Sulforaphane에 의한 HeLa 인체자궁경부함세포의 증식 억제 기전 연구

Sulforaphane, an isothiocyanate derived from hydrolysis of glucoraphanin in broccoli and other cruciferous vegetables, was shown to induce phase II detoxification enzymes and inhibit chemically induced mammary tumors in rodents. Recently, sulforaphane is known to induce cell cycle arrest and apoptosis in human cancer cells, however its molecular mechanisms are poorly understood. In the present study, we demonstrated that sulforaphane acted to inhibit proliferation and induce morphological changes of human cervical carcinoma HeLa cells. Treatment of HeLa cells with $10{\mu}M\;or\;15{\mu}M$ sulforaphane resulted in significant G2/M cell cycle arrest as determined by flow cytometry. Moreover, $20{\mu}M$ sulforaphane significantly induced the population of sub-G1 cells (9.83 fold of control). This anti-proliferative effect of sulforaphane was accompanied by a marked inhibition of cyclin A and cyclin-dependent kinase (Cdk)4 protein and concomitant induction of Cdc2, Cdk inhibitor p16 and p21. However, sulforaphane did not affect the levels of cyelooxygenases and telomere-regulatory gene products. Although further studies are needed, the present work suggests that sulforaphane may be a potential chemoprevetive/ chemotherapeutic agent for the treatment of human cancer cells.
한.중 소비자의 브랜드 원산지이미지와 브랜드 연상지식이 구매의도에 미치는 영향관계에 관한 비교연구 - 소비자 자민족중심주의성향 정도에 따른 상호조절효과 중심으로 -

This comparative study conducted to examine the differences between Korean and Chinese consumers. The specific goals of the study were as follows; First, It was to investigate the influences of COO(Country of Origin) image on the customer…s buying intention through the brand association knowledge. It is also aimed at analyzing the moderating effects of consumer's ethnocentrism on the influence of brand association knowledge on the buying intention. To test the hypotheses, 117 questionnaires were collected from university students in Korea and 119 questionnaires did from China and put to the test with SPSS 17.0 and AMOS 7.0. The results indicated the followings: For consumers in both countries, the COO image had not influences on the buying intention. For Korean consumers the COO image had positive impacts on the brand association knowledge, which in turn had positive impacts on the buying intention. For Chinese consumer, COO image did not have influences on the brand association knowledge, but the brand association knowledge had positive impact on the buying intention. In addition, the consumer's ethnocentrism had moderating effects on the influences of the brand association knowledge on the buying intention.

This study conducted to examine how consumer's perception of COO(Country of (Brand) Origin) image affects brand association knowledge, how coo image and brand association knowledge affect buying intention, how these relationship differs in hedonic and utilitarian products and the moderating effect of consumer's ethnocentrism in the influence of brand association to buying intension. The results indicated the followings: First, it was significant COO Image affect brand association knowledge, more strong significant in Hedonic products than in utilitarian products. But it was not supported COO Image affect buying intension significantly. Second, Brand association knowledge affected buying intention significantly. And the last, consumer's ethnocentrism mediated between brand association knowledge and buying intention strong significantly.

An experimental investigation has been carried out to examine the influence of injector design variables and operating conditions on the resultant drop size for triplet impinging streams injectors. The variables studied in this investigation are pressure drop, impinging angle, orifice length to diameter ratio, and impinging point distance. Droplet-size data are obtained using water as the propellant simulant by Malvern Particle Analyzer System. Drop size decreases with increasing impinging angle and pressure drop while other injector parameters remain constant at the same point. But it is found that there is no noticeable droplet-size change which results from change in orifice length to diameter ratio or impinging point distance within the investigated range.

일반적으로 변속기 전체에 대한 내구시험을 통하여 파킹 락 기어의 내구성을 평가한다. 파킹 락 기어 부품 하나의 내구시험에 대한 시험 기준은 부재한 상황이다. 따라서 본 연구에서는 파킹 락 기어의 내구시험 기준을 자체적으로 설정하여, 내구 성능시험을 수행하였다. 파킹 락 기어의 항복응력을 구하기 위해 정적 파단 시험을 수행하였고, 항복응력의 60%를 내구시험 조건으로 설정하였다. 설정된 시험조건으로 100만회의 반복내구 성능시험을 수행하였다. 또한, 내구 성능시험을 수행하기 위하여, 전용 지그를 제작하였다. 제작된 지그가 정적 파단시험에서 파손되어, 파손된 지그에 대한 유한요소해석을 수행하고 설계를 변경하였다. 수정한 지그를 제작하여 자체적으로 설정한 시험기준으로 내구 성능시험을 수행한 결과, 파킹 락기어는 본 조건에 충족하였다. 본 연구에서 수행된 시험과 해석을 통하여 파킹 락 기어의 성능 및 내구 한계에 대한 기초 데이터를 확보하였고, 이러한 기초 데이터는 파킹 락 기어의 설계와 개발에 중요한 기초 자료로 활용될 것으로 사료된다. Generally, the durability of the parking lock gear is evaluated through an endurance test for the entire transmission but no test standard exists for the durability test of one part of a parking lock gear. Therefore, in this study, the durability test standards of the parking lock gear were determined autonomously, and the durability performance test was conducted. A static fracture test was carried out to determine the yield strength of the parking lock gear, and a durability test condition was set to 60 % of the yield strength. The durability test of 1,000,000 times was conducted under the given conditions using a dedicated test jig manufactured for the test. Because the jig fractured during the static fracture test, finite element analysis for the jig was carried out and the jig was re-designed. The modified jig was manufactured and the durability test was carried out based on the test standard of autonomously. The basic data on the performance and endurance limit of the parking lock gear were obtained using these tests and analyses, and the basic data will be used as fundamental materials for the design and the development of the parking lock gear.

The aim of the study was to investigate the synthesis of a copolymer bearing cyclic carbonate and its miscibility with polymethylmethacrylate(PMMA). Poly [GMA(Glycidyl methacrylate)-St(Styrene)] was converted to poly[DOMA[(2-oxo-1,3-dioxolane-4-yl) methyl methacrylate]-St(Styrene)] by the polymer reaction with carbon dioxide using tetraoctyl ammonium chloride(TOAC) as a catalyst. The miscibility behavior various Poly(DOMA-St) with PMMA blend was studied using differential scanning calorimeter(DSC). Optical clarity test and DSC analysis showed that poly(DOMA-St) blends were miscible over the whole composition ranges with PMMA.

We propose the synthesis of photopolymers having pendant cinnamic ester and cyclic carbonate groups by the addition reaction of poly(glycidyl methacrylate)[poly(GMA)] with cinnamoyl chloride and CO₂. Quaternary ammonium salts showed good catalytic activity for this synthesis. Photochemical reaction experiments revealed that photopolymer bearing cinnamoyl groups has good photosensitivity, and photosensitizer enhenced the photosensitivity of the synthesized photopolymer.

항공기 레이다는 기총 사격 지원을 위하여 수 백 미터의 최소 거리까지 표적을 정확히 추적해야 하는 임무를 갖는다. 한편, 표적이 수 km 이내로 근접하는 경우 글린트 오차가 각도 정확성에 미치는 영향성이 커지며, 표적의 기동이 커서 표적 탐지에 어려움이 있으므로 이를 위한 파형 및 신호처리 설계가 필요하다. 본 논문에서는 이러한 사항들을 고려하여 항공기 탑재 레이다에서 수 km 이내의 기동 표적을 탐지하는 모드의 신호처리 설계에 대한 내용을 다룬다. 제안한 설계는 CPI 길이에 대한 coherence 제한사항 및 주파수 기민성을 사용하는 다중 Burst에 기반하여 파형을 운용함으로써 글린트 오차의 영향성을 줄이는 방법을 포함한다. 제시한 방법을 기반으로 수송기 비행시험을 통해 검증한 결과 표적을 정확하게 탐지하는 결과를 보였으며, 다양한 가중합에 의한 글린트 오차 감소를 분석한 내용을 제시하였다. Airborne radars should track an enemy aircraft from maximum to minimum range very accurately to assist in immediate action. Meanwhile, when the target aircraft enters into very close range, such as a few kilometers, the effect of the glint on the angle measurement error increases. In this study, the design of signal processing and waveform consideration of short-range tracking mode of airborne radars is described. The proposed mode design includes the coherence limitation of the CPI and usage of multiple bursts to reduce the glint effect. The proposed method is verified via flight tests, and it is confirmed that the signal processing works accurately with a reduced angle measurement error.

본 실험은 홍조해조류의 하나인 불등가사리를 추출 후 각 용매별로 분획하여 암세포 성장억제 효과와 QR 유도활성 효과 등 생리활성을 연구하였다. 불등가사리를 이용하여 4종의 인체 암세포주 HepG2, HeLa, MCF-7 및 HT-29와 정상 간세포에 대한 암세포 증식억제 실험을 한 결과 사용한 4종의 암세포주에서 모두 시료첨가 농도에 의존적으로 증식 저지 효과가 나타났고, 특히 불등가사리의 methanol 분획층인 GFMM에서는 매우 낮은 농도의 시료첨가에도 불구하고 괄목할 만한 높은 암세포 성장 억제효과를 나타내었으며, 정상 간세포에서는 모든 분획층에서 약한 세포증식 억제율을 나타내어 정상 세포에 미치는 독성이 낮음을 확인하였다. 특히 본 시료는 여성암의 대표적인 유방암과 자궁경부암의 암세포 성장저지효과가 탁월하였다. 한편, 사용한 4가지 암세포주 중 유일하게 QR을 가지고 있는 HepG2를 이용한 암예방지표인 QR 효소 유도 활성 여부를 측정한 결과 분획물 첨가농도를 10, 20, 30 및 40 ㎍/mL로 첨가하였을 때 GFMM의 첨가농도 20 ㎍/mL에서 대조군에 비해 약 2배 이상의 높은 QR 유도효과를 나타내어 최종농도 40 ㎍/mL에서는 2.6의 암예방 QR 유도효과를 나타내었다. The growth inhibitory effects on human cancer cell lines provide useful information regarding critical cellular targets. Reports on cytotoxicity of Gloiopeltis furcata (GF) to human cancer cell lines are conflicting. This study was performed to investigate the effects of cytotoxicity and quinone reductase activity of Gloiopeltis furcata on the human cancer cells. The four partition layers of methanol extracts (GFM) which are hexane (GFMH), methanol (GFMM), butanol (GFMB) and aquous (GFMA) were screened for their cytotoxic effects on HepG2, HeLa, MCF-7, HT-29, and normal liver cell lines. The GFMM showed the strongest growth inhibition effect on all cell lines we used. The GFMM showed the highest induction activity of quinone reductase on HepG2 cells among the other partition layers.
