        • Flavobacterium multivorum이 생산하는 덱스트란 분해효소의 특성에 관한 연구

          Characteristics of dextranase and cloning of the gene of Flavobacterium multivorum which was isolated from soil were studied. Optimum condition for the enzyme activity appeared to be at the 1% concentration of dextran. When NH4NO3ㆍNH4H2PO4 and beef extract was added to the culture media. highest activity of the enzyme was found. In the presence of Mg2+ in culture media, activity of dextranase was somewhat higher, but was inhibited Ca2+. Co2+, Hg2+ and Zn2+. The optimum temperature for the enzyme activity was 45-55℃. In the range of pH 4 to 10, activity of the enzyme appeared approximately similar.

          돌산갓의 부위별 sinigrin 함량

          돌산갓에서의 glucosinolates인 sinigrin의 분석법을 확립하기 위해 다양한 용매를 이용하여 추출 후 정성·정량하였다. 돌산갓을 뿌리, 줄기, 잎으로 구분하여 50% CH3CN, 10% NH4Cl, 60% CH2OH, 70% CH3OH 을 이용해 추출 시 50% CH3CN이 가장 sinigrin 함량이 높게 나타났으며 나머지 용매들은 큰 차이가 없었다. 또한 갓 부위별에서 sinigrin함량은 큰 차이가 없었으며 줄기 부위의 50% CH3CN추출물의 sinigrin함량이 728 ppm으로 다소 높게 나타났다. Sinigrin은 농도별로 standard curve를 작성하여 HPLC에 의한 돌산갓 부위별 sinigrin 함량을 정량화 하였고 HPLC로 분석시 UV detector 감도는 242 nm보다 228 nm에서 높게 나타났으며 228 nm에서는 줄기, 뿌리, 잎 순서로 sinigrin함량이 나타났다. 50% CH3CN로 추출한 돌산갓의 chromatogram은 표준물질인 sinigrin의 retention time과 일치하였으며, 잎에서 나타난 peak는 glucosinolates인 glucoraphanin으로 확인되었다. 본 연구의 결과로부터 돌산갓에 함유된 sinigrin 분석을 위한 추출용매는 50% CH3CN, UV detector 파장은 228 nm가 가장 적합하다는 것을 알 수 있었다. 또한, sinigrin의 추출 및 분석이 체계화됨으로써 돌산갓을 이용한 다양한 식품의 저장기간에 따른 sinigrin 함량변화 및 생리기능에 관한 연구에 활용 될 수 있을 것으로 판단된다. The aims of this study was to optimize the extraction conditions of sinigrin from Dolsan leaf mustard. Dolsan leaf mustard (Dolsan-eup, Yeosu-si) harvested during at May 2014 was used for sinigrin extraction. After the extraction of sinigrin using 50% CH3CN, 10% NH4Cl, 60% CH2OH, and 70% CH3OH, the sinigrin content was measured by HPLC analysis. The results showed that sinigrin content was highest with 50% CH3CN solvent extraction and UV detector sensitivity was greater at 228 nm rather than at 242 nm. The sinigrin concentrations of leaf, stem and root with 50% CH3CN extraction were 345 ppm, 728 ppm, and 539 ppm, respectively. After extraction of the different parts of Dolsan leaf mustard, The standard retention time by HPLC analysis of sinigrin content was 2.054, 2.032, 2.059, and 2.035 min from the root, stalk, and leaf, respectively. On the other hand, HPLC analysis showed that the leaf extracts contained glucoraphanin, one of glucosinolates. The optimum time and extraction solvent for the sinigrin extraction from Dolsan leaf mustard was found to be 24 hr with 50% CH3CN solvent. In addition, opotimum UV detector k at 228 nm. These results showed that the optimum extraction conditions for Dolsan leaf mustard were 24 hr extraction with 50% CH3CN solvent. In addition, the optimum wavelength of UV detector was determined to be 228 nm for sinigrin analysis. Therefore, this study could provide a useful information for sinigrin extraction and its systematic analysis during the storage.

          염색체 이상 환자의 세포 유전학적 연구

          Presented in this paper the data from a chromosome study of 397 patients referred for suspected chromosmal abnormalities. Karyotypes were obtained using short-term blood culture and direct method. Of these 238 patients had normal chromosome complements; 159 (40.1%) patients had chromosome abnormality. Among all patients with chromosome abnormalities, 82.4% (131/159) had aberrations of chromosome number, the others 17.60/0 (28/159\ had aberrations of chromosome structure. Ten had a chromosome rearrangement; Five of them were reciprocal and five Robertsonian translocations. Four patients with pericentric inversions and one with paracentric inversions and four with isochromosomes were observed. There were four patients with marker chromosome, two patients had a chromosome insertion; and three others. (additional abnormal chromosomes.) Thus the results of the present study indicate the importance of cytogenetic evaluation in clinically abnormal patients.

          Quality Characteristics of Mackerel Immersed in Sea Wind Mugwort Extract

          거문도 해풍쑥 추출물을 처리한 고등어의 저장 기간 동안의 품질 특성을 조사하였다. 5% 해풍쑥 추출물에 고등어를 2시간, 3시간, 4시간 처리한 후 개별 포장하여 $-20^{\circ}C$에 저장하여 실험하였다. 해풍쑥 추출물을 처리하지 않은 고등어(control)와 해풍쑥에 2, 3, 4시간씩 처리한 고등어의 염도는 2시간 처리한 고등어가 0.07%로 가장 낮은 수치를 나타냈다. pH는 모두 5.90~6.23를 나타냈고 산도는 15일째 감소 후 증가하는 경향을 나타냈다. 또한, 저장기간이 길수록 인장강도는 control은 증가하고, 2, 3, 4시간씩 처리한 고등어는 감소하였다. Volatile basic nitrogen(VBN) 함량은 control은 4.2~50.7 mg%, 2시간 처리한 고등어는 5.6~15.4 mg%로 control보다 가장 낮은 수치를 나타냈다. 2시간 처리한 고등어 제품에서 total polyphenol, total flavonoid 함량은 286.3~497.0 mg GAE/100 g, 177.5~385.6 mg QE/100 g를 나타냈다. DPPH, ABTS radical scavenging activity은 2시간 처리한 고등어 제품에서 50.6%, 61.3%를 각각 나타냈다. 전반적으로, 고등어를 SWM으로 단시간 처리하면 염도, pH, 산도, 경도, 항산화 활성 및 저장기간 연장과 같은 품질특성이 크게 향상되었음을 알 수 있다. This study examined the influence of sea wind mugwort extract treatment on quality characteristics of mackerel during storage. Mackerel were packaged individually and then immersed in 5% sea wind mugwort extract for 2, 3, or 4 hr and stored at $-20^{\circ}C$. The salinity of a control (no treatment) and that of mackerel immersed in sea wind mugwort for 2 hr was lowest (0.07%). pH of 5.90-6.23, and the change in acidity was in the opposite range. Immersion for 2, 3, and 4 hr led to a decrease in the tensile strength of the samples following storage, whereas the tensile strength of the control increased. The volatile basic nitrogen (VBN) content of the mackerel immersed for 2 hr was significantly lower than that of the control (5.6-15.4 mg% vs. 4.2-50.7 mg%). In the mackerel immersed for 2 hr, the total polyphenol and total flavonoid content was 286.3-497.0 mg gallic acid equivalents (GAE)/100 g and 177.5-385.6 mg quercetin equivalents (QE)/100 g, respectively. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2-azino-bis(3-ethyl-benzothizoline-6-sulfonic acid) (ABTS) radical scavenging activities of the mackerel immersed in sea wind mugwort for 2 hr were 50.6% and 61.3%, respectively. Overall, the immersion of mackerel in sea wind mugwort for a short time significantly improved quality characteristics, such as salinity, pH, acidity, hardness, antioxidant activity, and perceptible quality, following storage.

          Changes in Glucosinolate Content of Dolsan Leaf Mustard Kimchi during Fermentation and Correlation with Antioxidant, Antihypertensive, and Antidiabetic Activities

          The glucosinolate content, antioxidant activity, and antihypertensive and antidiabetic activities were measured in a crude extract of Dolsan leaf mustard kimchi (DLMK). The glucosinolate content was low at 6.41 and 7.92 mg/g in leaves and stems of DLMK after 21 days of fermentation. The total polyphenol and total flavonoid contents were more than 2 times higher in the leaves (211.7 mg GAE/g, 158.8 mg QE/g) than in the stem (53.7 mg GAE/g, 85.2 mg QE/g) during the fermentation period. The 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging activity and electron donating ability (EDA) were similar to those of the control group after 14 days of fermentation, while the ferric reducing antioxidant power (FRAP) was higher in the leaves after 14 days of fermentation when compared to the control group. The angiotensin converting enzyme (ACE) inhibitory activity showed similar or higher inhibitory activity in the leaves when compared to the control group (0.01% captopril), and the α-glucosidase inhibitory activity was higher in the leaves and stems when compared to the control group (0.05% acarbose). The glucosinolate content and the ABTS, ACE, and α-glucosidase inhibitory activity were correlated, as determined by the observed straight line plot with a positive grade. During the fermentation period, the detected glucosinolates were sinigrin, glucobrasicin, glucotropeolin, and progoitrin. The DLMK extract is therefore expected to be valuable as a functional food because of its effective antioxidant, antihypertensive, and antidiabetic activities.

          Effects of Dolsan Leaf Mustard Powder on the Quality of Frying Batter

          The anti-oxidant activity, sinigrin content, and sensory evaluation of frying batter treated with Dolsan leaf mustard powder (DLMP) were investigated. These endpoints were measured in the control sample (batter without DLMP) and seven other batters with the addition of DLMP in the following quantities: 0.03 g (S-1), 0.02 g (S-2), 0.06 g (S-3), and 0.3 g (S-4, S-5, S-6, and S-7 with different amounts of red pepper powder). The acid value (AV) and peroxide value (POV) were low in the S-4 and S-5 batters; the total polyphenol content was 190.97 mg GAE/g in the control sample and 721.17 mg GAE/g in S-4; and the total flavonoid content of S-6 was 67.82 mg QE/g which was higher than that of the control sample. The anti-oxidant activity was measured by EDA, ABTS, and FRAP assays and was seen to rise as the amount of DLMP increased. Moreover, sinigrin content was higher in S-4, S-5, S-6, and S-7 than in the control, S-1, S-2, and S-3, and there were no significant differences among S-4 to S-7 when just 0.3 g of DLMP was added. Sensory evaluation showed that color, flavor, taste, texture, and overall acceptability values were significantly higher in S-4 and S-5, and that there was no significant difference across the samples. The addition of DLMP is therefore expected to increase the anti-oxidant activity of frying batter which would be effective in improving the storage and quality of the product.

          Children seem to be more familiar with getting information from computer and internet than adults, and also it is true that information behavior in the internet has strong influences in the living of the children. When considering this fact, the public library which is the gateway to knowledge should provide a website for children services, allowing to do their life-long learning, read books and access information, without any restrains from time and space. The purpose of this study is to show the importance of public library website, especially for the children, and also to analyze trend of the country and present the solution to the problems. To achieve these goal, the analysis on the websites of the public libraries in Dagu and Gyengbuk area has been conducted. In detail, these were conducted about following criterions: existence of the website, structural system, search service, and contents. 오늘날 아동은 성인보다 컴퓨터와 인터넷을 통한 정보습득에 익숙하며 실제로 그들의 생활형태에 커다란 영향을 미치고 있다. 이에 공공도서관은 시공간에 제약 없이 아동들이 스스로 평생동안 학습하고, 책을 읽고, 정보에 접근할 수 있도록 웹사이트를 구축하여 제공해야 한다. 본 연구는 공공도서관 웹사이트의 중요성, 특히 아동을 위한 공공도서관 웹서비스의 중요성과 국내 동향을 분석하고 개선방안을 제시하고자 하는데 목적이 있다. 이를 위하여 대구.경북지역의 공공도서관 홈페이지를 대상으로 웹사이트의 구축여부, 구성체계, 검색서비스, 콘텐트를 분석하였다.

          Many visually handicapped people face difficulties with using libraries. Public libraries are thus required to meet the need of visually handicapped people. Public libraries should guarantee the visually handicapped the right to use appropriate services. If so, the visually handicapped can improve their life quality by decreasing information gap and increasing information comprehension or accessibility. This study is designed to analyze how visually handicapped people can use public libraries. Based on the analysis, this study can provide actual suggestions to improve opportunities for the visually handicapped to use libraries. Data are collected from visually handicapped people who are the members of Suseong Public Library, Daegu, Korea. In specific, this study can analyze various characteristics(i.e., information needs, selection and use behavior of information sources, difficulties and gaps to access information, use behavior and service satisfaction of public library) regarding their library use by telephone interviews. 오늘날 시각장애인은 정보접근과 도서관 이용에 많은 어려움을 겪고 있다. 이에 공공도서관은 장애인의 기본권(정보접근·이용권)을 보장하여 시각장애인의 지식정보격차를 해소하고 정보해득력을 강화시켜 삶의 질 향상과 사회참여·통합을 이룰 수 있도록 해야 한다. 본 연구는 시각장애인의 정보요구와 정보이용행태를 분석하고 그들이 도서관을 어떻게 이용하고 있는지를 살펴봄으로써 시각장애인을 위한 공공도서관 서비스의 구체적이고 실질적인 방안을 제시하고자 하는 데 목적이 있다. 이를 위하여 대구광역시립수성도서관에 정상회원으로 등록된 시각장애인을 대상으로 전화면담기법을 사용하여 그들의 정보요구, 정보원 선택 및 이용행태, 정보입수의 어려움과 격차, 공공도서관 이용행태, 공공도서관 서비스 이용의 만족도를 분석하였다.
