      • Characterization of Pure Organic Phosphors Assembled with Single Crystalline Ice

        Pure organic phosphorescence has a high quantum yield and a property of maintaining a long decay time even when irradiation disappears, so it is used for biochemical sensors, bio-imaging, and OLED materials. In particular, the single-crystalline ice facets that have highly ordered organized OH groups at the surface were suitable to control the assembly of molecules as a template. In addition, ice has a unique characteristic called a quasi-liquid layer, which makes it suitable for organizing molecules into organic phosphorescent materials. In this research, when organic phosphorescent materials were grown using ice as a substrate, it was found that ice induces the arrangement of the phosphorescent materials and increases the triplet concentration of the phosphorescent materials. We theoretically discovered that quasi-liquid layers (QLLs), a unique characteristic of ice through Molecular dynamics (MD) simulations, enhance the diffusion of Br6A particles to form a new crystal structure.

        Alteration of Gene Expression Profile by Melatonin in Endothelial Cells

        Melatonin is secreted by the pineal gland and gastrointestinal tract and plays a role in circadian rhythm synchronization. Melatonin is also reported to have various physiological effects on endothelial cells. Endothelial cells are responsible for physiologic homeostasis and maintaining blood vessel permeability in vivo. In this study, using gene expression profiling, we confirmed about 151 up-regulation and 1,023 downregulation expression genes, with an expression level difference of ¤1.5-fold, after exposure to 100 μM and 1 mM melatonin concentrations in endothelial cells. The genes were classified into 16 functional groups based on their functions, including cell adhesion, immune response, lipid metabolism, and response to stress. We analyzed the gene expression profiling data of melatonin- treated human umbilical vein endothelial cells(HUVECs), which indicated a broad spectrum of physiological functions, including anti-oxidative/anti-inflammatory effects and cell-to-cell junction maintenancerelated genes.

        Analysis of miRNA expression profiling in melatonin-exposured endothelial cells

        Melatonin is produced from tryptophan and is secreted by the pineal gland in periods of darkness; it has cytoprotective effects in endothelial cells. It has been reported that melatonin mediates the regulation of gene expression at the post-transcriptional level through its effect on miRNA expression. miRNAs are small non-coding RNAs that silence gene expression, regulate most cellular processes involved in development, and maintain cell functions. In this study, we confirmed the expression of 136 and 154 miRNAs when endothelial cells were treated with melatonin concentrations of 100 μM and 1 mM, respectively. We investigated anti-correlations in miRNA-target-mRNA pairs, and we identified a total of 13 and 44 mRNAs showing an expression difference of >1.5 fold, following exposure to melatonin concentrations of 100 μM and 1 mM, respectively. These profiling data revealed a variety of biological effects, including cell death, apoptosis and proliferation, via a Gene Ontology analysis of biological processes. The results of this study suggest that melatonin may regulate miRNA and gene expression and may have a cytoprotective effect on endothelial cells.

        Functional Screening of Altered MicroRNA Expression in 3-methylcholanthrene-treated Human Umbilical Vein Endothelial Cells

        The common environmental contaminant3-methylcholanthrene (3MC) is found in cigarettesmoke and is produced by incomplete combustion offat, wood and coal. 3MC is a poly aromatic hydrocar-bon that interacts with an aryl hydrocarbon receptorand causes inflammation and induces vascular dysfunc-tion; 3MC forms the DNA adducts structure and reg-ulates cell cycle, which increase oxidative stress andinflammation. MicroRNAs (miRNAs) are non-codingRNA molecules that negatively regulate gene expres-sion; these RNAs also play a role in cellular and mole-cular responses to toxicants, and can post-transcrip-tionally regulate gene expression. In this study, we ex-amined whether miRNAs affect the regulation of geneexpression in 3MC-treated human umbilical vein en-dothelial cells (HUVECs). We carried out pair-wisecorrelation analysis and identified 131 and 116 miRNAswith altered expression upon treatment of HUVECswith 100 nM and 1 μM 3MC, respectively. Furthermore,we identified 188 and 85 mRNAs with altered expres-sion upon treatment with 100 nM and 1 μM 3MC; wesubsequently analyzed their anti-correlations. TheGene Ontology (GO) enrichment analysis on the alter-ed expression of miRNA-related genes displayed sig-nificant enrichment for genes involved in certain bio-logical processes. Specifically, our results suggest thatchanges in miRNA expression caused by 3MC treat-ment are associated with inflammation of endothelialcells and may play a role in cardiovascular disease.

        Effect of Korean Red Ginseng treatment on the gene expression profile of diabetic rat retina

        Background: Korean Red Ginseng (KRG) is a herbal medicine used in Asian countries and is very popular for its beneficial biological properties. Diabetes mellitus (DM) and its complications are rapidly becoming a global public health concern. The literature on transcriptional changes induced by KRG in rat models of diabetic retinopathy is limited. Considering these facts, we designed this study to determine whether retinopathy-associated genes are altered in retinas of rats with DM and whether the induced changes are reversed by KRG. Methods: Male SpragueeDawley rats were intravenously injected with streptozotocin (50 mg/kg body weight) to induce DM, following which, KRG powder (200 mg/kg body weight) was orally administered to the KRG-treated DM rat group for 10 wks. The rats were then sacrificed, and their retinas were harvested for total RNA extraction. Microarray gene expression profiling was performed on the extracted RNA samples. Results: From among > 31,000 genes investigated, the expression of 268 genes was observed to be upregulated and that of 58 genes was downregulated, with twofold altered expression levels in the DM group compared with those in the control group. Moreover, 39 genes were upregulated more than twofold and 84 genes were downregulated in the KRG-treated group compared to the DM group. The expression of the genes was significantly reversed by KRG treatment; some of these genes were analyzed further to verify the results of the microarray experiments. Conclusion: Taken together, our data suggest that reversed changes in the gene expression may mediate alleviating activities of KRG in rats with diabetic retinopathy.

        Identification and Characterization of MicroRNAs in Acrolein- Stimulated Endothelial Cells: Implications for Vascular Disease

        MicroRNAs (miRNAs) have been implicated in the regulation of physiological and pathophysiological processes such as vascular disease. Analysis of miRNA expression profiles in vascular disease provides new insights into the possible mechanisms underlying and therapeutic targets of disease. Acrolein is a highly reactive component of cigarette smoke, which has been implicated in the development of vascular disease. In this study, we investigated whether miRNAs play a role in the modulation of various gene expression responses to acrolein in human endothelial cells. We analyzed whole-genome miRNA and mRNA expression and found that acrolein induced differential expression of 151 miRNAs in endothelial cells. We further classified a number of target genes of the differentially expressed miRNAs. Among the differentially expressed miRNAs, miR-342-3p and 422a were upregulated, and the expression levels demonstrated a high inverse correlation with their targets, the vascular disease-related genes CLDN1 and PON1. Thus, integrating specific patterns of miRNA and mRNA levels may improve the diagnosis of vascular diseases induced byexposure to environmental pollutants such as acrolein.

        사용후핵연료 습식저장시설 사고 안전성 평가 연구 현황 및 사고 사례 분석

        Spent nuclear fuel corresponds to high-level radioactive waste that has high decay heat andradioactivity. Accordingly, Spent nuclear fuel withdrawn from the reactor core is primarily stored andmanaged in a spent nuclear fuel pool in the nuclear power plant to reduce decay heat and radioactivity. In Korea, most nuclear power plant store all spent nuclear fuel in a spent nuclear fuel pool. For wetstorage, there are no defense in depth different with reactor core. The study related to spent nuclear fuelpool accident should be carried out to ensure safety. Therefore, it is necessary to analyze previous studycases related to safety of spent nuclear fuel pool and accident cases to build foundational knowledge. The Objective of this study is to analyze study cases of safety assessment and cases for spent nuclearfuel pool accident. For analyzing study cases of safety assessment, possible phenomena when spentnuclear fuel pool accident occurring identified, Subsequently, study cases for safety assessment abouteach phenomena were investigated, and materials & methods and results for each study are analyzed. For analyzing cases for spent nuclear fuel pool accident, we analyzed accident cases caused by loss ofcooling and loss of coolant in spent nuclear fuel pool. Subsequently, causes and change of water leveland temperature by each accident case are analyzed. As a result of the analysis on study cases of spentnuclear fuel pool accident, the results of the study conducted by each research institute were varydepending on the computer code, materials & methods of experiment and major assumptions used inthe study. As a result of analyzing cases for spent nuclear fuel pool accident, it was found that accidentcases for loss of cooling is more than cases for loss of coolant accident. Even though the types of accidentin spent nuclear fuel pool were similar, the specific causes were different by each accident case. All theaccident cases analyzed did not lead to severe accidents, such as nuclear fuel being exposed to the air. The result of this study will be used as fundamental data for study on spent nuclear fuel pool accident thatwill be conducted in the future.
