It was proved that the cell growth followed a photo-inhibition model in light-limited turbidostat cultivation, having 1.06 (1/h) of maximum specific growth rate and 0.00094(kcal/$cm^2$/h) and 0.063 (kcal/$cm^2$/h) as half saturation and light inhibition constants, repectively. ${\beta}$-carotene production showed a growth related porcess. And the activation energy of Dunaliella salina was roughly estimated as 12.36 (kcal/mole) in employing Arrhenius relationship. It should also point out that relatively much porduction of ${\beta}$-carotene was observed at hight light intensity with yieding 1.04 (mg-carotene/g-dry cell/day) of specific product production rate while the cell growth was decreased. The optimal conditions of producing ${\beta}$-carotene in turbiodostat cultivation were as follows: $7.5{\times}10^{-3}$(kcal/$cm^2$/h)of light intensity, 2 (mM) and 50(mM) of nitrate and sodium bicarbonate concentrations and 100(ml/h) of $CO_2$ flow rate.

8 X 10$^{6}$(viable cells/ml) of maximum cell density and 9000(IU/ml) of $\gamma$-IFN production were obtained at 55(ml/hr) of a perfusion rate by cultivating HSF cells using a moving membrane aeration bioreactor. This system proves to be an efficient culture process by maintaning 90% of viable cells during the whole cultivation periods. The metabolic molar quotient of glucose to lactate was 0.81 for overall ranges of glucose consumed while the evolution of ammonia was not linearly related to the consumption of glutamine. Low molar conversion ratio was observed in low consumptions of glutamine and high molar conversion ratio in high comsumptions. It also shows that the glutamolysis plays important role in the steady state conditions by evolving larger quantities of ammonia than lactate. At the above of 50 rpm, which is the optimum agitation speed for this bioreactor, the cell growth was severely affected while the IFN production was less decrea- sed, maintaing 1.5 X 10$^{-3}$(IU/cell/day) specific IFN production rate. The cumulatvie $\gamma$-IFN production was 7.2 X 10$^{8}$(IU) for 70 days of the cultivation, which yields 1 X 10$^{7}$ (IU/day) of IFN production rate. Therefore, a commercial production of $\gamma$-IFN by this culture process can be achievable by maintaining the above IFN productivity in a scaled-up culture system.
1-Deoxynojirimycin을 생산하는 Bacillus subtilis S10 배양액의 혈당강하 효과

본 연구는 α-glucosidase의 강력한 효소저해제인 1-deoxynojirimycin(DNJ)을 고효율로 생산하는 균주의 선발 및 동정, 배양최적화 및 균 배양액의 혈당강하효과 등에 관하여 조사하였다. 먼저 토양으로부터 α-glucosidase에 대해 강력한 저해능을 나타내는 9균주를 대상으로 이 중에서 DNJ 생산성이 가장 우수한 한 개 균주를 최종 선발하였고, 이 균주를 16S rDNA 염기서열분석으로 균주동정을 하여 Bacillus subtilis S10이라 명명하였다. 본 균주의 배양액을 이온교환수지법(Amberlyst 15 H? form, Dowex 1×2-100 OH? form, Amberlite CG-50 NH₃? form)에 의해 DNJ를 정제하고, 정제된 DNJ를 LC-MS로 분석한 결과 표준 DNJ와 동일한 물질임을 확인하였다. DNJ 대량생산을 위한 S10균주의 배양최적탄소원 및 최적질소원과 이들의 최적농도를 조사한 결과 각각 1% galactose, 1.6% polypeptone임이 밝혀졌으며, 확립된 최적화 조건에서의 DNJ 생산량은 0.75 g/L이었다. 고혈당 유도 마우스를 대상으로 각종 혈당강하제의 효과를 알아보기 위해 혈당치를 비교한 결과 무처리구의 혈당치 510±10.9 ㎎/dL에 비해 acarbose 처리구는 139.4±33.1 ㎎/dL, 누에분말 처리구는 209.1±19.6 ㎎/dL, 그리고 S10 균주 배양액 처리구는 208.6±39.8 ㎎/dL로써 무처리구에 비해 각각 72.7%, 59.0%, 그리고 59.1%의 혈당 억제율을 나타냈다. 이상의 연구결과 S10균주 배양액은 누에분말과 같은 수준의 고혈당 억제 및 완화효과가 있음이 확인되었으며 향후 S10균주의 대량생산을 통해 식후혈당조절을 위한 건강기능식품 개발이 기대된다 하겠다. 1-Deoxynojirimycin (DNJ) is a strong α-glucosidase inhibitor which inhibits hyperglycemia in animals. To select the Bacillus strains highly producing DNJ, 4,000 strains were isolated from soil and grain samples. By the inhibitory activity against α-glucosidase, nine Bacillus strains were selected and then identified by 16S rDNA sequencing. B. subtilis S10 was finally selected as the best strain for the production of DNJ. Various carbon sources and nitrogen sources in culture medium were evaluated for the highest production of DNJ. As the results, the optimized concentration of carbon source and nitrogen source was 1.0% galactose and 1.6% polypeptone and the concentration of DNJ produced was 0.75 g/L. The effect of culture supernatant of B. subtilis S10 on lowering blood glucose level was investigated in streptozotocin (STZ)-induced diabetic mice model. Mice were randomly assigned to control group (saline) and three test groups such as acarbose group, silkworm powder group and B. subtilis S10 group. After eight-week oral feeding, blood glucose levels of the B. subtilis S10 and silkworm powder groups were respectively 209.1±19.6 ㎎/dL (59.1%) and 208.6±39.8 ㎎/dL (59.0%) lower than 510±10 ㎎/dL of the control group. These results indicated that the culture supernatant of B. subtilis S10 was able to reduce the blood glucose level in STZ-induced diabetic mice.
1-Deoxynojirimycin 생산 균주 Bucillus subtilis MORI 3K-85의 단백질 분석

In our previous study, we isolated and characterized a 1-deoxynojirimycin (DNJ)-producing bacterium, Bacillus subtilis MORI, from chungkookjang, a Korean traditional food. B. subtilis MORI was subjected to γ-irradiation and the resulting bacteria were screened for increased DNJ production. A mutant was identified that produced 7.6 times more DNJ and named B. subtilis MORI 3K-85. In this study, the protein profiles of both strains were compared by one-dimensional and two-dimensional gel electrophoresis (1-DE and 2-DE, respectively) under both native and denaturing conditions. The 1-DE native-PAGE and 1-DE SDS-PAGE analyses identified 5 and 7 bands, respectively, that were found at higher concentrations in B. subtilisMORI 3K-85 than in B. subtilis MORI. Similarly, 2-DE analyses identified 20 protein spots which were found at higher concentrations in B. subtilis MORI 3K-85. The peptide mass profiles of these 20 proteins were analyzed by MALDI-TOF and compared with peptide sequences of B. subtilis and B. amyloliquefaciens in the MASCOT database. This screening suggested that three dehydrogenases, an aldolase, a synthetase, an isomerase, a reductase, and a peroxidase are elevated in B. subtilis MORI 3K-85. Based on this data, one or more of the elevated 8 enzymes might be related to the DNJ biosynthetic pathway.
바실러스 발효대사물이 함유된 신규 칼슘보충용식품의 성장기 쥐 장골 성장촉진효과

기존 칼슘보충용식품의 성장촉진효능을 증진시키기 위하여 칼슘흡수 관여 물질인 γ-PGA를 다량 생산하는 Bacillus subtilis SE4 균주를 선발하였다. 선발된 B. subtilis SE4의 발효대사물을 기존 칼슘보충용식품에 첨가하여 성장기 수컷 흰쥐에 28일간 경구투여 한 결과 발효대사물이 첨가된 신규칼슘보충용식품군에서 혈청 내 IGF-Ⅰ 함량이 대조군이나 기존칼슘보충용식품군에 비해 유의적으로 높게 나타났다(p<0.05). 일일증체량, 일일사료섭취량, 장골길이, 장골 무게, 장골폭 등의 성장관련 지표에서도 장골의 길이와 일부 부위별 두께에서의 유의적 결과를 포함하여 전반적으로 신규칼슘보충용식품군이 대조군과 기존칼슘보충용식품군에 비해 양호한 성적을 나타내었다. 이러한 결과는 기존칼슘보충용식품에 발효대사물을 첨가함으로써 나타난 칼슘의 생체흡수율 증대 및 성장호르몬의 분비활성화 등 여러 성장관련요인들이 흰쥐 성장에 긍정적으로 작용한 때문으로 생각되어진다. This study was conducted to investigate the effect of administration of calcium supplementary food containing fermented product of Bacillus subtilis SE4 highly producing poly-γ-glutamic acid on the growthparameters of adolescent male rats. Four-week old male Sprague-Dawley (SD) rats were fed for 4 weeks and assigned to the following 4 groups: two groups administered orally with new calcium supplementary food (such as 150 ㎎/㎏ and 300 ㎎/㎏) containing fermented product of B. subtilis SE4, one group administered with conventional calcium supplementary food product (150 ㎎/㎏) and one saline group as control. Daily weight gain and daily food intake in the two new food product groups were higher than those of conventional food product group and control group. Especially, the content of serum IGF-Ⅰ in the two new food product groups were significantly higher than those in conventional food product group and in control group (p<0.05). In addition, length and weight of longitudinal bone in the two new food product groups were longer and heavier than those of conventional food product group and control group. Therefore, the addition of fermented food product of B. subtilis SE4 into the conventional calcium supplementary food increased all the parameters examined for the growth of the adolescent male rats.
