Effect of Phenolic Compounds on Tobacco 6-Phosphogluconate Dehydrogenase

담배 callus (Nicotiana tabacum L. var. Virginia 115)에서 정제한 6-phosphogluconate dehydrogenase에 대한 여러 phenol 화합물의 영향을 조사해 본 결과, chlorogenic acid, esculin과, scopoletin은 모두 이 효소에 대하여 noncompetitive inhibitor로 작용하였으며 $K_i$ 값은 각각 $260\;{\mu}M$, $230\;{\mu}M$, $150\;{\mu}M$이었다. 또한 이 효소는 높은 농도의 caffeic acid와 ferulic acid에 의하여 약간 억제되었으며, 낮은 기질농도에서는 p-coumaric에 의해서도 억제되었다. 그 밖에 erythrose-4-phosphate, indole-3-acetic acid와 aromatic amino acids는 효소의 활성도에 아무런 영향을 미치지 못하였다. Effect of various phenolic compounds on the activity of tobacco 6-phosphogluconate dehydrogenase were studied. The enzyme was markedly inhibited by chlorogenic acid, esculin, and scopoletin noncompetitively with $K_i$ values of $150\;{\mu}M$, $230\;{\mu}M$, and $260\;{\mu}M$, respectively. The enzyme was also inhibited by caffeic acid and ferulic acid at high effector concentration (0.4 mM) and by p-coumaric acid at low 6-phosphogluconate concentration ($30\;{\mu}M$). All of the above phenolic compounds are known to be natural components of tobacco plants. Erythrose-4-phosphate, indole-3-acetic acid, and aromatic amino acids did not affect the activity of the enzyme significantly.

The statistical aspects of fatigue crack growth life of base metal (BM), weld metal (WM) and heat affected zone (HAZ) in friction stirwelded (FSWed) 7075-T651 aluminum alloy has been studied by Weibull statistical analysis. The fatigue crack growth tests were performedat room temperature on ASTM standard CT specimens under three different constant stress intensity factor range controls. Themain objective of this paper is to investigate the effects of statistical aspects of fatigue crack growth life on stress intensity factor rangesand material properties, namely BM, WM and HAZ specimens. In this work, the Weibull distribution was employed to estimate the statisticalaspects of fatigue crack growth life. The shape parameter of Weibull distribution for fatigue crack growth life was significantlyaffected by material properties and the stress intensity factor range. The scale parameter of WM specimen exhibited the lowest value atall stress intensity factor ranges.

6-Phosphogluconate dehydrogenase band II isoenzyme was isolated from a tobacco callus culture line, Nicotiana tabacum L., var. Virginia 115, utilizing ammonium sulfate precipitation, DEAF-cellulose chromatography, Red Sepharose CL-6B affinity chromatography followed by Sephadex G-150 gei filtration. N-terminal amino acid of the purified enzyme by dansylation method was lysine. Amino acid analysis indicated that the enzyme had 11 lysine and 17 arginine per 45,000 molecular weight subunit. Approximately 30 ninhydrin-positive spots were observed from tryptic peptide mapping study. Therefore, the results of N-terminal amino acid analysis, amino acid composition, and tryptic petide mapping suggest that the dimeric 6PG DH is composed of chemically identical subunits.

소프트웨어 개발주기에서 변경작업은 빈번하게 일어나며, 반드시 관리의 대상이 되어야 한다. 뿐만 아니라 변경작업에 대한 정량적인 측정을 통해 불필요하게 일어나는 변경작업의 원인을 찾아 제거해야 한다. 기존에 가장 널리 알려진 변경관리의 공수(Change Effort)를 추정할 수 있도록 제안된 모델은 개발자의 주관적인 요소가 포함되어 있어 모델의 성능을 검증해볼 수 없는 단점이 있으며, 그 외의 제안되어 있는 모델의 수가 적어 그 성능의 비교가 쉽지 않다. 따라서 본 논문에서는 NASA MDP Dataset 내의 defect data를 이용한 통계분석을 통해 모델을 객관적 요소로 구성하였고, 기존 모델에서 배재된 변경작업의 우선순위라는 새로운 변수를 추가했다. 본 논문에서 제안된 모델은 기존 모델에 비해 일반적인 적용이 쉽고, 개발 조직에서 변경 관리 노력을 추정함으로써 이를 통해 형상관리의 도입 시점을 예측할 수 있다. Changes occurs frequently in software life cycle, so should be managed. Moreover, unnecessary changing factors should be terminated by quantitative measurements. Current studies suggested to infer change efforts includes the subjective factors of development. Thus it is hard to be verified and be compared to a small number of suggested models. This paper proposes a new model consisting of objective factors through statistical analysis using defect data in NASA MDP Dataset. And we add a new factor priority of change to a model. The proposed model in this paper is more easier to apply and makes development organizations can estimate adoption time for Software Configuration Management(SCM) throughout inferring change effort.
