        Rhodiosin, an Antioxidant Flavonol Glycoside from Rhodiola rosea

        The EtOAc fraction of Rhodiola rosea ethanolic extracts showed a strong antioxidant activity. Through activity-guided fractionation and purification, we isolated two flavonol glycosides, which were identified as the well known flavonoids, rhodionin (1) and rhodiosin (2). To compare their antioxidant activities, we used an authentic aglycone compound, herbacetin (3). Among the compounds tested, rhodiosin (2) exhibited strong antioxidant activity, with IC50 values of 0.21 and 0.15 µM against ·OH and ·O 2, respectively. Rhodiosin (2) (100 mg/kg) reduced MDA content in the liver induced by irradiation when given prior to exposure of γ-radiation.

        Mass propagation of microtubers from suspension cultures of Pinellia ternata cells and quantitative analysis of succinic acid in Pinellia tubers

        The conditions for efficient tuber production from suspension cultures of Pinellia ternata cells which is one of medicinal herbs were established and succinic acid in tubers propagated in vitro was determined. Leaf explants formed white nodular structures and off-white calluses at a frequency of 90.6 % when cultured on Murashige and Skoog medium supplemented with 0.54 μM α-naphthaleneacetic acid (NAA); however, this frequency declined substantially with increasing NAA concentration, up to 16.2 lM, at which the frequency reached zero percent. In combination treatments with 4.44 μM 6-benzyladenine (BA) and NAA, however, the frequency of white nodular structure and off-white callus formation from leaf explants did not decrease, even at 16.2 μM NAA. Suspension cultures of P. ternata cells were established from leaf-derived off-white calluses in MS liquid medium containing 5.4 μM NAA and 4.44 μM BA. Upon plating onto MS basal medium, over 90 % of cell aggregates gave rise to microtubers and developed into plantlets. Regenerated plantlets were transplanted in potting soil and grown to maturity in a growth chamber, with a survival rate of >90 %. The highest succinic acid content in suspension culture-derived microtubers was 45 g/kg of extract. Compared with wild P. ternata medicinal tubers, the succinic acid content was very similar. The in vitro P. ternata microtuber proliferation system established in this study is thus an efficient alternative for the mass production of medicinal tubers.

        뽕잎 추출물을 이용한 동충하초(Paecilomyces japonica) 균사체 배양액의 멜라닌 합성 억제 효과

        본 연구는 뽕잎 추출물을 이용한 동충하초 균사체 배양액의 in vivo, in vitro멜라닌 생성 억제 효과를 조사한 것이다. 뽕잎 추출물을 이용한 동충하초 균사체 배양액은 10~50%의 농도 의존적으로 시험관 내에서 멜라닌 합성관련 효소활성의 저해를 나타냈다. 뽕잎 추출물 자체는 mouse melanoma cell (B16BL6)에 대한 세포독성을 보였으며 IC$_{50}$%값이 약 7% 농도로 나타난 반면, 뽕잎 추출물을 이용한 동충하초 균사체배양액은 50% 농도에서도 어떠한 세포독성도 관찰되지 않음을 확인할 수 있었다. 또한 B16BL6 mouse melanoma cell의 세포 내 멜라닌 생합성 억제에 대해서도 높은 활성을 나타내었다. 따라서, 뽕잎 추출물을 이용한 동충하초 균사체 배양액은 세포에 대한 독성이 전혀 없으며, 멜라닌 생합성 저해 작용이 우수하여 피부 미백용 화장료 조성물로서의 이용 가능성이 확인 되었다. This study was carried out to investigate the whitening effect of mycelial culture broth of P. japonica in the mulberry leaf extract. The culture broth inhibited melanin synthesis in vitro in the concentration dependent manner of 10 to 50%. The mulberry leaf extract itself has a cytotoxicity to the mouse melanoma cell, B16BL6 with the value of IC$_{50}$ of 7% concentration. But the culture broth of P. japonica in the mulberry leaf extract did not show any cytotoxicity up to 50%. Therefore, we concluded the mycelial culture broth of P. japonica in the extract of mulberry leaf can be used as a whitening cosmetic resource.

        뽕잎 추출물배지를 이용한 눈꽃동충하초(Paecilomyces japonica) 균사체 배양액의 여드름균(Propionibacterium acnes) 생육억제 효과

        This study was carried out to investigate the growth inhibition of Propionibacterium acnes by mycelial culture broth of Paecilomyces japonica in the Mulberry leaf extract. The growth inhibition effect of P. japonica mycelial culture broth against P. acnes in various concentration of Mulberry leaf extract was the most effective in 3% Mulberry leaf extract. The inhibition effect of P. japonica mycelial culture broth against P. acnes according to the culture time was the moust effective after 25 days mycelial cultivation. As the treating amount of the mycelial culture broth was increased, the growth inhibition effect against P. acnes was increased gradually. The growth inhibition effect of mycelial culture broth against P. acnes according to the time of heat treatment was active by 45min at 100℃, while it was inactive more than 60min at 100℃. This result suggested that the antibacterial materials are possible to be glycoside or peptides. Taken together P. japonica mycelial culture in the Mulbarry leaf extract has a possibility to be an element of skin -care cosmetics regulating the acnes.

        Tyrosinase Inhibitory Xanthones from Cudrania tricuspidata

        꾸지뽕나무(Cudrania tricuspidata(Carr.) Bureau) 뿌리껍질에서 3종의 xanthones 화합물을 분리하였으며, 분광학적인 자료를 바탕으로 cudraxanthone L (1), cudraxanthone D (2), 그리고 cudraxanthone M (3)으로 구조동정되었다. 분리된 화합물을 L-tyrosine이 기질로 작용할 때 tyrosinase 저해 활성을 측정한 결과 cudraxanthone M (3)의 $IC_{50}$ 값이 $16.5{\mu}M$로 가장 높은 저해 효능을 보였으며, kinetic type은 uncompetitive inhibition로 저해 상수($K_i$)는 $1.6{\mu}M$로 측정되었다. 또한 이 화합물은 $20{\mu}M$의 농도에서 lag time이 310초로 측정 되어 대조화합물인 kojic acid와 유사한 저해능을 나타내었다. The methanolic roots bark extract of Cudrania tricuspidata (Carr.) Bureau was chromatographed, which yielded three xanthones 1-3 by tyrosinase inhibitory activity-guided fractionation. The structures were fully characterized by analysis of physical and spectral data. Among them, furano prenylxanthone 3, never reported as tyrosinase inhibitor, showed potent activity with $IC_{50}$ value of $16.5{\mu}M$, and appeared to inhibit the polyphenol oxidase activity of tyrosinase in an uncompetitive inhibitor($K_i=1.6{\mu}m$) when L-tyrosine was used as a substrate. Moreover, potent inhibitor furano prenylxanthone 3 had an extended lag time of 310 sec at $20{\mu}M$, while lag time of kojic acid as positive control was prolonged with 350 sec at the same concentration.

        Metabolic Engineering for Resveratrol Derivative Biosynthesis in Escherichia coli

        We previously reported that the SbROMT3syn recombinant protein catalyzes the production of the methylated resveratrol derivatives pinostilbene and pterostilbene by methylating substrate resveratrol in recombinant E. coli. To further study the production of stilbene compounds in E. coli by the expression of enzymes involved in stilbene biosynthesis, we isolated three stilbene synthase (STS) genes from rhubarb, peanut, and grape as well as two resveratrol Omethyltransferase (ROMT) genes from grape and sorghum. The ability of RpSTS to produce resveratrol in recombinant E. coli was compared with other AhSTS and VrSTS genes. Out of three STS, only AhSTS was able to produce resveratrol from p-coumaric acid. Thus, to improve the solubility of RpSTS, VrROMT, and SbROMT3 in E. coli, we synthesized the RpSTS, VrROMT and SbROMT3 genes following codonoptimization and expressed one or both genes together with the cinnamate/4-coumarate:coenzyme A ligase (CCL) gene from Streptomyces coelicolor. Our HPLC and LC-MS analyses showed that recombinant E. coli expressing both ScCCL and RpSTSsyn led to the production of resveratrol when p-coumaric acid was used as the precursor. In addition, incorporation of SbROMT3syn in recombinant E. coli cells produced resveratrol and its mono-methylated derivative, pinostilbene, as the major products from p-coumaric acid. However, very small amounts of pterostilbene were only detectable in the recombinant E. coli cells expressing the ScCCL, RpSTSsyn and SbROMT3syn genes. These results suggest that RpSTSsyn exhibits an enhanced enzyme activity to produce resveratrol and SbROMT3syn cata-lyzes the methylation of resveratrol to produce pinostilbene in E. coli cells.

        Effects of different storage conditions on the metabolite and microbial profiles of white rice (Oryza sativa L.)

        Microbial populations in white rice (Oryzasativa L.) samples stored for 6 months in open or closedconditions were studied and their metabolite profiles analyzedusing GC/MS to elucidate the relationship betweenstorage and rice quality. Rice samples stored in openconditions at 25 C were contaminated by Aspergillustritici, Cladosporium cladosporioides, and Penicillium sp.,whereas the control stored in closed conditions at 5 C wasmainly contaminated by Hyphopichia burtonii and A. tritici. These differences resulted in significantly differentmetabolite profiles. Increased mold population decreasedthe levels of fresh rice flavor-associated volatile metabolitesand primary energy sources, but increased the levelsof metabolites associated with lipid oxidation, polyols, andenergy production. Thus, rice quality, especially flavor,could be significantly influenced by the increased moldpopulation caused by open storage at 25 C; volatilemetabolites and polyols are potential indicators of ricequality.
