      • PP-333, Brassinolide 및 기타 生長調節物質의 單用 및 混用이 녹두幼苗의 發根 및 胚軸伸長에 미치는 영향

        Experiments were conducted to evaluate and compare the response of mungbean seedlings as prepared for auxin bioassay to the recently introduced plant growth regulators used alone or in combination with other growth regulators. Results obtained can be summarized as follows. 1. Significantly larger number of roots were formed in the mungbean cuttings which were rooted in PP-333 solutions of 0.01-1.00 ppm. The higher the concentration, the more the number of roots formed. PP-333 was more effective in increasing the number of adventitious roots as compared to IAA. 2. S-07 was only slightly effective at 0.1 ppm or higher as compared to the very strong stimulatory action of similar substance, PP-333. 3. Brassinolide at 0.1 ppm or higher significantly reduced the number of adventitious roots, but promoted the epicotyl elongation, thus exhibiting very similar physiological activity as gibberellins. 4. The combined use of PP-333 and auxins showed mutual stimulation effect upto certain concentrations. The rooting was, on the other hand, markedly inhibited at higher concentrations. Theenhancing effect of rooting by PP-333 was markedly suppressed by gibberellic acid, thus showing clear antagonism between these chemicals. 5. Auxins such as IAA and IBA were effective in overcoming the inhibiting effect of root formation by brassinolide, thus exhibiting antagonism between these chemicals. Increase in epicotyl length was markedly enhanced only be the brassinolide treatments.

        Isolation of 2,4,5-Trichlorophenoxyacetic Acid-Degrading Bacteria

        화합물을 분해하는 우수균주 개발의 기초연구로서, 대전 근교 지역의 논과 밭에서 채취한 토양 표품으로부터 100균주의 세균을 분리하였고, 그 중 2,4,5-T를 단일 탄소원으로 하는 고체 최소 배지에서 잘 자라는 균주 19균주를 선별하였다. 이들 균주를 등정한 결과 Pseudomonas속이 11균주 Acinetobacter속이 4균주, Alcaligenes속이 1균주이고 3균주는 미동정되었다. Pseudomonas속으로 밝혀진 MU19와 MU92는 네가지 염소계 화합물들(2,4-D, 2,4,5-T, MCPA 그리고 3CB)을 모두 분해하는 것으로 나타났다. 최소 액체배지에서 배양한 경우 Acinetobacter로 동정된 MU38균주가 접종 48시간 후에 가장 높은 분해도를 나타내었고, MUl9, MU57, MU73과 MU92는 그 다음으로 높은 분해도를 나타냈다. 실험결과 선별된 19균주 중 Acinetobacter sp. MU38 그리고 Pseudomonas sp. MU19과 MU92는 염소계 방향쪽 화합물에 대한 넓은 분해능을 갖고 있으며, 특히 2,4,5-T에 대한 높은 분해도를 나타내는 것으로 조사되었다. 2,4,5-trichlorophenoxyacetic acid (2,4,5-T)-degrading bacterial strains were isolated from rice field and field in suburbs of Taejon. Of the total 100 isolates, 19 strains were selected by fast growth on solid minimal media containing 2,4,5-T as a sole of carbon and energy, and they were identified to genus level. 11 strains were identified as Pseudomonas, 4 strains as Acinetobacter, 1 strains were as Alcaliagenes and 3 strains were not identified. Strains MU19 and MU92 which were identified as Pseudomonas were capable of degradation for 4 kinds of chlorinated aromatic hydrocarbons, 2,4-D, 2,4,5-T, MCPA and 3CB. Acinetobacter sp. MU38 showed the highest degradability in liquid minimal media at 48 hours after inoculation, and Pseudomonas spp. MU19. MU57, MU73, and MU92 were able to degrade carbon source at higher rates. As the results Acinetobacter sp. MU38 and Pseudomonas spp. MU19 and MU92 were capable of biodegradation for broad range of halogenated aromatic hydrocarbons, and had higher rates of degradation for 2,4,5-T.

        Isolation and Identification of Pentachlorophenol-degrading Bacteria

        공해 문제를 유발하고 있는 Pentachlorophenol(PCP)와 2,4-dichlorophenoxyacetate(2,4-D)와 같은 난분해성의 염소계 방향족 화합물들을 폭 넓고 빨리 분해할 수 있는 우수 균주를 개발하기 위한 기초 연구로서, 대전, 청주, 전주 근교 지역에서 채취한 토양 표품으로부터 100균주의 세균을 분리하였다. 그 중 PCP를 단일 탄소와 에너지원으로 하는 고체 최소 배지에서 잘 자라는 균주 19균주를 선별하였다. 이들 균주를 동정한 결과 Pseudomonas속이 15균주, Acinetobacter속이 1균주, 3균주는 미동정되어 Pseudomonas속이 대부분을 차지하였다. 분해능이 비교적 좋은 것으로 알려진 Alcaligenes의 세균은 발견되지 않았다. Pseudomonas속으로 밝혀진 MU135, MU139, MU163과 MU184 네가지 염소계 화합물들(PCP, 2,4,-D, MCPA 그리고 3CB)을 모두 분해하는 것으로 나타났다. 최소 액체배지에서 배양한 경우 Pseudomonas sp. MU139 균주가 접종 72시간 후에 가장 높은 분해도를 나타내었고, Pseudomonas sp. Mu147, MU177, MU184 그리고 MU192는 그 다음으로 높은 분해도를 나타냈다. 실험결과 선별된 19 균주 중 Pseudomonas sp. Mu139와 Pseudomonas sp. MU184는 염소계 방향족 화합물에 대한 넓은 분해능을 갖고 있으며, 특히 PCP에 대한 높은 분해도를 나타내는 것으로 조사되어 우수 균주 개발의 좋은 재료로 이용될 수 있을 것으로 보인다. To develope the enhanced bacterial strains capable of biodegradation for various chlorinated aromatic compounds, 100 bacterial strains were isolated from soil samples of suburbs of Taejon, Cheongju, and Jeonju by the enrichment culture. These strains can degrade pentachlorophenol (PCP) which is a kind of wood preservatives. Nineteen strains of the isolates were selected by fast colony-forming rate on solid minimal media containing PCP as an only source of carbon and energy. These strains were identified to genus level. Fifteen strains were identified as Pseudomonas, 1 strain as Acinetobacter and 3 strains were not. Genus Alcaligenes strains were not found among them. Pseudomonas sp. MU135. MU139, MU163 and MU 184 were able to degrade for 4 kinds of chlorinated compounds, PCP, 2,4-D, MCPA and 3CB. Pseudomonas sp. If was observed that MU139 exhibits the highest degradability in liquid minimal media at 72 hours after inoculation. Pseudomoans sp. MU147, MU177, MU184 and MU192 also degraded the compounds at higher rates. As the results, Pseudomonas sp. MU139 and unidentified strain MU184 had biodegrability for broad range of chlorinated compounds and higher rates of degradation for PCP.

      • 이중형질전환에 의한 웅성불임유기유전자의 발현억제

        This study was carried out with the purpose of looking into the regulation of gene related to anther development, and applying their promoter to induce male sterile plants and restore their fertility. Progenies of transgenic male sterile tobacco plants containing anther specific promoter (BAN215) and diphteria toxin A (DTx-A) gene were analyzed by danamycin screening and pollen observation. DTx-A gene in male sterile plants was confirmed by PCR with the two synthetic primers, 5'-AACTTTTCTTCGTACCAC-3' and 5'-GCTTTCGCCTGTTCCCAG-3', from the sequence of DTx-A genes. The fragment of 350bp was appeared when the PCR product of 428 bp was digested with Rsal. pDTx-2 containing DTx-A gene was used as a control (DTx). Some of them were double transformed with the construct of BAN215 promoter and antisense DTxA (pKDA215b) to test restoration of fertility.

        채소작물의 형질전환을 위한 재분화체계 확립 및 항생제 검정

        This study was conducted to determine the concentrations of plant growth regulators required for regeneration and the concentrations of antibiotics for the selection of transformed regenerants from lettuce, musk melon and tomato. The optimal concentrations of plant growth regulators for shoot formation were NAA 0.1 mg/$\ell$ +BA 0.1 mg/$\ell$ for lettuce, NAA 0.01 mg/$\ell$ +BA 2.0 mg/$\ell$ for melon and NAA 0.1 mg/$\ell$ +BA 0.5 mg/$\ell$for musk melon. Shoot induction from tomato, lettuce and melon was completely inhibited by 30 mg/$\ell$ or higher concentrations of kanamycin. Shoot formation from mu나 melon was not affected by kanamycin up to 40 mg/$\ell$, but was reduced in the presence of 50 mg/$\ell$ and completely inhibited by 100 mg/$\ell$. Shoot formation of all four crops was completely inhibited by higromycin at 10 mg/$\ell$. Both carbenicillin and cefatoxinme did not show any negative effects on shoot formation.
