In the present investigation we have studied the effect of bear's gall on mammalian cells and demonstrated that COS-7 cells, which were derived Monkey kidney cells, had shown almost same extent of growth with 78 hrs in 10% FCS Dulbecco's Modified Eagle's medium with bear's gall and without bear's gall. But the hybridoma cells which were fused murine myeloma cells and the rat spleen cells for monoclonal antibody production died almost within 48 hrs. To investigate the effect of biosynthetic mechanism, cDNA were transfected to COS-7 cells, and it was shown that cDNA-transfected COS-7 cell had produced 30-40% less the amount of recombinant protein than the medium without bear's gall.

In this study, the immunemodulatory activities of arabinogalactans-type polysaccharide (APS) from Cordyceps militaris grown on germinated soybeans was evaluated in THP-1 human monocytes. The cellular size and the number of intracellular organelles of THP-1monocytes were increased by APS. Also, APS-treated THP-1 cells significantly developed cellular adherence and macrophagic differentiation to the culture plate surface. APS noticeably enhanced phagocytic activity of THP-1cells against IgG-FITC-latex beads. APS induced the level of TNF-α, IL-12 p40, and IL-8 as well as TLR2 and TLR4mRNAs. APS can be developed as a promising immunmodulating agent with macrophage-activating properties.

Antrodia camphorata (AC) has been used as a traditional medicine to treat food and drug intoxication, diarrhea, abdominal pain, hypertension, pruritis (skin itch), and liver cancer in East Asia. In this study, we investigated anticancer activities of AC grown on germinated brown rice (CBR) in HT-29 human colon cancer cells. We found that the inhibitory efficacy of CBR 80% ethanol (EtOH) extract on HT-29 and CT-26 cell proliferation was more effective than ordinary AC EtOH 80% extract. Next, 80% EtOH extract of CBR was further separated into four fractions; hexane, ethyl acetate (EtOAc), butanol (BuOH), and water. Among them, CBR EtOAc fraction showed the strongest inhibitory activity against HT-29 cell proliferation. Therefore, CBR EtOAc fraction was chosen for further studies. Annexin V-fluorescein isothiocyanate staining data indicated that CBR EtOAc fraction induced apoptosis. Induction of G0/G1 cell cycle arrest on human colon carcinoma cell was observed in CBR EtOAc fraction-treated cells. We found that CBR decreased the level of proteins involved in G0/G1 cell cycle arrest and apoptosis. CBR EtOAc fraction inhibited the b-catenin signaling pathway, supporting its suppressive activity on the level of cyclin D1. High performance liquid chromatography analysis data indicated that CBR EtOAc fraction contained adenosine. This is the first investigation that CBR has a greater potential as a novel chemopreventive agent than AC against colon cancer. These data suggest that CBR might be useful as a chemopreventive agent against colorectal cancer.

Given the growing interest in damage control training for the naval ships and their organizations, expectations for a new concept of training program have also increased. The existing training programs and its concept focus on training crew to be more proficient and skilled so that they can respond better to damage situations, i.e., fires and flooding. This paper suggests a development procedure of damage control training scenarios using the survivability analysis results as a new concept of damage control training programs employing advanced systems such as damage control console, automation system, and kill cards. This approach could help the decision maker not only enhance his or her capability but also improve the reacting capability of crew members for complex situations induced by a weapon hit.

Phellinus linteus (PL) and ginseng (Panax ginseng, PG) have been widely used as traditional herbal medicines in East Asia. In this study, we evaluated whether the extract of P. linteus grown on ginseng (PGP) exhibited an more enhanced anti-proliferative activity on HT-29 human colon carcinoma cells than PL or PG extract. PGP EtOAc extract inhibited HT-29 cancer cell viability stronger than PG or PL EtOAc extract. FACS analysis revealed that PGP EtOAc extract increased sub-G1 (apoptotic) HT-29 cell population. The nuclear structures of HT-29 cells were disrupted, resulting in apoptotic fragments (45 kDa) of lamin B after treating PGP EtOAc extract. PGP EtOAc extract induced activation of caspase-8 and -9 in HT-29cells. These results suggest that the PGP EtOAc extract has a direct anti-cancer effect through apoptosis and cell cycle blockade in HT-29 human colon carcinoma cells.
