        압축잔류응력장을 전파하는 피로균열의 개구거동의 유한요소법을 이용한 해석적 검토

        The finite element analysis was performed for the cracks existing in residual stress fields in order to investigate the effects of configuration of residual stress distribution to the fatigue crack opening behaviour. And the variation of stress distributions adjacent to the crack caused by uploading was examined. The finite element model with contact elements for the crack plane and plane stress elements for the base material and the analytical method based on the superposition principle to estimate crack opening behaviour and the stress distribution adjacent to the crack subjected to uploading were used. The results of the analysis showed that crack opening behaviors and variations of stress distribution caused by uploading were changed depending on the configuration of residual stress distribution. When the crack existed in the region of compressive residual stress and the configuration of compressive residual stress distribution were inclined, a partial crack opening just behind of a crack tip occurred during uploading. Based on the above results, it was clarified that the crack opening behaviour in the residual stress field could be predicted accurately by the finite element analysis using these analytical method and model.

        In Vitro Propagation of Myco-heterotrophic Gastrodia elata

        Gastrodia elata must establish symbiotic associations with different compatible fungi at seed germination and during vegetative growth. One of the major problems associated with its field production is the degeneration of tubers, which is mainly caused by multiple regeneration via vegetative propagation using immature tubers. To overcome this problem, an in vitro asexual propagation method via artificial pollination was developed. Seeds were first germinated on medium containing a leaf-disc from four different Quercus species, previously infected with Mycena osmundicola. Among the four species tested, Q. acutissima was determined to be the best organic resource and the germination rate reached up to 40.4% when a leaf-disc (2 × 2 cm) was provided. Subsequent infection with Armillaria mellea allowed the protocorms to further develop into immature tubers, which can be directly used for field production. In this study, we established an in vitro symbiotic propagation system of G. elata, which might be an efficient way to prevent degeneration of G. elata tubers and the reduction of yield in the field.

        In vitro symbiotic germination of myco-heterotrophic Gastrodia elata by Mycena species

        Symbiotic germination was analyzed in themyco-heterotrophic orchid Gastrodia elata. Seeds wereplaced on water agar medium containing a leaf-disc ofQuercus accutissima that had been previously inoculatedwith one of six different Mycena species. Among the sixfungi, KFRI1212 (HQ662845) and KFRI2121 (HQ662846)germinated 60.1 and 47.0 % seeds, respectively, whileothers germinated less than 3.5 %. Although KFRI1212induced a significantly higher germination rate thanKFRI2121, initiation of protocorm development wasinduced much faster by KFRI2121 than by KFRI1212. Molecular identification and phylogenetic analysis showedthat the two fungi belonged to one clade that includesMycena chlorophos, M. amicta and M. tenerrima, suggestingthat seed germination of G. elata depends on anarrow taxonomic range of fungi. In conclusion, this studynot only showed fungal preference of G. elata for seedgermination but also confirmed molecular identities ofmycorrhizal fungi for the first time, which will allow us tobetter understand the process of symbiotic events at thegermination stage of G. elata.

        치주 큐렛의 절단 연 마모도 평가

        The quality of periodontal instrument cutting edge is a basic element of effective root planing procedure. Using instruments, the sharp edge is changed into blunt or beveled edge. With the blunt instrument, the periodontal treatment can't be carried into accuracy and effective. The study on the wear of periodontal curet is insufficient, there are few publications about the change of sharpness of cutting egde after using instrument and a certen reports were published on the study of scanning electron microscope(SEM) examination. In this study, to declare the number of strokes for sharpening of instruments, the changes of cutting edge is measured by the clinical methods, tactile sensitivity examination and refraction light-white line test after scaling strokes and root planing strokes. SEM test was added for defined the changes of cutting edges. The 7/8 Gracey curets that have been never used was tested. Maxillary molars which were extracted from the School of Dental Medicine, Dankook University was used. Subjected teeth had attachment loss more than 6 mm in bucca-lingual surface and sufficient calculus of a band type in cervical area. The strokes of curet were executed 3, 5, 7, 9, 11, 13 times on scaling stroke and 10, 15, 20, 25, 30, 35 times on root planing stroke. A resident has periodontal experience over 3 years carried out the clinical examinations those tactile sensitivity examination and refraction light-white line test 5 times. The case there being tactile sensitivity certenly is 2, the case being felt tactile sensitivity is 1, and the case there not being tactile sensitivity is 0. The visual examination was recorded as following. The case that refracted white line is not recognised is 2, the case that uncerten is 1, and the case that acknowledged is 0. The results were obtained as follows. 1. After scaling strokes, the tactile sensitivity was reduced after 11 strokes and disappeared in 13 strokes. 2. In tactile sensitivity after root planing procedures, sensitivity was reduced after 25 strokes and disappeared in 35 strokes. 3. In case of visual examination, the detection of refracted white line was increased after 9 strokes of scaling procedures and the accuracy of wear wasn't showed after root planing procedures. 4. In SEM, metal projection was observed on new periodontal curet cutting edge and it was disappeared after scaling procedures. 5. In SEM, the cutting edge was showed changing linear into an aspect of the surface after 5 strokes of scaling procedures and 10 strokes of root planing procedures and showed beveled edge in 11 strokes of scaling procedures, 25 strokes of root planing procedures. The results of 3-type examination indicated that the sharpening of curet should be performed after 11 strokes of scaling procedures and 25 strokes of root planing procedures.

        용접잔류응력장에서 피로균열의 전파에 따른 잔류응력 재분포에 대한 해석적 평가

        An investigation was performed to predict residual stress redistribution for the crack propagation initially through tensile residual stress field. The analytical method, which is based on Dugdale model by finite element analysis using elastic analysis method considering the superposition principle, was proposed to estimate the redistribution of residual stress caused by crack propagation. The various aspect of distribution of residual stress caused by crack propagation was examined based on the configuration change of specimen. The analysis results show that the aspect of redistribution of residual stress caused by crack propagation depends on the width of the specimen provided that the initial distribution of residual stress is identical.

        In this study. the procedures to predict the behaviour of a fatigue crack is studied by the finite element method. The crack is closed by the residual stresses or the plastic zone and increasing tensile load. Former studies show that a crack propagates after the opening of the crack. So the mechanical behaviour of the ligament is important and investigated in this study. A general purpose finite element package. ABAQUS is used to predict the behaviour of the ligament. Stresses and strains of the ligament under various states of crack faces and tips are analyzed.

        Quantitative Effects of Various Tree Species on Tuber Growth and Pharmacological Compositions of Gastrodia elata

        Gastrodia elata is a myco-heterotrophic orchid that has been used in traditional medicine to treat number of human illnesses. It establishes symbiotic associations with compatible mycorrhizal fungi, such as Mycena species and Armillaria mellea, depending on its developmental stages. G. elata tubers contain two major pharmacological substances; gastrodin (GA) and ergothioneine (ERG). Here we investigated whether the use of different tree species could enhance the growth of G. elata tubers, or of the content of GA or ERG. A. mellea infected all 30 tree species but its infection levels varied among species. However, only 14 species stimulated the growth of G. elata immature tubers; Ulmus davidiana produced the biggest tubers (7.8 g FW) while Abies holophylla generated the least significant changes on the growth of tubers reaching their weights to 0.9 g. In contrast to the tuber growth, the infection levels of A. mellea in those trees were similar, indicating that the colonized levels of A. mellea on different tree hosts were not correlated with the yield of G. elata tubers. Interestingly, the weights of immature tubers developed on 14tree species were negatively correlated with the levels of ERG, but not with those of GA. In conclusion, our study suggested that the use of U. davidiana instead of Quercus species, which has been used for traditional cultivation of G. elata, might enhance the yields of G. elata tubers, but reduce the ERG levels while maintaining the GA levels.
