Retinol Binding Protein-Albumin Domain III Fusion Protein Deactivates Hepatic Stellate Cells

Liver fibrosis is characterized by accumulation of extracellular matrix, and activated hepatic stellate cells (HSCs) are the primary source of the fibrotic neomatrix and considered as therapeutic target cells. We previously showed that albumin in pancreatic stellate cells (PSCs), the key cell type for pancreatic fibrogenesis, is directly involved in the formation of vitamin A-containing lipid droplets, inhibiting PSC activation. In this study, we evaluated the anti-fibrotic activity of both albumin and retinol binding protein-albu-min domain III fusion protein (R-III), designed for stellate cell-targeted delivery of albu-min III, in rat primary HSCs and investigated the underlying mechanism. Forced expression of albumin or R-III in HSCs after passage 2 (activated HSCs) induced lipid droplet formation and deactivated HSCs, whereas point mutations in high-affinity fatty acid binding sites of albumin domain III abolished their activities. Exogenous R-III, but not albumin, was successfully internalized into and deactivated HSC-P2. When HSCs at day 3 after plating (pre-activated HSCs) were cultured in the presence of purified R-III, spontaneous activation of HSCs was inhibited even after passage 2, suggestive of a potential for preventive effect. Furthermore, treatment of HSCs-P2 with R-III led to a significant reduction in both cytoplasmic levels of all-trans retinoic acid and the subsequent retinoic acid signaling. Therefore, our data suggest that albumin deactivates HSCs with reduced retinoic acid levels and that R-III may have therapeutic and preventive potentials on liver fibrosis.

노래굿 「공장의 불빛」의 문화사적 의미 연구 ― 문화운동 1세대의 특수성과 공연성을 중심으로

「공장의 불빛」공연 녹화본이 보여주는 현대성은 당대 진보적인 연행적 생산물들의 생산 주체였던 대학생 집단의 위치적 특수성과 이들이 ‘민족’과 ‘민중’으로 대변된 사회의 민주화된 사유들을 전유하고 현장과의 만남을 시도한다는 것이 발생시켰던 다층적인 문화적 의미들을 다시 사유하게 해준다. 이에 본고는 「공장의 불빛」을1970년대를 중심으로 한 문화운동의 맥락 위에서 살피면서 그 속에 나타난 문화운동1세대의 특수성과 공연 녹화본이 보여주는 모던한 공연 형식의 의미를 논구하고자 한다. 먼저 2장에서 「공장의 불빛」의 창작자인 김민기와 녹화본 공연의 연출 및 안무를 담당한 채희완을 중심으로 한 문화운동 1세대가 탄생하는 장면을 재조명하면서 그들의 특수성이 예술적 기량과 민중 지향성 사이에서 만들어 지고 있음을 분석한다. 김지하와의 만남 속에 이루어진 문화운동 1세대의 민중적 지향은 이후 그들의 활동에서 일종의 강박과도 같이 작용한다. 특히 이 과정 속에서 초창기 문화운동에서 분명한 존재감을 드러냈던 전문적인 기량과 예술적 자질들이 이후의 작업 속에는 당사자들에 의해 ‘민중’으로 대표되는 사회 참여와 이념의 문제에 대한 강조에 가려진다. 이후 3장에서는 1978년 공연 녹화본에 나타난 연극성의 지표들을 동작과 소리를 중심으로 검토하고 이것이 동시기 전위적인 공연들이 지향하고자 한 현대성과 비교하여 예술사적 위치를 재조명한다. 공장의 불빛」공연 녹화본은 특별한 무대 장치와 대사가 없이 노래에 맞춘 배우들의 춤과 마임이라는 비언어적인 표현 양식만으로 이루어져 있다. 특히 녹화본의 동작은 심리적인 상태를 형상화하거나 기계화된 인간의 모습을 보여주는 춤과 마임으로 구성됨으로써 특정 행동을 과장하고 왜곡하는 표현주의적 특성을 보인다. 이를 통해 연극성을 높이고, 비인간적인 노동환경을 극대화시켜 보여준다. 「공장의 불빛」에서 배우들의 몸짓을 통해 구현되는 연극성은 서구의 현대 무용들이 추구하고자 한 현대성의 지표와 당대 드라마센터 출신 연출가들의 행로와도 겹치며 이 공통성은 둘 사이의 거리가 반드시 ‘예술적으로’ 반대의 입장에 있었던 것만은 아니라는 것을 보여준다. 마지막으로 4장에서는 「공장의 불빛」을 통해 살펴볼 수 있는 대학에서 출발한, 연행예술을 중심으로 한 문화운동의 생산물들에 나타난 아비투스의 연속성과 흔들림이 지닌 다층적인 문화적 의미에 접근한다. 음원과 공연 녹화본이 보여주는 형식적 현대성은 민중을 ‘향한’ 실천 속에 그들이 의식하지 못했음에도 계층/계급적 차이에 기반한 아비투스들이 존속하고 있었음을 보여준다. 채희완의 탈춤과 임진택의 연극적 기량에서 확인할 수 있듯이 1970년대 문화운동의 연행적 성과에서 대학이라는 학습자본과 예술적 기량이라는 문화자본은 중요한 역할을 했다. 「공장의 불빛」의원 작자인 김민기가 카세트테이프라는 매체를 선택한 것, 그리고 국악의 기법으로 만들어진 <두어라 가자>가 노동자들의 공연에서 차용되는 과정은 문화운동 주체들의 예술적 기량의 문제가 현장적 전환, 실질적인 노-학의 ‘만남’을 통해 특히 새로운 의미를 만들어 내고 있음을 보여주는 장면들이다. 「공장의 불빛」이라는 하나의 문화적 생산물은 대‘학’생 출신 창작자와 ‘노’동자들의 아비투스의 차이가 서로를 매혹하고 변화시 ... The video-recorded theatrical performance of Kim Min-ki’s 1978 musical play Lights of a Factory demonstrates such modernity that enables us to revisit the unique position of university students in Korea back then as producers of progressive-minded folk performances, while also guiding us through the multiple layers of cultural meaning arising from the students’ role as the bastion of democratic discourses represented by the concepts “nation“and “people“ and their attempt to engage with the people on the ground in a society that was still languishing under the military dictatorship. In this light, within the context of cultural activism, a trend especially prominent in the 1970s, I explore the uniqueness of first-generation cultural activists and the meaning of the modern format in the recorded performance. I begin Chapter II by revisiting the emergence of first-generation cultural activists, for which Kim Min-ki, the author of Lights of a Factory, and Chae Hee-wan, who directed the play and oversaw its choreography, played a leading role. In this context, I argue that the two figures’artistic prowess and orientation towards the people situate them in a unique position. The people-focused mind set among the first-generation cultural activists, formulated through Kim and Chae’s meetings with Kim Jiha, became something of an obsession in the former two figures’ future activities. As aresult of this process, their professional capacities and artistic talent, once clearly manifest at the initial stages of cultural activism, were over shadowed by their emphasis on socialengagement and ideology, as represented by the focus on the people, in their later works. Then, in Chapter III, I examine the indices of theatrical qualities in the video-recorded performance from 1978 with particular attention to motions and sound and simultaneously cast renewed spotlight on its position in the history of performing arts by comparing its modern qualities with the modernity that contemporary avant-garde plays were aiming for. The video-recorded version of Lights of a Factory solely features non-verbal modes of expression, i.e., actors and actresses’ dances and mime along with songs, without any special stage devices or spoken lines. In particular, the motions in the recorded performance consist of dances and mime that represent a psychological state or illustrate a mechanized human being, there by demonstrating expressionist characteristics that exaggerate and distort specific actions. This enhances the theatrical quality and maximizes the impression of the inhumane labor environment on spectators. The theatrical qualities, expressed through actors’gestures in Lights of a Factory, overlaps with the indices of modernity pursued by Western modern choreography as well as the career paths of the directors hailing from the then Drama Center. These commonalities demonstrate that the theatrical qualities of Lights of a Factory are not necessarily contrary to what the directors of the Drama Center pursued from an artistic perspective. Finally, in Chapter IV, I explore through Lights of a Factory the multi-layered cultural meaning in the continuity and undermining of habitus shown in the products of cultural activism, the movement that began from university campuses with the focus on traditional performing arts. Its formal modernity as displayed in sound-recorded and video-recorded versions demonstrates the continued presence of habitus arising from the gap between classes, albeit inadvertently, despite the artists’ practical orientation towards the people. As we can see in Chae Hee-wan’s mask dances and Lim Jin-taek’stheatrical prowess, the academic capital in the form of universities and cultural capital called artistic prowess played a crucial role in the achievements of the 1970s’ cultural activism in folk performances. Purpose: This study was conducted to identify the most effective method in cell therapy by comparing and analyzing the level of wound healing after the application of various cell delivery methods. Methods: Human mesenchymal stem cells were administered using 5 different methods on full thickness skin defects which were deliberately created on the back of 4-week-old mice using a 8 mm punch. Different modes of administration, cell suspension, local injection, collagen GAG matrix seeding, fibrin, and hydrogel mix methods were used. In each experiment group, 4×105 mesenchymal stem cells were administered according to 5 deferent methods, except for the corresponding control group. Results: The wound healing rate was fastest in the local injection group. The wound healing rate was relatively slow in the collagen matrix group, however, the number of blood vessels or VEGF increased most in this group. Conclusion: For rapid wound healing through wound contraction, it is advantageous to administer MSC by the local injection method. For the healing process of a wide area, such as a burn, the seeding of cells to collagen matrix is thought to be effective.

The effcets of adenosine 5'-triphosphate(ATP) were investigated on the uterine smooth muscle motility in the pig. The results were summarized as follows: 1. The effects of the porcine uterine smooth muscle and the contractile responses increased between the concentration of ATP $10^{-5}$ and $10^{-3}$ M with a dose-dependent manner. 2. The contractile response induced by ATP($10^{-4}$ M) was not blocked by pretreatment with cholinergic receptor blocker, atropine ($10^{-6}$ M) 3. The contractile response induced by ATP ($10^{-4}$ M) was not blocked by pretreatment with $\alpha$ -adrenergic receptor blocker, phentolamine(10$^{-6}$ M) and ${\beta}$-adrenergic blocker, propranolol ($10^{-6}$ M). 4. The contractile response induced by ATP($10^{-4}$ M) was not appeared in 4Ca^{++}$ -free medium. As the concentration of $Ca^{++}$ in $Ca^{++}$ -free medium was increased, the contractile response induced by ATP ($10^{-4}$ M) was enhenced but was completely inhibited by pretreatment with $Ca^{++}$ -channel blocker, papaverine($10^{-6}$ M) or verapamil($10^{-6}$ M). From these results, it was conclued that the effects of ATP were the contraction mediated by purinergic receptor in uterine smooth muscle of pig.

The effects of histamine were investigated on the uterine smooth muscle motility in the pig. The results were summarized as fellows : 1. Histamine caused the contraction of the porcine uterine smooth muscle and the contractile responses increased between the concetration of histamine $10^{-8}$ and $10^{-5}$ M with a dose-dependent manner. 2. The contractile response Induced by histamine ($10^{-6}$ M) was completely blocked by pretrevatment with $H_1$-histaminergic receptor blocker, pyrilamine($10^{-6}$ M) 3. The contractile response induced by histamine($10^{-6}$ M) was increased by pretreatment with $H_2$-histaminergic receptor blocker, cimetidine($10^{-6}$ M) From these results, it was concluded that the effects of uterine smooth muscle by histamine were the contraction mediated by $H_1$-histaminergic receptor and the relaxation mediated by $H_2$-histaminergic receptor in pig.
