      • CO₂고정을 위한 화학독립영양미생물 Aeromonas sp. strain JS-1의 분리 및 특성

        The increase of CO2 in the atmosphere by fossil-fuel combustion has been recognized as a major cause of global warming. Thus many studies on CO2 removal from the combustion gas have been performed, and recently the techniques for biological CO2 fixation by photoautotrophs have also been developed, In this study, extensive screening was conducted to obtain micoorganism with high capability in fixing CO2, and its characteristics was investigated. As a result, a chemoautotrophs identified as a Aeromonas sp. strain JS-1 was isolated from the fresh water. The isolated strain used the molecular hydrogen and CO2 as an energy and a carbon source, respectively. Growth characteristics, which will be used in the development of a continuous culture system related in CO2 fixation, were examined in batch cultivation under conditions of various CO2 and NaCl concentrations, temperature, and pH. The results of the examination showed that the isolated strain had a good growth to high CO2 concentration(40%), indicating applicable in removing CO2 discharged from industries.

        절임조건에 따른 돌산갓의 이화학적 특성 변화

        To investigate the optimal salting conditions, the physicochemical properties of Dolsan leaf mustard were compared under various salting times and using various washing methods and storage temperatures at 10% saline. On the other hand, the cell number of lactic-acid bacteria in Dolsan leaf mustard salted under the optimal conditions was also examined during the 50-day storage. The results of the sensory evaluation of the crispness value of the Dolsan leaf mustard treated with stepwise washing after 8h salting showed the optimal conditions for long-term preservation. The pH of the salted Dolsan leaf mustard decreased with the increase in the lactic-acid bacteria during the storage at 4℃. The cutting energy slowly increased during the storage, but the cutting energy of the salted Dolsan leaf mustard treated with stepwise washing remained at the initial values. The color of the salted Dolsan leaf mustard did not change during 40-day storage, but the mustard turned brown after 50-day storage, for which reason it could not be made into Dolsan leaf mustard kimchi. These results could be recommended as the optimal salting conditions and storage time of salted Dolsan leaf mustard for making kimchi.

        흑미 및 흑미미강을 이용한 티백차의 품질특성

        Black rice bran is a potential source of the dark purple anthocyanin pigments. This study examined the feasibility of using black rice and black rice bran as an ingredient in tea-bags. The antioxidative activities of black rice and black rice bran extracts were investigated. The contents of total phenolic compounds in black rice and black rice bran extracts were 452 ± 0.84 and 418 ± 0.60 mg/100 g, respectively. Mixed tea-bags were prepared with green tea, mistletoe and arrowroot flower, and quality characteristics were then analyzed. The soluble solid and a value of the mixed tea-bag extracts increased with increasing black rice bran contents in the mixed tea-bag. From the results of the sensory evaluations, the formula of mixed tea-bags were selected as follows : BR (black rice 1.0 g, black rice bran 2.0 g), BG (black rice 1.4 g, black rice bran 1.0 g, green tea 0.6 g), BM (black rice 1.2 g, black rice bran 1.0 g, mistletoe 0.8 g), and BAF (black rice 1.2 g, black rice bran 1.0 g, arrowroot flower 0.8 g). These results suggest that mixed tea-bag processed with black rice and black rice bran has the potential to become a commercial product.

        천연추출물의 발모촉진 및 탈모 예방에 미치는 영향

        본 연구는 검은콩, 은행, 녹차를 포함한 천연추출물이 두발의 발모 촉진과 두개피부에 미치는 영향을 조사한 연구이다. 연구 방법은 검은콩, 건조은행, 녹차잎을 2 : 1 : 1의 비율로 10 배수의 증류수에 침지하고 100℃에서 2시간 열수추출한 후, Eclipse Plus C18(4.6×250 mm, 5 μm)를 사용하여, 이동상은 초순수로 희석한 70% Methanol을 이용하였다. 추출물의 항균 활성은 Malassezia Furfur를 이용하였으며 항산화 활성은 DPPH에 대한 전자공여능 측정으로 평가하였다. 인간 모유두세포 및 섬유아세포의 증식 활성은 96 Well Plate에 세포를 3×104 (100 ㎕/well)로 분주하고 24시간 배양한 후 MTT Assay를 실시하였다. 천연추출물을 농도별로 투여하고 배양이 끝나면 DMSO 용액을 넣고, Plate Shaker에서 30분 추출 후, 570 nm에서 ELISA를 이용하여 흡광도를 측정하였다. 연구 결과 항균 효과 조사에서 천연추출물의 모낭염 유발균인 Pityrosporum Ovale와 지루성피부염 유발균인 Malassezia Furfur에 대해 성장억제를 확인하였고, DPPH 라디컬 소거능 측정을 통해 항산화 활성을 확인하였다. 또한, 인간 모유두세포(HFDPC)와 섬유아세포(NIH3T3)의 세포증식 효과에 대한 실험에서 천연추출물 0.10% 농도에서 모유두세포의 40% 이상 증식 효과, 섬유아세포는 0.02% 농도에서 약 10% 증식 효과가 있음을 확인하였다. 이들 3종의 천연추출물은 발모 촉진 및 탈모 예방 효과를 목적으로 하는 두개피부 및 모발보호 제품개발을 위한 기초자료로 활용될 수 있음을 시사하는 바이다. This study examined the effects of natural extracts including black bean, Ginkgo biloba L. fruits, and green tea, on hair growth and scalp improvement. For the research method, black beans, dried ginkgo biloba, and green tea leaves were immersed in 10 times distilled water at a ratio of 2 : 1 : 1, extracted with hot water at 100 ° C for 2 hours, and Eclipse Plus C18 (4.6 × 250 mm, 5 μm) was used. As the mobile phase, 70% methanol diluted with ultrapure water was used. The antibacterial activity of the extract was evaluated using Malassezia furfur, and the antioxidant activity was evaluated by measuring the electron donating ability to DPPH. For proliferative activity of human dermal papilla cells and fibroblasts, 3×104 (100 μl/well) of cells were aliquoted in a 96 well plate and cultured for 24 hours, followed by MTT assay. After administration of natural extracts by concentration, and after incubation, DMSO solution was added, and after 30 minutes extraction on a plate shaker, absorbance was measured using ELISA at 570 nm. Antimicrobial activity of natural extracts on Pityrosporum ovale and Malassezia furfur were investigated. In antioxidative activities of natural extracts, DPPH radical scavenging activity was about 62.40±4.8 % at 5.0 mg/mL. On the other hand, cell proliferation in NIH3T3 cells and HFDPC cells were determined. Natural extracts promoted the proliferation of NIH3T3 cells, a human epidermal cells by about 10 % at 0.02 % concentration. HFDPC cells, a hair follicle dermal papilla cells were significantly stimulated in cell proliferation by above 40% at 0.10% concentration. These results suggest that natural extracts have antimicrobial activity, antioxidant effects, induce cell proliferation, and could be used for hair loss prevention and scalp improvement.

        여성 골다공증의 위험요인에 관한 환자-대조군 연구 - 서울지역 1개 병원 내원환자를 중심으로 -

        Until now there are few available epidemiologic data of osteoporosis in Korea, and the severity of osteoporosis-related health problem has not been widely recognized yet. But the numbers of the old people are increasing in Korea, and in 2000, the proportion of people over 65 will be up to about 6.8% of total population. Therefore, osteoporosis, one of the most common metabolic bone disease among the old people, will be one of the most important public health problem. On this background this study was performed to find out risk factors of the development of osteoporosis in Korean women through case-control approach. The subject of this study were selected among the women who visited menopause clinic in one general hospital in Seoul and were checked bone density from Sep. 1988 to Sep. 1993. Those who were diagnosed to have hypertension, diabetes mellitus, thyroid disease, breast disease, or liver disease, which are thought to influnce bone density, were excluded. Also excluded those who are age-unknown. Finally the subjects were 2,139 women aged between 18 and 79. We operatively defined patient group as those whose bone density is below $1.039/cm^2$, 90% of average bone density of women of 4th decade who visited the same hospital. And we defined control group as whose bone dendity is above $1.15g/cm^2$. We randomly selected 201 women from the patient group and 202 from the control. As independent variables we chose age, menarche age, menopause age, menopause type, the number of siblings, the number of pregnancies, body mass index, taking oral pill or not, feeding type, and educational state. Multiple logistic regression analysis was done to see the influence of these variables on the risk of osteoporosis. Results are as follows; 1. menopausal status was statistically significant risk factor to all women irrespective of her age, while obesity and later menopause age were found to be statistically significant protective factors. 2. The more siblings and pregnancies, the greater tile risk of osteoporosis, but these factors were not statistically significant. This result is not consistent with other studies. Further studies are strongly needed.
