        Genome sequence of Prevotella intermedia strain originally isolated from cervicofacial actinomycosis

        Anaerobic Gram-negative bacterium Prevotella intermedia is a part of normal flora of the oral cavity and associated with various types of oral and systemic diseases. We present here a draft genome sequence of P. intermedia ATCC 15032, originally isolated from cervicofacial actinomycosis. The genome is 2,848,426 bp in length and has a GC content of 43.45%. The genome includes 2,358 protein-coding genes, 5 rRNAs, and 43 tRNA. The sequence information will provide important clues in understanding the genome diversity within the bacterial species, and genetic basis for phenotypic differences among P. intermedia strains.

        Genome sequence of Bifidobacterium dentium strain ATCC 15424 originally isolated from pleural fluid of an empyema patient

        We present here a draft genome sequence of Bifidobacterium dentium strain ATCC 15424, originally isolated from pleural fluid of an empyema patient. The genome is 2,625,535 bp in length and has a GC content of 58.5%. The genome includes 2,154 protein-coding genes, 4 rRNAs, and 55 tRNAs. Unlike other B. dentium strains isolated from human dental caries, ATCC 15424 carries 247 strain-specific genes, including prophage remnants and type III/IV secretion system proteins, N-acetylmuramoyl-L-alanine amidase, and PRTRC system protein E. The sequence information will contribute to understanding of the natural variation of B. dentium as well as the genome diversity within the bacterial species.

        In vitro effects of N-acetyl cysteine alone and in combination with antibiotics on Prevotella intermedia

        N-acetyl cysteine (NAC) is an antioxidant that possesses anti-inflammatory activities in tissues. In the field of dentistry, NAC was demonstrated to prevent the expression of LPS-induced inflammatory mediators in phagocytic cells and gingival fibroblasts during the inflammatory process, but the effect of NAC on oral pathogens has been rarely studied. Here, we examined the effect of NAC against planktonic and biofilm cells of Prevotella intermedia, a major oral pathogen. NAC showed antibacterial activity against the planktonic P. intermedia with MIC value of 3 mg/ml and significantly decreased biofilm formation by the bacterium even at sub MIC. NAC did not affect the antibiotic susceptibility of planktonic P. intermedia, showing indifference (fractional inhibitory concentration index of 0.5–4) results against the bacterium in combination with ampicillin, ciprofloxacin, tetracycline or metronidazole. On the other hand, viability of the pre-established bacterial biofilm exposed to the antibiotics except metronidazole was increased in the presence of NAC. Collectively, NAC may be used for prevention of the biofilm formation by P. intermedia rather than eradication of the pre-established bacterial biofilm. Further studies are required to explore antibacterial and anti-biofilm activity of NAC against mixed population of oral bacteria and its modulatory effect on antibiotics used for oral infectious diseases.

        Genome sequence of Actinomyces georgiae KHUD_A1 isolated from dental plaque of Korean elderly woman

        Gram-positive anaerobic bacilli Actinomyces spp. commonly reside on mucosal surfaces of the oropharynx, gastrointestinal tract, and urogenital tract. Here, we first report the draft genome sequence of Actinomyces georgiae KHUD_A1, isolated from dental plaque of a Korean elderly woman. The genome is 2,652,059 bp in length and has a GC content of 68.06%. The genome includes 2,242 protein-coding genes, 9 rRNAs, and 64 tRNA. We identified 157 KHUD_A1 strain-specific genes, including genes encoding CPBP family intramembrane metalloprotease, bile acid: sodium symporter family protein, Txe/YoeB family addiction module toxin and Phd/YefM family antitoxin. The sequence information of A. georgiae KHUD_A1 will help understand the general characteristics of the bacterial species and the genome diversity of the genus Actinomyces.

        국제상사중재와 미국법상 디스커버리의 교차점에 관한 연구

        국내 기업이라면 미국 내 소송은 꺼려질 수밖에 없다. 왜냐하면 통상 미국 내 소송은 미국법상 증거개시(디스커버리)에 막대한 비용과 시간이 소요되기 때문이다. 미국 내 소송에서 디스커버리에 대한 부담을 피하고자 미국 관련 거래에서 발생하는 분쟁의 해결을 미국 국내 법원이 아니라 국제상사중재를 통하여 해결하려는 이유가 바로 여기에 있다고 해도 과언이 아니다. 한편 미국 연방법전 제28편 제1782조는 국제상사중재에서 당사자가 중재판정부와 관할 내 미국 법원의 승인을 받으면 미국에 소재하고 있는 특정인을 상대로 미국 법원의 도움을 받아 미국법상 디스커버리를 가능하게 허용하고 있다. 즉 제1782조는 미국 내 소송의 디스커버리와 국제상사중재상 증거조사를 이어주는 다리의 역할을 한다. 이 논문은 바로 이 부분을 다룬다. 구체적으로 이 논문은 제1782조가 국내 기업에 적용될 수 있는 사례를 제시하고, 미국에서 논점이 되었던 제1782조 관련 과거 및 최신 주요판결을 정리한다. 정리한 내용을 바탕으로 국제상사중재에서 제1782조의 적용 여부는 각 연방순회항소법원마다 다르므로, 국내 기업이 소재한 지역에 따라 적용여부가 달라질 가능성이 있다고 분석한다. 한편 우리나라 기업이 상대방만이 제1782조를 활용하여 수집한 증거를 이용하여 최종적으로 패소한 중재절차에서 상대방의 중재판정 집행신청이 들어올 경우, 뉴욕협약상 집행거부사유를 활용하여 중재판정의 집행을 다툴 방어 논리를 제시한다. 마지막으로는 점차 자국 우선주위를 내세우며 보수화하고 있는 미국 사회의 분위기를 고려하면, 미국 연방대법원 입장에서는 미국과 관련된 국제상사중재에서 미국 연방법인 제1782조를 적용하는 것이 자국의 이익 증진에 부합한다고 판단할 여지가 다분하기에 장차 제1782조가 국제상사중재에도 적용될 수 있다고 확인할 가능성이 높다는 결론으로 끝맺음한다. Any Korean company is bound to be wary of being embroiled in a litigation in the U.S. This is largely attributable to their concern that litigating in the U.S. could be costly and time-consuming. Indeed, uneasiness surrounding discovery in the U.S. may very well be one of the reasons why international commercial arbitration is celebrated as a method for resolving disputes involving transactions with U.S. parties. But against this backdrop, 28 U.S.C. § 1782 may allow a party in an international commercial arbitration to conduct U.S.-style discovery on a party residing in the U.S. with the help of a U.S. court with jurisdiction if the tribunal and the court both approve the request. In that sense, Section 1782 provides a point of intersection between discovery as it is practiced in the U.S. and document production in international commercial arbitration, which is what this paper addresses. Specifically, this paper starts out by offering a hypothetical situation in which Section 1782 could apply to Korean companies, and then summarizes notable U.S. case law on Section 1782. Afterwards, this paper concludes that since the applicability of Section 1782 in international commercial arbitration varies with the concerned federal court of appeals, its applicability depends on which federal district of the U.S. a Korean company is located in. Next, under the premise that the counterparty in an international commercial arbitration prevails against a Korean company by relying on evidence collected pursuant to Section 1782 and seeks to enforce the arbitral award, this paper offers lines of defense for Korean companies to contest the enforcement of the award by utilizing grounds for refusal of enforcement under the New York Convention. Finally, this paper ends on the note that in light of how the U.S. is growing increasingly conservative and prioritizing its own interests, there is a substantial likelihood that the Supreme Court of the U.S. would confirm that Article 1782 also applies to international commercial arbitration since it very likely might deem the application of Article 1782, as U.S. federal law, in international commercial arbitration to enhance the state interest of the U.S.

        Antibacterial effects of sodium tripolyphosphate against Porphyromonas species associated with periodontitis of companion animals

        Porphyromonas species are closely associated with companion animal periodontitis which is one of the most common diseases in dogs and cats and leads to serious systemic diseases if left untreated. In this study, we evaluated the antimicrobial effects and mode of action of sodium tripolyphosphate (polyP3, Na5P3O10), a food additive with proven safety, using three pathogenic Porphyromonas species. The minimum inhibitory concentrations (MICs) of polyP3 against Porphyromonas gulae, Porphyromonas cansulci, and Porphyromonas cangingivalis were between 500 and 750 mg/L. PolyP3 significantly decreased viable planktonic cells as well as bacterial biofilm formation, even at sub-MIC concentrations. PolyP3 caused bacterial membrane disruption and this effect was most prominent in P. cangingivalis, which was demonstrated by measuring the amount of nucleotide leakage from the cells. To further investigate the mode of action of polyP3, high-throughput whole-transcriptome sequencing was performed using P. gulae. Approximately 30% of the total genes of P. gulae were differentially expressed by polyP3 (> 4-fold, adjusted p value < 0.01). PolyP3 influenced the expression of the P. gulae genes related to the biosynthesis of thiamine, ubiquinone, and peptidoglycan. Collectively, polyP3 has excellent antibacterial effects against pathogenic Porphyromonas species and can be a promising agent to control oral pathogenic bacteria in companion animals.

        측두하악관절 골관절염 환자의 진단에서 단층촬영과 골스캔 검사의 유용성에 대한 예비연구

        TMJ Osteoarthritis is a degenerative disease caused by overload on joint tissue, and often accompany with local tenderness on the joint, crepitus by mandibular movement, restriction of mandibular movement, and anterior openbite. In general, panoramic radiography, TMJ panoramic radiography, and transcranial radiography are conducted to diagnose osteoarthritis after clinical examination, however, these radiographic evaluations are limited in detecting minute bony changes of early pathologic lesion. The aim of this pilot study was to evaluate the limitation and usefulness of several TMJ imaging techniques, so we compare the findings from clinical examination, plain film radiographs, tomograph, and bone scan from 81 patients with temporomandibular joint disorder. The proportion of patients showing same findings in plain film radiographs, TMJ tomograph, and bone scan was high, however, it is desirable that clinician should conduct phased imaging examinations according to the clinical findings due to the possibilities of false negative findings in diagnosis of osteoarthritis.

        Complete genome sequence of Granulicatella adiacens KHUD_009 isolated from subgingival biofilm

        Granulicatella adiacens is known to participate in several oral diseases including periodontitis, caries and endodontic infection. However, complete reference genome sequences for G. adiacens have only been analyzed for non-oral isolates. We sequenced the complete genome of G. adiacens KHUD_009 originated from the subgingival biofilm of a Korean with periodontitis, using the PacBio Sequel II platform. The genome is 1,949,834 bp with a GC content of 37.81% and includes 1,830 protein-coding genes, 15 rRNAs, and 58 tRNA. We identified that KHUD_009 carries several genes encoding virulence-associated proteins such as phosphocarrier and acyl carrier proteins and type VII secretion proteins.

        Preparation of antibacterial chitosan membranes containing silver nanoparticles for dental barrier membrane applications

        The materials of the dental barrier membranes (DBM), which is widely used for effective alveolar bone reconstruction, require high biocompatibility and additional functions. In this study, we prepared a chitosan/polyurethane (CSP) nanofibrous membrane incorporated silver nanoparticles (AgNPs) within the membrane. Characterization of CS-AgNP/polyurethane composite (AgCSP) nanofiber membranes showed that AgNPs were well embedded in chitosan membranes. In addition, biocompatibility and antibacterial effect against Porphyromonas gingivalis of the AgCSP was demonstrated by viability analysis and growth inhibition test. Collectively, AgCSP nanofiber membrane with antibacterial activity as well as biocompatibility may be useful in DBM and other tissue engineering applications.
