        공액 이중 결합을 갖는 Adenosine과 Uridine 유사체의 합성

        7'-Aldehyde-nucleoside 유사체(2a, 2c)을 6-N-benzoyl-2',3'-O-isopropylideneadenosiner과 uridine으로부터 합성하였다. 2와 Wittig reagent인 ethoxycarbonylmethylene을 축합시켜 ethoxycarbonyl group을 갖는 탄소수를 증가시킨 공액 이중 결합을 지닌 nucleosi de 유사체, ethyl-1',5',6',7',8'-pentadeoxy-1'-(adenin-9-yl)-$\beta$-D-ribo-nnona-5'(E).7'(E)-dienofuranuronate(4b), ethyl-1',5',6',7',8'-pentadeoxy-1'-(uracyl-1-yl)-$\beta$-D-ribo-nona-5'(E),7'(E)-dienofuranuronate(4c)을 얻었다. 또한 2와 CBr4, Ph3P을 반응시켜 공액 이중 결합을 지닌 9-[8',8'-dibromo-5',6',7',8'-tetradeoxy-$\beta$-D-ribo-octa-5'(E),7'(E)-diene]nucleosides (6b,6c) 유사체를 각각 합성하였다. 7'-Aldehyde-nucleosides analogue (2a, 2c) were synthesized from 6-N-benzoyl-2',3'-O-isopropylideneadenosine and uridine. The condensation of 2 with ethoxycarbonylmethylene Wittig reagent produced the adenosine and uridine analogues containing extended conjugated diene and ethoxycarbonyl group, ethyl-1',5',6',7',8'-pentadeoxy-1'-(adenin-9-yl)-$\beta$-D-ribo-nona-5'(E),7'(E)-dienofuranuronate (4b) and ethyl-1'. 5',6',7',8'-pentadeoxy-1'-(uracil-1-yl)-$\beta$-D-ribo-nona-5'(E),7'-(E)-dienofuranuronate (4c). 9-[8',8'-dibromo-5',6',7',8' -tetradeoxy-$\beta$-D-ribo-octa-5'(E),7'(E)-diene]nucleosides (6b, 6c) were also prepared from 2 with similar method.

      • Oligodeoxyribonucleotides의 합성을 위한 유기염기의 보호기 도입

        In this study the starting materials, 2'-deoxyriboadenosine and 2'-deoxyribocytidine, were protected selectively as the tritylation of the 5'-hydroxyl group and the N-acylation of the amino group with 4,4'-dimethoxytrityl chloride and benzoyl chloride, in a oneflask procedure. Protection of the amino group and the 5'-hydroxyl group in 2'-deoxyguanosine can be performed directly by isobutyric anhydride and 4,4'-dimenthoxytrityl chloride pyridine to obtain 2-N-isobutyryl-5'-O-dimethoxytrityl-2'-deoxyguanosine in a one-flask procedure. The protected deoxyribonucleotides have been crharacterized on UV, IR and ^1H-NMR spectroscopy.

      • 5´-Chloro-2´,5´-dideoxycytidine의 합성

        2'-Deoxycytidine 4 was prepared from cytidine by treatment of the cytidine with 4-bromocarbonyl-1-methylethylacetate followed by radical de- bromination with Bu_3SnH, AIBN, and NH_3/MeOH. 5'-chloro-2',5'-dideoxy- cytidine 6 was synthesized regioselective in 93% yield by reaction of 4 with SOCl_2/HMPA. COSY spectrum and HETCOR spectrum for 5'-chloro-2',5'-dideoxycytidine were good. 1-(β-D-ribofuranosyl)-4(1.2.4-triazole-1-yl)pyrimidine-2-one 7 was also synthesized from cytidine.

      • 1,2-Dihydroxybenzene, 1,3,5-Trihydroxybenzen과 요오드 사이의 전하전이 착물

        Charge transfer complexes of 1, 2-Dihydroxybenzene and 1, 3, 5-Trihydroxybenzene with lodine in dimethylsulfoxid have carried out by Ultraviolet Visible-Spectrophotometer. The stability constants and the molar extinction coefficients of the complexes have been determined by Benesi-Hildebrand equation, and the least square methods. Thermodynamic parameters for the formation for the charge transfer complexes were calculated from these values. The results are as follows. 1) The charge transfer complexes have maximum of absorption at 282 nm, and the charge transfer complex of 1, 3, 5-Trihydroxybenzene with lodine reveals the formation of the one to one molecular complex. 2) The maximum of absorption of 1, 3, 5-Trihydroxybenzene-Iodine C.T complex does not move as the concentration of the donor and the temperature increase. 3) The molar extinctions coefficients of 1, 3, 5-Trihydroxybenzene-Iodine C.T complex are about ten thousands. 4) The stability constants are relatively large, and decreased with temperatues rasing. 5) The enthalpy, free energy and entropy are small, and the positive entropy is decreased with temperatues raising.

      • Phosphotriester 법에 의한 Oligodeoxyribonucleotides의 고상합성

        After the protected deoxyribonucleotides attached to the Whatmann chromatography paper used as support, the two deoxyribonucleotides, 5'-d(CAGATCTG)-3' and 5'-d(GAGGCCTC)-3' have been synthesized on the disks by the phosphotriester method. The average yield of trityl analysis after detritylation of the two deoxyribonucleotides is 68.3% and 74.5%, respectively, for overall yield, and is 93.8% and 94.3% for stepwise yield. purification and separation of the synthesized oligoribonucleotides were treated by HPLC.

        The decamers an the undodecamers were synthesized on succinated support-nluceoside via. a phosphite triester method, By using tetrazol as catalyst for the activation of nucl-osidecyanoethyl phosphoramidite, the coupling reaction was carried out very rapidly 5 minutes. The time required for one cycle was only 25 minutes. The avarae yield of trityl analysis for 12mers was 76%, that for 10mers was 86%. The crude oligodeoxribonucleotides were purified by reversed phase H.P.L.C. and were guantified by snake venom phosphodiest-erase.

        Paper Disk를 지지체로 이용한 Oligodeoxyribonucleotides 화학적 합성

        Whatmann chromatography paper(3MM)을 지지체로 사용하여 phosphite-triester법에 의해 미량의 염기가 부착된 5'-d(GAATTCCGGCCA)와 5'-d(GGAGCTGTC)의 두 oligodeoxyribonucleotide를 합성 하였다. Trityl yield 분석에 의하면 76.1${\%}$와 86.5${\%}$의 평균 coupling 수율을 각각 얻었다.두 oligodeoxyribonucleotide들을 HPLC를 이용하여 분리 정제할 수 있었고 snake venom phosphodiesterase와 bacterial alkaline phosphatase에 의해 분리 정량화할 수 있었으며 이때의 염기의 수는 합성을 시도한 염기수와 일치했다. 이처럼 paper를 지지체로 이용한 oligodeoxyribonucleotide의 합성은 값싸고 동시에 많은 염기가 부착된 미량의 DNA를 빠르고 짧은 기간에 수행할 수 있는 장점이 있다. The two deoxyribonucleotides, 5'-d(GAATTCCGGCCA) and 5'-d(CGAGCTGTC), were synthesized on disks of Whatmann chromatography paper (3MM) by the phosphite-triester method. The average yield of the dodecamer and the nonamer that was measured by trityl analysis is 76.1${\%}$ and 86.5${\%}$ respectively. Separation of the synthesized oligodeoxyribonucleotides were purified by HPLC. The two oligodeoxymers'bases were quantified by HPLC with snake venom phosphodiesterase and bacterial alkaline phosphatase. The numbers of bases were equal to the designed ones. The advantage of using a paper disk as a support allows us to synthesize many different oligodeoxyribenucleotides during one synthesis. This method is the most convenient synthetic technique to synthesize oligodeoxyribonucleotides easily and inexpensively in a relatively short time.

        광주·전남지역 공통과학 교과 지도 및 공통과학 교과서에 대한 의견 조사

        This investigation into the actual condition of instruction with the General Science textbook was carried out by conducting a questionnaire survey of a General Science teachers at high schools in the Gwangju-Jeonnam area. As a result of analysis, it was found that the percentage of four different teachers teaching the four science subjects was 62.5% of the whole, and the percentage of one teacher teaching all four science subjects was 7.5% in Gwangju, with the percentages being 21.1% and 36.6%, respectively, for Jeonnam. An most appropriate type of General Science textbook instruction is for four different teachers to teach the for different subjects (75%) according to teachers in the Gwangju-Jeonnam area. The teachers that teach the General Science textbook want the course to include the following sections in this order in the chemistry field in the new General Science textbook: atoms, molecules, and ions; chemical reactions, chemical periodicity, and acids and bases 본 연구는 광주광역시${\cdot}$전남 지역 고등학교 공통과학 담당 교사를 대상으로 공통과학 담당 교사의 배경과 공통과학 운영실태, 공통과학 부전공 연수, 공통과학 교과서 교재 내용을 설문지로 조사 연구하였다. 공통과학 지도 교사 중 약 45.9% 정도가 공통과학 연수를 받았다. 설문지 응답자 학교 중 광주 지역에서는 전공 분야별로 4명의 과학 교사가 분야별로 공통과학을 지도하는 경우가 62.5% 이였고 1명이 지도하는 경우는 7.5% 이였다. 전남 지역에서는 21.1% 정도가 4명의 교사, 36.6% 정도가 1명의 교사가 공통과학을 지도했다. 공통과학 지도의 바람직한 형태는 전공 분야별로 4명의 교사가 지도를 해야 한다는 경우가 75%에 이르고 있었다. 공통과학 교과서 내용중 화학 분야에 포함되었으면 하는 단원은 원자 분자 이온, 화학 반응, 원소의 주기적 성질, 산과 염기 순이었다.

      • 산, 염기 및 산-염기 반응에 대한 학생들의 오개념 연구

        This study is to grasp the pattern of misconception according to the pattern of questionnaire and to supply the substantial information in order that teachers might correct the misconception effectively. As for the questionnaire used in the research, it used by making it as four patterns by editing the questionnaire with prerequisite. And the survey was executed on the object of 138 students in the first year in High school. The result of analyzing is as follows. 1. The rate of correction in the question 1 about the pattern of existence of strong acid in the aqueous solution was relatively high. In particular, the percentage of BGa was highest to 70.7%, and the one of AGa was lowest to 42.4%. 2. The rate of correction in the question 2 about the pattern of existence of strong base in the aqueous solution was also presented to similar to the rate of strong acid. And the percentage of BGa was highest to 76.5%, and the one of AGa was lowest to 42.4%. 3. The rate of correction in the question 3 about the pattern of existence of weak acid in the aqueous solution was highest to 58.8% in BGa and was lowest to 22.2% in BNa. 4. The rate of correction in the question 4 about the pattern of existence of mixed solution after neutralized reaction was highest to 72.2% in BNa and was lowest to 37.0% in ANa. 5. The question 1, 2, 3 was found the higher rate of correction in the figure type questionnaire than the narrative type questionnaire.
