        오가피(Acanthopanax sessiliflorus Seeman) 열매로부터 분리한 페놀 화합물의 항산화활성

        The fruits of Acanthopanax sessiliflorus Seeman (Araliaceae) were extracted with 70% aqueous ethanol at room temperature. The concentrated extract was partitioned with ethyl acetate (EtOAc), n-butyl alcohol, and H2O, successively. From the EtOAc fraction, two compounds were isolated through the repeated silica gel, octadecyl silica gel, and Sephadex LH-20 column chromatographies. According to the results of physicochemical and spectroscopic data including NMR, mass spectrometry, and infrared spectroscopy, the chemical structures of the compounds were determined as 3,5-dihydroxycinnamic acid (1) and protocatechuic acid (2). Compound 1 was isolated from the fruits of A. sessiliflirus Seeman for the first time. And the compounds were evaluated for the radical scavenging the antioxidant capacity using 2,2`-azino-bis(3-ethylbenzthiazoline-6- sulfonic acid)diammonium salt, 1,1-diphenyl-2-picrylhydrazy, and oxygen radical absorbance capacity assay.

        OneSAF 모델을 위한 명중률 데이터 변환 방법

        OneSAF 모델의 활용이 본격화될 것으로 판단되는 현재 시점에서 가장 중요한 요소는 입력 데이터의 확보이다. 특히 모델 운용자들이 가장 결정하기 어려운 입력 데이터는 명중률과 살상률 데이터이다. 이러한 데이터들은 개발시험평가와 운용시험평가 기간 중에 실시되는 실사격 실험들을 통하여 결정되므로 많은 자원과 시간이 소요된다. 따라서 상대적으로 효율적이면서 비교적 정확한 방법들이 필요하다. 본 논문에서는 명중률 데이터를 확보할 수 있는 가능한 방안들을 검토하고, 방안별로 어떻게 데이터를 산출할 것인가에 대한 방법론을 소개하였다. 먼저 JMEM 도구들을 사용하여 산출된 결과로부터 OneSAF 모델의 명중률 데이터로 변환하기 위하여 필요한 방법을 제시하였다. 다음은 기존의 분석용 모델인 AWAM 모델에 입력 데이터로 존재하는 %로 표시된 명중률 데이터를 오차(mil)형태로 변환하는 방법인 오차요소법을 적용할 수 있음을 보였다. 더불어 소개된 오차요소법을 사용하여 명중률을 산출한 결과를 수행하였다. 오차요소법을 사용하여 공개된 사진 자료로부터 투사면적을 계산하여 보다 정확한 명중률을 계산하였다. 최종적으로 정확한 명중률 산출의 중요성을 명중률이 전투효과에 미치는 민감도 분석을 실시하여 입증하였다. 본 논문에서는 명중률 데이터 결정의 중요성과 꾸준한 명중률 데이터의 수집 및 분석으로 M&S 체계의 신뢰성이 향상되어야 하는 것을 강조하였다. To use the OneSAF model for the analysis of Defence M&S, the most critical factor is the acquisition of input data. The model user is hard to determine the input data such as the probability of hit(P<sub>h</sub>) and the probability of kill(P<sub>k</sub>). These data can be obtained directly by live fire during the development test and the operational test. Therefore, this needs more time and resources to get the P<sub>h</sub> and P<sub>k</sub>. In this paper, we reviewed possible ways to obtain the P<sub>h</sub> and P<sub>k</sub>. We introduced several data producing methodologies. In particular, the error budget method was presented to convert the P<sub>h</sub>(%) data of AWAM model to the error(mil) data of OneSAF model. Also, the conversion method which can get the adjusted results from the JMEM is introduced. The probability of a hit was calculated based on the error budget method in order to prove the usefulness of the given method. More accurate data were obtained when the error budget method and the projected area from the published photo were used simultaneously. The importance of the P<sub>h</sub> calculation was demonstrated by sensitivity analysis of the P<sub>h</sub> on combat effectiveness. This paper emphasizes the importance of determining the P<sub>h</sub> data and improving the reliability of the M&S system though steady collection and analysis of the P<sub>h</sub> data.

        새싹 밀의 추출용매에 따른 생리활성 평가

        The purpose of this study was to examine biological activities, including total contents of polyphenol, antioxidant activities, inhibitory activities of tyrosinase, and protective effect against oxidative stress in the HepG2 cells of ethanol extracts from wheat sprout. The antioxidant activity of extracts was determined by ABTS and DPPH radical scavenging activities. Ethanol extracts were tested using different ethanol concentrations (0%, 30%, 50%, 80% and 95%, respectively). The highest amount of total polyphenol was extracted by 50% and 80% ethanol which was 26.3 and 26.8 mg gallic acid equivalents/g sample, respectively. High levels of ABTS and DPPH radical scavenging activity were found in 50% ethanol (26.7 and 15.0 mg TEAC/g sample, respectively) and 80% (24.3 and 16.1 mg TEAC/g sample, respectively) ethanol extracts. Also, 50% and 80% ethanol extracts indicated higher inhibitory activities of tyrosinase compared with other extracts. In the cell-based assay, pre-treatment of the HepG2 cells with wheat sprout extracts prevented the cell damage induced by TBHP (tert-butyl hydroperoxide). The results of this study indicate that wheat sprout has significantly higher diverse biological activities and apparently has significant health benefits.

        재배기간에 따른 쓴메밀(Fagopyrum tataricum Gaertner)싹의 항산화 활성 및 생리활성 평가

        Buckwheat (Fagopyrum species) has long been an excellent functional food. Besides, buckwheat sprouts contain various functional substances. In this study, we investigated the antioxidant activity of buckwheat sprouts in the context of cultivars harvested after different cultivation periods (0, 3, 5, 7, 9, 13, and 15 days after planting). Buckwheat sprouts were cultivated at 25℃ for up to 15 days and then extracted with ethanol. Antioxidant components were then extracted from sprouts and leaves using a freeze dryer. The total polyphenolic content, flavonoid content, and antioxidant activity were then analyzed. The total polyphenol content increased from 32.26 mg GA eq/100 g for raw buckwheat to 114.75 mg GA eq/100 g after 7 days of cultivation. Also, the flavonoid content increased from 20.61 mg catechin eq/100 g (0 days) to 56.54 mg/g after 9 days of cultivation. The DPPH radical scavenging activity (concentration of extract at 0.25 mg/mL) increased from 7.89% at day 0 to 53.48% after 9 days of cultivation. Additionally, the ABTS radical scavenging activity increased from 10.26% at day 0 to 32.89% after 7 days of cultivation; of note, the activity decreased afterward. These results suggest that the best buckwheat sprouts with higher biological activities are those cultivated for 7-9 days. For a complete understanding of the potential of buckwheat sprouts as functional foods, we plan to further analyze their antioxidant activity in the future.

        구증구포 처리 대두 추출물의 항산화 활성 및 Angiotensin-I Converting Enzyme 저해 효과

        To increase the functional material content of soybean, a repetitive steaming and drying process was used. We investigated the changes in the total polyphenol content, the antioxidant activity, and the angiotensin-I converting enzyme (ACE) inhibition in soybean following nine rounds of steaming and drying. Soybean was steamed 9 times for 2 h and then dried 9 times from 55℃ to 73℃ for 3 h. The total polyphenol content in the soybean reached a maximum value of 60.47 mg GAE eq./100 g at 73℃ while the total polyphenol content in the raw soybean reached 25.17 mg GAE eq./100 g. In the raw soybean samples, the DPPH radical scavenging activity (5 mg/mL) was 8.04% but it increased by 43.29% after drying 9 times to 73℃. ABTS radical scavenging activity also improved following 9 rounds of steaming and drying. ACE inhibitory activity of the soybean dried 9 times at 73℃ was 58.94% at 10 mg/mL. These results showed that steaming and drying soybean 9 times enhanced the antioxidant activity and the ACE inhibitory activity of soybean. Therefore, more research on the biological and anti-hypertensive activity of soybean using this steaming and drying method is necessary.

        염화나트륨 처리 및 재배방법이 새싹밀의 항산화 성분 및 활성에 미치는 영향

        Sprouts have various health benefits. Specifically, wheat sprouts are rich in bioactive compounds, such as vitamins and polyphenols. Elicitation induces and enhances secondary metabolite biosynthesis in plants. Therefore, in this study, we investigated the effects of sodium chloride (NaCl) treatments on the growth profile, free amino acid content, and antioxidant activity of germinated wheat (Triticum aestivum). Wheat seeds were germinated at 20℃ for 10 days and treated with 0, 2, 4, 7.5, and 10 mM of NaCl 10 days before harvesting. Treating the soil bed with NaCl increased the nutritional component amounts, such as free amino acids and γ-aminobutyric acid. The chlorophyll a and b concentrations were the highest in the hydroponic system treated with 7.5 mM NaCl. In addition, the polyphenol and flavonoid contents of sprouts treated with 2 and 7.5 mM NaCl were 1.94 and 1.34 times higher than that of the control sprouts (0 mM NaCl, water only), respectively. These results suggest that 2 to 4 mM NaCl treatments improve the nutritional and food quality of wheat sprouts more than water only.

        콩발아배아추출물이 난소적출 랫트의 기억력 손실, 체중 증가 및 골다공증에 미치는 영향

        많은 폐경기 여성들이 기억 감퇴, 체지방 증가, 골다공증 및 에스트로겐 결핍에 의한 고지혈증과 같은 증상으로 고통을 받고 있는데, 이러한 증상을 완화하기 위한 많은 연구가 이루어지고 있다. 본 연구에서는 난소 적출 rat 모델을 이용하여 SGE의 갱년기 증상 개선 평가를 진행하였다. SGE를 4주간 13 ㎎/㎏, 26 ㎎/㎏ 및 51 ㎎/㎏의 농도로 처리하면 OVX에 의한 체지방, 렙틴 및 심혈관 관련 지표의 증가를 유의적으로 억제할 수 있었으며, 이는 SGE가 에스트로겐 결핍으로 인한 비만을 억제할 수 있음을 나타낸다. 또한 OVX 모델에서 기억력 및 인지기능이 현저히 저해되는데, 본 연구를 통해 기억 인지기능은 OVX군에 비해 모든 SGE군에서 개선되었고 SGE 농도 의존적으로 개선됨을 확인할 수 있었다. 이러한 결과는 SGE가 OVX로 인한 기억 인지기능 감퇴를 억제할 수 있음을 나타낸다. 골다공증은 폐경기여성의 대표적인 증상 중 하나이며, 이 증상은 OVX rat에서도 확인된다. 뼈건강의 생화학적 지표인 ALP는 골다공증의 병리학적 지표로 간주하는데, SGE 투여군에서 OVX군에 비해 ALP 수준이 유의하게 감소했으며, 특히 대두이소플라본 섭취군보다 통계 유의적으로 개선됨을 확인하였다. 이러한 결과는 SGE가 ALP의 농도를 낮추고 골다공증을 개선하여 뼈 건강에 도움을 줄 수 있음을 보여준다. 추가로 단층 촬영을 이용하여 뼈의 미네랄 밀도를 측정했으며, 골다공증 진단에 사용되는 진단기준인 BMD는 OVX군에서 현저히 감소하였다. SGE를 4주 동안 처리하면 OVX의 효과가 반전되고 BMD가 유의하게 증가했다. 또한 골 강도의 평가를 위해 BV/TV를 측정하였는데, 모든 SGE 투여군에서 BV/TV가 OVX군과 비교하여 유의하게 증가하였음을 관찰하였다. 결론적으로 SGE는 OVX로 인한 기억력 감퇴 및 체지방 개선에 효과적이며, 특히 골다공증에 있어서는 대두이소플라본보다 통계 유의적 개선을 보여주어, 향후 여성갱년기 증상 개선 기능성 소재로써 개발 가능성이 충분할 것으로 기대된다. It is well-known that soybeans relieve hormone deficient symptoms in postmenopausal women. In recent studies, germination of soy germ was shown to increase bioactive components such as aglycone isoflavones, γ-amino butyric acid, and soyasaponins. Therefore, this study was conducted to explore the effects of germinated soy germ extract (SGE) on memory loss, obesity, and osteoporosis in an ovariectomized (OVX) animal model. We found that oral administration of SGE improved memory function in OVX rats using a passive avoidance test. Levels of leptin and alkaline phosphatase and adipocyte size were significantly decreased in the SGE-treated group. Moreover, administration of SGE significantly prevented OVX-induced bone loss in OVX rats compared to the isoflavone-fed group. These results indicated that SGE is effective for treatment of memory loss, obesity, and osteoporosis caused by ovariectomy in rats.

        재배지역에 따른 검정콩 부위별 추출물의 항산화 활성

        With the aim of developing region specialized crops, this study was conducted to clarify effects of variant and cultivation region on antioxidative activities in various black soybean (Glycine max L.) seed tissues. Three black soybean varieties (SCEL-1, Wonheug, and Cheongja 3) were each cultivated in 4 different regions (Jeonju, Pyeongchang, Paju, and Cheonan). Harvested seeds were used to assess DPPH and ABTS radical scavenging activity, and total polyphenol, flavonoid and anthocyanin content. SCEL-1 soybean hull contained higher DPPH and ABTS radical scavenging activity (61% and 85% respectively) compared to Wonheug (40% and 50% respectively). SCEL-1 cultivated in Pyeongchang displayed the highest total polyphenol and flavonoid content (1,189 mg GAE/100g sample and 951 mg CTE/ 100g sample, respectively). Total anthocyanin content was ranked in the following order: SCEL-1>Wonheug>Cheongja 3. All black soybeans showed much higher antioxidant activity in the soybean hull than in the dehulled soybean. The antioxidant activity of black soybeans cultivated at high latitudes was high. These results suggest that the best black soybean variant for high beneficial biological activities is the SCEL-1 variant. For a complete understanding of the potential of black soybean as functional foods, we plan to further analyze their antioxidant activities in future studies.

        흑찰거대배아미를 이용한 팝라이스와 흑미차의 품질 특성

        거대쌀눈의 특성과 검정쌀 그리고 찹쌀의 특성을 보유한 흑찰거대배아미인 ‘눈큰흑찰’을 이용한 팝라이스 제조와 이를 이용한 현미차로 이용하였을 경우의 주요 성분의 분석 결과, 눈큰흑찰을 이용한 최적의 팝라이스 가공조건은 157℃에서 5 ~ 20분 볶는 조건이 상품성이 가장 적당함을 확인하였다. 눈큰흑찰 팝라이스는 대조인 일미에 비해 단백질(11.3%)과 지방(3.7%)이 높음을 보였고, 당 함량과 조성에서도 일미에 비해 다량 함유함을 보였다. 멜라노이딘 색소 함량에서 일미에 비해 4배(0.2 OD) 증가하였고, 폴리페놀, 가바(GABA), 총 아미노산 등 주요 영양과 기능성분에서 가공에 의한 눈큰흑찰의 특성이 좋음을 확인하였다. 현미차로 눈큰흑찰 팝라이스를 우려내었을 경우도 글루코오스, 슈크로오스, 라피노오스, 총 당함량에서 현미와 일미에 비해 다량 용출됨을 보였고, 특히 당도와 가장 밀접한 관계가 있는 슈크로오스는 현미에 비해 4배 이상 증가함을 보였다. 항산화성 분석을 통한 생리활성 검정에서도 눈큰흑찰 팝라이스는 현미에 비해 4배(DPPH, SC50, 0.24 mg/ml) 이상의 항산화력이 있음을 보여주었다. Recently developed black waxy rice with a giant embryo (‘Nunkeunheukchal’, BGE) was selected and processed to produce high quality nutritional food. BGE contains high levels of several phytochemicals with antioxidant activities, as well as other reported health beneficial properties. In addition, the giant embryo has high protein, lipid, and amino acids contents. Within the free amino acids, γ-aminobutyric acid (GABA), a major inhibitory neurotransmitter, has long been used for treating the aftereffects of brain injuries and stroke. A method for manufacturing pop-rice and black rice tea by popping process in BGE is provided to increase a taste, nutrition and functionality. The produced ‘pop-rice’ showed increased protein (11.3%) and lipid (3.7%) contents compared with control variety, IB (‘Ilmibyeo’). In addition, melanoidin related products, polyphenol and functional amino acid contents were increased by the popping process. Pop-rice tea made of BGE showed the highest extraction of total sugar, glucose, raffinose and sucrose (4 times higher than brown rice) by hot water. Scavenging activity (SC50) of processed BGE rice powder showed strong antioxidative activity of 0.24 mg/ml using DPPH and 1.82 mg/ml using ABTs method. Thereafter, these results suggested that the popping processed rice of BGE could be one of the promising materials for healthy food development.
