2003년 8월 3일부터 8월 5일까지 전라북도 군산시에 속한 고군산군도의 5개 섬을 대상으로 조간대 및 조하대와 어선의 그물에서 채집된 십각류를 관찰한 결과 새우류 2과 4종, 집게류 4과 9종, 게류 11과 29종으로 모두 17과 42종이 동정, 분류 되었다. 이 가운데 새우류 2종, Athanas sp., Alpheus sp.는 한국미기록종이었으며, 이들을 포함하여 10과 20종-새우류 1과 2종, 집게류 3과 6종, 게류 6과 12종-이 고군산군도 해역에서 처음 보고 되는 종들이었다. 따라서 과거 이 지역에서 보고 된 19과 52종에 10과 20종을 추가하여 총 72종이 보고 된다. 조사된 십각류 중에서 게류가 11과 29종으로 가장 다양했으며, 생물량은 새우류에서 줄새우아재비 [Palaemon (Palaemon) serrifer], 게류에서 칠게 (Macrophthalmus japonicus)와 엽낭게(Scopimera globosa), 집게류에서 가시투성어리게 (Hapalogaster dentata)가 가장 많았다. A faunal study on the marine decapods of Gogunsan Islands located in the western part of South Korea was performed during the period from August 3 to August 5 in 2003. As a result of the present study, a total 42 species in 17 families (Macrura: four species in two families, Anomura: nine species in four families, Brachyura: 29 species in 11 families) were identified. Of these, two species of shrimp, Athanas sp. and Alpheus sp., are new to Korea. Including these two species, total 20 species in 10 families are newly known to Gogunsan Islands. Fifty two species in 19 families were previously known to occur in this area. Therefore, a total 72 species in 24 families of decapods are now known to occur in Gogunsan Islands. Of the species collected in the present study, Palaemon (Palaemon) serrifer (Stimpson, 1860) in shrimps, Macrophthalmus japonicus De Haan, 1835 living at mud area and Scopimera globosa De Haan, 1835 at sand beach in crabs, and Hapalogaster dentata (De Haan, 1849) in hermits crab were most dominant.

새만금에 서식하는 십각류 조사는 1차 조사가 2003년 8월 21일에서 23일 사이에, 2차 조사가 2003년 9월 20일부터 28일 사이에, 3차 조사가 10월 24일과 25일 양일에 걸쳐 모두 3회, 9개 지점에서 실시되었다. 본 조사 결과 십각류 17과 44종이 동정 되었는데, 이중 새우류가 7과 15종, 집게류가 2과 3종, 게류가 8과 26종이었다. 조사결과 3종(Alpheus sp.,Athanas sp., Asthenognathus sp.)이 한국미기록종으로 밝혀졌으며, 한국 해역에서 새만금에서만 서식하고 있는 희귀종인 뿔눈새우(Ogyrides orientalis)를 확인할 수 있었고, 게류 중에서 희귀종인 높은 등옆길게 (Tritodynamia horuathi)와 큰애기비단게 (Acmaeopleura balssi)가 새만금 조하대에서 다수 서식하고 있음을 확인할 수 있었다. A faunal study on the marine decapods of Saemangeum located in the western part of South Korea was performed during the period from August 21, 2003 to October 25, 2003. Forty four species in 17 families were identified through the present investigation, which were represented by 15 species of shrimps in seven families, three species of anomurans in two families, and 26 species of crabs in eight families. Three species of the identified species were new to Korea: Alpheus sp. and Athanas sp. in shrimps, and Asthenognathus sp. in crab. Of the species collected in the present study, Ogyrides orientalis (Stimpson, 1860) in shrimp, and Acmaeopleura balssi Shen, 1932 and Tritodynamia horvathi Nobili, 1905 in crabs are rare species in the western coast of South Korea.
Crustacean Decapods of Jindo Island, Korea

A taxonomic survey was carried out to see the decapod fauna of Jindo Island and its adjacent islet, Korea in June 2004. The 35 species in 13 families were identified in this study and of which 12 species of caridean shrimps in three families, one thalassinidean species, two anomuran species in one family, and two crab species in two families were newly added to the decapod faun3 of the study area. With the previously known 58 species in the Jindo Island and its adjacent islets, a total of 75 species are listed with some brief remarks. Distribution patterns of species are also discussed based on the composition of geographical distribution forms. 진도의 십각류 종류상을 밝히기 위해 분류학적 연구를 수행하였다 그 결과 13과 35종이 동정되었으며, 이 가운데 새우류 3과 12종, 쏙류 1종, 집게류 1과 2종, 게류 2과 2종이 진도의 십각류 종류상에 새로이 추가되었다. 과거기록을 포함하여 총 75종의 분류목록을 작성하였으며, 주요 분류군들에 대해서는 간략한 고찰을 하였다. 또한 이미 분포상이 밝혀진 십각류 73종의 분포형과 종 출현 양상을 근거로 진도와 그 인근도서의 십각류 분포 양상을 논하였다.
Construction of Expression Vectors Containing Portable PL-Promoter of $\lambda$ Phage

본 실험에서는 prokaryotic promoter중에서 강력하다고 알려진 phage $\lambda$의 PL-promoter를 끼어 넣은 유전자 운반체를 개발하여 온도로써 이의 발현을 조절할 수 있는 대장균 속에 이주시켜 유전자 발현이 강력하면서도 온도로써 그 발현을 조절할 수 있는 host-vector system을 확립하였다. $\lambda$ phage의 PL-promoter와 N gene이 함유된 BamHI--BglII fragment가 pBR322의 BamHI site에 삽입된 pPL81에서 N gene의 일부를 포함하는 부위인 899bp를 ClaI을 처리하여 제거하고 다시 연결시켜 pPL82를 개발하였으며, pPL82는 PL-promoter와 N gene의 일부를 가지므로 fused protein을 얻을 수 있다. 또한 pPL81을 HpaII로 처리하면 PL-promoter와 operator만을 갖는 273 bp가 나오며 이를 cohesive end가 같은 pBR322의 ClaI site에 삽입하였다. PL-promoter가 시계반대방향으로 들어간 것을 pPL83, 시계방향인 것을 pPL84이라 명명 하였으며 이들 plasmid는 EcoRI과 HindIII를 처리하면 PL-promoter만 있는 약 300 bp의 DNA조각을 얻을 수 있다. PL-promoter와 구조 유전자와의 거리에 따른 PL-promoter의 효율을 $\beta$-lactamase 효소의 활성도로 조사한 결과 pPL81보다는 pPL82가 pPL82보다는 pPL83이 높았다. In this study, series of expression vectors that contain PL-promoter of phage lambda were constructed. The PL-promoters in those plasmids were able to be controlled by temperature in the host Escherichia coli which contained temperature sensitive CI857 gene on the chromosome. Plasmid pPL81, which was constructed with the fragment containing the N gene and its PL-promoter of phage lambda into the BamHI site of pBR322, was digested with ClaI for the deletion of 899 bp and self-ligated to construct pPL82. The plasmid pPL82 is useful for constructing gene fusions whose product contained amino acids encoded by N gene. Besides the construction of pPL82, HpaII fragment containing only oLpL region of pPL81 was inserted into the ClaI site of pBR322. The plasmid with the PL promoter in the counterclockwise orientation was named as pPL83, and that in the clockwise orientation was named as pPL84. These plasmids carry portable promoter that can be separated by codigestion with EcoRI and HindIII restriction endonucleases. Since the, $\beta$-lactamase genes of pPL81, pPL82 and pPL83 are located at the downstream of the PL-promoter, the $\beta$-lactamase can be induced at $42^{\circ}C$. The comparison of the production of $\beta$-lactamase indicated that PL-promoter of pPL82 was more efficient than pPL81, and that of pPL83 was the most efficient. These results indicated that when the structural gene is closely located to the PL-promoter, the influence of PL-promoter is more effective.
Providencia stuartii 164로부터 Pst ⅠMethylase 의 분리 및 특성에 관한 연구

In this report, the purification and characterization of Pst I methylase from Providencia stuartii 164 strain was described. Pst I methylase was purified by the following procedures; ammonium sulfate fractionation, DEAF-cellulose chromatography, Phosphocellulose chromatography and Heparin agarose chromatography. Pst I methylase was a site-specific methylase which has been proven by the facts that pBR322 DNA cleaved by Pst I restriction endonuclease was not methylated by Pst I methylase, and pBR322 DNA methylated by Pst I methylase was not cleaved by Pst I restriction endonuclease. The plasmid pRL829, which contains Pst I restriction-modification genes and thus was fully methylated in vivo, was not methylated by Pst I methylase. These results strongly suggest that the methylase has a site specificity on Pst I recognition site. Pst I methylase activity has optima at pH 7.5 and in the presence of 50 mM NaCl. In addition, Pst I methylase has also been found to have methylation activity on single-strand ΦX174 DNA.
