In this experiment, we examined the the effect of Laetrile Oil to the immune system and tried to disclose the anti-cancer mechanism of this. The result is listed as below. 1. After per-os, sub-cutaneous and into the peritoneal injection of Laetrile Oil to the SPF mice, the LD50 is above 5000mg/kg at even group. 2. Mean survival days of mice treated with laetril oil, after S-180 cells transplantation into the peritoneal cavity decreases 1.5 days(-6.8%) compared with the Mean survival days of the control group(22days.) 3. Effects of laetril oil on natural killer cell activity at Effector/Target Cell Ratio with 100:1, 50:1, 10:1 into methotrexate-pretreated mice is like this.: Compared with $29.22\pm12.7\%$ Cytotoxicity of the control group, sample group's Cytotoxicity had $38.83{\pm}12.5%$ of meaningful acrease. At 100:1 Effecr/Target Cell Ratio. At 50:1 Effector/Target Cell Ratio, control group has $20.02{\pm}9.6%$ Cytotoxiciy and sample group had $31.53{\pm}13.4%$ Cytotoxicity. At 25:1 Effector/Target Cell Ratio control group has $13.60{\pm}6.6%$ Cytotoxicity and sample group had $20.81{\pm}9.8%$ Cytotoxicity According to the above results, the Laetrile Oil represents nontotoxic to a SPF mice. non-effective to transplantable Sarcoma 180 tumors, and activaion in NK cell activity.
청리자감탕(淸離滋坎湯)의 폐암 세포주 A549의 invasion activity 억제 효과

By applying in vitro invasion assay model, we examined the anti-metatstastic effect of ChunghjagamTang(CLJGT). In 3H-thymidine incorporation assay, CLJGT treated groups showed the decreased DNA synthesis rate compared with control group. Gelatin zymogram assay showed that CLJGT decreases the gelatinolytic activity of MMP-9 from A-549, at the concentration of $800{\mu}g/ml$. We examined whether CLJGT inhibits the invasion of A-549 cells through the matrigel precoated transwell chamber. The results showed that CLJGT effectively inhibited the invasion of A-549 as compared with the control (+PMA) groups. From our research, part of mechanism underlying anti-metastastic effect of CLJGT was proven in vitro.
혈부축어탕(血府逐瘀湯)이 암전이(癌轉移) 억제(抑制)에 미치는 영향(影響)

To examine the effect of Xuefuzhuyutang on the metastasis of cancer, the following experiments were carried out. Before the main experiments, the cytotoxicity was measured by putting Xuefuzhuyutant sample in HT1080. Then zymography was made to examine the change of gelatinolytic activity. Western blotting was carried out to examine the changes of Fos, Jun, Ets, Erk, md JNK. In vitro invasion assay with transwells coated by collagen and matrigel was carried out. From the above results the following conclusions were obtained. 1. The experimental result about cytotoxicity of Xuefuzhuyutang agaitst HT1080 was a below. The stained cell count after being treated by by Xuefuzhuyutang sample $400{\mu}g/ml$ for 24 hours was 0.9% of total cells, and the stained cell count by Xuefuzhuyutang sample $100{\mu}g/ml$ was 1.5% of total cells. Both were near the level of control group which showed 0.6% stained. 2. The result of collagenase assay was as below. In Xuefuzhuyutang sample $400{\mu}g/ml$, MMP2 was reduced as compared with TPA control group, and the band of MMP-9 induced by TPA disapappeared. In Xuefuzhuyutang samle $800{\mu}g/ml$ both bands of MMP-2 and MMP-9 disapeared. 3. The results of western blots for Jun, Fos, Ets, Erk, JNK were a below. In Xuefuzhuyutang sample $200{\mu}g/ml$, Ets was reduced, and Jun, Fos were increased. 4 The result of invasion assay was as below. The number of cells which migrated across transwell membrane in Xuefuzhuyutang-treated group was less than that of control(+TPA) group. From the above results, it was concluded that Xuefuzhuyutang might inhibit the activity of collagenase not by the MMP-2, MMP-9 promoter but by the other way.
활혈거어약물(活血祛瘀藥物)이 항혈관신생(抗血管新生)에 미치는 영향(影響)에 관(關)한 연구(硏究)

Object When angiogenesis is excessive, Cancer, RA, Blindness, Psoriasis, Hemangioma, Diabetic retinopathy, Granulation, etc are induced. On the contrary, when it is insufficient, Stroke, Heart disease, Ulcer, Infertility, Scleroderma, artherosclerosis, delay of the wound recovery, etc occur. In recently, the methods which is control of abnormal angiogenesis are researching actively in relathion to anticancer research. This study is search for effective drugs which suppress this angiogenesis, in the ingredients of the herbs that invigorate and dispel blood stasis using to treat intravascular coagulation in the oriental herbal medicine. Methods We maked 80 % methanole extracts of Cnidii Rhizoma, Olibanum, Myrrha, Corydalidis Tuber, Curcumae Radix, Curcumae longe Rhizoma, Zedoariae Rhizoma, Salviae miltiorrhizae Radix, Polygoni cuspidati Rhizoama, Leonuri Herba, Persicae Semen, Carthami Flos, Trogopterorum Faeces, Achyranthis Bidentatae Radix, Manitis Squama, Eupolyphaga, Hirudo, Tabanus, Lycopi Herba, Artemisiae anomalae herba, Vaccariae Semen, Sappan Lignum, Gleditsiae Spina, Draconis Resina, Leonunari Semen, Selaginelliae Folium, Spatholobi Caulis, and these extracts were tested for MTT viabilaty test, BrdU incorporation, Tube foramtion assay on ECV304(immotalized human umbilical vein endothelial cell) at the concentration of $50{\mu}g/ml$, $100{\mu}g/ml$, $200{\mu}g/ml$, $400{\mu}g/ml$ Results All extracts except Draconis Resina have no cytotoxicity at the $100{\mu}g/ml$, and in BrdU incorporation test, proliferation rate were reduced below 60% at the concentaraion of $100{\mu}g/ml$ by Zedoariae Rhizoma, Sappan, Lignum Gleditsiae, Spina Draconis Resina Vaccariae Semen. Zedoariae Rhizoma Sappan Lignum Gleditsiae Spina Draconis Resina Vaccariae Semen Olibanum, Achyranthis Bidentatae Radix showed inhibition effects on tube formation of ECV304 at the concentration of $100{\mu}g/ml$. Conclusion At the concentration of $100{\mu}g/ml$ in which cytotoxicity is not found, Zedoariae Rhizoma, Sappan Lignum, Gleditsiae Spina, Vaccariae Semen showed the inhibition effect on proliferation and tubeformation of ECV304.

In the present studies, decursinol angelate, decursin isolated from Angelica gignatis radix and oil fraction of Cnidii rhizoma was analyzed by normal phase HPLC and GC/MS respectively. The standardized water extracts of Angelica gignatis radix, Cnidii rhizoma and its complex named Gung-gui-tang was tested the anti mutagenic effects by in vitro genotoxicity using Salmonella reversion assay (Ames test) and micronucleus test in chinese hamster ovary(CHO) cells. Angelica gignatis radix, Cnidii rhizoma and Gung-gui-tang was not exhibited the antimutagenic effects in the Salmonella reversion assays with or without metabolic activation. However, the micronucleus test assays, Angelica gignatis radix and Gung-gui-tang was showed the antimutagenic effects significantly. The maximum inhibition observed with Gung-gui-tang was reduced by 59% in the micronucleus test without metabolic activation. In this paper, results are presented on the availability of potential antimutagenic activity of the water extracts of Gung-gui-tang.
가미자도환(加味慈桃丸) 구성약물(構成藥物)의 혈관신생(血管新生) 억제효과(抑制效果)에 관한 연구(硏究)

Jiaweicitaowan (JWCTW) has been used to inhibit recurrence and metastasis of cancer in clinical practice, Further study has shown its anti-metastatic and anti-angiogenic effects. By applying in vitro and in vivo anti-angiogenic evaluation model, the author assayed the each ingredients of JWCTW. The author performed the following experiments and the results are listed as below: 1. Cell viability assay showed that the viability was almost identical between that of the control and that of the ingredients extract $40{\mu}g/ml$ treated, except of hexane fraction of Curcumae Radix $(40{\mu}/ml,\;2.0\%\;of\;control)$, ethyl acetate fraction $(40{\mu}g/ml,\;26.7%)$, butanol fraction $(20{\mu}g/ml,\;87.2%;\;40{\mu}g/ml,\;12.5%)$ of Cremastrae appendiculatae Tuber, water fraction of Persicae Semen $(40{\mu}g/ml,\;82.7%)$, ethylacetate fraction of Hippocampus $(40{\mu}g/ml,\;85.3%)$. 2. The results of gelatin zymogram assay showed that the ingredients of JWCTW decreases the gelatinolytic activity of MMP-9 from ECV304, at the concentration of $10{\mu}g/ml$. 3, In in vitro invasion assay, the ingredients of JWCTW effectively inhibited the invasion of cancer cells as compared with the control (+PMA) groups. 4. In capillary-like tube formation assay, the hexane and ethylacetate fractions of Curcumae Radix, Cremastrae appendiculatae Tuber and Persicae Semen showed the dramatic inhibition effects on tube formation of ECV304 at the concentration of $40{\mu}g/ml$. 5. In ex vivo rat aortic ring assay, the hexane and ethyl acetate fractions of Curcumae Radix, Cremastrae appendiculatae Tuber and Persicae Semen showed the inhibition effects on angiogenesis of rat aorta at the concentration of $40{\mu}g/ml$. According to the above research, the anti-angiogenic effects of the ingredients of JWCTW was proved and it suggested that the more effective prescription for anti angiogensis could be developed.
