Immunogenicity of Escherichia coli pilus and LT were evaluated in 20-week-old hens. The antigens were consisted of K88, K99, 987p pilus and heat labile toxin purified from enterotoxigenic Escherichia coli. The durations of antibody titers in sera and egg yolk were investigated by an enzyme-linked immunosorbent assay(ELISA). After first inoculation, antibody titers in sera reached at peak 2 weeks postinoculation. However, peak antibody titers in egg yolk were detected 4 weeks postinoculation, indicating that transfer of immunoglobulin from serum to egg yolk took about two weeks period. Although there were slight reduction in titers, the specific antibodies in egg yolk lasted up to 3 months. Immune responses against monovalent and combined antigens were showed as almost same patterns. The transfer rate of antibodies from serum to egg yolk didn't show any significant differences among three pilus antigens in this study. Considering the concentrations of antigens in each inoculated group, multivalent antigens containing heat labile toxin of E coli were found to be more immunogenic than monovalent antigen in producing specific antibodies. From this experiment, it was demonstrated that multivalent antigens containing three pilus and heat labile toxin could be a promising candidate for the production of egg yolk antibodies for prophylactic use in preventing swine colibacillosis in future.

The present study describes the effectiveness of egg yolk antibodies (IgY) against enteric colibacillosis and edema disease in piglets. The antibodies were gained from the egg yolk of hens immunized with k88, k99, 987p fimbrial adhesin and heat-labile toxin antigens of enterotoxigenic Escherichia coli (ETEC). Orally-administered egg yolk antibodies solution protected against experimental challenge with ETEC $K88^+$ and $k99^+$ strains in neonatal piglets and mice. In field trial, a total of 598 diarrheal piglets were orally treated with 3ml of antibody once a day to determine for the therapeutic effect. Of them, 582 (97.3%) piglets were recovered from diarrhea in 3 days. We also experimentally treated with the egg yolk antibodies twice a day for 5 consecutive days for 94 weaning piglets with edema disease for the determination of therapeutic effects. Seventy four piglets (78.7%) were recovered from clinical edema signs. Theses findings indicate that egg yolk antibodies against k88, k99, 987p and LT of ETEC are useful source of passive immunity for enteric colibacillosis and edema disease of piglets.

The safety of Brucella abortus strain RB51(SRB51) was investigated in dairy cows and Korean native cattle of 4~7th month of gestation. From experimentally inoculated cattle, 18 of 25 (72.0%) dairy cows, and 3 of 10 (30.0%) Korean native cattle were aborted or delivered premature fetus. There were no significant differences in the incidence of abortion depending on the inoculation route (intramuscular and subcutaneous) and dosages ($1{\times}10^9$, $2.8{\times}10^9$, and $4.0{\times}10^9$ CFU). The antibodies to the SRB51 were measured by a dot blot enzyme-linked immunosorbent assay. The highest titers to SRB51 were detected between 5~7 weeks after inoculation and the specitic antibody could be detected up to 28 weeks after inoculation. The SRB51 was isolated from amnio-allantoic fluid, bronchial lymph node, mammary gland, and supramammary lymph node in 5 of 25 dairy cows during 4 weeks after either abortion or delivery. Although SRB51 was isolated from 4 of 24 aborted fetus or normally delivered calves at parturition time, it was not isolated during 4 weeks afterward. Eleven of twentyfive dairy cows showed the endometritis and/or necrosis until 6 weeks after delivery, no lesions were seen at 8 weeks after delivery and uterus from control dairy cows. The results of present study revealed that SRB51 might induce the clinical signs of brucellosis in the pregnant cattle at 4~7th month of gestation.
부루세라백신(RB51)의 안전성에 관한 연구 I. Brucella abortus RB51 백신균주의 생화학적 및 유전학적 성상비교

Biochemical and genetic analysis were carried out to investigate the potential recovery of pathogenecity or related mutations of Brucella abortus RB51 vaccine strains. RB51 strains were recovered from commercial vaccines, including related seed stocks from private companies in Republic of Korea, strain from USA, a reference strain from C university and a field isolate (Daehungjin) from aborted dairy cow after RB51 vaccination were compared with two identified virulent wild strains (S2308 and a field strain isolated from dairy cow in Korea) at the same conditions. All the strains examined, except identified pathogenic strains, revealed the identical characteristics to the original RB51 in biochemical properties, antigen and bacteriophage typing. Outer membrane protein (OMP) profiles from strains of RB51 showed the same patterns with standard RB51 in SDS-PAGE. In addition, Western blotting with the brucella specific monoclonal antibody also indicated that all the vaccine strains were completely deficient in their LPS compared to the pathogenic Br abortus strains. The differences in DNA structures among strains were also possible to detect after PCR. All vaccine strains, except S19, S1119-3, S1075, S544 and Br suis, were amplified a 178bp DNA fragment of eri-gene, and 364bp of IS711 elements. In contrast, 498bp DNA product was only found with Br abortus. Overall evidences in the present study confirmed that the RB51 strains for vaccine production in Korea did not originated from the phenomena of possible recovery of pathogenicity or related to any potential mutation event at all.

The pathogenicity of Br abortus RB51 strains, producted by commercial vaccine companies in Republic of Korea and USA, were evaluated in mouse and guinea pig. BALB/c and ICR mice were intraperitoneally inoculated with RB51 vaccines or virulent field strain and the existence of RB51 including its ratio of spleen to weights and persistence in spleens were examined. Groups of guinea pigs on day 55-58 of received subcutaneously with various RB51 vaccines, RB51 field isolates (Daehungjin) or virulent field isolates(Sangju) to compare the histopathogenicity in uterus. All the mice received RB51 vaccines or RB51 field isolates survived for 10 days, but the groups of mice received virulent field isolates died 5 from 11 (45.5%) in case of BALB/c mice and 12 from 12 (100%) in ICR mice, respectively. The number of RB51 in the groups of mice given with vaccine strains and RB51 field isolates were declined rapidly were in spleens between 12 and 20 days after inoculation. In contrast the mice given with the virulent field isolates rose in number of bacteria up to 20 days after inoculation. In the groups of mice infected with virulent field isolates, the ratio of spleen weights to body weight were significantly higher than those in control or in the groups inoculated with RB51 strain, including RB51 field isolates, at 12 and 20 days after inoculation. At ten days after inoculation, placentas of both the pregnant and non-pregnant guinea pigs were conducted for histopathological examination. Although any abnormal lesions were not observed in non-pregnant guinea pigs, all the strains caused the inflammation of the placenta, implying pathogenecity of RB51 in pregnant guinea pigs.

In October 2005, ROK Army established the Army's Force Structure Reforming Program focusing on simplifying force structure, and unit mobilization and applying IT technique in the framework of Defense Reforming 2020. The needs of verifying and validating the program with logical and scientific way were increased. This article detailedly shows the concept, procedure and contents of Total Army Analysis(TAA) conducted by the ROK Army's Analysis and Evaluation Group from Oct 2005 to July 2006 and suggests major outcome and further study required. The result of TAA could be referred and used when reforming force structure to prepare for the future change in international defense environment and suggesting force requirements. The integrated analysis system suggested in this article is expected to contribute to the effective allocation and usage of the limited defense budget and the establishment of an optimal force structure ’05년 10월 육군에서는 ‘국방개혁안’과 연계하여 군 구조의 단순화, 기동화, 첨단 정보화에 초점을 두고 ‘육군 전력구조 혁신안’을 수립하였으나, 이에 대한 논리적이고 타당성 있는 검증이 제한되었다. 따라서 본고에서는 이러한 제한사항을 극복하기 위해 실시한 “총전력 분석”의 수행개념과 절차, 분석 내용 및 방법 등을 상세히 소개하고, 주요 성과 및 향후 발전방향 등을 제시하였다. 이러한 “총전력 분석” 결과는 향후 주변국과의 관계변화에 따른 전력구조 변경시 이를 실증적으로 검증하거나, 전력증강을 위한 소요요청시 유용하게 활용될 수 있을 것이며, 발전방향에 제시된 종합적인 분석체계가 구축된다면 제한된 국방재원의 효율적 배분 및 전력구조 완전성 달성에 크게 기여할 수 있을 것이다.
