        Penicillium daejeonium sp. nov., a New Species Isolated from a Grape and Schisandra Fruit in Korea

        Two isolates of monoverticillate Penicillium species were collected from a grape and schisandra fruit in Korea. Multigene phylogenetic analyses with the nuclear ribosomal internal transcribed spacer (ITS) region and genes encoding β-tubulin (benA) and calmodulin (cmd), as well as morphological analyses revealed that the two isolates are members of the P. sclerotiorum complex in Penicillium subgenus Aspergilloides, but different from species of the P. sclerotiorum complex. The isolates are closely related to P. cainii,P. jacksonii, and P. viticola in terms of their multigene phylogeny,but their colony and conidiophore morphologies differ from those of closely related species. The name P. daejeonium is proposed for this unclassified new species belonging to the P. sclerotiorum complex in subgenus Aspergilloides.

        In the study the bearing capacity of the eccentrically loaded shallow footing is estimated by means of the upper bound method of limit analysis. In the case of applying the upper bound, the results depend on the failure mechanism. So the failure surface is correctly studied through the model test using sand. New method is proposed to estimate the bearing capacity of the eccentrically loaded shallow footing by means of the upper bound method to apply the failure mechanism based on the model test. The propriety of new method is verified by the results of various methods. And the influences of the variables eccentricity, embedment depth, variations of contact width factor by model test using sand are also studied in this paper

        Phylogenetic Analysis, Morphology and Pathogenicity of Penicillium spp. associated with Blue Mold of Apple in Korea

        Blue mold is the most important postharvest disease of apples in Korea. Apple fruits with blue mold symptoms were collected from storages in different locations in Korea and were investigated for their association with Penicillium species. A total of sixty five isolates of Penicillium were sampled from the collected apples. Based on DNA sequence analysis of ${\beta}$-tublin gene and ITS and lsu rDNA (ID region) and morphological characteristics, they were identified as P. crustosum, P. expansum, P. italicum, P. solitum and P. sp.. P. sp. which is closely related to P. hirsutum is a new species, not reported before. P. expansum (35%) was predominant species followed by P. crustosum. The phylogenetic tree inferred from combined ${\beta}$-tublin and ID region sequence showed good correlation with species that are defined by morphological characteristics. In pathogenicity test, apples were wound-inoculated with conidial suspension and incubated at $20-22^{\circ}C$. The most severe and destructive species was P. expansum. The species caused a decayed area 42-50mm in diameter after 8-10days. Decayed area caused by P. crustosum and P. sp. was 26-32mm and 20-26mm, respectively. This is the first record of P. crustosum, P. italicum and P. sp. from apple in Korea.

        Two Novel Talaromyces Species Isolated from Medicinal Crops in Korea

        Two novel biverticillate Talaromyces species, T. angelicus and T. cnidii, were collected from the medicinal crops Angelica gigas and Cnidium officinale, respectively, in Korea. Phylogenetic analyses with the nuclear ribosomal internal transcribed spacer (ITS) region and the β-tubulin gene as well as morphological analyses revealed that the two species differ from any known Talaromyces species. Talaromyces angelicus is related to T. flavovirens in the phylogeny of the ITS region,but the new species is grouped together with Penicillium liani and T. pinophilus in terms of its β-tubulin phylogeny,and its growth rate on Czapek yeast autolysate differs from that of T. flavovirens. Talaromyces cnidii is phylogenetically similar to T. siamensis, but exhibits differences in the morphologies of the colony margin, metulae, and conidia.

      • Plant disease management using SDHI fungicides and mechanisms of SDHI resistance

        The succinate dehydrogenase inhibitor (SDHI) is a fungicide class containing 12 chemical groups and 24 different compounds and is widely applied to manage plant diseases caused by plant pathogenic fungi. SDHI fungicides inhibit fungal respiration by blocking the ubiquinone-binding site of the mitochondrial succinate dehydrogenase complex composed of four subunits, SdhA, SdhB, SdhC, and SdhD. Currently, SDHI fungicides have been treated to many different crops, fruits, turf, and vegetables and provide effective control of plant diseases. For example, one of the SDHI fungicides, fluopyram, has been used as a seed or foliar treatment for plant disease control and the soybean seed treatment of fluopyram effectively managed soybean sudden death syndrome caused by Fusarium virgulifrome. In addition, this fungicide displayed a good efficacy against the SDHI fungicide boscalid resistant Botrytis cinerea isolates, causing gray mold of grapes. Despite the effective disease control by SDHI fungicides, resistance to SDHI fungicides has been reported in many phytopathogenic fungal species such as Alternaria solani, Botrytis cinerea, Clarireedia homoeocarpa, and Zymoseptoria tritici. Most fungal strains resistant to SDHI fungicides contain mutation(s) in SdhB, SdhC, or SdhD gene and some fungal pathogens exhibited reduced sensitivity to SDHI fungicides harbor overexpression of ATP-binding cassette (ABC) or Major facilitator superfamily (MFS) transporters. In this presentation, the current status of plant disease management by SDHI fungicide applications and molecular mechanisms of SDHI resistance will be discussed.

      • 土壓과 輪壓에 따른 淺層地中理設管의 變形量 分析

        In this paper, indoor soil tank is used to measure the deformation of flexible pipe in the soil mass on the theory of Bossinesq's stress and Marston's Ditch Conduit case. Besides, field compaction test is perfomed for compaction effect of the real construction field. In compression test of the pipe in the soil mass, the flexible pipe has 3.5% of the allowed defomation, and 220kg of the allowed compression load. With theories and soil tank test in this study, the allowed deformation ratio of the flexible pipe is determined to be safe at the depth over 40cm. Consequently, it is considered that the quality of the pipe should be maintained, and the development of the high quality pipe is needed for the safe and economical construction. Especially in compaction, inappropriate compaction could be dangerous.
