      • Clostridium acetobutylicum 무포자 변이주와 야생주의 protonmotive force와 용매 생산의 상관 관계

        해리되지 않은 분자상의 유기산에 대한 Clostridium acetobutylicum의 내산성 특징을 연구하기 위하여, C. acetobutylicum KCTC 1037과 그 균주의 용매 생성능이 향상된 변이주인 C. acetobutylicum KCTC 1724의 protonmotive foroe를 측정하였다 C. acetobutylicum의 protonmotive force는 50 mM의 아세트산이 있을 때 산 생산기 세포들은 -189~-232 mV, 용매 생산기 세포들은 -174~-204mV로 유지되었다. 25 mM의 아세트산과 부티르산이 있을 때 용매 생산기 세포들의 protonmotive force는 -135~-163 mV로 낮아졌는데 변이주 KCTC 1724의 pH gradient는 야생주 보다 더욱 감소하여, 부티르산의 투과성이 야생주보다 높은 것으로 측정되었다. 변이주에서 용매 생산성이 높은 것은 용매생산의 전구물질이 되는 유기산이 세포안으로 쉽게 들어갈 수 있기 때문으로 생각된다. Wild strain and asporogenous mutant of Clostridium acetobutylicum were used in a study to investigate the effect of organic acids on the maintenance of protonmotive force and on solventogenesis. In the presence of 50 mM acetate both of the strains maintained the protonmotive force of -174~-232 mV and the internal pH of 5.46-6.52. The protonmotive force determination in the presence of 25 mM acetate and 25 mM butyrate showed that normal magnitude of protonmotive force was maintained by the acidogenic culture of the mutant and by the wild strain. On the other hand the solventogenic culture of mutant strain maintained the protonmotive force of -135~-148 mV in the presence of 25 mM acetate and 25 mM butyrate. From these results it is concluded that the higher solventogenic ability of the mutant strain is due to the increased permeability of butyrate into the cell.

        일차산물 부티르산이 Clostridim acetobutylicum 의 Protonmotive force 에 미치는 영향

        In order to understand the acid tolerance mechanism in Clostridium acetobutylicum against the undissociated weak acids, a two stage continuous culture of C. acetobutylicum KCTC 1037 was made to supply acidogenic and solventogenic phase culture for a study on the protonmotive force and fermentation profile in the presence of butyrate, the acidic fermentation product. Undissociated butyric acid was permeable to the membrane of acidogenic cell, while the influx of the acid was minimal into the solventogenic cell. Acidogenic cell had higher pH gradient and protonmotive force than solventogenic cell. The protonmotive forces decreased linearly with the increase in butyrate from 25 to 75 mM. Acidogenic culture maintained the internal pH of 5.6-6.2 in the presence of 75 mM butyrate, and the internal pH of the solventogenic cultures was 5.3-5.7 in the similar conditions. Though the acidogenic culture was found to maintain the protonmotive force in the presence of 75 mM butyrate in the short term experiment, a long term incubation showed that the acidogenic culture was unable to ferment sugar in the similar conditons. On the other hand the solventogenic culture maintained active protonmotive force and fermented sugar in the presence of 75 mM butyrate. The protonmotive force of acidogenic culture was dissipated by 75 mM butyrate in the presence of ATPase inhibitor, dicyclohexylcarbodiimide, whilst the protonmotive force of solventogenic culture was maintained a similar condition. From these results, it is concluded that the acidification of cytoplasm by butyric acid is prevented by proton pumping action of the membrane bound ATPase which maintains the protonmotive force for a short-term period in the acidogenic cultures, and that solventogenic culture possess the ability to reduce the influx of organic acid in long term fermentation. It is suggested that C. acetobutylicum maintains the protonmotive force by the investment of ATP during the fast-fermenting acidogenesis; on the other hand, the solventogenic culture can reduce the influx of the acidic fermentation products to maintain the protonmotive force.

        영아교사의 교사리더십이 영아-교사 상호작용에 미치는 영향 : 조직내 의사소통과 학습조직문화의 매개효과

        본 연구의 목적은 영아교사의 리더십이 영아-교사 상호작용에 미치는 영향에서 조직 내 의사소통과 학습조직문화의 매개효과를 알아보는 것이다. 이를 위해 서울시에 소재한 어린이집의 영아교사 290명을 대상으로 설문조사를 실시하였다. 본 연구의 주요결과를 살펴보면 다음과 같다. 첫째, 영아교사의 리더십과 조직 내 의사소통, 학습조직문화가 영아-교사 상호작용에 유의미한 영향을 미친 것으로 나타났다. 둘째, 영아교사의 교사리더십과 영아-교사 상호작용의 관계에서 조직 내 의사소통과 학습조직문화는 각각 부분 매개효과가 있는 것으로 나타났다. 본 연구의 결과를 바탕으로 영아-교사 상호작용을 촉진하기 위한 다양한 방안에 대해 논의하였다. The purpose of this study is to investigate the mediating effect of intra-organizational communication and learning organizational culture in the influence of infant-teacher leadership on infant-teacher interaction. To this end, a survey was conducted on 290 infant teachers at daycare centers located in Seoul. The main results of this study are as follows. First, it was found that there was a positive correlation in all the variables of infant teacher leadership, infant-teacher interaction, intra-organizational communication, and learning organizational culture. Second, it was found that the leadership of infant teachers, intra-organizational communication, and learning organizational culture had a positive effect on infant-teacher interaction. Third, in the relationship between infant teacher leadership and infant-teacher interaction, intra-organizational communication and learning organizational culture had an indirect mediating effect. Based on the results of this study, various measures to promote infant-teacher interaction were discussed.

        탁주 제조 시 율무 첨가에 따른 휘발성 성분의 분석

        탁주의 기능성 강화를 위하여 율무를 첨가하여 제조한 탁주의 탁주로서의 적합성을 조사하였다. 우선 기존 쌀코오지를 이용하여 전분원으로서 율무를 15, 30, 60와 100% 첨가했을 경우, 효모의 균수와 ethanol의 생성량은 각각 9.5×107~2.3×10^8 cell/mL과 13.6~15.2%로 시료 간의 큰 차이를 보이지 않아 율무의 알콜 발효원으로 적합성을 보여주었다. 한편 고급알콜인 iso-amyl alcohol은 첨가된 율무량에 따라 1,150ppm으로부터 1,206, 1,213, 1,293, 1,604 ppm으로 각각 증가되고, acetaldehyde와 iso-butanol도 율무의 첨가량에 따라 증가하였다. 한편 Aspergillus kawachii(AK)와 Rhizopus japonicus(RJ)를 이용하여 율무누룩주를 만든 결과 ethanol 생성량은 쌀코오지로 만든 율무주에 비해 14%로부터 각각 11.2%(AK), 7.5%(RJ) 그리고 10.4%(AK+RJ)로 감소하였고, 효모의 수는 7.5×10^7~2.6×10^8 cell/mL이었으며, isoamyl alcohol 함량이 1,840 ppm으로부터 각각 632[AK], 855[RJ], 792 ppm[AK+RJ]로 감소하였고, n-propanol, isobutanol 등의 fusel oil도 감소하였으나, acetaldehyde가 57 ppm으로부터 99[AK], 138[RJ], 131 ppm[AK+RJ]으로 각각 증가되었다. 전체적으로 탁주제조에 있어 율무의 이용은, 전분원으로서 사용하는 경우 알콜 발효와 관능검사에는 큰 영향을 미치지 않았으나 fusel oil의 함량이 증가하였고, 누룩을 원료로서 사용하는 경우 ethanol의 생성과 fusel oil의 생성이 감소하고, acetaldehyde의 함량이 현격히 증가하였으며, 관능검사 결과 기호도가 낮아졌다. In order to use nutraceutical components in Yulmoo (Coix lachryma-jobi L. var. ma-yuen Stapf), the acceptability of Yulmoo for Takju (Korean rice wine) was examined in terms of the production of volatile components. When Yulmoo was added to the Takju made by commercial koji as a substitute of starch at 0 (rice 100%), 15, 30, 60, and 100%, the ethanol content slightly increased to 13.6, 15.2, 15,2, 14.1, and 13.8%; the production of iso-butanol and iso-amyl alcohol increased as the amount of Yulmoo increased. While the use of Yulmoo to the Takju as the nuruk (Korean traditional koji), inoculated with Aspergillus kawachii (AK) and Rhizopus japonicus (RJ), decreased the contents of n-propanol, iso-butanol, and iso-amyl alcohol as well as the contents of ethanol from 14.3% to 11.2 [AK], 7.5 [RJ], and 10.4% [AK+RJ]. The Yulmoo nuruk in Takju increased the production of acetaldehyde and ethyl acetate indicating the negative effect in the organoleptic evaluation. It was determined that the addition of Yulmoo as a starch source or as nuruk has a critical effect on the production of fusel oils and other volatile compounds in Takju fermentation.

        저염된장 제조시 에탄올 첨가효과

        저염된장 제조시 에탄올 첨가효과를 알아보기 위하여 된장의 염분함량을 8%. 4%로 낮추고 에탄올을 1%.2% 첨가하여 발효시킨 결과. 적정산도/총질소와 암모니아태질소/총질소와 염분과 에탄올은 1%유의수준의 역상관을 나타냈다. 보존실험및 관능검사결과. 봄철이나 여름철의 된장제조시 염분8%-에탄올1%첨가시료가 저염된장으로 실용가능성이 있었다. Preservation effects of the addition of ethanol (0-2% w/w) in low salt (4, 8% w/w) Doenzangs were investigated. Salt and ethanol suppressed remarkably the content of titrable acidity and ammoniacal nitrogen during the fermentation for 30 days at $30^{\circ}C$. In storage test after aging, 1% ethanol and 4% salt showed almost similar antiseptic effect and in the sensory evaluation, 4% salt-1% ethanol added sample had the most overall acceptability.
