        유통 중인 물휴지의 안전성 조사 연구

        The purpose of this study was to investigate and evaluate the safety of the wet tissues. In this study, we analyzed sterilizing preservatives and the presence of harmful substances in 62 wet tissue samples in the market. The contents of preservatives, formaldehyde and methanol were analyzed by HPLC and headspace-GC, respectively. Cetylpyridinium chloride was detected as 7-13 ppm in 5 samples. Sodium benzoate was detected in 46 samples ranging from 200 ppm to 3500 ppm, and 9 ppm of methylparahydroxy benzoate was detected in 1 sample. Propylparahydroxy benzoate was not detected in any samples. 5 ppm of methylchloroisothiazolinone and 140 ppm of methylisothiazolinone were detected in 1 sample. Formaldehyde was detected as 0.0069-1.796 μg/g in 59 samples. Methanol was detected ranging from 2 ppm to 51 ppm in 22 samples, and 4 samples showed more than 20 ppm of the legal limit. The pH of the wet tissues was 4.0 to 8.2. Continuous investigation and monitoring are necessary to ensure safe distribution of products.

        How Do Oroxylin A and Spinosin Exert Their Activities on Cognitive Function?

        Flavonoids are mainly contained in the vegetables and medicinal herbs. Until now, over 5,000 kinds of flavonoid have been identified and their biological activities have been reported. Among them, we are interested in oroxylin A and spinosin because of their specific structures having bulky group at C-6 of ring A. Oroxylin A is contained in the Scutellaria baicalensis and exhibits cognitive enhancing activity as a GABAA receptor antagonist, which is different from those of mainly contained in the S. baicalenis, baicalein or wogonin. Spinosin is isolated from Zizyphus jujuba var. spinosa and mainly studied as a hypnotic or anxiolytic agent because of traditional knowledge about its original herb. As far as we know, the cognitive function of spinosin was first identified by our group. In this review, we discuss how such flavonoids exert their pharmacological activities associated with cognitive function based on the receptor binding study and behavioral studies. Traditional knowledge and reverse pharmacology may be addressed in the research field of phytochemical pharmacology and useful to unveil the secret of phytochemicals.

        향신식물의 잔류농약 실태조사

        This study was conducted to research the status of pesticide residues in a total of 114 herbs and spices obtained from January to October 2020. 341 pesticide residues were analyzed by the multi class pesticide multiresidue methods using GC-MSMS, GC-ECD, GC-NPD, LC-MSMS, LC-PDA, and LC-CAS. As a result of analysis, 36 pesticide residues were found, and detection rate was 31.6%. Of them, seven samples were detected over Maximum Residue Limits (MRLs) and the unsuitable level in pesticide was 6.1%. The herbs and spices exceeding MRLs include coriander (2 times), mint (2 times), basil (once), rosemary (once), and boraye (once). According to an analysis of 341 pesticide residues, 22 pesticides were detected 52 times and 8 pesticides were found to exceed the MRLs. The pesticides exceeding MRLs were ingredients such as etofenprox, flufenoxuron, fluquinconazole, iprodione, lufenuron, paclobutrazol, phenthoate, and spiromesifen. 본 연구는 2020년 1월부터 10월까지 향신식물 114건을 수거하여 잔류농약 실태조사를 실시하였다. GC-MSMS, GC-ECD, GC-NPD, LC-MSMS, LC-PDA, LC-CAS를 이용하여 다종농약 다성분분석법으로 잔류농약 341종을 분석하였다. 조사 결과 36건의 검출이 있었으며 검출률은 31.6%이었다. 이 중 7건은 농약 잔류허용기준을 초과하였으며 부적합률은 6.1%이었다. 농약 잔류허용기준을 초과한 품목은 고수(2건), 민트(2건), 바질(1건), 로즈마리(1건), 서양자초(1건)이었다. 잔류농약 341종을 분석한 결과 22종의 농약 성분이 52회 검출되었고 농약 잔류허용기준을 초과한 성분은 8종이었다. 농약 잔류허용기준을 초과한 성분은 etofenprox, flufenoxuron, fluquinconazole, iprodione, lufenuron, paclobutrazol, phenthoate, spiromesifen 이었다.

        고령화 사회에 따른 한일 장례문화 비교 연구

        최근 한국에서 저출산, 핵가족화, 1인 가족 증가 등의 사회적 병리 현상과 더불어 고령화가 급속히 진행되고 있다. 한국의 고령화 추이는 일본의 고령화 추이와 유사하게 나타나고 있어, 향후 더 심화된 고령화가 나타날 것으로 보인다. 그러나 한국은 아직 고령화 문제에 대한 연구가 매우 부족하여 향후 고령화가 급속도로 진행시 많은 문제가 발생할 것으로 예상된다. 이에 고령화에 대해 대비하고 있는 일본의 장례문화 중 한국의 장례에 도입할 수 있는 적합 한 문화를 비교하여 고령화에 맞춰 대비할 필요가 있다. 일본의 고령화에 따라 나타나는 장례문화는 슈카츠(終活), 직 장(直葬), 가족장(家族葬) 등이 있다. 이 중, 핵가족화 및 1인 가족 증가 등에 따른 소규모 장례문화가 확산될 것으로 예상되어 일본의 ‘가족장(家族葬)’이 한국에 도입하기에 적합할 것으로 보인다. 급속히 고령화되어가는 한국 사회와 고령화에 따른 변화된 장례문화의 제시는 필수적이며, ‘가족장(家族葬)’ 제도의 도입은 향후 발생할 한국 내의 고령화 에 따른 장례문화를 선도하는데 중요한 지침이 될 것으로 기대한다. Social pathologic phenomenon such as low fertility, nuclear family and one person households seems to be on the rise as Korea moves towards being an aging society. We think many trouble is expected to be exit as aging society progresses, but there were few studies about an aging society, especially funeral culture. Appearance of the aging society in Korea similarly follows the course of Japan. So we studied about similar funeral culture of aging society in Japan and prepared for the funeral culture in Korea. This study will suggest a gaze on realistic objective point of view, through the way to illustrate exactly the concept of “funeral for family (家族葬)” among Japanese funeral culture(Syukacheu(終活), Chyokusou(直葬) and funeral for family(家族葬)) and will be expected to be an important guide for leading the funeral culture in Korea occur due to aging in future. And this study will give a chance to ruminate on our’s funeral attitude about the deceased’s dignity.

        복합처방인 승마갈근탕(升摩葛根湯)의 항산화 및 항균효과

        Biological activities such as anti-oxidative and anti-microbial of the Seungmakalgeuntang, a tradi-tional prescription, were evaluated. The electron donating ability of water, ethanol, supercritical fluid and 1,3-butylene glycol extract of Seungmakalgeuntang showed more than 50% at a 100 ppm con-centration. At a 1000 ppm concentration, the superoxide dismutase-like activities of ethanol and supercritical fluid extract of Seungmakalgeuntang showed less than 50%. xanthine oxidase inhibition effect of the supercritical fluid extract showed more than 70% at a 1,000 ppm concentration, which was higher than vitamin C. From the measurement on lipid oxidation, the Fe 2+ chelating abilities of the supercritical fluid extract of Seungmakalgeuntang was more than 60% at a 100 ppm concentration. Also the Cu 2+ chelating abilities of supercritical fluid extract Seungmkalgeuntangwas showed more than 60% at a 500 ppm concentration. Clear zones formed by sample against the human skinresident microflora such as Staphylococcus epidermidis, Staphylococcus aureus and Propionibacterium acne of ethanol and supercritical fluid extract of Seungmakalgeuntang showed the highest among all the extracts tested using a 4㎎/disc. The minimum inhibitory concentration (MIC) against both S. epidermidis and S. aureus showed 2,500 ppm in the extract of the supercritical fluid. 복합처방인 승마갈근탕을 이용하여 화장품 및 식품에 응용하고자 항산화 및 항균효과를 검토하였다. 승마갈근탕의 전자공여능은 모든 추출물 100 ppm에서 50% 이상의 효과를 나타내었고, 특히 초임계 추출물의 경우 500 ppm에서 88%로 가장 높은 효과를 나타내었다. SOD 유사활성은 승마갈근탕 1,000 ppm에서 50% 이하의 다소 낮은 경향을 보였으나, 그 중 에탄올과 초임계 추출물의 경우 각각 46, 45의 효능을 나타내었으며, xanthine oxidase 저해활성의 경우 초임계 추출물 1,000 ppm에 서 76%로 대조군인 vitamin C의 39%보다 높은 효과를 나타내 었다. 승마갈근탕의 지방산패 억제능을 측정하기 위해 산화촉 진제인 Fe2+, Cu2+ 이온을 첨가하여 측정한 결과 초임계 추출물 의 Cu2+ 첨가 시 500 ppm에서 68%의 효과를 나타내었고, Fe2+를 첨가 시 100 ppm에서 60%의 지방산패 억제능을 나타내었다. 승마갈근탕의 clear zone 형성을 관찰한 결과, 추출물 모두 Escherichia coli를 제외한 모든 균주에 대해서 항균 효과가 나타내었다. Staphylococcus aureus와 Propionibacterium acnes의 경우 4 ㎎/disc에서 초임계 추출물에서는 1.45, 2.10 ㎝로 가장 높은 항균 효과를 나타내었고, Staphylococcus epidermidis의 경우, 에탄올 추출물이 1.82 ㎝의 항균효과를 나타내었다.

        과일류의 부위별 잔류농약 함량 비교

        In this study, we tested for pesticide residues in 114 samples representing 20 different types of fruit circulating in Gyeonggido. A total of 341 pesticides were identified via a multi-residue method using GC-NPD, GCECD, GC-MSMS, and LC-MSMS. In whole fruit, pesticides were detected in 48 cases, with a concentration range of 0.0025-0.1663 mg/kg. In the peels, pesticides were detected in 85 cases, with a concentration range of 0.0026-4.7185 mg/kg and a higher average concentration than was found in whole fruit. In the pulp, pesticides were detected in 8 cases, with a concentration range of 0.0021-0.0993 mg/kg. In most of the peels, pesticides were detected below MRL. In the kiwi peel, deltamethrin (with an MRL of 0.05 mg/kg) was detected at 0.1020 mg/kg. Since MRLs are applied to whole fruit, these levels are not unsuitable for ingestion. However, washing is recommended before ingestion to promote healthier and safer inclusion of the peel in the diet. Continuous monitoring of the peel and pulp is indicated. 본 연구에서는 경기도내 유통매장 및 온라인에서 유통중인 과일류 18품목과 박과과채류 2품목 114건의 시료를구입하였다. 과피와 과육을 분리한 후 GC-NPD, GC-ECD, GC-MSMS, LC-MSMS를 이용하여 341종 잔류농약 분석을 진행했다. 과일 전체(과피+과육)를 분석했을 때 48건의시료에서 검출이 되었고 부적합은 없었으며 0.0025- 0.1663 mg/kg의 검출범위를 나타냈다. 과피에서는 85건에서 0.0026-4.7185 mg/kg의 검출범위를 보이며 분리 전과비교했을 때 높은 검출량을 보였다. 과육에서는 8건의 검출이 있었으며 검출범위는 0.0021-0.0993 mg/kg 로 나타났다. 대부분의 과피에서 농산물의 잔류허용기준 이하의 농약이 검출되었고 키위 과피에서 기준 0.05 mg/kg인deltamethrin이 0.1020 mg/kg의 검출량을 보였다. 잔류허용기준은 과일 전체에 대한 기준이기 때문에 섭취에 부적합한 것은 아니지만 더욱 안전하고 건강한 과피 섭취를 위해 세척 후 과일 그대로를 섭취할 것을 권장하며 과피와과육에 대한 지속적인 모니터링이 필요한 것으로 보여진다.

        난소 절제 동물모델을 이용한 경옥고의 갱년기 증후군 개선 효과

        Kyung-Ok-Go (KOK) is a traditional prescription used for debilitating natural aging and post-illness debilitation. KOK has been used in a variety of ways because it strengthens immunity, prevents illness, and helps recovery in case of illness. In particular, recent research has revealed that KOK helps improve memory and cognition. Therefore, in this study, we investigated whether KOK was effective in improving memory decline and depression-state observed during menopause. In the present study, we employed ovariectomized mouse as an animal model for measuring menopausal syndrome. The administration of KOK for 8 weeks, the object recognition memory and working memory were improved in novel object recognition test and Y-maze test. And in the forced swimming test, the immobility time were decreased. Additionally, the expression level of mature brain derived neurotropic factor (mBDNF) was increased by KOK administration in ovariectomized mouse hippocampus. These results suggested that KOK could improve cognitive decline and depression during menopausal period, and it might be come from enhancing expression level of mBDNF in hippocampus.

        출아효모의 세포주기동안 DNA 상해에 의한 발현 유도에 미치는 DPB11 유전자의 영향

        S기 checkpoint기작은 DNA복제 저해나 DNA상해 등에 반응하여, S기 세포주기 정지를 일으키거나 상해 회복에 관련된 유전자들의 전사가 유도됨으로서 진핵세포에서의 유전적인 안정성을 유지한다. 이러한 반응에 대한것ba11 변이주의 결손을 확인하기 위해서, nPB11 (DNA polymerase B possible subunit)유전자의 과다발현 효과에 대해 조사하고, HU (Hydroxyurea)와 MMS (Methyl methanesulfonate)에 대한 감수성 및 DNA상해 물질에 의한 RNR3 (Ribonulectide reductase) mRNA의 전사 유도를 조사하였다. RNR3 mRNA의 전사는 DNA합성 저해에 의해 발생한 스트레스나 화학물질에 의한 직접적 인 DNA상해 등에 의해 유도되어진다. 그 결과, dpb11-1변이주는 DNA상해 물질에 감수성을 나타내었고, RNR3 mRNA전사유도 또한 야생형 균주에 비해 약 40% 정도 감소를 나타내었다. 더욱이 dpb2-1 균주에서도 이와 동일한 결과를 얻었다. 그러므로 DPB2와 DPB11 유전자는 복제에 대한 sensor로서, 복제 정지 요인에 대한 세포주기 반응과 전사 조절에 모두 작용하는 것으로 사료된다. The S-phase checkpoint mechanisms response to DNA damage or inhibition of DNA replication for maintenance of genetic stability in eukaryotic cells. These roles include cell cycle control arrest at S-phase and Iranscriptional induction of repair genes. To characterize the defects of dpbll mutant for both these responses, we examined the over-expression effect of DPBll gene, the sensitivity to HU, MMS, and the transcriptional pattern by DNA damage agent for RNRS mRNA. RNRS transcript is induced in response to a wide variety of agents that either damage D7A directly through chemical modification or induce stress by blocking DNA synthesis. As results, dpbll-1 cells are sensitive to DNA damage agents and the level of RNR3 mRNA is reduced approximately 40% than wild type cells. Moreover, we found the same results in dpb2-1 cells. Therefore, we propose that DPB2 and DPBll act as a sensor of replication that coordinates the transcriptional and cell cycle responses to replication blocks.

        Aster glehni Extract Ameliorates Scopolamine-Induced Cognitive Impairment in Mice

        The leaves of Aster glehni Fr. Schm. (Asteraceae) have been used to treat insomnia in Korea. Insomnia is a common adverse effect of therapeutic agents for Alzheimer's disease (AD), and the control of sleep disturbance may prevent dementia. We hypothesized that the leaves of A. glehni can attenuate cognitive dysfunctions observed in AD. We observed the ameliorating effects of the ethanolic extract of leaves of A. glehni (AG-D) on memory dysfunction through the Morris water maze test, the passive avoidance test, and the Y-maze test. We performed acetylcholinesterase (AChE) activity assay and Western blotting to determine the mechanism of action of AG-D. AG-D significantly attenuated memory dysfunction observed in the above behavior studies and inhibited the activity of AChE. AG-D also increased the levels of phosphorylation extracellular signal-regulated kinase (ERK), cAMP response element-binding protein (CREB), phosphatidylinositol 3-kinase (PI3K), protein kinase B (Akt), and glycogen synthase kinase 3β (GSK-3β) and the expression levels of brain-derived neurotrophic factor (BDNF) in the hippocampi. These results suggest that AG-D ameliorates memory impairments by AChE inhibition and activation of ERK-CREB-BDNF and PI3K-Akt-GSK-3β signaling pathways. Taken together, this study suggests that AG-D could be used as a potential treatment for cognitive dysfunction.
