2022 Perinatology Vol.33 No.1

Situs inversus totalis is a mirror reversal of the normal asymmetrical arrangement of the viscera. The incidence is approximately 0.3 in 10,000 live births. Situs inversus totalis is commonly seen in a condition known as primary ciliary dyskinesia, and can be associated rarely with various congenital abnormalities, such as polysplenia syndrome, biliary atresia, congenital heart disease, and midgut malrotation. The association of neonatal intestinal obstruction with situs inversus totalis, especially duodenal obstruction, is extremely rare with only few cases reported in the literature to date. We report one case of situs inversus totalis with duodenal obstruction and treated successfully by gastro-duodenostomy without complications.

Genetic Structure and Diversity of the Moon Jellyfish Aurelia coerulea Polyp Population in Jaran Bay, Korea, Revealed by Mitochondrial COI and 16S rRNA Genes

2021 Ocean science journal Vol.56 No.1

Blooms of the moon jellyfish (Aurelia coerulea) have been responsible for huge economic losses and environmental disruptions in oceans around the world. The mass occurrence of these jellyfish may be highly related to the benthic polyp populations; however, studies supporting this are scarce. Here, we analyzed the genetic diversity and structure of Korean A. coerulea, using two mitochondrial genes, COI and 16S rRNA, in 51 polyps collected from Jaran Bay (South Korea) that is greatly affected by the mass occurrence of the jellyfish. Genetic diversity analysis identified 20 and 9 haplotypes in COI and 16S rRNA genes, respectively. TCS network analysis and haplotype distance matrix revealed a significant genetic structure in both genes. In addition, two distinct haplogroups (A and B) coexisted in Jaran Bay, and a single haplotype (COI-H4 or 16S-H2) dominated. These findings suggest a strong population differentiation among the polyp population that may have an effect on the jellyfish blooms in Jaran Bay.

2020 Ocean and Polar Research Vol.42 No.3

The distribution and genotypes of the sea nettle Chrysaora pacifica have been reported in the South Sea of Korea; however, little research work has been attempted in the East Sea. Here, we collected similar jellyfishes from the East Sea coasts (Goseong, Yangyang and Sokcho), and identified them to the sea nettle morphologically. In addition, the genotypes of these sea nettle were compared with those from the South Sea (Tongyeong and Geoje). Phylogenetic analysis by using the mitochondrial COI sequences showed that the genus Chrysaora was clearly separated from other taxa to be formed a monophyletic group, with each species distinctly separated. C. pacifica in the East and South Seas was separated geographically by the COI phylogeography, representing potentially different populations. The COI gene of the Korean C. pacifica had approximately 7 times more genetic variation than the nuclear ITS rDNA, and thus it might be considered as a useful marker for genetic analysis of the jellyfish population.

Recent Trends in Biosensors Based on Electrochemical and Optical Techniques for Cyanobacterial Neurotoxin Detection

2022 BioChip Journal Vol.16 No.2

Cyanobacteria that cause algal blooms are distributed worldwide and have been reported for a long time. Further, cyanobacterial growth is increasingly spreading because of anthropogenic eutrophication; this results in increasing damages by the production of a neurotoxin called cyanotoxin. Neurotoxins have adverse effects in humans, including disrupting signal transmission between neurons and muscles in various ways resulting in paralysis and death within minutes. To detect neurotoxins, analytical techniques such as high-performance liquid chromatography (HPLC), liquid chromatography–mass spectrometry (LC–MS), and enzyme-linked immunosorbent assay (ELISA) are being implemented, but have limitations as they are difficult to measure quickly in the field and are difficult to miniaturize. Introduction of biosensors for toxin detection provides a new direction as applying bioreceptors such as aptamers, antibodies, and nanomaterials can be used to create various sensors, without the limitations of conventional detection methods. In this review, we discuss the continued development of neurotoxin biosensors that are based on electrochemistry and optics.

Vol.37 No.3

Two aerophytic cyanobacteria from the rockwall of Haje port located in Geum river, Korea, were isolated in unialgal cultures and submitted to polyphasic evaluation. The filaments of the populations presented solitary or several to many parallel arranged. The straight trichomes were not attenuated with rounded apical cell. Phylogenetic analyses based on 16S rDNA sequences indicated that these populations formed the same clade with Wilmottia murrayi and had 99% or greater DNA similarity. Through the ultrastructure of the TEM, these populations showed parietal thylakoid arrangement, which coincides with family Coleofasciculaceae. From the above results, we reported the newly recorded genus Wilmottia, and species W. murrayi in Korea.

Vol.56 No.1

Microcystis protocystis was sampled from the Nakdong River, Geum River, and freshwaters of Yongin and Incheon. Within 3 days after sampling, morphologies of colonies and mucilage were observed using India ink. The colony of M. protocystis has irregular outlines and lacks distinct lobes and holes. Cells are distributed sparsely and irregularly within indistinct and homogeneous slime, which is diffuse at the margin. Three unispecific cultured strains from the So-ok stream were classified as Microcystis by phylogenetic analysis using 16S rRNA sequences. Three Korean isolates of M. protocystis showed 16S rRNA sequences completely identical to each other. The phylogenetic tree of the 16S rRNA sequences showed that Korean M. protocystis formed narrow clusters with other Microcystis members and M. protocystis with 99% or greater similarities. Additional trees of RNA polymerase beta-subunit gene (rpoB) sequences separated the Korean M. protocystis from other Microcystis. Through these results, M. protocystis was reported as a new species in Korea.

Evaluation of the complete nuclear rDNA unit sequence of the jellyfish Cyanea nozakii Kishinouye (Scyphozoa: Semaeostomeae) for molecular discrimination

2021 Animal cells and systems Vol.25 No.6

The harmful jellyfish Cyanea nozakii Kishinouye has frequently occurred on Korean coasts, and its blooms have caused serious ecological and economic damages. DNA sequences of the C. nozakii for molecular detection and discrimination are relatively scarce. In this study, we determined the complete sequence of a single unit of tandemly repeated ribosomal DNA (rDNA) of the Korean C. nozakii and characterized the molecular features of the rDNA. The complete rDNA contained 8,003 bp (48.4% GC) with the same gene arrangement (18S, ITS1, 5.8S, ITS2, 28S, and IGS) to the typical eukaryotes. Dot plot analysis showed that the coding regions (18S, 5.8S, and 28S) were highly conserved, while the non-coding regions (ITS1, ITS2, and IGS) were more variable and parsimony-informative. The IGS contained a putative transcription termination signal (poly(T) tract) and four repeats of block minisatellites. Phylogenetic analyses using 18S and 28S rDNA revealed well-resolved relationships of C. nozakii within the order Semaeostomeae, separating it from other Cyanea species. The complete rDNA sequence provides various options for the selection of jellyfish taxonomic markers and may be useful for discriminating between species of C. nozakii and phylogeny reconstruction with close relatives.
