        Polyamine에 의한 토끼 정낭 평활근의 이완반응

        Purpose: This study was designed to investigate the effects of polyamines on rabbit seminal vesicular contractility. Materials and Methods: The polyamines; putrescine, spermidine and spermine, were added to deepithelized and precontracted seminal vesicle strips, with either 10-4M norepinephrine(NE), 10-4M acetylcholine(ACh) or 70mM KCl, in organ chambers to obtain cumulative concentration response curves. A whole cell mode patch clamp study was also performed to observe the effects of the polyamines on the L-type calcium channel activities. Results: The polyamines elicited concentration-dependent relaxations of the precontracted strips with the NE, ACh and KCl. The spermine showed the most potent relaxation response. Both extracellular and intracellular application of the spermine decreased the L-type calcium channel currents. Conclusions: Spermine more potently inhibited the seminal vesicle contraction than putrescine or spermidine, which suggests the polyamines may play a role in maintaining the basal tonicity of seminal vesicle in a flaccid state. The spermine-induced relaxation response seems to be related with an inhibition of the L-type calcium channel activities.

        Interleukin-1beta가 전립선암세포 PC-3 및 DU-145의 증식력과 Interleukin-6 및 Interleukin-6 수용체 mRNA 발현에 미치는 영향

        Purpose: IL-1 is a multifunctional proinflammatory cytokine. As the proliferative effects of IL-6 and IL-6 receptor expressions on prostatic cancer cells in response to IL-1 have not been determined, the effects of IL-1 on prostatic cancer cell lines were investigated. Materials and Methods: PC-3 and DU-145 cells were cultured in RPMI-1640 medium containing 10% fetal bovine serum. Cell cultures were supplemented with various concentrations of IL-1(0, 1, 10, 20 and 40ng/ ml), and the MMT growth assay performed. PC-3 and DU-145 cells were treated for 2, 4, 8, 12 and 24 h both with and without IL-1. IL-6 and IL-6 receptor(IL-6R) mRNA expressions were investigated using RT-PCR, and the IL-6 levels in cultured supernatant measured by ELISA. Results: The viability of both PC-3 and DU-145 cells decreased after IL-1 treatment(10, 20 and 40ng/ml). With 40ng/ml the IL-1, IL-6 and IL-6RmRNA expressions were lower in PC-3 cells, but unchanged in DU-145 cells, whereas the IL-6 protein production was higher in both PC-3 and DU-145 cells. Conclusions: IL-1 inhibited the proliferation of both PC-3 and DU145 cells. In the PC-3 cells, IL-1 decreased the expressions of IL-6 and IL-6R mRNA, but paradoxically increased the IL-6 production. In the DU-145 cells, IL-1 treatment did not affect the IL-6 or IL-6R mRNA expressions, but the IL-6 production was increased. This discrepancy between IL-1-induced IL-6 mRNA and protein production may be mediated by modification to the protein synthesis or an increased cellular excretion. (Korean J Urol 2004; 45:810-816)

        인슐린에 의한 가토 정상 음경해면체 평활근의 내피세포 의존성 이완 기전

        Purpose: Insulin is known to induce relaxation in various vasculatures by increasing the release of nitric oxide or the expression of nitric oxide synthase. However, its action mechanisms on the corpus cavernosum remain to be uncovered. This study aimed to investigate the relaxative responses and the mechanism of normal cavernous smooth muscles to insulin. Methods and Materials: Rabbit corpus cavernous tissues were prepared in 2x2x8 mm sized strip for isotonic tension recording. The dose-dependent relaxation responses of norepinephrine(10-4M)-precontracted strips to insulin (10-6M) were measured. The relaxation responses of NE(10-4 M)-precontracted strips to insulin(10-6M) were measured under low to high glucose concentrations(0, 0.1, 1 and 11mM) in physiological solution. The relaxation responses of the NE-precontracted strips to insulin(10-6M) were also observed after endothelial denudation, 30-minute preincubation with L-NAME(5mM) or 30-minute preincubation with indomethacin(10-4 M). Results: The cavernosal strips were relaxed by insulin in a dose-dependent manner. The insulin-induced relaxation was dose-dependently increased by glucose. The endothelial denudation or indomethacin pretreatment almost abolished the insulin-induced relaxation, but L-NAME rarely affected the relaxation. Conclusions: Insulin induces an endothelium-dependent relaxation of rabbit cavernous smooth muscles, which is mostly NO-independent, but seems to be related with prostaglandins or their metabolites.         β-아드레날린성 수용체 아형에 대한 효현제 및 길항제가 토끼 정낭의 수축 및 이완 반응에 미치는 영향

        Purpose: This study was aimed at investigating the role of β-adrenoceptor subtypes in mediating the relaxation and contraction of seminal vesicles in rabbits. Materials and Methods: Relaxation or contractile responses of epithelium- removed muscle strips of a rabbit seminal vesicle, which were precontracted with 10-5M norepinephrine, to selective β subtypes-adrenoceptor agonists were observed in an organ bath. The contractile responses induced by isoproterenol were also observed after application of selective antagonists. Results: Isoproterenol showed a concentration-dependent contractile response, but the contractility was weaker than those with phenylephrine and norepinephrine. The β selective-agonists(xamoterol for β1, clenbuterol for β2 and BRL37344 for β3) alone evoked neither contraction nor relaxation. However, the β1- and β3-agonists inhibited the contraction of the precontracted strips with 10-5M norepinephrine, while the β2-agonist enhanced the contraction. The pretreatment with a β2-antagonist(ICI118551) reduced the tension of the strips developed by 10-4M isoproterenol, but the β1-(atenolol) and β3-(SR59230A) antagonists showed no changes in the response. Conclusions: β1- and β3-adrenoceptors seem to mediate the relaxation of the seminal vesicle, while the β2-adrenoceptor may have a supplementary role in contraction. (Korean J Urol 2004;45:726-731)

        The testicular volume measured by a Prader orchidometer was compared with sperm count in semen, the levels of serum FSH, LH, testosterone, prolactin, estradiol and progesterone in 59 nonobstructive infertile men. 1. The causes of infertility were primary hypogonadisms in 50 patients (35 unknown, 9 Klinefelter syndromes, 6 varicocels) and secondary hypogonadism in 9 (5 isolated FSH deficiencies, 1 hyperprolactinerriia, 3 pituitary hypogonadisms). 2. Decreased levels of serum FSH (less than 4 mIU/ml) did not correlate with testis volume but increased level of serum FSH (more than 20 mIU/ml) were mostly noted in the testis less than 10ml. 3. Decreased level of serum testosterone (less than 3 ng/ml) were distinguishably noted in the atrophied testis less than 5 ml. 4. There was no correlation between the testicular volume and the levels of serum prolactin, estradiol, and progesterone. Coclusively, testicular volume less than 11 ml suggests poor spermatogenesis, but normal testicular volume dose not nessarily rule out poor spermatogenesis. Function of Leydig cell is relatively well preserved in atrophied testis of 5 to 10 ml comparing with that of seminiferous tubule.

        사람 정관의 Maxi-K 통로 활동성에 미치는 성호르몬의 효과

        Purpose: We investigated the effects of sex steroids on the maxi-K channel activities of the single smooth muscle cells of the human vas deferens. Materials and Methods: Human vas deferens cells were isolated using proteolytic enzymes(collagense and papain), and then they were employed for an electrophysiological study with using the inside-out patch clamp configurations. The intracellular calcium concentrations were adjusted between 0.1μM to 1μM, and the membrane potentials were depolarized. Results: The maxi-K channels were identified by their single channel conductance and calcium dependency. The application of 10ng/dl estrogen induced a significant increment of the maxi-K channel activities. The effects were not affected by the change of the intracellular calcium concentration. The other sex steroids(progesterone and testosterone) rarely affected the maxi-K channel activities. Conclusions: Estrogen has stimulatory effects on the maxi-K channel activity of human vas deferens. This suggests that estrogen may relax the human vas deferens via maxi-K channel activation.
