하이퍼텍스트를 이용한 재사용 라이브러리의 이해도 증진에 관한 연구

재사용 시스템인 CARS 1.0에서 부품선택시 이해의 과정은 검색 단계에서의 이해 그리고 수정, 합성 단계에서의 이해로 나뉘어진다. 본 연구에서는 부품 선택시 이해도 증진을 위해 구현 기술로서 하이퍼텍스트를 적용하여 재사용 시스템의 한 부분인 이해 시스템을 설계, 구현 하였다. 하이퍼텍스트 기반 이해 시스템에서는 객체모델링 단계 에서의 정보와 재사용 시스템 사용시 발생하는 경험적 유의사항등을 annotation하는 기능을 제공 하므로 사용자의 부품선택시 이해의 증진을 이루고자 하는 시스템이다. 본 이해 시스템의 이해를 위해 제공하는 뷰 로서는 모델링 카드 뷰, 코맨트 뷰, 원시 코드 뷰, 상속 뷰, 그리고 계층구조 뷰가 있다.

Caffeic acid phenethyl ester의 cytochrome P450 저해 활성 평가

Caffeic acid phenethyl ester (CAPE) is one of the major active components of propolis showing inhibitory activ-ity against cytochrome P450 (CYP). The purpose of this study was to determine CYP inhibitory potential and metabolic sta-bility of CAPE using human liver microsomes (HLM) for characterization of its metabolic properties. Among CYP isoforms including CYP1A2, 2A6, 2B6, 2C8, 2C9, 2C19, 2D6, 2E1 and 3A4, CAPE exhibited strong inhibitory potential against CYP1A2 and 2C9, moderate against CYP2E1, 2B6, 2C8, 2C19 and 3A4, and weak against CYP 2D6 and 2A6, based on the IC 50 values. The inhibition of CYP1A2 but not CYP2C9 was increased by preincubation with CAPE and NADPH, suggesting that the CAPE-induced CYP1A2 inhibition may be metabolism-dependent. In metabolic stability study using HLM, CAPE was rapidly hydrolyzed to caffeic acid in a NADPH-independent manner. Caffeic acid exhibited week CYP inhibitory activity, relative to CAPE. These results raise the possibility that CAPE plays a role in propolis-mediated CYP inhibition.

홍삼 Ginsenoside의 Cytochrome P450 저해 활성 평가

In the present study we evaluated comparative herb-drug interaction potential of red ginseng total powder, ginsenoside Rg1, and Rb1 by inhibition of CYP isoforms including CYP1A2, CYP2A6, CYP2B6, CYP2C9, CYP2C19, CYP2D6, CYP2E1 and CYP3A4 using pooled human liver microsomes (HLMs). As measured by liquid chromatography-electrospray ionization tandem mass spectrometry, red ginseng total powder inhibited significantly activities of CYP1A2, CYP2A6, CYP2B6, CYP2C9, CYP2C19, CYP2D6, CYP2E1 and testosterone 6-beta hydroxylation by CYP3A4, but the IC50 values were higher than 556 μg/ml. Activities of CYP2B6, CYP2C9, CYP2D6 and CYP3A4 were inhibited by ginsenoside Rb1. Also, activities of CYP1A2, CYP2A6, CYP2B6, CYP2C9, CYP2C19, CYP2D6 and testosterone 6-beta hydroxylation by CYP3A4 were inhibited by ginsenoside Rg1. The IC50 values of ginsenoside Rb1 and Rg1 were higher than 200 μg/ml. Based on IC50 values against CYP isoforms, ginsenosides-drug interactions by CYP inhibition may be very low in clinical situations.

Tetravalent Bispecific 항체 분자인 Di-diabody의 제조 및 표적 단백질에 대한 항염증 영향

TNF-α and VCAM-1 play a pivotal role in the pathogenesis of rheumatoid arthritis, and the development of drugs targeting these molecules has extended the therapeutical approaches to rheumatoid arthritis patients. Bispecific antibodies combine the antigen-binding sites of two antibodies within a single molecule and thus they are able to bind to two different epitopes simultaneously. A specific bispecific antibody format termed “Di-diabody” was made for the efficient approach to anti-inflammation. In this study, the DNA vector construct of Di-diabody was built up against two antigens, VCAM-1 and TNF-α. For evaluating this Di-diabody as a bispecific antibody on the efficacy of anti-inflammation, the proteins were analyzed according to each antigen binding affinity and cell based assay related separate molecules. The 7H/Humira Di-diabody produced in this study interacted with its ligands, VCAM-1 and TNF-α, respectively. Also, this antibody exhibited the similar functional activities as compared to 7H-IgG in respect to inhibition of hVCAM-1-induced cell adhesion and Humira-IgG in respect to inhibition of TNF-α induced cytotoxicity. Further study to elucidate the pharmacological significance of the Di-diabody is warranted using experimental animals.

인간 HER-2 재조합 단백질을 사용한 항 HER-2 항체 단백질의 ELISA 정량 방법 개발

HER-2 (Human Epidermal Growth Factor Receptor-2) is a protein giving higher aggressiveness in human breast cancers. Trastuzumab is a monoclonal antibody that targets HER-2 and is known to extend survival across all stages of HER2-positive breast cancer. In this study, we attempted to development of a quantitative ELISA (Enzyme-Linked ImmunoSorbent Assay) for evaluating anti HER-2 antibodies using human HER-2 recombinant proteins to support antibody producing processes and pharmacokinetic studies. We established direct or indirect ELISA method for the trastuzumab-like protein combined human recombinant HER-2. The ELISA method will prove to be great value in quantitating anti-HER-2 antibodies levels for developing anticancer antibodies.

제주도 <허궁애기본풀이>의 의미와 의의

이 글은 제주도에 전승되는 <허궁애기본풀이>의 자료적 가치와 의의를 밝히는 것을 목표로 한다. 이 자료는 현재까지 자료적으로 온전하게 조명되지 않았고, 이로 인해 자료가 지닌 다양한 특성에 대한 본격적인 논의가 진척되지 못하고 있다. 이 글에서는 <허궁애기본풀이>가 본풀이와 설화, 그리고 서사민요로 전승되는 다면적 측면을 지니고 있는 것을 주목하고 그 양상을 살폈다. 또한 자료에 따라 주인공을 ‘허궁애기’라고 하는 특정한 명칭을 지닌 경우도 있고, 이와 달리 일반적인 예사 명칭으로 여성주인공이 제시된 경우도 있어서 일률적이지 않음이 확인된다. 그러나 이 본풀이의 근간은 제주도 무속제의에서 무당에 의해 구연되던 본풀이일 것으로 추정하고 그 가능성을 탐색하였다. 이후 전승 과정에서 무당의 본풀이가 일반 대중에게까지 침투되면서 다양한 문학 갈래의 형태로 구현되는 양상을 확인할 수 있다. 아울러 이 연구에서 이승과 저승을 오고가는 ‘허궁애기’라는 존재를 주목하여 제주도 무속의 이계관을 해명하는 작업도 함께 병행하였다. 이승과 저승을 오고가는 이야기는 본풀이를 비롯하여 일반적인 신앙적 근거를 제공하고 있는 점에서 주목할 만하다. 이 본풀이에서 제시되는 이계관을 통하여 제주도 무속의 일반적 이해는 물론하고 이승과 저승을 통해서 구현하는 여러 가지 근간을 다시 점검할 수 있었다. 본 연구를 통해서 <허궁애기본풀이>라는 자료에 대한 새로운 접근, 본풀이에서 시작된 특정한 서사적 전통이 민요와 설화로까지 파급되는 양상, 제주도 무속에 나타난 이계관 등에 대한 해명이 시도되었다. 이를 토대로 차후 제주도 본풀이의 세계관적 저변에 대한 탐색이 더욱 확대될 것으로 기대된다. The purpose of this paper is to investigate data value and significance of <Heogungaegi Bonpuri> handed down in Jeju Island. This material has not been addressed soundly as data up to now, which makes earnest discussion of various features the material has hard to show progress. This study paid attention to the fact <Heogungaegi Bonpuri> has many sides, passed down as the Bonpuri, folk tale and descriptive folk song, and investigated their aspects. Calling the main character varies according to data, sometimes specially calling the main character as ‘Heogungaegi’, while proposing the heroine as the generally normal name. The foundation of this Bonpuri, however, was presumed to be the one, narrated by the shaman in shaman ritual of Jeju Island, and its possibility was explored. It can be identified shaman’s Bonpuri was permeated since even into the general public in the process of transmission and realized as the form of varied literary prongs. Moreover, by taking note of the being, ‘Heogungaegi’, who comes and goes to this world and the other, this research clarified a viewpoint of different worlds of Jeju Island shamanism. The story of coming and going to this life and afterlife is worth paying attention to, in that it offers basis of general faith, including Bonpuri. Through the viewpoint of different world suggested in this Bonpuri, not only general understanding of Jeju Island shamanism, but also various foundations realized through this world and the other could be examined again. Through this study, new approach to the material <Heogungaegi Bonpuri>, the aspect that specific descriptive tradition started from Bonpuri spread even to folk song and folk tale, and explanation of the viewpoint of Jeju Island shamanism were tried. It is expected that exploration into the base of the world view of Jeju Island bonpuri will be expanded based on this research in the future.
