Pink Rot on melon and White Stain Symptom on grape are caused by Trichothecium roseum, one of the most important diseases of grape and melon. These diseases have been occurred in national-wide in Korea and causes irreversible damage on the grape and the melon at harvest season. This research presents the evaluation of the capacity of Bacillus subtillis HK2 to protect both melon and grape against T. reseum and establishes its role as a biocontrol agent. In this study, we isolated a Bacillus strain HK2 from rhizosphere soil, identified it as Bacillus subtillis by 16S rRNA analysis and demonstrated its antifungal activity against T. roseum. Under I-plate assay it was observed that the effect of hyphal growth inhibition was not due to production of volatile compounds. The optimum culture condition of HK2 was found at 30℃ and initial pH of 7.0. Application of HK2 culture suspension reduced 90.2% of white stain symptom on grape as compared to control, resulting in greater protection to grape against T. roseum infestation. Butanol extract of HK2 culture purified using flash column chromatography. The antifungal material was a polar substance as it showed antifungal activity in polar elute. Therefore, our results indicated a clear potential of B. subtilis HK2 to be used for biocontrol of Pink rot in melon and white stain symptom on grape caused by T. roseum. Key words - Bacillus subtillis, Grape, Melon, Pink rot, Trichothecium roseum, White stain symptom 적 요2012년부터 2013년에 걸쳐 식물체 근권과 비근권 토양을 수집하였다. 수집한 토양을 희석평판법을 이용하여 총 782개의세균을 분리하였고, 분리한 세균을 실험실내에서 Trichothecium roseum과의 대치배양을 통해 균사생장억제율이 80% 내외로우수한 길항력을 나타내는 균주를 선발하였다. I-plate에서HK2균주와 T. roseum 두 균을 격리하여 밀폐배양 시 모두 균사생장억제 효과는 없기 때문에 휘발 물질은 아닌 것으로 판단되었다. NB배지에 길항균주와 T. roseum을 액체배양 한 결과88% 이상 균사생장억제 효과를 보였다. HK2균주를 동정하기위해 16S rDNA 염기서열분석과 API 50 CHB Kit (BioMerieux, France)를 이용하여 생화학적 특성을 분석한 결과 Bacillus subtilis로 동정되었다. HK2 균주가 생산한 항진균물질을 butanol 로 추출한 후 flash column chromatography를 이용하여 항진균물질을 정제한 결과 methanol 80%의 조건에서 잘 분리되었으며 향후 분리 순화를 통한 화학구조분석이 필요하다.

A total of 65 isolates of Penicillium were isolated from decayed post-harvest fruits of apple pear citrus and grape. The Penicillium species isolated from the apple were idnetified as P. aurantiogriseum and P. expansum those from the pear were P. crustosum and P. expansum and those from the grape were P. aurantiogriseum and P. expansum, From decayed citrus fruits. P. digitatum and P. italicum were isolated. Citrinin and patulin from these species in the YES(yeast extract sucrose) broth were extracted with ethyl acetate and purified by thin-layer chromatography(TLC) and high performance liquid chromatography(HPLC) Among 51 isolates of Penicillium from apple pear and grape 7 isolates produced citrinin 13 isolates produces patulin and 12 isolates produced citrinin and patulin also. All 14 isolates of Penicillium from citrus produced only patulin.

A sex cord tumor with annnular tubules is a relatively rare ovarian neoplasm. The cytologic findings from a fine needle aspiration biopsy of neck metastasis of a sex cord tumor with annnular tubules are described. The origin of the neck metastasis was the right ovary, and the tumor was diagnosed six years ago. The cytologic findings were characterized by tumor cells arranged in solid or follicular patterns. The tumor cells formed rosette-like or complex tubular structures with central rounded or coalesced hyaline materials. It was difficult to distinguish this tumor cytologically from granulosa cell tumor, thyroid follicular neoplasm, Sertoli-Leydig cell tumor, and Brenner tumor, but complex tubular structures were helpful in discriminating between these tumors.

A few authors have already investigated microbial contamination of leafy vegetables in distribution condition and examined the effect of temperature abuse on microbial safety or product quality. But this study analyzed proliferation of indicator microorganisms and food poisoning bacteria in real situation in Korean agroindustry and investigated washing effect of micobial contamination on leaf surface. Leafy vegetables were found to be contaminated with aerobic bacteria at 0.1~0.32 × 103 CFU/g. Staphylococcus aureus and Bacillus cereus were detected in leafy vegetables at 0~0.54 × 103 CFU/g. However, Salmonella spp., Coliforms, Clostridium spp. and Listeria spp. were not detected in any samples. Indicator microorganisms and food poisoning bacteria on leafy vegetables increased at room temperature when simulatively distributed condition. After clean-up, the indicator microorganisms and food poisoning bacteria of kimchi cabbage, lettuce and perilla leaf were decreased to a very low level. This study suggests that it is necessary to improve the level of hygiene management such as use of cold chain system and hygiene management of transport tools during the distribution process for fresh leafy vegetables.
재배농법에 따른 국내산 배추의 위해미생물 및 중금속 오염평가

Kimchi cabbage is one of the four major vegetable crops in Korea. The total annual production of kimchi cabbage, the main material of kimchi, was 20,559 tons in 2015. Kimchi cabbage is one of the majer crops produced by farmers which accounts for about 80% of the total leaf vegetable production in Korea. As the consumption of environmental-friendly agricultural products increases, food safety is one of the major public health concerns. We analyzed the biological hazards of kimchi cabbage produced by two types of cultivation methos such as organic farming and conventional farming using various culture media and microscopy. A total of 432 samples were analysed for presence of sanitary indicator microorganisms (aerobic plate count, coliform count, yeast & mold) and food-borne pathogens (Staphylococcus aureus, environmental Listeria, Bacillus cereus). The population of sanitary indicating microorganisms and food borne pathogens was under 5 Log CFU/g in all tested samples. The results of total microorganism numbers of leaf surface showed a positive correlation to those of soil samples. Additionally, we examined chemical factors such as pesticide residues and heavy metals in soil samples. All tested samples did not shown contamination levels higher than the standard limit.

This study was conducted to evaluate the microbiological safety of leafy vegetables (perilla leaf and lettuce) in relation to cultivation methods. A total of 2,304 samples were collected from plants, harvesting tools and soil mulching film during the production and harvest stages from organic- and conventional- farms. From the samples, sanitary indicator microorganisms (total aerobic bacteria, coliforms, E. coli., Environmental Listeria, and yeast and mold) and pathogenic microorganisms (S. aureus, B. cereus, Salmonella spp., Clostridium spp., and L. monocytogenes) were analyzed. In the production stage of leafy vegetables, the sanitary indicator microorganisms was not detected regardless of cultivation method or it was detected to be less than 3.4 Log CFU/100 cm2. B. cereus and S. aureus were found to be 0.22~1.55 Log CFU/g in perilla leaf and lettuce produced by organic farms, and S. aureus was not detected and B. cereus was found to be 0.42~2.19 Log CFU/g in conventional farms. There were no significant differences between two cultivation methods. In the harvesting tools and soil mulching film, the contamination levels of sanitary indicator microorganisms and pathogenic microorganisms was low regardless of the cultivation method. However, there was a positive correlation (R2 = 0.4526) in that the higher the microbial contamination level in the harvesting tool, the higher the microbial contamination on the surface of the plant. In addition, sanitary indicator microorganisms and pathogenic microorganisms were not detected or low in soil mulching during the production of organic leafy vegetables. As a result of this study, microbial hygiene control by soil mulching and harvesting tools was more important than difference of cultivation method in production of leafy vegetables.
