        Novel Discovery of LINE-1 in a Korean Individual by a Target Enrichment Method

        Long interspersed element-1 (LINE-1 or L1) is an autonomous retrotransposon, which is capable of inserting into a newregion of genome. Previous studies have reported that these elements lead to genomic variations and altered functions byaffecting gene expression and genetic networks. Mounting evidence strongly indicates that genetic diseases or variouscancers can occur as a result of retrotransposition events that involve L1s. Therefore, the development of methodologies tostudy the structural variations and interpersonal insertion polymorphisms by L1 element-associated changes in an individual genome is invaluable. In this study, we applied a systematic approach to identify human-specific L1s (i.e., L1Hs)through the bioinformatics analysis of high-throughput nextgeneration sequencing data. We identified 525 candidatesthat could be inferred to carry non-reference L1Hs in a Korean individual genome (KPGP9). Among them, we randomlyselected 40 candidates and validated that approximately 92.5% of non-reference L1Hs were inserted into a KPGP9 genome. In addition, unlike conventional methods, our relatively simple and expedited approach was highly reproduciblein confirming the L1 insertions. Taken together, our findings strongly support that the identification of non-reference L1Hsby our novel target enrichment method demonstrates its future application to genomic variation studies on the risk of cancer and genetic disorders.

        학사행정관리자의 대학 교양교육과정 만족도 영향 요인 분석

        Studies focusing on liberal education in a college have been done with a macroscopic perspective. There is little research on administrative staff and/or administrative in a college who mainly manage liberal education in a college practically. Thus, this research aimed to investigate factors influencing the satisfaction of administrative staff on liberal education in a college. Survey instrument was developed and distributed to do this study. Descriptive and correlation analyses showed the recognition of administrative staff on liberal education. Regression analysis was done to verify factors that influence the satisfaction of liberal education from the perspective of administrative staff. The results indicated that course organization, support facilities, college self-evaluation, student background, and course management have effects on the satisfaction. In addition, we discussed that administrative staff have enormous interest on the management of liberal education in a college, how they can play a pivotal role in the management of liberal education in their college, and curriculum management system is need to be introduced. 대학의 교양교육과 관련된 연구들은 교양교육에 관해 주로 거시적으로 접근해왔다. 실제 대학에서 교양교육을 운영하는데 핵심적인 역할을 수행하는 학사행정관리자들에 대한 연구는 거의 이루어지지 않았다. 이에 본 연구에서는 현재 4년제 대학에서 학사행정을 담당하고 있는 학사행정관지라들을 대상으로 교양교육과정에 대한 전반적인 만족도에 영향을 미치는 요인에 대해서 살펴보고자 하였다. 이를 위하여 설문조사를 실시하였고 기술통계량과 상관분석 등을 통해 문항 및 요인에서 나타나는 학사행정관리자들의 교양교육과정에 대한 인식을 살펴보았다. 또한, 교양교육과정의 만족도에 가장 큰 영향을 미치는 요인을 알아보기 위해서 실시한 회귀분석의 결과, 강좌구성, 지원시설, 대학평가, 학생배경, 강좌운영방식이 교양교육과정의 만족도에 영향을 미치는 것으로 나타났다. 결과 분석을 통해 나타난 대학 내에서 학사행정관리자들의 교양교육과정 운영에 대한 관심과 그들의 역할에 대한 논의를 통해 학사행정관리자들의 교양교육과정 운영에 대한 권한과 전문성의 부여 및 교육과정운영시스템의 필요성을 제기하였다.

        Post lumbar puncture headache: Case report of a serious adverse event in first-in-human study

        A lumbar puncture can be used to measure the concentrations of drugs and/or pharmacodynamicbiomarkers during clinical trials of central nervous system drugs. We report a case of a post lumbarpuncture headache (PLPH) in a first-in-human study, which was reported as a serious adverseevent. A 20-year-old man received 200 mg of the investigational product (IP) for 7 days and underwenta lumbar puncture for cerebrospinal fluid sampling before IP administration (Day 1, pre-dose)and after 7 days and multiple IP administrations (Day 7, 1 hour post-dose). After discharge on Day8, the subject complained of headache, nausea, vomiting, neck stiffness, and numbness of the extremities. The symptoms occurred when he got up and disappeared after he remained in the supineposition for several minutes. Five days later, he visited the neurology clinic of the main hospital. The neurologist recommended hospitalization for further evaluation and symptom management,and the subject was then admitted to the hospital. There were no abnormal findings in vital signs,laboratory results, or brain-computed tomography. His symptoms disappeared during the hospitalizationperiod. It was important to distinguish whether the headache was IP-related or lumbarpuncture-related. Therefore, knowledge of clinical characteristics and differential diagnosis of PLPHis paramount. Furthermore, if severe PLPH occurs, a consultation with a neurologist and imagingstudies should be considered for a differential diagnosis of PLPH.

        Bioequivalence of the pharmacokinetics between tofacitinib aspartate and tofacitinib citrate in healthy subjects

        Tofacitinib is an oral disease-modifying anti-rheumatic drug to selectively inhibit Janus kinases. Tofacitinib is a representative small molecule inhibitor that is used to treat many diseases including rheumatoid arthritis and various autoimmune conditions. Unlike biological agents, tofacitinib has several advantages, including the ability to be administered orally and a short half-life. This study aimed to evaluate the bioequivalence of the pharmacokinetics (PK) between tofacitinib aspartate 7.13 mg (test formulation) and tofacitinib citrate 8.08 mg (reference formulation; Xeljanz®) in healthy subjects. A randomized, open-label, single-dose, 2-sequence, 2-period, 2-treatment crossover trial was conducted in 41 healthy volunteers. A total of 5 mg of tofacitinib as the test or the reference formulation was administered, and serial blood samples were collected up to 14 hours after dosing for PK analyses. The plasma concentration of tofacitinib was determined by ultraperformance liquid chromatography-tandem mass spectrometry. A non-compartmental analysis was used to estimate the PK parameters. A total of 35 subjects completed the study and the study drug was well-tolerated. The mean maximum concentration (Cmax) and area under the concentration-time curve from time zero to the time of the last quantifiable concentration (AUClast) for the test formulation were 52.67 ng/mL and 133.86 ng∙h/mL, respectively, and 50.61 ng/mL and 133.49 h∙ng/mL for the reference formulation, respectively. The geometric mean ratios (90% confidence intervals) of the Cmax and AUClast between the 2 formulations were 1.041 (0.944–1.148) and 1.003 (0.968–1.039), respectively. Tofacitinib aspartate exhibited bioequivalent PK profiles to those of the reference formulation.

        로봇활용교육에 대한 교사들의 테크놀로지내용교수지식 분석

        Robotics has been increasing its role among factors affecting teachers' technological, pedagogical, and content knowledge(TPACK) in education. This paper reviews these factors and analyses each to identify those most influential: teachers' individual (gender, teaching experiences), professional development(PD) for technology integration, psycho-philosophical characteristics (attitude toward technology integration, constructive belief), and school characteristics.Those having the most significant influence on teachers' TPACK of robotics were PD for technology integration from individual characteristics, attitudes toward technology integration, educational belief from psycho-philosophical characteristics, and schools' atmosphere and support from school characteristics.Thisreview confirmed that PD for technology integration and school atmosphere are the two most influential factors among all factors considered. Practical implications on precautions for effective integration of robotics in education were identified. 본 연구는 초등교사들을 대상으로 로봇활용교육의 테크놀로지내용교수지식(TPACK)에 영향을 미치는 요인들을 알아보고자 하였다. 이를 위해 교사들의 개인적 배경(성별, 교직경력, 테크놀로지 활용교육 연수)과 심리·철학적 배경(테크놀로지 활용교육에 대한 태도 및 신념) 및 학교 배경(테크놀로지 활용교육과 관련된 학교의 분위기, 지원, 시설) 및 에 대해서 살펴보고, 이 중 중요한 요인이 어떤 것인지 살펴보았다. 설문조사는 온라인 도구을 통해서 이루어졌으며, 총 121명의 초등학교 교사들이 참여하였다. 연구결과, 개인적 배경요인에서 테크놀로지 활용교육 연수와 학교의 분위기 및 지원이 유의미한 영향을 미치는 것으로 나타났다. 또한, 개인의 심리철학적 배경인 테크놀로지 활용교육에 대한 태도와 교육적 신념 모두 유의한 영향을 미치는 것으로 나타났다. 마지막으로 본 연구에서 고려한 요인 중 어떠한 요인이 가장 큰 영향을 미치는지 살펴본 결과, 테크놀로지 활용교육에 대한 학교의 분위기와 교사들의 연수가 교사들의 TPACK수준에 가장 큰 영향을 미치는 것으로 나타났다. 본 연구를 통해 현직 교사들의 로봇활용교육에 대한 TPACK수준을 알아볼 수 있었으며, 추후 로봇활용교육을 효과적으로 진행하기 위해서 교사들로 하여금 어떠한 준비를 해야 하는지와 어떻게 지원할 수 있는지에 대한 근거를 제시할 수 있을 것으로 기대한다.

        Characterization of Spastic Ankle Flexors Based on Viscoelastic Modeling for Accurate Diagnosis

        Characterization of the musculoskeletal system is essential for diagnosis providing the implications for therapy corresponding to causes of the diseases. This paper presents a characterization of an ankle neuromuscular system of patients with spasticity, to provide quantitative pathological level of the ankle spasticity with biomechanical and neurological disorders. Measurements from manual spasticity evaluation combined with a suggested neuromuscular model and parameter optimization process enabled a reliable characterization of the spastic ankle flexors. The model included two non-neural parameters representing the viscoelasticity of the muscle and four neural parameters showing the dynamics of muscle activation and corresponding force only using the measured joint angle and resistance torque. Torque contributions from non-neural parameters especially elastic properties of muscle was greater than 50% of the overall torque, common in both patients with spasticity and healthy controls. Among subgroups of the patients, subjects with short post diseases period less than 5 years, had higher torque contribution level from neural components more than 50% of the overall torque compared to the patients with longer post diseases period more than 10 years who had overall torque less than 30% of the total estimated torque. We concluded that proposed model based ankle flexor characterization served as a tools for diagnosing the patients with spasticity corresponding to their causes of diseases with both quantified neural and non-neural parameters.
