        This study is to know the concept and function of Vital Gate. The results are summarized as follows, 1. Vital Gate is the original of life, which is essential to life. 2. A physiological function if Vital Gate controls the physiological activity of the whole body through Triple Heater. 3. It is said that a partial establishment of Vital Gate is the concept of Hwang jeong(黃庭) as the central point of the body. 4. It is effective that a clinical application of Vital Gate uses on the basis of Zoagyuyeum(左歸飮) and Woogyuyeym(右歸陰). 5. Fire of the Vital Gate means a original activity of life-activity and is different from the concept of the fire of Pericardium as a substitute for King Fire. 6. It is said that to explain the essence of Vital Gate in relation of kidney is to say the generative function. According to the above results, Vital Gate is understood as the control of physiological activity of the whole body as the original point of life-activity.

      • Ethanol-유도 위점막손상에 대한 葛花解酊湯의 항산화 효과

        Oxygen radicals are associated with organ injury in a wide spectrum of human disease. Ethanol-induced experimental gastric injury may also involve the generation of oxygen radicals. This Study was carried out to investigate the antioxidant effects of GalHwaHaJungTang(GHT) on the gastric lesions induced by ethanol in rats. Rats were pretreated with GHT extract 1.7㎖/kg, 3.4㎖/kg, 5㎖/kg respectively, 30min prior to exposing the stomach to 100% ethanol. The animals were killed lhr after ethanol treatment. Ethanol treatment increase lipid peroxidation catalase(CAT) levels and decreased superoxide dismutase(SOD), glutathione(GSH). On the other hand pretreatment of GHT extract significantly inhibited ethanol-induced lipid peroxidation and also induced an increase of activity of SOD, CAT and GSH levels. These results suggest that antioxidant activities of GHt are effective in prevent of ethanol-induced gastric lesions formation, and more study are required.

        Mixture of Wild Panax Ginseng and Red-Mold Rice Extracts Activates Macrophages through Protection of Cell Regression and Cytokine Expression in Methotrexate-Treated RAW264.7 Cells

        Objective:In this study, the immunomodulatory activity of a mixture of wild Panax ginseng and red-mold rice extracts (MPR) on RAW 264.7 macrophage cells in the presence and absence of methotrexate (MTX), an anti-cancer drug, was investigated. Methods and Results:In the cell viability, MPR showed a significant cell proliferation and inhibited cell regression by red-mold rice (RMR) alone or MTX alone. MPR induced moderate increase in nitric oxide (NO) production. NO production and inducible nitric oxide synthase (iNOS) mRNA expression by LPS decreased after MPR treatment. In addition, MPR slightly induced COX-2 mRNA expression, but it did not affect the expression of COX-2 mRNA by LPS treatment. In RT-PCR analyses, MPR induced IL-1α, IL-1β, IL-6, and TNF-α mRNA expression, but had no effect on IL-10 and TGF-β, regardless of MTX treatment. Furthermore, MPR did not interfere with the cytotoxicity of MTX against MCF-7 human breast carcinoma cells. Conclusions:MPR is efficacious in protecting against MTX-induced cell regression as a result of macrophage activation, resulting in induction of cytokine expression, implying that MPR could be considered an adjuvant in MTX-chemotherapy.

        The relation between the physiological revelation of the five viscera and the five circuit spirits, and of the physiological system has been studied. The results of this study are as follows ; 1. Life phenomenon of Oriental Medicine has been observed from the relation to the correspondence of heaven and man(天人相應), and the basic rules of the correspondence of heaven and man have been based to the principles between the male and female and the arrangement of the five elements(陰陽五行). 2. The physiological function of the five viscera become a center of viscera phenomenon reflects the characteristics of the five circuit spirits. 3. According to the base on the principles of the male and female the arrangement of the five elements, the theory of visceral phenomenon, by classifying the human body as the standard of the five elements, systematizes the physiological system of the central five viscera, and this is recognized as an organic whole 4. Viscera phenomenon is the basic theory of oriental medicine physiology, so the physiological function of the five viscera is to be inferred from a sign reveling from the function system. As the results of this study : The physiological revelation of visceral phenomenon is to be systemized by the characteristics of the five circuit spirits. As the physiological system of the five viscera reflects the function change of the five viscera, the five visceral systems are to be suitable for the existence of visceral phenomenon.

        HMC05의 경구투여 소핵시험 및 복귀돌연변이 시험

        Objectives: We investigated genetic toxicity of HMCO5 using the Micronucleus Test in bone marrow cells of mice and Bacterial Reverse Mutation Assay in plate incorporation method according to OECD Guidelines and KFDA Guidelines. Methods: 1. Micronucleus test: The male rats were divided into 5 groups, respectively; G(1), treated with distilled water: G(2), treated with 1250mg/kg HMC05: G(3), treated with 2500mg/kg HMC05, G(4), treated with 5,000mg/kg HMC05; G(5), treated with Cyclophosphamide·H2O. Sterilized distilled water and HMC05 were administered for two consecutive days. Cyclophosphamide·H2O was administered once on the day of 2nd administration. 2. Bacterial Reverse Mutation Aassay: Experimental groups were divided into two groups: with S-9mix(+S) or without S-9mix(-S). Each group treated with sterilized distilled water only, HMCO5(62, 185, 556, 1,667, 5,000㎍/plate) and, positive vehicles(Sodium azide, 2-Aminoanthracene, 4-Nitroquinoline N-oxide, ICR 191), respectively. Results: HMC05 did not show any changes in the number of micronucleated polychromatic erythrocytes(MNPCE) among 200 polychromatic erythrocytes compare to negative control. However, there were significant (p<0.01) increase with CPA in MNPCE. In Bacterial Reverse Mutation Aassay, no significant increases in the number of revertant colonies compared to (삭제) negative control were detected in all concentrations of HMC05. Conclusions: These results indicate that HMC05 did not show any genotoxicity against in Micronucleus test and Bacterial Reverse Mutation Aassay.

        당고특대황(唐古特大黃)의 주증(酒蒸) 여부가 혈관이완 기전에 미치는 영향

        Objectives: The aim of this study was to evaluate the differential mechnism of vasodilation of alcohol steamed Rhei Tangutici Radix et Rhizoma. (ART) and Rhei Tangutici Radix et Rhizoma. (RT) in rat thoracic aorta. Methods: Rat aortic ring preparations were mounted in organ baths with oxygenated (95% O2-5% CO2) Krebs-Ringer bicarbonate solutions at 37±0.5 ℃and subjected to contractions or relaxations. Results: ART exerted vasorelaxation on phenylephrine(PE)-induced contraction in a dose dependent manner. Vasorelaxation effects of ART and RT were endothelium-independent. In the Ca2+-free high KCl (60 mM) baths, the contraction of aortic rings induced by accumulative addition of Ca2+ (0.3-10.0mM) was significantly reduced by pre-treatment with both ART and RT for 10 min. The magnitude of vasodilatation was biggerin ART. Moreover, verapamil (0.001uM) and diltiazem (10uM), voltage operative Ca2+channel blockers, attenuated the relaxation effect of ART but not that of RT. In the absence of extracellular Ca2+, pre-incubation of the aortic rings with RT (1.0㎎/㎖) significantly reduced the contraction caused by PE but not that of ART. K+ channel inhibitors such as glibenclamide (Gli, 10-5M), tetraethylammonium (TEA, 1mM) and 4-aminopyridine (4-AP, 0.2mM) significantly reduced the ART`s vasorelaxation efficacy, but not that of RT. However, the relaxation effects of ART and RT were not inhibited by pre-treatment with indomethacin (10-5M), and atropine (10-6M). Conclusions: These results suggest that the endothelium-independent relaxation is due to inhibition of Ca2+ influx via the suppression of Ca2+ release from intracelluar store in RT but via both voltage operative Ca2+channel blockage and K+ channel activation in ART.

        HMC05의 배양 Chinese Hamster Lung 세포를 이용한 염색체이상 시험

        Objectives: We investigated genetic toxicity of HMCO5 in relation to chromosome aberration test on Cultured Chinese Hamster Lung (CHL) in the presence and absence of S-9 mix. Methods: Experimental groups were divided into two groups: with S-9mix (+S) or without S-9 mix (-S). -S group was also divided 2 series by treatment hours (6 hr: 6-S; or 24 hr; 24-S). Each group treated with vehicle only (complete culture medium), HMCO5 (1,250, 2,500, 5,000 ㎍/㎖), and cyclophosphamide monohydrate (CPA) and ethylmethanesulfonate (EMS), respectively. Results: HMC05 did not show any aberrant metaphase. However, there were significant (p<0.01) aberrant metaphases with CPA in S+ and with EMS in S-. Conclusions: These results indicate that HMC05 fomula does not show any toxicity in chromosome aberration test.
