        Crystal Structure of MJ0684 from Methanococcus jannaschii,a Novel Archaeal Homolog of Kynurenine Aminotransferase

        MJ0684 from Methanococcus jannaschii is a hypothetical protein belonging to the subfamily Ig amino acid aminotransferases. In the present study, the crystal structure of MJ0684 has been determined at 2.2 resolution. It reveals that MJ0684 has an overall structure similar to subfamily Ig aminotransferases and its active site architecture is most similar to that of kynurenine aminotransferases among several kinds of aminotransferases in the subfamily Ig?. It has two hydrophobic active site residues conserved in the kynurenine aminotransferases for recognizing hydrophobic substrates. In addition, the absence of any basic residue for recognizing the side chain carboxylic group of the aspartate in the active site rules out the possibility that MJ0684 would act as an aspartate aminotransferase. These structural observations collectively imply that MJ0684 is a novel archaeal homolog of the subfamily Ig kynurenine aminotransferase.

        품질기능전개(QFD) 기법을 적용한 건설프로젝트 설계VE 준비단계 업무 개선 및 체계화

        건설 프로젝트의 설계VE는 비용 절감 및 품질 향상을 위한 주요 수단이며 최근 그 중요성이 부각되고 있다. 특히 올 하반기부터는 건설공사비 100억원 이상 프로젝트까지 VE검토가 확대 적용되어 더욱 더 활성화될 것으로 예측된다. 따라서 설계VE를 체계적이고 효율적으로 수행할 수 있는 방법이 요구되지만 국내에선 아직까지는 이에 미치지 못하고 있다. 따라서 본 연구에서는 설계단계 YE를 효율적으로 실시할 수 있는 방법을 모색하고자 한다. 그 방법은 설계YE 준비단계 업무에 QFB(Quality Function Deployment)를 적용하는 것으로 이는 발주자 요구사항을 체계적으로 반영하고 기능분석을 효과적으로 실시하는데 기여할 것으로 판단된다. The importance of design value engineering in construction projects recently is rising, which deal about in reduction of cost and improvement of quality. Especially, From half of this year 2005, It is predicted that the value engineering is necessary to be carried out in the projects having project cost more than 10 billion won. In increasing importance of value engineering, we require a way by which we accomplish value engineering systemically and effectively though it is not common in domestic situation. So, this case study shows a way in carrying out value engineering effectively. As the way is to apply QFD(Quality Function Deployment) in pre-study work of design value engineering, this is expected to contribute as function analysis and reflect the requirement of users or owners.

        VE 품질모델 기법을 활용한 공동주택 노후화 평가 기준 추출 및 AHP 중요도 분석

        With the increasing number of aging apartment complexes, there's a growing need for systematic maintenance to address potential facilityissues over time. This study aims to establish maintenance standards for aging apartment complexes using the value engineering quality modeltechnique. Through a combination of prior research analysis and interviews with industry experts, 17 evaluation items were identified andcategorized into 4 groups using the affinity diagram method. Expert surveys and importance analysis demonstrated consistent significanceacross factors. To further understand relative importance, an AHP analysis was performed, presenting results aimed at supporting the efficientmaintenance of aging apartment complexes. 노후화된 공동주택은 지속적으로 증가하고 있으며, 시간이 경과함에 따라 시설물에 다양한 문제점들이 발생 가능하다. 따라서 노후 공동주택은 유지관리를 체계적으로 수행할 수 있는 업무 기준이 요구된다. 이에 본 연구에서는 가치공학 품질모델 기법을 활용하여 노후화된 공동주택의 유지관리 기준을 추출하였다. 이를 위하여 선행연구 분석 및 관련 실무전문가 인터뷰를 실시하였다. 그 결과 17가지 평가항목을 도출하였으며, 친화도기법을 활용하여 4개 카테고리로 그룹핑하였다. 다음으로 전문가 설문조사를 실시하여 중요도 분석을 실시하였으며, 그 결과 요인별 중요도에 편차가 거의 없는 것으로 나타났다. 따라서, 본 연구에서는 AHP 분석을 실시하여 평가요인 간 상대적 중요도를 분석하여 결과를 제시하였다. 제시된 결과는 노후화된 공동주택의 효율적 유지관리 업무를 지원해 줄 것으로 기대된다.

        Utilization of Contradiction for Creating Design Alternatives in Construction Value Engineering

        The design review in a Value Engineering (VE) workshop generates ideas for alternative construction designs that, if adopted,would improve the function of the original design. Each alternative conceived during idea generation will usually carry somebeneficial and some adverse effects that must be traded off against each other when deciding whether or not to adopt the design. Theobjective of this paper is to investigate the characteristics of contradiction in idea creation, and use them to create better designalternatives in construction VE. An Idea Breakdown Structure (IBS) is applied as the principle of problem solving. A contradictiontable is then set up to overcome contradictions in construction design. A case study shows that balanced contradiction produces betteralternatives in construction VE.

        Selection and Management Factor Analysis of Urban Infrastructure for U-City Construction

        U-City (Ubiquitous City) projects are being actively pursued, where ubiquitous technology is applied to large-scale new city development. U-City is all about creating a ubiquitous environment which, on the basis of Information Technology (IT)infrastructure, is combined with construction technology to make our lives easier and more comfortable. More recently, the concept of a U-Eco City (Ubiquitous Eco-friendly City) is widely discussed to incorporate the notion of a ‘green city’ into the U-City concept, with relevant research efforts also underway. In accordance with this tendency, this paper sets the criteria for selecting the key infrastructure required to build an effective U-City and conducts expert surveys and a Delphi analysis. For roadway facilities on which the highest priority is known to be placed, this paper analyzes their required VE-based functions and extracts management factors. The AHP technique is employed to analyze the relative importance of the nine core management factors which are required to build the U-City in roadway facilities. Among these key factors, ‘Establishment of a Ubiquitous System’ is shown to be of the greatest importance, followed by ‘Analysis of an Automation System’, ‘Establishment of Factor Technology’ and ‘Ideation of an Eco-Friendly City’. Formulating U-City establishment plans on the basis of high-priority management factors will enable the effective construction of roadway facilities. Hence, this paper sets work standards for individual management factors and proposes development guidelines.

        건설VE 아이디어 창조 기법 적용효과 비교분석

        Value Engineering (VE) applied to construction projects is the effective method which reduces input costs and improves their capacity and quality. Creating ideas is that value engineering of construction projects play an important role to suggest a new proposal. For this reason, an approach to method to accomplish creating ideas effectively is demanded, but doesn't play a role as facilitation. This study, analyzes and suggests the way to bring about effective results of many ideas. The method is to compare and analyze the effectiveness applied to a design VE case study and then suggest the result, which provides the base to carry out creating ideas effectively.         SOC (Social Overhead Capital) 프로젝트의 효율적 투자를 위한 타당성분석 데이터베이스 적용 방안

        SOC 프로젝트는 대규모의 재원과 시간이 소요되는 프로젝트이다. 따라서 초기의사결정을 위한 타당성분석의 신뢰도는 상당히 중요하므로 이에 대한 지속적인 연구개발이 요구된다. 이에 본 연구에서는 SOC 프로젝트의 타당성분석 체계에 관한 고찰과 연구자료 분석을 통해 문제점을 도출하고, 그에 따른 개선방안을 제안하고자 한다. 그 개선방안은 프로젝트별 정보축적을 통한 타당성분석 데이터베이스 시스템을 구축하는 것이다. 이는 합리적 의사결정을 지원할 뿐 아니라 프로젝트 진행과정에서 발생 가능한 리스크를 초기 예측함으로서 효율적 수행을 지원할 것이다. A SOC(Social Overhead Capital) project needs a large scale of financial resources and time. Therefore, it needs a constant investigation development, because the trust of feasibility analysis for an early decision-making is very important. In this study, we will extract a problem through data analysis and an inquiry into the feasibility analysis system of SOC project, and suggest improvement ways for it. The appropriate ways is to construct a feasibility analysis database system through the accumulation of information by project. This way will help as to predict a risk that may be generated in process of the project, as well as support a rational decision-making.

        FLIP as an Anti-Cancer Therapeutic Target

        Suppression of apoptosis is one of the hallmarks of carcinogenesis. Tumor cells endure apoptotic pressure by overexpressing several antiapoptotic proteins, and FLICE inhibitory protein (FLIP) is one of the important antiapoptotic proteins that have been shown to be overexpressed in various primary tumor cells. FLIP has two death-effector domains in tandem, mimicking the prodomain of procaspase-8. It is recruited to Fadd in death-inducing signaling complex, thereby preventing the activation of procaspase-8. To date, three isoforms of human cytosolic FLIP (c-FLIP) and six viral homologs (v-FLIP) have been identified. Recently, the crystal structure of v-FLIP MC159 was determined for the first time as an atomic-detail FLIP structure, which revealed that two death effector domains are packed tightly against each other mainly through conserved hydrophobic interactions. The overexpression of c-FLIP in tumor cells has been shown to be the determinant of the tumor’s resistance to death ligands such as FasL and TRAIL. It has also been shown that the down- regulation of c-FLIP results in sensitizing resistant tumor cells. Therefore, the agents directly targeting c-FLIP at mRNA and protein levels are expected to be developed in near future and tested for the potential as a new class of anti-cancer drugs.
