        어리연꽃 (Nymphoides indica (L.) O. Kuntze) 화경 배양으로부터 체세포배발생을 통한 식물체 재생

        어리연꽃의 화경조직으로부터 형성된 캘러스로부터 체세포배발생을 통한 기내 식물체 재생체계를 확립하였다. 2,4-D가 첨가된 배양배지에 4주 배양 후 어리연꽃의 화경화경조직으로부터 연황색의 구형세포과가 발달하였으며 생장조절제가 첨가되지 않은 1/2MS 배지로 옮걱 명배양한 결과, 약 배양2주 후부터 녹색의 체세포배 유사구조의 발달이 관찰되었으며 정상적인 식물체로 발달하였다. 따라서 캘러스 발달 초기에 형성된 구형 세포괴 및 캘러스는 초기 단계의 체세포배이며 캘러스는 배발생캘러스임을 알 수 있었다. 화경조직으로부터 배발생캘러스 형성빈도는 0.1-0.3 mg/L 2,4-D가 첨가된 1/2MS 배지에서는 거의 100%이었으며 2,4-D 농도가 증가할수록 배발생캘러스의 형성빈도는 반대로 감소하였고 고농도 2,4-D 처리구에서는 캘러스 형성이 전혀 이루어지지 않았다. 한편 BA는 처리 농도에 상관없이 조직의 갈변 및 고사를 유발함으로써 체세포배 형성에 저해적임을 알 수 있었다. 어리연꽃의 잎조직의 경우는 화경조직에 비하여 연황색 구형 세포괴 형성 빈도가 매우 낮아 0.3 mg/L 2,4-D 처리구 (배발생캘러스 형성빈도 9.5%)를 제외하고는 나머지 모든 처리구에서 배발생캘러스의 형성을 관찰할 수 없었다. 현재 배발생캘러스의 현탁배양 체계를 확립중이며 본 연구에서 확립된 어리연꽃의 식물체 재생체계는 어리연꽃의 기내 대량증식 수단은 물론, 유용 형질 도입을 통한 분자육종의 수단으로 활용이 가능할 것으로 기대된다. Plant regeneration system from floral stem of Mymphoides indica via somatic embryogenesis was established. After four weeks of culture onto 1/2MS medium containing 2,4-D, pale-yellow globular structures and calluses were formed on the cut surface of floral stem explants. Upon transfer to 1/2MS basal medium, pale-yellow globular structures were developed into somatic embryos and normal plantlets. These results indicated that pale-yellow globular structures and calluses from floral stem were globular embryos and embryogenic calluses, respectively. The frequency of embryogenic callus formation from floral stem was reached to nearly 100% when floral stem was cultured onto 1/2Ms medium supplemented with low concentration of 2,4-D (0.1 to 0.3 mg/L). However, the higher concentration of 2,4-D resulted in decrease of the frequency of embryogenic callus formation. In this study, low concentration of 2,4-D had a stimulative role in embryogenic callus formation, whereas BA showed inhibitory role in callus formation. In comparison to floral stem, leaf explants showed low frequency of embryogenic callus formation. The highest frequency of embryogenic callus formation from leaf explants was 9.5% when leaf explants were cultured onto 1/2MS medium supplemented with 0.3 mg/L of 2,4-D. The plant regeneration system of Nymphoides indica established in this study, might be applied to mass proliferation, conservation of genetic resources and genetic transformation for molecular breeding.

        요통환자의 사상체질과 양도락의 상관관계

        Objectives : The aim of the study is to investigate the correlation of Ryodoraku score and four constitution in the patients with chronic low back pain. Methods : Using Pearson's correlation test, we divided 79 patients into two groups(normal and chronic low back pain group) and analyzed the correlation between Ryodoraku score and major constitution rate of four constitution. Results : 1. Ryodoraku score in the chronic low back pain group was mostly found to be below 40 ${\mu}A$, regardless of constitution. 2. There was negative correlation on F2 in lesser Yin person of chronic low back pain group, and there was positive correlation on H4 H5 F2 F4 and F5 in greater Yin person of chronic low back pain group. 3. There was negative correlation on H1 and H6 in lesser Yang person of normal group. 4. There was negative correlation on H6 in lesser Yang person of total group including normal and chronic low back pain group. Conclusion : These results suggest that roydoraku should be correlated to four constitutional diagnosis in part and available to increase accuracy of four constitutional or Ryodoraku diagnosis.

        퇴행성 슬관절염 환자의 뜸 치료가 양도락 점수에 미치는 영향

        Objectives : This study was done for reporting the effect of moxibustion therapy on Ryodoraku score of the patients with degenerative arthritis of knee joint. Methods : We investigated 65 cases of patients with degenerative arthritis of knee joint, and devided patients into two groups : One group treated by moxibustion therapy, which was not applied to the other group we analyzed of each group the Ryodoraku score(F1, F6) of each group before and after moxibustion therapy and compared it. Results : 1. In moxibustion therapy group compared with baseline, at final, Ryodoraku score(F1, F6) was significantly increased. 2. At final, moxibustion therapy group showed significant increase on Ryodoraku score(F1, F6) score compared with non moxibustion therapy group. Conclusions : It is suggested that Ryodoraku score(F1, F6) should be available for diagnosing degenerative arthritis of knee joint.

        제주상사화 (Lycoris chejuensis K. Tae et S. Ko) 잎 및 뿌리 절편으로부터 소자구 형성을 통한 식물체 재생안

        제주상사화의 잎 및 뿌리 조직으로부터 형성된 캘러스 및 구형 소자구로부터 효율적인 기내 식물체 재생체계를 확립하였다. 2,4-D가 첨가된 B5 배양배지에서 12주 배양 후 제주상사화의 잎 조직으로부터 백색의 구형 세포괴 및 캘러스가 동시에 발달하였으며 그 빈도는 32.1%이었다. 그러나 3 mg/L 이상의 고농도 2,4-D 처리구에서는 캘러스 형성빈도가 14%로 크게 감소하였으며 10 mg/L 2,4-D 처리구에서는 캘러스 형성이 전혀 이루어지지 않았다. 잎 조직과 달리 뿌리 절편의 경우 3 mg/L 2,4-D 처리구에서 캘러스 형성빈도가 36.1%로 가장 높았으며 BA 단독처리구나 2,4-D와 BA의 혼용처리구에서는 그 빈도가 감소하였다. 형성된 백색의 구형세포괴는 생장조절제가 첨가되지 않은 B5배지에 배양하면 소자구 발달 경로를 거쳐 소식물체로 발달이 이루어졌다. 소식물체는 생장조절제가 첨가되지 않은 1/2MS 기본배지로 옮겨 명배양한 결과, 약 배양 2주후부터 녹색의 잎이 신장되는 것을 관찰 하였으며 4주 후 뿌리 발달이 이루어지면서 정상적인 식물체로 발달하였다. 재생된 소식물체는 배양기내에서 순화과정을 통해 토양이식이 가능하였다. 본 연구에서 확립된 제주상사화의 식물체 재생체계는 제주상사화의 대량증식 수단은 물론, 유용 형질 도입을 통한 분자육종의 수단 및 유용 유전자원의 장기보전을 위한 초저온 보존 연구의 직접적인 연구소재로 활용이 가능할 것으로 예상된다. Plant regeneration system from leaf and root segments of Lycoris chejuensis via bulblet formation was established. Surface-sterilized leaf and root segments were cultured on the B5 medium containing 2,4-D. After 12 weeks of culture onto B5 medium containing 2,4-D, white globular structures and white calluses were formed on the cut surface of the explants. The highest frequency of globular structures and calluses formation from leaf explants was 32.1% when leaf explants were cultured onto B5 medium supplemented with 1 mg/L of 2,4-D. However, the higher concentration of 2,4-D (over than 3 mg/L) resulted in decrease of the frequency. In comparison to leaf explants, root segments showed the highest frequency at a rate of 36.1% when root explants were cultured onto B5 medium supplemented with 3 mg/L of 2,4-D. These structures and calluses were sub-cultured and proliferated onto the same culture medium. Upon transfer to B5 basal medium, white globular structures were developed into bulblets and normal plantlets. After 4 weeks of incubation in the light, plantlets were successfully rooted over the frequency of approximately 90%. Rooted plantlets were successfully transferred to potting soil and acclimatized in the growth chamber. The plant regeneration system of Lycoris chejuensis established in this study, might be applied to mass proliferation, conservation of genetic resources and genetic transformation for molecular breeding.

        유근피(楡根皮)가 전신적(全身的) 및 국소적(局所的) 아나필락시스에 미치는 효과(效果)

        Ulmi radicis cortex is a herb medicine which has been used for the treatment of such allergic disease as urticaria, allergic rhinitis and athma. To assess the contribution of an aqueous extract of Ulmi radicis cortex(URC) in systemic anaphylaxis, we used compound 48/80 as a fatal anaphylaxis inducer in rats. URC inhibited anaphylactic shock 100% with a dose of 1.0 mg/g body weight (BW) 1 hr before injection of compound 48/80. URC significantly inhibited serum histamine levels induced by compound 48/80. URC (1.0 mg/g BW) also inhibited to 79.1% passive cutaneous anaphylaxis activated by anti-dinitrophenyl (DNP) IgE. URC dose-dependently inhibited the histamine release from the rat peritoneal mast cells (RPMC) by compound 48/80. Moreover, URC had a significant inhibitory effect on anti-DNP IgE-induced histamine release or tumor necrosis $factor-{\alpha}$ production from RPMC. The level of cAMP in RPMC, when URC was added, significantly increased compared with that of normal control. These results indicate that URC may possess strong antianaphylactic effect.

      • 조리교육기관의 교육서비스품질이 교육만족도, 진로준비행동에 미치는 영향 연구

        The recent changes in the perception of eating out due to dietary changes and the influence of various media and the Internet are specializing and diversifying cooking education to cultivate talents with the development of the food service industry. However, the number of private education institutes is increasing amid the decline in the school-age population and the wave of educational opening, while the number of students is gradually decreasing. Therefore, in this study, the relationship between empathy, tangible, certainty, reliability, and information gathering behavior, which are sub-factors of educational service quality, was analyzed. For empirical analysis, a total of 381 response samples were organized to analyze demographic characteristics, student use patterns, validity and reliability, correlation, and hypothesis verification. The hypothesis verification results showed that all constituent factors of educational service quality had a positive effect on educational satisfaction, and educational satisfaction had a significant effect on all career preparation behaviors. The implications of the hypothesis verification results are that existing education service quality research continues, but research on education service quality by private cooking education institutions centered on practitioners is insufficient, so it is meaningful to provide basic data for education service quality research by private cooking education institutions.

        GIS를 이용한 돌발홍수의 한계유량과 유역특성인자의 적합도 분석

        최근 홍수피해의 문제점을 분석해 보면 기상 이변으로 집중호우가 발생하고 특히, 국지적인 호우에 의한 돌방홍수 (flash flood)로 인한 피해가 빈번하게 발생하고 있다. 본 연구에서는 하천 지역의 돌발홍수의 특성을 밝혀 보고자 GIS 기법을 이용하여 하천 유역의 수문학적 돌발홍수 예측모형을 지형기후학적순간단위유량도를 적용하여 구축하였다. DB는 GIS를 사용하여 구축하였으며 DEM으로부터 유역과 하천을 추출하였다. 연구지역의 하천은 소, 중, 대규모 유역을 포함하였다. 돌발홍수정보에 관한 기준설정을 위해 첨두홍수량과 한계유량을 결정하였다. 한계유량은 지형기후학적 단위유량도 모델로 계산되었고 지형인자와의 적합도를 분석하였다. Recently, local heavy rain for a short term is caused by unusual changing in the weather. This phenomenon has, several times, caused an extensive flash flood, casualties, and material damage. This study is aimed at calculating the characteristics of flash floods in streams. For this purpose, the analysis of topographical characteristics of water basin through applying GIS techniques will be conducted. The flash flood prediction model we used is made with GCIUH (geomorphoclimatic instantaneous unit hydrograph). The database is established by the use of GIS and by the extraction of streams and watersheds from OEM. The streams studied are included small, middle and large scale watersheds. For the first, for the establishment of criteria on the flash flood warning, peak discharge and trigger runoff must be decided. This study analyzed the degree of aptitude of topographical factors to the trigger runoff calculated by GCUH model.

        수영운동이 비만 zucker rat의 골격근 TNF-α 및 백색지방조직 UCP-2 발현에 미치는 영향

        The purpose of this study was to investigate the expression on TNF-α of skeletal muscle and UCP-2 of white adipose tissue with glycometabolism according to 12 weeks of swimming in Zucker rat. Zucker rat were randomly divided into two groups (n=7 in each group): control, swimming group. Animals in swimming group were subjected to swim. Swimming exercise was conducted daily in swimming pool for 30min five days a week for 12 consecutive weeks (with free weight). Rats were sacrificed 24 hrs after the last bout of exercise. Soleus muscle and white adipose tissue were extracted to analyze for TNF-α and UCP-2, respectively. TNF-α and UCP-2 variables were analyzed using the Western Blotting technique. A independent t-test was used to examine the difference between groups. The present results demonstrated that in swimming group, fasting glucose levels significantly decreased in swimming group compared to control (p<.001). But no significant differences in levels of serum insulin were found between groups. UCP-2 induction levels in white adipose tissue further increased in swimming group compared to control groups (p<.001). Also swimming exercise significantly decreased expression of TNF-a in soleus muscle of Zucker rats. The present data suggested that low-intensity swimming exercise may improve glycometablism control by up-regulating UCP-2 expression and down-regulation the TNF-α of skeletal muscle in Zucker rats.
