawater compared to that of average $14.8{\pm}4.2$ in the bottom seawater during sampling periods. The dissolved inorganic nitrogen might act as a limiting factor of the growth of phytoplankton because the DIN:DIP ratio (on average $8.35{\pm}4.67$) was low during the spring season.

        적외선용 광학소자의 초정밀 절삭특성

        Machining technique for optical crystals with single point diamond turning tool is reported in tills paper. The main factors influencing the machined surface quality are discovered and regularities of machining process are drawn. Optical crystals have found more and more important applications in the field of modern optics. Optical crystals are mostly brittle materials of poor machinability The traditional machining method is polishing which has many shortcomings such as low production efficiency, poor ability to be automatically controlled and edge effect of the workpiece. SPDT has been widely used in manufacturing optical reflectors of non-ferrous metals such as aluminum and copper which are easy to be machined for their proper ductility. But optical crystals being discussed here are characterized by their high brittleness which makes it difficult to obtain high quality optical surfaces on them. The purpose of our research is to find the optimum machining conditions for ductile cutting of optical crystals and apply the SPDT technique to the manufacturing of ultra precision optical components of brittle materials. As a result, the cutting force is steady, the cutting force range is 0.05-0.08N. The surface roughness is good when spindle is above 1400rpm, and feed rate is small. The influence of depth of cut is very small.

        악성 종양 , 만성 염증성 질환 , 및 철 결핍성빈혈 환자에서의 혈중 Erythropoietin 및 Tumor Necrosis Factor α

        N/A Objectives: The aim of this study was to determine the serum erythropoietin (S-EPO) level in anemia of chronic disorders (ACD) and the influence of tumor necrosis factor α (S-TNFα) to S-EPO. We also investigated the relationship between S-EPO and S-TNFα levels and other factors, such as fever and consumptive syndrome. Methods: Serum erythropoietin (S-EPO) and tumor necrosis factor alpha (S-TNFα) levels were measured by radioimmunoassay in 35 patients with anemia, classified into three groups according ta their etiology: neoplastic (n=17), chronic inflammatory (n=9), iron deficiency (n=9), and 10 normal controls. Resultr: The iron deficiency anemia group showed a higher mean S-EPO level (mean value±SEM, 229,05±50.50mU/ml) than the neoplastic (25.09±3.85 mU/ml; p<0.0001), chronic inflammatory (24.38±4.98 mU/ml; p<0.0001), and normal control groups (11.96±0.99 mU/ml; p<0.0001). The neoplastic group showed a higher mean S-TNFα level (6.22±0.33 fmol/ml) than the iron deficiency (3.26±0.18 fmol/ml; p<0.0001) and normal control groups (4.06±0.12 fmol/ml; p<0.0001). The chronic inflammatory disorder group showed a higher mean S-TNFα level (5.39α0.52 fmol/ml) than the iron deficiency anemia group (p<0.0001). There were no significant differences of S-EPO and S - TNFα Level of ACD group in febrile subjects (9 patients with fever and 17 patients without fever) and in weight loss (10 patients with weight loss and 16 patients without weight loss). Conclusion: These data suggested that the cause of ACD is thought to be the inappropriate erythropoiesis and TNFα may preferentially inhibits erythropoiesis.

        골수이형성증에서 장기골수배양과 In Situ Nick End Labeling 법을 이용한 조혈장애와 아포토시스에 관한 연구

        N/A Objectives: The myelodysplastic syndromes (MDS) are a group of acquired clonal hematopoietic disorders characterized by peripheral cytopenias and a hypercellular or normocellular dysplastic bone marrow. The mechanisms responsible for development of MDS are not known. We performed this study to evaluate the hematopoietic abnormalities and apoptosis in MDS. Methods: Long-term bone marrow culture (LTBMC) was performed for colony assays, cobblestone area assay, stromal morphologic changes from 7 patients with MDS and 7 normal controls. In situ nick end labeling (ISNEL) method was performed for detection of apoptosis from LTBMC in 7 patients with MDS and 7 normal controls. ISNEL method also performed in bone marrow cell bloc samples in 36 patients with MI3S. Results: Viability of nonadherent cells from LTBMC of patients with MDS was not decreased compared with normal controls at 1 week, but significantly decreased at 2 and 3 weeks compared with normal controls (P<0.0001). Formation of the cobblestone areas from patients with MDS was slightly decreased compared with normal controls at 1st week, but significantly decreased at 2nd and 3rd weeks compared with normal controls (P<0.0001). Slightly decreased compared with normal controls at 1 week, but significantly decreased at 2 and 3 weeks compared with normal controls (P<0.0001). Stromal layers produced in LTBMC of normal controls and 1 patient with MDS were detected at 1 week and were formed confluent stroma from 3 weeks, but another patients with MDS who did not form a confluent stroma. Patients with MDS had significantly lower colony forming unit granulocyte-macrophage (CFU-GM) compared with normal controls at 1 (P<0.01) and 2 weeks (P<0.001) of LTBMC. Two weeks of LTBMC resulted more profound inhibition of CFU-GM formation than 1 week (P<0.0001). Apoptotic cell death was absent in adherent and non adherent cells from normal controls at 1 and 2 weeks, but massive apoptotic cell death was found in adherent and non adherent cells from patients with MDS at 1 and 2 weeks and the degree of apoptosis was profound at 2 weeks compared with 1 week. Among the 36 patients, fifteen patients demonstrated varying degrees of apoptosis positive cells, 4 having low, 8 intermediates, and 3 high scores. Remaining 21 patients showed absent apoptosis or only occasional positive cells. Conclusion: Hematopoietic abnormalities such as a failure of differentiation are caused by the stromal defects and the biologic basis of the apparent paradox of peripheral cytopenias in the face of hypercellular (or normocellular) marrow is related by intramedullary apoptotic cell death of the stromal and hematopoietic cells.

        A study on the use of various additives to polymer-based solid electrolytes for all-solid-state batteries

        Lithium-ion batteries are currently widely used secondary batteries, but as the use environment becomes harsher and larger, there is a problem in stability, and it is believed that the possibility of developing energy density is limited. Therefore, there is a need for an all-solid-state battery that can solve this problem by replacing the conventional liquid electrolyte to the solid-state electrolyte. In particular, the polymer-based solid electrolyte has been treated as the promising candidate for all-solid-state batteries due to the excellent mechanical properties and inexpensive fabrication process. However, overcoming low ion conductivity is an inevitable issue for enabling the practical application of polymer-based electrolyte. To compensate the limitation of ion conduction in polymer layers, the strategies of introducing the additives in polymers have been suggested through the hybridization and functionalization of polymers for enhancing the ion transportation. In this review, the recent research progress of various type of additives for polymer electrolyte is presented and the active/passive/polymer type additives are studied to obtain the design principle dealing with the improvement of ionic conductivity and properties of polymer solid electrolytes.
