산화스트레스와 염증은 간 손상의 진행과정에 중요한 인자로 작용한다. 굴가수분해물의 항산화 및 항염증 활성은 지질대사, 혈압 및 혈당, 면역기능의 조절과 같은 다양한 기능에 관여한다. 그러나 급성 간 손상 모델에서 굴가수분해물의 효과를 확인한 연구 결과는 아직 확인된 바 없다. 본 연구는 LPS/D-GalN에 의해 유도된 급성 간 손상 생쥐 모델에서 굴가수분해물의 효과를 확인하기 위해 수행되었다. 실험군은 대조군(생리식염수), LPS/D-GalN 간 손상군, LPS/DGalN과 굴가수분해물(100 mg/kg, 200 mg/kg, 400 mg/kg)의 병합투여군 및 LPS/D-GalN과 silymarin(25 mg/kg) 병합투여군으로 나누었다. 급성 간 손상 모델은 1 μg/kg의 LPS와 400 mg/kg의 D-GalN으로 유도되었다. 먼저 시료의 항산화 및 항염증 활성을 분석한 결과 굴가수분해물은 농도 의존적으로 높은 DPPH 및 ABTS 라디칼 소거 활성을 보였으며, 인간 정상 간세포주(Chang)에서 과산화수소에 의한 세포 내 활성산소의 생성을 유의적으로 감소시켰다. 또한, 굴가수분해물은 농도 의존적으로 높은 COX-2 및 5-LOX 억제능을 보였으며, LPS에 의해 활성화된 생쥐 대식세포주 RAW264.7에서 발현되는 TNF-α, IL-6 및 IL-1α의 염증성 사이토카인의 mRNA 발현률을 감소시켰다. 굴가수분해물 투여는 LPS/D-GalN에 의한 혈청 ALT 및 AST증가를 유의적으로 감소시켰으며, 간 조직의 출혈 및 간세포의 자멸사를 감소시켰다. 또한, 간 균질의 TNF-α, IL-1β 및 IL-6 함량을 감소시켰으며, 감소한 catalase의 활성을 유의적으로 증가시켰다. 이상의 결과로부터 굴가수분해물은 간 보호 효과를 가지는 것으로 판단되며, 급성 간 손상의 예방 및 치료에 도움이 될 수 있는 시료로 활용될 수 있을 것으로 기대된다. Oxidative stress and inflammation are key factors responsible for progression of liver injury. A variety of functions of oyster hydrolysate (OH) are affected by their antioxidant and anti-inflammatory activities. However, little is known regarding the effects of OH on a liver injury model. This study was performed to evaluate the effects of OH on acute liver injury induced by lipopolysaccharide/D-galactosamine (LPS/D-GalN) in mice. Experimental groups were divided into six groups as follows (each group, n=10): control (saline), LPS/D-GalN, LPS/D-GalN+OH (100 mg/kg), LPS/D-GalN+OH (200 mg/kg), LPS/D-GalN+OH (400 mg/kg), and LPS/D-GalN+silymarin (25 mg/kg, positive control). The experimental acute liver injury model was induced with LPS (1 μg/kg) and D-GalN (400 mg/kg). We first analyzed antioxidant and anti-inflammatory activities in OH. OH showed high DPPH and ABTS radical scavenging activities and reduced ROS generation in Chang cells in a dose-dependent manner. In addition, OH showed anti-inflammatory activities, such as inhibition of cyclooxygenase-2 and 5-lipooxygenase. Treatment with OH down-regulated tumor necrosis factor (TNF)-α, interleukin (IL)-6, and IL-1α expression levels in LPS-stimulated RAW264.7 cells. OH significantly reduced LPS/D-GalN-induced increases in the concentrations of alanine transaminase and aspartate aminotransferase in serum. In the LPS/D-GalN group, liver tissues exhibited apoptosis of hepatocytes with hemorrhages. These pathological alterations were ameliorated by OH treatment. Consistently, hepatic catalase activity was low in the LPS/D-GalN group compared to the control group, and catalase activity was significantly restored by OH treatment (P<0.05). Furthermore, OH markedly reduced the LPS/D-GalN-induced increase in TNF-α, IL-1β, and IL-6 levels in liver tissue. Taken together, these results show that OH has hepatoprotective effects on LPS/D-GalN-induced acute liver injury via inhibition of oxidative stress and inflammation, suggesting that OH could be used as a health functional food and potential therapeutic agent for acute liver injury.
액체종균에 의한 표고의 수확 주기에 따른 이화학적 특성 및 항산화 활성

The physicochemical characteristics and antioxidant activities of Lentinula edodes cultivated with liquid spawn were compared according to the harvest cycle (1-3 cycles). There was no significant difference in moisture content. The crude lipid and crude protein contents tended to decrease according to harvest cycle. The content of β-glucan was the highest in mushrooms after one cycle. The β-glucan content in cycles 2 and 3 was 68.5% and 62.3% that of the content after cycle 1, respectively. Mineral content in the pileus and stipe was similar. The mineral content was highest in the pileus after two cycles. Composition and contents of free amino acids tended to decrease according to the harvest cycle. Contents of essential amino acids were higher in mushroom after one cycle. Total phenol content was highest in the pileus (31.96 mg/100 g) and the stipe (21.10 mg/100 g) after one cycle and tended to decrease with subsequent cycles. The flavonoid content was 11.13 mg/100 g after one cycle and significantly decreased according to the harvest cycle. Antioxidant activities were also highest after one cycle and significantly decreased according to the harvest cycle. When compared to L. edodes cultured for one cycle, L. edodes cultivated with liquid spawn for three cycles was considered to have similar marketability in terms of quality characteristics.
숙성 조건에 따른 흑생강의 이화학적 특성 및 항산화 활성

The physicochemical properties and antioxidant activities of aged ginger (Zingiber officinale Roscoe) were investigated every four days during the twenty days of aging process and compared with fresh ginger. Browning intensity and redness value were significantly increased compared to fresh ginger; however, lightness and yellow values were significantly decreased. Total sugar content was the highest during the second step and decreased significantly after the third step. The content of gingerol decreased with aging whereas 6-shogaol increased from 17.7 to 34.1 times the baseline. Total phenolic and flavonoid contents were the highest during the second step and decreased in the subsequent steps. The radical scavenging activities and reducing power were the highest during the second and third steps, respectively, and then they gradually decreased. The antioxidant activities of aged ginger during the aging process showed a high correlation with browning intensity, 6-shogaol, total sugar, and total phenolic content. Therefore, our study suggested that aging during the second and third steps were effective in optimizing antioxidant activity.

This study analyzed food component characteristics and antioxidant activities in five kinds of domestic commercial shiitake mushrooms (Lentinula edodes). Moisture content (79.17-82.90 g/100 g) showed little difference, but there was a significant difference in crude protein content. Total mineral content showed a significant difference, and potassium (K) content was the highest. Total and essential amino acid contents were higher in mushroom A than in the other samples. Nonetheless, the ratios of essential amino acids, aspartic acid, and glutamic acid to total amino acids were similar in all samples. 𝛽-Glucan content was in the range of 10.28-5.68 g/100 g, exhibiting a significant difference. The color intensity and overall texture of the mushrooms showed no remarkable difference. Total phenol and flavonoid contents were in the range of 19.92-30.77 mg/100 g and 6.95-10.39 mg/100 g, respectively; the ratio of flavonoids to total phenols ranged from 31.53% to 38.21%. DPPH and ABTS radical scavenging activities were 74.92-79.04% and 80.47-84.97%, respectively, and showed little difference. However, the reducing power varied between 195.23 𝜇M and 317.85 𝜇M, displaying a significant difference in all samples. Therefore, this study observed similar trends in the food component characteristics and antioxidant activities of domestic commercial shiitake mushrooms.
참비름의 라디칼 소거활성 및 콜레스테롤 급이 흰쥐에 대한 혈액 지질성분에 미치는 영향

참비름의 in vitro 라디칼 소거활성 및 콜레스테롤 급이 흰쥐에 보충 급이할 경우 혈액 중 지질 성분의 함량에 미치는 영향을 분석하였다. 참비름 물 및 80% 에탄올 추출물의 ABTS, DPPH 및 nitric oxide 라디칼 소거활성은 물 추출물보다 에탄올 추출물에서 유의적으로 높았다. 콜레스테롤을 급이한 대조군에 비해 5%(HCA1) 및 10%(HCA2)의 참비름 분말 급이 시 혈청 총 지질, 총 콜레스테롤 함량, 동맥경화 지수 및 심혈관질환 위험지수는 시료의 첨가량이 증가됨에 따라 유의적으로 감소하였다. 중성지방은 HCA2군에서만 유의적인 감소를 보였으며, HDL-콜레스테롤은 대조군에 비해 참비름 분말 급이 시 증가하였으나, 첨가량에 따른 유의차는 없었다. 혈중 GPT 및 γ-GTP 활성은 대조군에 비하여 참비름 분말 급이군에서 유의적으로 감소하였다. 혈중 지질과산화물 함량도 같은 경향이었다. 따라서 참비름은 식이성 콜레스테롤에 의한 질환의 조절에 유효할 것으로 생각된다. To evaluate of biological activity of Chambirum (Amaranthus lividus) in vitro and in vivo, we investigated the free radical scavenging activity of its extracts in vitro and the effect of lyophilized powder on the serum lipid profile of rats fed cholesterol. ABTS, DPPH, and NO radical scavenging activities were tested from water and 80% ethanol extracts of Chambirum, and biological activities of the ethanol extracts were significantly higher than the water extracts. The total lipid and total cholesterol content of serum, atherogenic index (AI), and cardiac risk factor (CRF) were decreased significantly for the groups fed with a 5% and 10% supplement of Chambirum powder (HCA1 and HCA2) in comparison with the group fed cholesterol (HC). Triglyceride content decreased drastically in the HCA2 group, while its content was not decreased in the other group. HDL-cholesterol content was elevated in the HCA1 and HCA2 groups, but was not significantly different to the supplemented amount of Chambirum powder. GPT and γ-GTP activities were decreased significantly in the groups fed with Chambirum powder compared to the HC group. And the content of the lipid peroxide level was the same trend. Therefore, these results give evidence that Chambirum might be useful in the control of induced disorders by dietary cholesterol and/or lipids.
명월초, 여주 및 울금을 포함한 돼지감자 복합물의 항산화 및 AMPK 활성화를 통한 항당뇨 활성

명월초, 여주 및 울금의 물 및 70% 에탄올 추출물에서 항산화 및 α-glucosidase 저해활성을 측정하였으며, 돼지감자 농축액에 이들 식물류의 에탄올 추출물을 첨가한 5종의 돼지감자 복합물(JA1~5)에서 항산화 및 항당뇨 활성을 비교하였다. 명월초, 여주 및 울금의 에탄올 추출물은 물 추출물에 비해 총 페놀 및 플라보노이드 함량이 많았다. DPPH, ABTS 라디칼 소거활성 및 FRAP법에 의한 환원력은 총 페놀 및 플라보노이드 함량에 의존적이었으며, 특히 명월초와 울금의 에탄올 추출물은 항산화 활성이 비슷한 수준이었다. α-Glucosidase 저해활성은 에탄올 추출물의 2,000 μg/ml 농도에서 50% 이상이었다. 5종의 복합물은 돼지감자 농축액에 비해 총 페놀 및 플라보노이드 함량이 많았다. 또한 항산화 활성 및 α-glucosidase 저해활성도 복합물에서 모두 증가되었으며, 특히 JA1의 활성이 유의적으로 높았다. 돼지감자 복합물은 인체 정상 간세포주인 Chang세포에 대해 세포독성이 없었으며, Chang세포에서 AMPK 활성화 및 근육세포인 C2C12세포에서 세포 내 당 유입을 유의적으로 증가시켰다. 이로써 명월초, 여주 및 울금의 추출물이 혼합된 돼지감자 복합물은 항산화 및 항당뇨 활성 증대에 효과적인 것으로 판단된다. This study was performed to identify the antioxidant and α-glucosidase inhibitory activities of water and 70% ethanol extracts of the three following herbs: G. procumbens, M. charantia, and C. longa. In addition, the antioxidant and antidiabetic activities of five types of Jerusalem artichoke composites (JA1–5), which were prepared by adding ethanol extracts of several herbs to Jerusalem artichoke concentrate, were studied and compared. The results showed that the total phenol and flavonoid contents of the ethanol extracts were higher than those of the water extracts. The DPPH and ABTS radical scavenging activities and reducing power depended on the total phenol and flavonoid contents. The antioxidant activities of ethanol extracts from G. procumbens and C. longa were comparable. Moreover, the α-glucosidase inhibitory activity of the ethanol extracts (2,000 μg/ml) from each herb was found to be over 50%. In contrast, the five types of JA composites showed higher total phenol and flavonoid contents than those of JA concentrate. In addition, increased antioxidant and α-glucosidase inhibitory activities were observed, with that of JA1 being the highest. However, all concentrations (1~100 μg/ml) of JA tested did not affect the cell viability of Chang cells. In addition, JA induced the activation of AMP-activated protein kinase (AMPK) in Chang cells and significantly increased the glucose uptake in C2C12 cells. Therefore, it could be concluded that the JA composites (JA1–5) mixed with G. procumbens, M. charantia, and C. longa extracts were effective in increasing the extracts’ antioxidant and antidiabetic activities.
