        단삼음(丹蔘飮)이 고지혈증(高脂血症)에 미치는 영향(影響)

        Objectives : In order to study effects of Dansamyeum on Hyperlipidemia which causes Hypertension, Arteriosclerosis, Cerebral vascular disease and Ischemic heart disease Methods : The changes of serum total lipid, triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol, body weight and organ weight were observed after the liquid extracts of Single-dosage Dansamyeum and Double-dosage Dansamyeum were administered p.o to the Hyperlipidemic rats induced by 1% cholesterol diet for 10, 20 and 30 days. Results : The body weight was significantly decreased in Double-dosage Dansamyeum group for 20 and 30 days compared with control group. The contents of total lipid were significantly decreased in Single-dosage Dansamyeum group for 20 days, but Double-dosage Dansamyeum group did not show any significant change compared with control group. The contents of total cholesterol were significantly decreased in Single-dosage Dansamyeum group for 20 days, and Double-dosage Dansamyeum group showed significant changes for 10 and 20 days compared with control group. The contents of triglyceride were significantly decreased in Single-dosage Dansamyeum group for 30 days, and Double-dosage Dansamyeum group showed significant changes for 10 days compared with control group. The contents of HDL-cholesterol were significantly increased in Single-dosage Dansamyeum group and Double-dosage Dansamyeum group for 10 days compared with control group. The contents of LDL-cholesterol were significantly decreased in Single-dosage Dansamyeum group for 10, 20 and 30 days, and Double-dosage Dansamyeum group showed significant changes for 10, 20 and 30 days compared with control group. The liver weight was significantly decreased in Single-dosage Dansamyeum group and Double-dosage Dansamyeum group compared with control group. The kidney and testis weight was significantly decreased in Single-dosage Dansamyeum group compared with control group. The spleen weight was significantly decreased in Double-dosage Dansamyeum group compared with control group. Conclusions : It was thought that Single-dosage Dansamyeum and Double-dosage Dansamyeum could be applied effectively to the Hyperlipidemia.

        청상견통탕(淸上甄痛湯))이 고지혈증에 미치는 영향

The contents of total lipid were significantly decreased in Double-dosage Chungsanggyuntong-tang group for 20, 30 days, but Single-dosage Chungsanggyuntong-tang group did not show any significant change compared with control group. The contents of total cholesterol were decreased in Single-dosage Chungsanggyuntong-tang group and Double-dosage Chungsanggyuntong-tang group, but did not show any significant changes compared with control group. The contents of triglyceride were significantly decreased in Double-dosage Chungsanggyuntong-tang group for 10, 30 days, but Single-dosage Chungsanggyuntong-tang group did not show significant changes compared with control group. The contents of HDL-cholesterol were significantly increased in Single-dosage Chungsanggyuntong-tang group for 10 days compared with control group. The contents of LDL-cholesterol were significantly decreased in both Chungsanggyuntong-tang group for 10 and 30 days compared with control group. The liver and spleen weight were significantly increased in control group compared with normal group. The liver weight was significantly decreased in Single-dosage Chungsanggyuntong-tang group compared with control group. The liver, spleen and kidney weight were significantly decreased in Double-dosage Chungsanggyuntong-tang group compared with control group. Conclusions : It was thought that Single-dosage Chungsanggyuntong-tang and Double-dosage Chungsanggyuntong-tang could be applied effectively to the Hyperlipidemia.

        대시호탕가미방(大柴胡湯加味方)이 고혈압(高血壓) 및 고지혈증(高脂血症)에 미치는 영향(影響)

        The aim of the study was the experiment of the effect that Kami-Daesihotang had on the essential hypertension and hyperlipidemia. Rats were orally administered with Kami-Daesihotang for 30days and the constituent of the plasma and serum were analysed at the 10th, 20th and 30th day from the first day of experiment, respectively. The heart rate, blood pressure, plasma renin activity, plasma level of aldosterone, catecholamine, sodium and angiotensin II were measured after an oral administration of Kami-Daesihotang in SHR. In addition, serum levels of total cholesterol, triglyceride, HDL-cholesterol, LDL-cholesterol and total lipid were measured with cholesterol-fed rats. The results were summarized as following ; 1. Single-dosage Kami-Daesihotang & double-dosage Kami-Daesihotang remarkably decreased the blood pressure in SHR. 2. Double-dosage Kami-Daesihotang were recognized as having the effect on the decreased of the pulse rate in SHR. 3. Plasma renin activity was significantly decreasd in SHR after single-dosage Kami-Daesihotang & double-dosage Kami-Daesihotang treatment. 4. Double-dosage Kami-Daesihotang considerably reduced the plasma angiotensin level in SHR. 5. Noticeable decreased of plasma norepinephrine level was showed in SHR, after single-dosage Kami-Daesihotang & double-dosage Kami-Daesihotang treatment. 6. Single-dosage Kami-Daesihotang & double-dosage Kami-Daesihotang noticeable reduced body weight in hyperlipidemia rats which had fed with 1% cholesterol. 7. Single-dosage Kami-Daesihotang & double-dosage Kami-Daesihotang had a significantly decreasing effect on serum total cholesterol in hyperlipidemia rats which had fed with 1% cholesterol. 8. Serum triglyceride level was importantly decreased in hyperlipidemia rats which had fed with 1% cholesterol, after single-dosage Kami-Daesihotang & double-dosage Kami-Daesihotang treatment. 9. Remarkable decreased of serum low density lipoprotein cholesterol level was found in hyperlipidemia rats which had fed with 1% cholesterol, after single-dosage Kami-Daesihotang & double-dosage Kami-Daesihotang treatment. 10. Double-dosage Kami-Daesihotang was showed a significantly decreasing effect on serum total lipid level in hyperlipidemia rats which had fed with 1% cholesterol. 11. Single-dosage Kami-Daesihotang noticeably reduced organ weight of liver, kidney, spleen and testis in hyperlipidemia rats which had fed with 1% cholesterol. Double-dosage Kami-Daesihotang significantly decreased organ weight of liver, kidney and spleen in hyperlipidemia rats. These Findings suggest a possible anti-hypertensive and hyperlipidemic effect of Kami-Daesihotang.

        분심기음(分心氣飮)이 고혈압 백서와 인간유래 혈관내피세포주(ECV 304)에 미치는 영향에 대한 연구

        Object : This study was designed to research whether the protection and inhibitory effects of cardiovascular diseases in L-NAME induced rat or ECV 304 cell lines through the Cell morphological pattern, Tunel assay, LDH activity, heart rate, blood pressure and immunohistochemistric analysis by Boonsimgieum water extract Methods : Nitric oxide(NO) play an important role in normal and pathophysiological cells including as a messenger molecule, neurotransmitter, microbiocidal agent, or dilator of blood vessels and artheriosclerosis, hypertension, myocardial infarction, respectively. Endothelial cell products can modulate the magnitude of a response to a vasoconstrictor, as evinced by the greater constriction after endothelium removal or NO synthesis blockade. To investigate that Boonsimgieum in the potential contribution of the levels of nitric oxide generated by endothelial nitric oxide synthase (eNOS) and the mechanisms of protection against NG-nitro-L-arginine methyl ester (L-NAME), human ECV 304 cells, which normally do not express eNOS, were expressed by L-NAME. L-NAME stimulated rat or cells were found to be resistant to injury and delayed death following the Boonsimgieum. Inhibition of nitric oxide synthesis abolished the protective effect against L-NAME, thrombin and collagen exposure. Interestingly, such effects have been observed during stimulation with agents such as phenylephrine and KCl on L-NAME mediate rats, were damaged by the NOS inhibitor L-NAME. Result : As the result of this study, In group, the anti-apoptosis and necrosis in the cardiovascular system have a potential capacity for prevented, protected and treating the diseases of cardiovascular system, against the necrosis of rat and ECV 304 cells with Caspase 3 and calpain expression by L-NAME is promoted. Conclusion : these results demonstrate neuroprotective and memory enhancing effects of ZIBU, suggesting its beneficial actions for the treatment of AD.

        CT105로 유도된 신경모세포종 세포주에서 지미탕(指迷湯)의 항치매효과

        Alzheimer's disease(AD) is a geriatric dementia that is widespread in old age. In the near future AD may be the biggest problem in public health service. Although a variety of oriental therapies in the study of Jimitang have been traditionally utilized for the treatment of AD, their pharmacological effects and active mechanisms have not been fully elucidated. This study in an investigation of effects of Jimitang on apoptotic cell death induced by CT105 overexpression in SK-N-SH neuroblastoma cell lines. DNA fragmentation, neurite outgrowth assay and LDH activity assay were examined. The regeneratory and inhibitory effects on Alzheimer's disease in pCT105-induced neuroblastoma cell lines by Jimitang water extract were examined. Findings from these experiments have shown that Jimitang inhibits the synthesis or activities of CT105, which has neurotoxicities and apoptotic activities in cell lines. In addition, pretreatment of $Jimitang(>50\;{\mu}g/mL\;for\;12\;hours)$ partially prevented CT(105)-induced cytotoxicity in SK-N-SH cell lines, and were inhibited by pretreatment. $Jimitang(>50\;{\mu}g/mL\;for\;12\;hours)$ repaired CT(105)-induced neurite outgrowth when SK-N-SH cell lines were transfected with CT(105). Results of this study show that. in the Jimitang group, the apoptosis in the nervous system in inhibited, the repair against the degerneration of neuroblastoma cells by CT105 expression is promoted. In addition, Jimitang was found to inhibit DNA fragmentation induced by CT105 overexpression, and promote neurite outgrowth. These findings suggest that Jimitang is beneficial for the treatment of AD.

        전탕시간(煎湯時間)에 따른 생대황(生大黃) 및 주대황(酒大黃)이 어혈병태모형(瘀血病態模型)에 미치는 영향(影響)

        In order to investigate the effects of Alcohol-Steamed Rhei Rhizoma and Row Rhei Rhizoma on varied extract time in both endotoxin-induced blood stasis model(hereafter Endotoxin Model) and hydrocortisone acetate-induced blood stasis model (hereafter HA Model), Half of rats were treated with endotoxin(0.4mg/kg, single Ⅳ, into caudal vein) for Endotoxin Model. Thereafter, they were orally administrated water extract of Alcohol-Steamed Rhei Rhizoma or Row Rhei Rhizoma, which were boiled during 30, 60, 120 minute, respectively. Finally, the number of platelet, fibrinogen, prothrombin time, hematocrit, the number of RBC and WBC were measured after sacrifice. The remainder rats were treated with hydrocortisone acetate(10mg/kg, daily IM for 7 days into the muscular rump) for HA significantly decreased. Together, they were orally administrated for 7 days water extract of Alcohol-Steamed Rhei Rhizoma or Row Rhei Rhizoma that were boiled above methods Finally, the number of platelet, fibrinogen, prothrombin time, hematocrit, the number of RBC and WBC were measured after sacrifice. The results were summarized as follows : 1. The number of platelet was significantly increased in boiled water extract for 30 min of Row Rhei Rhizoma group as compared with that of control group in Endotoxin Model. 2. Fibrinogen level was significantly increased in all administration groups as compared with that of control group in Endotoxin Model. It was significantly increased in all administration groups except boiled water extract for 30 min of Alcohol-Steamed Rhei Rhizoma group as compared with that of control group in HA Model. 3. Prothrombin time was significantly shortened in boiled water extract for 60 min of Alcohol-Steamed Rhei Rhizoma group and boiled water extract for 120 min of Alcohol-Steamed Rhei Rhizoma group as compared with that of control group in Endotoxin Model It was significantly shortened all administration groups as compared with that of control group in HA Model. 4 Hematocrit was significantly increased in all administered groups except boiled water extract for 60 min of Alcohol-Steamed Rhei Rhizoma group and boiled water extract for 30 min of Row Rhei Rhizoma group as compared with that of control group in Endotoxin Model. It was significantly increased in all administration groups as compared with that of control group in HA Model. 5. The number of RBC was significantly decreased in boiled water extract for 60 min of Alcohol-Steamed Rhei Rhizoma group, boiled water extract for 120 min of Alcohol -Steamed Rhei Rhizoma group and boiled water extract for 30 min of Row Rhei Rhizoma administered group in Endotoxin Model. It was significantly increased boiled water extract for 30 min of Row Rhei Rhizoma group and boiled water extract for 60 min of Row Rhei Rhizoma group in HA Model as compared with data of control group. 6 The number of WBC was significantly decreased in all administered groups except boiled water extract for 30 min of Alcohol-Steamed Rhei Rhizoma group and boiled water extract for 60 min of Alcohol-Steamed Rhei Rhizoma group as compared with that of control group in HA Model.

        온담탕이 고혈압 백서와 인간유래 혈관내피세포주(ECV 304)에 미치는 영향

        Nitric oxide(NO) play an important role in normal and pathophysiological cells including as a messenger molecule, neurotransmitter, microbiocidal agent, or dilator of blood vessels and artheriosclerosis, hypertension, myocardial infarction, respectively. To investigate that Ondamtang in the potential contribution of the levels of nitric oxide generated by endothelial nitric oxide synthase (eNOS) and the mechanisms of protection against L-NAME, human ECV304 cells, which normally do not express eNOS, were expressed by L-NAME. L-NAME stimulated rat or cells were found to be resistant to injury and delayed death following the Ondam-tang. Inhibition of nitric oxide synthesis abolished the protective effect against L-NAME, thrombin and collagen exposure. Interestingly, such effects have bee observed during stimulation with agents such as KCl on L-NAME mediate rats, were damaged by the NOS inhibitor NG-nitro-L-arginine methyl ester (L-NAME). Cardiovascular diseases is one of the blood vessels and renin-angiotensin system dynfunction. So we studied on herbal medicine that have a relation of vessels endothelium necrosis. In Oriental Medicine, Ondam-tang has been used for disease in relation to cardiovascular system. We studied on the protection and inhibitory effects of cardiovascular diseases in L-NAME induced rat or ECV304 cell lines through the Cell morphological pattern, Tunel assay, LDH activity, heart rate, blood pressure and immunohistochemistric analysis by Ondam-tang. As the result of this study, In group, the anti-apoptosis and necrosis in the cardiovascular system have a potential capacity for prevented, protected and treating the diseases of cardiovascular system, against the necrosis of rat and ECV304 cells with eNOS and calpain expression by L-NAME is promoted.

        CT105로 유도된 인간신경아세포종 세포주에서 전매단의 항치매 효과

        Alzheimer's disease(AD) is a geriatric dementia that is widespread in old age. In the near future AD will be the biggest problem in public health service. Although a variety of oriental prescriptions in study Jeonmaedan have been traditionally utilized for the treatment of AD, their pharmacological effects and action mechanisms have not yet fully elucidated. It has been widely believed that A${\beta}$ peptide devided from APP causes apoptotic neurotoxicity in AD brain. However, recent evidence suggests that CTl05(carboxy terminal 105 amino acid peptide fragment of APP) may be an important factor causing neurotoxicity in AD. In addition, AD is one of brain degeneration disease. So we studied on herbal medicine that have a relation of brain degeneration. In Oriental Medicine, Jeonmaedan has been used for disease in relation to brain degeneration. As the result of this study, in Jeonmaedan the apoptosis in the nervous system is inhibited, the repair against the degerneration of SK-N-SH cell lines by CT105 expression is promoted. So Jeonmaedan may be beneficial for the treatment of AD.
